The Effect of Soybean Galactooligosaccharides on Nutrient and Energy Digestibility and Digesta Transit Time in Weanling Piglets

  • Eight $12.4{\pm}0.6kg$ initial body weight crossbred barrows were used to determine the effect of soybean galactooligosaccharides on nutrient and energy digestibility, and digesta transit time. Four dietary treatments were utilized in this trial. Treatment one was a corn-soybean meal based diet (SBM) containing raffinose and stachyose at the levels of 0.16% and 0.75%, respectively. Treatment two (control) was a corn-HP300 (soybean concentrate protein) diet. In treatments three and four, 1.1% and 2.2% commercial stachyose was added to the control diet to provide total dietary stachyose at the levels of 1% and 2%, respectively. The soybean galactooligosaccharides (raffinose + stachyose) level in treatment one was slightly lower compared to that in treatment three. Three collection periods were run with two pigs for each treatment/period. There was a 4 d adjustment period followed by a 3 d collection period. The results showed that the nitrogen retention (86.79%) of pigs fed treatment two diet was higher than that of pigs fed treatment one by 5.2% (p<0.05). The nitrogen retention of treatment three was intermediate 83.09%. The apparent fecal digestibility of all amino acids in treatment two was numerically highest, followed by treatments three and four. However, there were no significant difference among groups (p>0.05). The dry matter (DM), organic matter (OM), crude protein (CP), and crude fiber (CF) digestibility numerically decreased as the soybean galactooligosaccharides level increased, but were not significantly different (p>0.05). Chromium content in feces (from the inclusion of 0.3% chromic oxide in the diets) differed among treatments (p<0.05) at 15 h, 18 h, and 21 h after eating. This showed that the digesta transit time was differed significantly among treatments. Treatment four was the shortest, followed by treatment three, SBM and control. The results demonstrated that in the absence of antinutritional factors and soybean antigen protein, inclusion of 1% and 2% stachyose in corn-HP300 diet has no significant effect on the digestibility of DM, OM, CP, CF and amino acids. When the soybean galactooligosaccharide level in diet one and diet three were adjusted to be almost the same, antinutritional factors such as trypsin inhibitor and soybean antigen protein could decrease the nutrient digestibility and nitrogen retention rate of diet. High levels of soybean galactooligosaccharides shortened the digesta transit time in the intestinal tract. This trial suggested that the total level of soybean galactooligosaccharides (stachyose+raffinose) in the weanling piglet diet is better not to exceed 1% when common soybean meal is used as main protein source.

  • Soybean curd residue (SCR), known as a major waste product of soybean processing, is the water-insoluble fraction which is removed by filtration during soymilk production. For these reasons, SCR was usually considered as food waste. SCR might have a good potential as a functional food material, value-added processing and utilization. SCR contains high-quality protein and consists of a good source of nutrients, including protein, oil, dietary fiber, minerals, along with un-specified monosaccharides and oligosaccharides. Also, SCR might be a potential source of low cost protein for human consumption. However, SCR could be a source of bacterial contamination when during food processing. This study was aimed to investigate antibacterial capacity of natural product through detecting relationship between SCR and microbial. We isolated five bacterial strains from SCR and elucidated antibacterial activity of nature medicines to extend storage capacity of food made with SCR. Thus, the extract which showed antibacterial effects in Corynebacterium calloonae and Raoultella amithinolytica is a combination of seven kinds of extracts: Glycyrrhiza uralensis, Cudrania tricuspidata, Salvia miltiorrhiza Bunge, blueberry, Acorus gramineus, Ginkgo biloba L., Camellia sinensis. This study suggested that antibacterial activities of natural medicines could be used for extension of storage capacity in SCR-contained food.

Effects of Legume Supplementation on the Glucose and Lipid Metabolism and Lipid Peroxidation in Streptozotocin-Induced Diabetic Rats (두류의 첨가가 당뇨병 쥐의 내당능과 지질대사 및 지질과산화에 미치는 영향)

  • The present study was conducted to evaluate the usefulness of common Korean legumes as a high-fiber supplement in therapeutic diets for diabetic patients. Streptozotocin-induced diabetic rats were used as animal models and four kinds of legumes, black soybean (BS), yellow soybean (YS), green pea (GP) and soybean curd residue (SCR) were tested as high-fiber supplements. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of four legumes or purified cellulose and pectin for 6 weeks. The effects of the legumes on the glucose and lipid metabolism of diabetic rats was examined and compared with the effects of cellulose and pectin. The legume supplementation did not show any beneficial effect on glucose tolerance, however, it exhibited a plasma cholesterol-lowering effect in diabetic rats. The cholesterol-lowering action was especially strong in BS and the degree of the effect was comparable to that of pectin. The levels of total lipids, cholesterol, and triglyceride in the hepatic tissues of rats fed legume diets were similar to those of the pectin group. All legume supplements induced an increase in fecal steroid excretion. The fecal cholesterol contents were significantly high following the supplementations of YS and SCR (p < 0.05). The excretion of fecal bile acids in the BS and YS groups was significantly higher than it was in the pectin group (p < 0.05). Concentration of lipid peroxidation products in the blood and urine of diabetic animals was lower in the legume groups than in the cellulose group. The levels of hepatic lipid peroxidation products were significantly lower in the BS and YS groups than in the pectin group (p < 0.05). From the results of this study, the plasma cholesterol-lowering effect of BS is possibly due to the significant (p < 0.05) in-crease in fecal steroid excretion, which suggests that BS could be beneficial in improving abnormal lipid metabolism in diabetic rats. Rapid Soybean-Milk Preparation with Dehulled Soybean and Its Quality Properties (탈피대두를 이용한 신속 두유 제조 및 품질 특성)

