Effect of Choice Feeding on Performance, Gastrointestinal Development and Feed Utilization of Broilers

  • The experiment was conducted to study the effect of choice feeding on growth performance, carcass quality, gastrointestinal development and feed utilization of 22-49 days old broilers. One hundred and forty four 22-day-old broilers were randomly allocated to 3 treatments with 4 replicates per treatment and 12 birds per replicate. Three feeding regimes are complete diet (control), ground corn and protein concentrate (treatment I), and soybean meal and balancer (treatment II). Protein concentrate is the residue part of complete diet without corn, and balancer is the residue part of complete diet without soybean meal. Treatment I and II are designed for the broilers to freely choose the two parts of diet. The results showed that: (1) broilers under choice feeding (treatment I and II) had lower performances compared with the control; (2) gastrointestinal development and the efficiency ratios that broilers converted dietary crude protein and lysine to body weight gain were improved in treatment I (p<0.05); (3) there were no significant differences in the apparent metabolizabilities of dietary dry matter, crude protein and gross energy, and deposition ratios of dietary nitrogen and energy, and carcass quality among three feeding regimes (p>0.05).

Application of the Microwave-assisted Process to the Fast Extraction of Isoflavone from the Waste Residue of the Soybeans

  • Due to the importance of isoflavone content in soybean extracts, the microwave-assisted process (MAP) was compared to the conventional extraction methods. For comparison of the three methods, all extraction parameters (solvent, sample to solvent ratio, temperature, etc.) were kept the same; the microwave extractor was operated at 187.5 W with an emission frequency of 2450 MHz under atmospheric pressure conditions and the extractions were carried out at 75 ${^{\circ}C}$ for 3 min while the conventional reflux was at 75 ${^{\circ}C}$ for 3 h. Total yield and crude isoflavone content were determined by ultraviolet spectrophotometric and compared with the three methods. Results indicated that the MAP was comparable to the conventional method in its capability to extract target compounds without causing any degradation; in addition it dramatically reduced the extraction time from 3 h to a few minutes, suggesting that it can be an alternative technique to the time-consuming conventional reflux method.

HPLC-UVD/MS를 이용한 농산물 중 bentazone의 분석법 확립 (Analytical Method of Bentazone Residue in Agricultural Commodities Using HPLC-UVD/MS)

  • HPLC-UVD/MS를 이용하여 농산물 중 benzothiadiazole계 선택적 접촉 제초제인 bentazone의 잔류분석법을 확립하였다. 농산물 시료에 인산용액으로 pH를 1로 조절한 acetone을 가하여 추출하였고, bentazone의 잔류분은 dichloromethane 액-액 분배법과 florisil 흡착크로마토그래피법으로 정제하여 분석대상 시료로 하였다. Bentazone의 정량적 분석을 위한 최적 HPLC 분석 조건을 확립하였고, $C_{18}$ 칼럼을 이용한 HPLC 분석 시 불순물의 간섭은 없었으며, 대표 농산물 중 bentazonc의 분석정량한계(LOQ)은 0.02 mg/kg 이었다. 전체 농산물에 대해 정량한계, 정량한계의 10배 및 50배 수준에서 회수율을 검토한 결과 모든 처리 농도에서 82.0~97.9% 수준을 나타내었으며, 농산물 시료 및 처리수준에 관계없이 10%미만의 분석오차를 나타내어 잔류분석 기준이내를 만족하였다. 본 연구에서 확립한 benzothiadiazole계 제초제인 bentazone의 잔류분석법은 검출한계, 회수율 및 분석오차 면에서 국제적 분석기준을 만족할 뿐만 아니라, LC/MS SIM을 이용한 잔류분의 재확인과정 및 회수율 검증의 결과를 종합해 볼 때 분석과정의 편이성 및 신뢰성이 확보된 공정분석법으로 사용이 가능 할 것으로 판단된다.

