Development of Threshing Cylinder for Reduction of Soybean Seed Damage (콩 탈곡손상 절감을 위한 콩 탈곡통 개발)

  • This study was carried out to develop soybean thresher which is able to reduce the soybean threshing damage in comparison to the conventional thresher. A threshing cylinder with different diameter of 480 and 384 mm at each end and with one quarter disc pegs of 60 mm radius was developed and attached to the prototype thresher. A conventional thresher which has a threshing cylinder with $\wedge$ type threshing pegs and same diameter of 480 mm at each end was used for comparative test. A series of comparative performance test was conducted using sun-yu and chung-ja soybean. For sun-yu bean, which is white and usually used for soybean paste and soy sauce, the ratio of damaged beans of prototype ranged 2-3% for 330-360 rpm which is recommended cylinder speed by manufacturer. The ratio of damaged beans of conventional thresher was 3-4% for the same range of cylinder speed. chung-ja beans with black color usually shows high damaged ratio compared with white beans, thus cylinder speed of 250-300 rpm is recommended by manufacturer to reduce the damaged ratio. For this range of cylinder speed, the damaged ratio of prototype was 1.3-1.4% and it was 2.7-6.1% for the conventional thresher. Thus prototype is able to reduce the damaged ratio 1.5-5.0% compared with conventional thresher. Prototype shows 0.4% of unthreshed soybean ratio for sun-yu bean in the optimum range of cylinder speed and it was 0.87% for the conventional thresher. For chung-ja bean, the ratio of unthreshed soybean was almost same for both prototype and conventional thresher with the value of 4.0%. The reason of high unthreshed soybean ratio for chung-ja bean compared sun-yu bean is due to the high seed moisture content of 29.11% which is much higher than that of the recommended.

  • Soybean grain, the main source of protein and lipid, has been used for various purposes in our food resources. Therefore, to abtain and supply protein in safety, soybean must be considered as more and more important and it must be made many efforts in breeding for good quality and high protein. Contents of seed protein of the recommeded soybean cultivars in Korea were above 40 persents. Soybean seed protein was divided four fractions by solubility differences. Contents of fraction-albumines, globulines, prolamines and glutelines- were 7.59%, 71.43%, 1.32%, and 9.11% to total protein in Jangyeobkong. The major components of amino acids were glutamic acid, aspartic acid and arginine in order, cystein, methionine, and tyrosine were the minor components in soybean seed protein. Soybean seed protein is constituted mostly of 11s and 7s proteins. Contents of methionine and cystein, sulfur containing amino acid, in 11s protein has already identified higher than 7s protein's The yields of soybean curd were positively correlated with the soluble protein contents of the soybean varieties.

Fungi Associated with Soybean Seed, their Pathogenicity and Seed Treatment (콩 종자(種子)에서 분리(分離)한 사상균(絲狀菌), 그 병원성(病原性) 및 종자(種子) 소독(消毒)에 관한 연구(硏究))

  • Alternaria tenuis, Arthrobotrytis sp., Aspergillus spp., Cephalosporium sp., Cladosporium sp., Cylindrocarpon sp., Fusarium equiseti, F. moniliforme, F. semitectum, F. solani, Penicillium spp., Rhizopus sp. were saprophytic fungi and Cercospora kikuchii, Colletotrichum truncatum, Diaporthe phaseolorum var. sojae and Fusarium oxysporum were pathogenic fungi detected from 14 seed samples of soybean. Initial symptoms caused by C. kikuchii, C. truncatum, D. phaseolorum sojae and F. oxysporum on seedlings from naturally infected seed by the test tube agar method have been described and discussed. Soybean seeds infected with C. truncatum, D. phaseolorum sojae and F. oxysporum reduced germination of seeds and have influenced on the growth of soybean seedling caused by C. kikuchii. Surface-sterilized soybean seedlings became diseased in the test tube agar artificially inoculated with C. kikuchii, C. truncatum and D. phaseolorum sojae isolated from naturally infected soybean seeds. F. oxysporum showed very weak pathogenicity. Seed disinfectants of Benlate-T, Homai, Tecto and Sisthane have effective to C. kikuchii, C. truncatum, D. phaseolorum sojae and F. oxysporum. Arasan, Captan, Busan-30 and Mercron were inferior to C. kikuchii but effective against others. Seed disinfectants treated in this experiment have increased seed germination campared with non-treatment.