  • The purpose of this study was to investigate the quality of dehulled soybean (DHSB), and the rapid preparation possibility of soybean milk with DHSB (SM-DHSB), and then the quality of SM-DHSB. In DHSB, the moisture content decreased, the crude protein, crude fat, minerals, and carbohydrate contents increased, and the isoflavone (daidzein, genistein and glycitein) content was similar to that of soybean (SB). The water absorption rate of DHSB for soybean milk preparation was higher than that of SB. In the results of SM-DHSB and soybean milk (SM) qualities, the crude protein content, total solid content, and the viscosity of SM-DHSB were higher, the yield and the proximate composition (except crude protein) were similar, and the Biji production rate, and total dietary fiber content of SM-DHSB were lower compared to the SM. In terms of the isoflavone contents of SM-DHSB, daidzein and genistein content were similar, and glycitein content was lower compared with the SM. Consequently, these results suggest the possible use of DHSB for rapid SM-DHSB preparation, because the soaking time was decreased by the high water absorption rate of DHSB in the SM preparation, and the quality of SM-DHSB improved compared to those of the SM.

Effects of Soybean Powder and $Chungkukjang$ Powder on Quality Characteristics of Bread (대두분과 청국장분 첨가가 제빵특성에 미치는 영향)

  • The effects of soybean powder and $chungkukjang$ powder on the quality characteristics of bread were investigated. The nutritional elements of the soybean powder, $chungkukjang$ powder and wheat flour differed remarkably. The average crude protein, ash, crude lipid, and fiber of soybean powder and $chungkukjang$ powder were approximately 3, 15, 8 and 5 times higher than those of the wheat flour, respectively. The properties of the dough to which soybean powder and $chungkukjang$ powder were added, i.e., its water absorption (%), development time (min) and stability (min) were measured with a farinograph. The water absorption, and development time of the farinogram increased, but its stability decreased when soybean powder and $chungkukjang$ powder were added to white wheat flour. As long as the mixture rate of soybean powder and $chungkukjang$ powder increased, the loaf volume of the bread declined, but the weight significantly increased. The L value decreased, but the a and b values increased in bread crumb that was made from soybean powder and $chungkukjang$ powder. The white wheat flour to which 10% soybean powder and 5% $chungkukjang$ powder were added had the highest scores in flavor, color, texture, taste, and overall acceptability in the sensory evaluation.

Effect of Soybean Milk Residues Powder on the Quality of Dough (두유박 분말 첨가가 식빵 반죽에 미치는 영향)

  • The rheological properties of dough made the substitution of wheat flour(composite flour) at the levels of 0%, 5%, 10% and 15% soymilk residue flour, with addition of vital wheat gluten at the levels of 3, 6 and 9% were investigated. And nutrition contents of soymilk residue flour were analyzed. The results were as follows; Principal components of soymilk residue flour were 22.0% crude protein, 13.2% crude lipid, 54.3% carbohydrate, 27.2% dietary fiber and $220{\mu}g/g$ isoflavones. Free amino acid component of soymilk residue were L-glutamic acid, L-leucine, L-lysine, L-valine, L-phenylalanine, L-isoleucine, L-threonine, L-methionine and L-cystine. Total dietary fiber content of bread with soymilk residue and wheat flour were 5% soymilk residue; 3.50%, 10% soymilk residues; 4.65%, 15% soymilk residues; 5.96%, and wheat flour bread: 2.1% respectively Mixing water absorption capacity was increased by increasing amounts of added soymilk residue and vital wheat gluten. Dough development time was increased by increasing amounts of added soymilk residues, while decreased by increasing amounts of vital wheat gluten. The dough volume of composite flour with 5%, 10% and 15% soymilk residue flour were the smaller than wheat flour dough. But the dough volume was increased by added vital wheat gluten, and the composite flour with 5% soymilk residue flour and 9% vital wheat gluten was better than the others. This study proved that the dough volume of composite flour with 5% soymilk residue flour and 9% vital wheat gluten was better than the others. On the other hand, the soymilk residue flour contains dietary fiber, isoflavone, protein, lipid and carbohydrate. Therefore the soymilk residue flour will be very useful as food material.

