대두부산물의 지방세포분화 유도유전자의 발현저해 및 전지방세포 분화 억제 효과 (Suppressive Effects of By-Product Extracts from Soybean on Adipocyte Differentiation and Expression of Obesity-Related Genes in 3T3-L1 Adipocytes)

  • 대두는 여러 종류의 파이토케미컬을 함유하고 있으며 이에 의한 항산화, 항염증, 항비만 효능 등이 있음이 잘 알려져 있다. 대두 부산물 성분의 비만억제효능을 확인하기 위하여 Oil-Red O 염색법과 정량 PCR을 통하여 지방세포분화억제 및 지방생성억제 효능을 분석하였다. 분화유도물질인 isobutylmethylanthine (IBMX), dexamathasone 및 insulin 처리에 따른 지방세포 분화유도와 함께 대두부산물-침지수, 순물, 두유-의 처리는 세포손상 없이 지방세포분화 연관 유전자-PPAR${\gamma}$, Fabp4, Scd1, adipsin, apolipoprotein (APOE), adiponectin (ADIPOQ)-의 발현을 감소시키는 효능을 보였다. 또한 대두에서 잘 알려진 isoflavone -daidzein과 genistein- 분석을 통하여 대두 부산물에 포함된 두 종류의 isoflavone의 함량을 확인하였다. 결과적으로 대두부산물에는 지방세포분화유도 유전자의 발현억제를 통한 전지방세포의 분화 및 지방세포의 지방생성을 억제하고 있음을 확인하였다. 따라서 두부생산 시 발생하는 대두부산물은 지방세포분화 억제 효능을 바탕으로 이를 이용하는 건강기능 식품제조에 활용될 수 있을 것으로 판단된다.

PCR 방법과 면역학적 분석법을 이용한 두부와 비지에서 GM 콩의 검출법 (Detection of Genetically Modified Soybean in Tofu and Biji using PCR and Immunological Methods)

  • GM 콩으로 제조된 두부와 비지 등 콩 가공제품들의 효율적인 모니터링을 위하여 GM 콩이 0%, 1%, 3%, 5%, 100% 함유된 두부와 비지를 제조하여 이들의 삽입유전자인 5-enolpyruvylshikimate-3-phosphate synthase(epsps)를 검출하기 위한 PCR 방법과 EPSPS 발현 단백질 검출을 위한 western blotting과 lateral flow strip을 사용하여 검출 감도를 비교하였다. PCR 결과 산물의 크기가 큰 600 bp의 증폭에서 두부의 경우에는 100% GM으로 제조된 두부에서만 확인이 가능하였고, 비지에서는 5%, 100%에서 확인이 되었다. 크기가 작은 123 bp가 생성되는 실험에서는 0%를 제외한 모든 두부와 비지에서 검출되었다. Western blot 결과는 100% 두부, 비지에서만 검출이 되었고, lateral flow strip test에서는 100% 비지에서만 검출되었다. 이러한 결과로부터 GM 콩 가공식품 검출은 면역학적 방법보다는 PCR이 더 높은 민감도가 나타내는 것을 확인하였고, 또한 가공식품에서 효율적인 GMO 검출을 위해서는 100 bp 정도의 작은 크기의 PCR 산물을 이용해야 민감한 검출 결과를 보여주는 것을 확인하였다.

Ethanol Extract of Fermented Soybean, Chungkookjang, Inhibits the Apoptosis of Mouse Spleen, and Thymus Cells

  • Apoptosis is a step of the cell cycle which is important in the regulation of immune cell populations. Chungkookjang is a Korean traditional fermented soybean containing microorganisms, enzymes, and bioactive compounds which was used in the treatment of mouse spleen as well as thymus cells (CH1-fermented soybean containing barley, wormwood, and sea tangle; CH2-fermented soybean) and was found to exhibit substantially reduced small DNA fragmentation. An MTT assay showed that the treatment of CH1 and CH2 into the mouse splenocytes and thymocytes sharply increased their survival. Moreover, a FACS analysis also showed that CH1 and CH2 are effective at suppressing the apoptosis of splenocytes and thymocytes. The fermented soybean isoflavone concentrations, which are implicated in lowering breast and prostate cancers, lowering the risk of cardiovascular diseases, and improving bone health, were determined using Capillary Electrophoresis-Electrochemical Detection (CE-ED). The amount of Daidzein in fermented soybean significantly increased by 44-fold dramatically, compared with those in unfermented soybean. In this study, we demonstrated that ethanol extracts of Chungkookjang promote the survival of the mouse spleen and thymus cells in culture by suppressing their apoptotic death. Future studies should investigate which genes are related to apoptosis of the immune cells.