비지에서 분리된 젖산균의 동정 및 발효특성 (Characterization and Fermentation Characteristics of Lactic Acid Bacteria Isolated from Soybean Curd Residue (Biji))

  • 비지로부터 두 균주를 순수분리하여 동정한 결과 Ent. faecium LL과 L. rhmnosus LS로 확인되었다. 분리된 균을 대두 미세분말용액에서 발효정도를 알아보기 위하여 pH, 산도, 생균수 및 유기산 함량을 측정하였다 Ent. faecium LL은 대두미세분말 용액에서 발효시 pH 4.9, 산도 0.38%, 생균수 1.8$\times$$10^{9}$ CFU/$m\ell$로 젖산생성 및 높은 생균수를 나타내었다. L. rhamnosus LS는 대두미세분말 용액에서는 4.6$\times$$10^{8}$ CFU/mL의 생균수를 나타냈지만 젖산의 생성은 매우 미흡하였다. 그러나 대두미세분말 용액에 당을 첨가하거나 skim milk를 첨가할때 산생성이 급격히 증가되었다. 대두미세분말과 skim milk 4 : 1 혼합액에서 Ent. faecium LL과 L. rhamnosus LS에 의한 젖산발효는 37$^{\circ}C$에서 20시간 안에 curd를 형성하였으며, 각각 0.33% 및 0.77%의 산도와 $10^{8}$ ~$10^{9}$ CFU/$m\ell$정도의 생균수를 보였다. HPLC 분석에서 생성된 젖산의 농도는 L. rhamnosus LS가 600 mg%로 Ent. faecium LL의 350 mg%보다 높았다.

Effect of feeding tamarind kernel powder extract residue on digestibility, nitrogen availability and ruminal fermentation in wethers

  • Objective: This study was to examine in vivo digestibility, nitrogen balance and ruminal fermentation of tamarind (Tamarind indica) kernel powder extract residue (TKPER) compared to soybean products and by-products in wethers. Methods: Four wethers with initial body weight (BW) of $51.6{\pm}5.5kg$ were assigned in a $4{\times}4$ Latin square design to investigate nutritional characteristics of TKPER, dry heat soybean (SB), dry soybean curd residue (SBCR) and soybean meal (SBM) feeding with ryegrass straw (R) at a ratio of 1:1 at 2% of BW in dry matter (DM) on a daily basis. Results: The digestibility of DM, crude protein, and ether extract (EE) of TKPER-R diet were 57.0%, 87.0%, and 86.0%, respectively. Higher non-fiber carbohydrates digestibility was observed in TKPER-R diet (83.2%) than in SB-R diet (73.9%, p<0.05). Wethers fed the TKPER-R diet had lower retention of nitrogen (N) and ruminal ammonia nitrogen ($NH_3-N$) contents at 4 h after feeding than those fed the SBM-R diet (p<0.05), which had values similar to the SB-R or SBCR-R diet. The TKPER feeding had higher propionate (C3) and lower butyrate content, as well as lower acetate to propionate ratio (C2:C3) in rumen fluid than SBM feeding at 4 h after feeding (p<0.05). Conclusion: TKPER did not bring any side effect to the wethers although it was lack of fiber, and could be used as a high protein and energy ingredient in concentrate with appropriate roughage to meet the fiber requirement for ruminants.

High Performance Liquid Chromatographic Method for Determination of Metazosulfuron Residue in Representative Crops

  • BACKGROUND: This study was performed to develop a single residue analytical method for new herbicide metazosulfuron in crops. METHODS AND RESULTS: Brown rice, apple, mandarin, Kimchi cabbage and soybean were selected as representative crops, and clean-up system, partition solvent and extraction solvent were optimized. Instrumental limit of quantitation (ILOQ), linearity of calibration curve and method limit of quantitation (MLOQ) were determined based on the chromatography and whole procedures. For recovery tests, brown rice, apple, mandarin, Kimchi cabbage and soybean samples were macerated and fortified with metazosulfuron standard solution at three levels (MLOQ, 10 MLOQ and 100 MLOQ). And then those were extracted with acetonitrile, concentrated, and partitioned with ethyl acetate. Then the extracts were concentrated again and cleaned-up through $NH_2$ (aminopropyl) SPE cartridge with acetone : dichloromethane (1% acetic acid) (20 : 80, v/v) before concentration and analysis with HPLC. CONCLUSION(S): ILOQ of metazosulfuron was 2 ng (S/N${\geq}$10) and good linearity was achieved between 0.05 and 12.5 mg/Kg of metazosulfuron standard solutions, with coefficients of determination of 0.9999. MLOQ was 0.02 mg/Kg. Good recoveries from 74.1 to 116.9% with coefficients of variation (C.V.) of less than 10% were obtained, regardless of sample type, which satisfies the criteria of Korea Food and Drug Administration (KFDA). Those results were reconfirmed with LC-MS (SIM). The method established in this study is simple, economic and efficient to be applied to most of crops as an official and general method for residue analysis of metazosulfuron.