  • The structure of the seed of Lupinue angustifolius was studied in order to investigate the Food quality of lupin seed in comparision with soybean. The cotyledonary cells of lupinseed was in egg-like shape and much (more than 4 times) larger than those of soybean. The microstructure of cotyledonary cells of lupinseed was characterized with thick cell wall having distinct pit-pairs. The protein bodies in lupinseed cotyledon cell contained numerous crystaloids, which was absent in soybean. The middle lamella of soybean cell was partially disintegrated by excessive heat treatment ($120^{\circ}C$, 20 min), whereas those of lupinseed did not change much by heting at $120^{\circ}C$ for 130 min.

  • Soybean [Glycine max (L.) Merrill] crops, grown in a rice soybean rotation, can suffer when grown in soil with excessive moisture. The objective of this work were to determine the reduction in growth and yield, responses of vegetative and reproductive growth of soybean to excessive soil moisture achieved by prolonged irrigation. Responses of different cultivars were determined at growth stages from V6 to R8 to clarify the sensitive growth stages or characteristics to excessive soil moisture. Cultivar differences in response to excessive soil moisture condition were conspicuous in seed dry weight and harvest index (HI) but not in the response of seed number or pod number per plant. The timing of irrigation causing the condition of excessive soil moisture influenced the vegetative or reproductive traits. Soybean plants were more affected by irrigation commencing at the pre-flowering than at the post-flowering stage. Post-flowering irrigation did not reduce growth of vegetative organs significantly; in fact the growth of stems and leaves was facilitated by the prolonged irrigation commencing at flowering. Differences between cultivar response to prolonged irrigation were assumed to relate to the reduced amount of assimilates translocated to the reproductive organ.

QTL Mapping for Major Agronomic Traits across Two Years in Soybean(Glycine max L. Merr.)

  • The agronomic traits, such as days to flowering and maturity, plant height, 100-seed weight and seed filling period, are quantitatively inherited and important characters in soybean(Glycine max L. Merr.). A total of 126 $F_5$ recombinant inbred lines(RILs) developed from the cross of PI 171451$\times$Hwaeomputkong were used to identify quantitative trait loci(QTLs) for days to flowering(FD), days to maturity(MD), plant height(PH), 100-seed weight(SW), number of branches(NB) and seed filling period(FP). A total of 136 simple sequence repeat(SSR) markers segregated in a RIL population were distributed over 20 linkage groups(LGs), covering 1073.9 cM of the soybean genome with the average distance between adjacent markers of 7.9 cM. Five independent QTLs were identified for FD, three for MD, two for PH, three for SW, one for NB and one for FP. Of these, three QTLs were related to more than two traits of FD, MD, PH, NB and FP and mapped near the same positions on LGs H and O. Thus, these traits could be correlated with biologically controlled major QTLs in this soybean RIL population.

  • Isoflavone concentration(daidzein, glycitein, and genistein) of soybean(Taekwangkong and Sowonkong) were analyzed for studying the relationship between seed size and isoflavone concentration and seed coat color and isoflavone concentration. The isoflavone concentration was the greatest in daidzein then followed by genistein, however glycitein was the smallist of all. The isoflavone concentration was increased with the increasing of the ripening degree and seed size.