Quality Characteristics and Consumer Acceptability of Brownies with Rice Bran Dietary Fiber (현미 식이섬유를 대체한 브라우니의 품질 및 관능적 특성)

  • This study investigated the optimal percentage of brownies substituted with rice bran dietary fiber (3%, 6%, 9%, and 12%). The sample and control were compared for quality characteristics, specific gravity, viscosity, pH, moisture content, water activity, specific volume, color, textural characteristic, consumer acceptance, and CATA (check-all-that-apply). The specific gravity of the control sample was not significantly different among the samples. The viscosity was highest at the brownie containing 12% of rice bran dietary fiber. Moisture content and water activity were highest in the 6% sample. pH of control sample batter was highest, but reduced with increasing amounts of rice bran dietary fiber. For colors, lightness of the 6% sample batter was highest at 25.31, and redness and yellowness significantly increased with increasing amounts of rice bran dietary fiber. For textural characteristics, hardness, and chewiness significantly increased with increasing amounts of rice bran dietary fiber. Cohesiveness and resilience were not significantly different between samples. In the sensory evaluation, overall acceptability was highest in the control group and lowest for the 12% sample. In the frequency analysis of the CATA, as rice bran dietary fiber was added, the amount of bran flavor, oliy, and unpleasant increased while frequency of bitterness, sweetness, and chocolate taste decreased. In the principal component analysis, characteristics of unpleasant, oily, soybean flavor, and sweetness were strongly detected in the 12% sample. The control sample showed strong sweetness and bitterness. The results indicate that 3% to 6% rice bran dietary fiber is appropriate for production of brownies.

Effects of DHA-rich Fermented Soybean Meal as a Dietary Protein Replacement for Fish Meal in the Parrot Fish Oplegnathus fasciatus (돌돔(Oplegnathus fasciatus) 사료의 어분대체원으로서 DHA가 다량 함유된 발효대두박의 효과)

  • This study determined the level of fermented soybean meal (FSM) that could be substituted for fish meal in the diet for parrot fish Oplegnathus fasciatus. Fish meal (FM) was used the sole protein source in the control diet. FSM was substituted for 20, 40, 60, 80, and 100% of the fish meal in the experimental diets. The FSM resulted in increased crude protein, crude fat, and crude ash, but decreased crude fiber. The methionine and tryptophan contents in the FSM increased, while lysine and threonine decreased. The 18:2n-6 fatty acid content decreased from 55.30% to 28.67%. Fermentation increased 20:5n-3 (EPA) and 22:6n-3 (DHA) by 2.03% and 15.54%, respectively, although the differences were not significant. Based on growth performance, we concluded that FSM could replace up to 60% of FM for the maximum growth of juvenile parrot fish.

A Study on the Protein Bioavailability in Rats Fed Fermented Soybeans (발표대두를 섭취한 흰쥐에서의 단백질의 생체 이용율에 관한 연구)

  • Thirty mael Sprague Dawley rats of 3 weeks of age were adopted to investigate changes in nutritive quality of the traditional, fermented soybean foods, Meju and Chungkugjang prepared by culturing with Aspergillus oryzae and the rice straw, respectively. The levels of each dietary protein were set at 10% . The effect of supplementation of fermented soybean on the rat diet was evaluated by measuring growth rate, feed efficiency. weight of the pancreas and biological values, such as NPU, PER and NPR on day 10. After incubation of soybean with Aspergilllus oryzae and the rice straw, the contents of crude protein, crude fat and crude ash were increased, but the levels of crude fiber and nitrogen-free extract were reduced. The amounts of feed intake, body weight gain an feed efficiency were greater for the rats fed dietscontaining heated unfermented soybeans or Meju compared to rats fed the diet containing raw soybeans. Pancreating enlargement was observed in the rats fed diets containing raw soybeans. The values of NPR and PER in rats fed diets containing Meju and Chungkugjang were significantly higher than those of rats fed diets containing raw soybeans. The values of NPU in rats fed diets containing raw soybans, heated unfermeted soybeans. Meju and Chungkugjang were 39.40%, 40.60%, 45.00% and 46.205, respectively, demonstrating no significant differences.

Antigenotoxic Effect of Paecilomyces tenuipes Cultivated on Soybeans in a Rat Model of 1,2-Dimethylhydrazine-induced Colon Carcinogenesis

  • We evaluated the effect of soybean dongchunghacho [SD, cultivated dongchunghacho fungus (Paecilomyces tenuipes) on soybeans] on dimethylhydrazine (DMH)-induced DNA damage and oxidative stress in male F344 rats. The animals were divided into 3 groups and fed a casein-based high-fat, low fiber diet without (DMH group) or with 13%(w/w) of soybean (DMH+S group), or SD (DMH+SD group). One week after beginning the diets, rats were treated weekly with DMH (30 mg/kg, s.c.) for 6 weeks; dietary treatments were continued for the entire experiment and endpoints measured at 9 weeks after the first DMH injection. SD supplementation reduced DMH-induced DNA damage in colon cells and reduced plasma lipid peroxidation. Thus, SD may have therapeutic potential for early-stage colon carcinogenesis.