Nutrient Balances and Soil Properties Affected by Application of Soybean and Barley Residues

  • An accurate analysis of nutrient balance in different cropping systems is necessary for improving soil fertilities, causing higher crop yields and quality. This study was carried out to investigate the nutrient balance, changes in soil properties, and their effects on crop yield in long-term field cultivation under mono- and rotation-cropping systems (MCS and RCS, respectively). The analytical results of the soil properties showed that the application of mineral fertilizers alone in the MCS leads the reduction of soil CEC, exchangeable Ca, and microbial biomass C and N. Compared with the MCS of soybean, the RCS of soybean and barley significantly improved the soil properties, which increased crop yield. It might be due to the barley residue added to the RCS soil. Mean nutrient balances for 4 years were -55.9 kg N, +34.7 kg $P_2O_5$, and -0.3 kg $K_2O$ $ha^{-1}$ for the MCS and +19.7 kg N, +107.4 kg $P_2O_5$, and -48.6 kg $K_2O$ $ha^{-1}$ for the RCS, respectively. These nutrient imbalances mean that conventional fertilizer recommendations were inadequate for maintaining soil nutrient balance. From these results, we can conclude that the crop rotation may change comprehensive physical, chemical, and biological soil properties. These changes could affect the nutrient balance and then the crop yield.

쌀, 콩나물, 물고기의 수세(水洗) 및 조리(調理)중 수은(水銀) 함량의 변화 (Changes in the Mercury Content of Some Foods during the Washing and Cooking Processes)

  • 식품에 수은이 오염(汚染)되었을 경우 수세(水沈) 및 조리(調理)중 수은함량에 어떠한 변화가 일어나는가를 알아보기 위해 수은 오염이 우려되는 쌀, 콩나물, 물고기가 사용되었으며 수은의 검출은 무염화(無炎火) 원자흡광법에 의해 측정되었다. 수은을 인위적으로 1ppm수준으로 오염시킨 쌀을 3회 수세시 전체 수은량의 24%가, 4회 수세시 31%가 감소되었으나 취반(炊飯)에 의해서는 유의적인 함량변화를 보이지 않았다. 수은 80ppm으로 오염된 콩나물을 3회 수세한 경우 수은잔류량의 약 26%가 감소하였으나 가열조리에 의해서는 전체 수은량의 감소가 없었다. 콩나물의 가열조리시 수은은 고형물에서 국물로 용출(溶出)되었으며 용출량은 가열시간, 소금농도와 첨가하는 물의 양에 따라 23-41%의 범위에서 다르게 나타났다. 수은 1ppm으로 오염된 물고기의 가열조리시 전체 수은량의 감소는 없었으나 이때 수은은 고형물에서 국물로 용출되었으며 용출량은 조리용수(調理用水)의 소금농도에 의해 2-10%의 범위에서 달리 나타났다. 물고기를 조리후 비가식부(非可食部)를 버리는 경우 오염(汚染)된 수은의 32%가 제거되었다.

두유박 고오지를 이용한 밀된장 제조 (Utilization of Soymilk Residue for Wheat Doenjang as Koji)

  • 두유 제조에서 얻어지는 두유박으로 밀가루 두유박 혼합 고오지를 이용하여 밀된장을 제조한 결과, 된장 숙성중 환원당의 변화는 전 시험구가 숙성 50일경까지 증가하다가 그 이후에는 감소하였고 시험구별로는 두유박의 배합비가 많아질수록 환원당이 감소하였으며, amino 태질소 함량이 높았다. 총산함량은 40일까지는 현저하게 증가하나 그 이후는 둔화되었으며, 각 시험구별로는 그 차이가 적었다. 숙성 80일 되는 각 시험구의 색도는 두유박의 배합비가 많을수록 대체로 명도가 감소하며 적색이 강해지고 황색도 강하게 나타났다. 숙성 90일 되는 된장에 대해 관능검사를 한 결과 콩나물 된장국은 대조구 [두유박 밀가루 혼합고오지 100(두유박 : 밀가루 ; 100 : 0), 대두 100]가 가장좋고 두유박의 배합비가 많을수록 맛은 떨어지나 대조구와 시험구 A[두유박 밀가루 혼합고오지 125(두유박 : 밀가루 ; 100 : 25), 대두 75]사이, 시험구 B[두유박 밀가루 혼합고오지 150(두유박 : 밀가루 ; 100 : 50), 대두 50]와 시험구 C[두유박 밀가루 혼합고오지 175(두유박 : 밀가루 ; 100 : 75), 대두 25] 사이, 그리고 시험구 C와 시험구 D[두유박 밀가루 혼합고오지 200(두유박 : 밀가루 ; 100 : 100), 대두 0] 사이에는 유의차가 없었다.