  • Soybean varieties derived from Korea are classified into four groups on the basis of their food types such as soybeans for vegetable, sprout, sauce and paste and soybeans with colored seed coat. This study was carried out to know the differences in storage protein concentrations among these four groups. There were differences in storage protein concentrations among four groups. In 7S protein, the $\alpha$'-and $\alpha$-subunit concentrations did not vary among four groups, while a $\beta$-subunit concentration greatly varied. 7S protein concentration was the highest(40.6%) in soybean for sauce and paste and the lowest(37.7%) in soybean for vegetable, while 11S protein concentration was the highest (62.3%) in soybean for vegetable and the lowest (59.4%) in soybean for sauce and paste. In view of the fact that 11S protein has much higher sulfur containing amino acids than 7S protein, it was shown the soybeans for vegetable may have higher nutrition value than other groups.

Response of Leaf Water Potential and Growth Characteristics to Irrigation Treatment in Soybean

  • Soybeans [Glycine max (L.) Merr.] are frequently exposed to unfavorable environments during growing seasons and water is the most important factor limiting for the production system. The purpose of this study was to determine the leaf water potential changes by irrigation, and to evaluate the relationships of leaf water potential, growth and yield in soybeans. Three soybean cultivars, Hwangkeumkong, Shinpaldalkong 2, and Pungsannamulkong, were planted in growth chamber and field with irrigated treatments. Leaf water potential of three soybean cultivars was positively correlated with leaf water content during vegetative and reproductive growth stages in growth chamber and field experiments. Leaf water potentials measured for three soybean cultivars under growth chamber were higher than those of under field conditions. Higher leaf water potential with irrigated plots under field was observed compared to conventional plots during reproductive growth stages. Leaf water potentials of three soybean cultivars were continually decreased during reproductive growth stages under field and there was no significant difference among them. Number of leaves, leaf water content, pod dry weight, number of seeds and seed dry weight with irrigated plots were higher than those of conventional plots. The results of this study suggested that leaf water potential could be used as an important growth indicator during the growing season of soybean plants.

  • Soyfoods have potential roles in the prevention and treatment of chronic diseases, most notably cancer, osteoporosis, and heart disease. There is evidence that carcinogenesis are supressed by isolated soybean derived products in vivo such as a protease inhibitor, phytic acid, saponins and isoflavones. It is believed that supplementation of human diets with soybean products markedly reduces human cancer mortality rates. Especially, recent papers recognize the potential benefit of soybean isoflavone components for reducing the risk of various cancers. Isoflavones exhibit a multitude of medicinal effects that influence cell growth and regulation, which may have potential value in the prevention and treatment of cancer. In addition to potential biological effects, soybean isoflavones have the important physiological functions such as the induction of Bradyrizobium japonicum nod genes and the responses of soybean tissues to infection by Phytophthora megasperma as well as biochemical activities such as antifungal and antibacterial actions. Genistin, daidzin, glycitin and their aglycone (genistein, daidzein, glycitein) are the principal isoflavones found in soybean. Malonyl and acetyl forms have also been detected but they are thermally unstable and are usually transformed during the processing in glucoside form. Most soy products, with the exception of soy sauce, alcohol-extracted soy protein concentrate, and soy protein isolate, have total isoflavone concentrations similar to those in the whole soybean. Soybean-containing diets inhibit mammary tumorigenesis in animal models of breast cancer, therefore, it is possible that dietary isoflavones are an important factor accounting for the lower incidence and mortality from breast cancer. Of the total soybean seed isoflavones, $80\~90\%$ were located in cotyledons, with the remainder in the hypocotyls. The hypocotyls had a higher concentrations of isoflavones on a weight basis compared with cotyledons. Isoflavone contents were influenced by genetics, crop years, and growth locations. The effect of crop year had a greater impact on the isoflavone contents than that of location. The climate condition might be the attribution factor to variation in isoflavone contents. Also, while the isoflavone content of cotyledons exhibited large variations in response to high temperature during seed development, hypocotyls showed high concentration in isoflavone content. So, it is concluded that one of the factors affecting isoflavone content in soybean seeds is temperature during seed development. High temperature, especially in maturity stage, causes lower isoflavone content in soybean seed. It is also suggested that there may exist a different mechanism to maintain isoflavone contents between cotyledon and seed hypocotyls. In a conclusion, soy foods may be able to have a significant beneficial impact on public health.