콩, 두부 및 두부부산물중의 Isoflavone함량 및 항산화효과 (Isoflavone Contents and Antioxidative Effects of Soybeans, Soybean Curd and their By-Products)

  • The content of genistein and daidzein which were known to be major antioxidative compounds in soybeans were detected by $C_{18}$ reverse phase HPLC. Most of isoflavones in soybeans were detected in the methanol extract but much less amount of isoflavones in the water extract. Among the four different kind of soybeans, the isoflavone content was highest in brown soybean, followed by yellow, small black and black soybean, in the order. These isoflavones were known to be soluble in hot water, which means transfer of isoflavone content was highest in brown soybean, followed by yellow, small black and black soybean, in the order. These isoflavones were known to be soluble in hot water, which means transfer of isoflavones in soy curd into whey during soy curd processing. To identify the change of isoflavone content during processing of soybean curd, soybean curd were made from yellow, brown and black soybean and isoflavone content were determined in each soybean curd, curd residue and whey. Most of soflavones were remained in the whey, it means most of useful antioxidative compounds were wasted. Thus, it is necessary to develop new technology to collect these isoflavones lost during soybean curd processing.

장류산업의 현황과 향후 발전 방안 (Current status of the Jangryu industry and future development direction)

  • Traditional food is the basis of Korean food, but in the process of industrialization, Japanese soybean fermented product making method became standardization process of Korean soybean fermented product as a factory type. As a result, traditional fermented food was pushed behind the industrialization. At present, there is anxiety in the development of the fermented soybean product industry due to the gap in management level between the manufacturers, the decrease in consumption of Jangryu due to changes in dietary life, and the negative image as high salt food. In order to overcome these problems and lead continuous growth, governmental industrial development policies such as traditional liquor and Kimchi are inevitably needed. By laying the legal and institutional foundation and making good use of it in industry, it will be the foundation for continuous development in the market where fierce competition is accelerated.

천연 항균복합제재용액을 침지 및 재배용수로 처리한 콩나물의 선도유지 효과 (Preservative Effect of Soybean Sprouts Pre-soaked and Cultivated in the Solution of Natural Antimicrobial Mixture)

  • 본 실험에서는 식품의 미생물작용에 의한 변질의 저해 효과가 뛰어난 식물성천연항균제품(botanical antimicrobial agent)에 천연보조제(인삼추출물, 알로에, 매실추출물)를 첨가하여 paper disk법에 의한 항균성 검사를 실시한 결과, 인삼추출물을 첨가한 천연복합항균제재(BAAG)에서 생육저해환이 가장 크고 뚜렷하게 나타났다. 한편, 천연 항균복합제재 용액에 콩나물용 콩을 농도별로 각각 침지 처리하여 냉장 및 상온에 저장하면서 대장균수, 총균수의 변화를 살펴보고, 외관상의 변화에 미치는 영향을 수도수로 침지처리한 대조구와 비교하면서 조사하였다. 실험 결과, 대장균은 BAAG의 침지처리에 의해 성장이 억제되었으며 처리농도가 증가시킬수록 균증식 억제 효과가 뚜렷하였고, 총균수도 BAAG 처리구의 콩나물이 가장 낮은 수치를 나타내었다. 외관상의 변화와 상품 가치를 측정한 결과는 수도수처리구인 대조구 콩나물이 저장 3일 후부터 부패취와 함께 표피의 점질성 갈변이 형성되어 상품가치가 크게 떨어진 상태였다. 위와 같은 결과, BAAG용액 속에서 콩나물 콩을 발아시켜 침지 처리함으로써 콩나물의 유통기간을 상당기간 연장할 수 있을 것으로 기대된다.

유기 반추동물 전용 수입산 유기사료 및 국내산 유기 부산물의 사료적 가치 평가 (Nutritional Evaluation of Imported Organic Feeds and Locally Produced Agricultural By-products for Organic Ruminant Farming)

  • This study was conducted to evaluate the nutritional value of locally produced organic agricultural by-products to substitute imported organic feeds for organic ruminant farming. Imported organic feeds (corn grain, soybean meal, soybean seed, oat grain, barley grain, wheat grain, buckwheat, sunflower seed meal) and byproducts (rice bran, grape seed meal, rice straw, soybean hull, soybean curd, rice hull, green kernel rice, and crushed rice grain) were analyzed for chemical composition and NDF, ADF, mineral, and amino acid contents and anti-nutritional factors. Dry matter, NDF and ADF contents in organic feeds were higher than those in conventional feeds. Especially, the 9.65% fat content of organic soybean meal was 6 times higher than the 1.95% fat content of conventional soybean meal. Fat contents of rice bran, grape seed meal, green kernel rice, and crushed rice grain were 25.66, 6.09, 3.57 and 1.59%, respectively. Protein contents of soybean hull and soybean curd were 14.68 and 19.87%, respectively, which are highest among organic by-products. Levels of aflatoxin in all feeds were below the safety level. Therefore, organic rice bran, green kernel rice and crushed rice as energy source, and soybean hull and soybean curd as protein source could partial replace imported feeds for organic ruminant farming.

산업체 급식소에서 제공되는 콩국수의 미생물적 품질관리에 관한 연구( I ) (A Study on Microbiological Quality & Safty Control of Cold Sybean Noondles serviced by an Industry Foodservice Establishment)

  • This study is written to look into microbiological quality by passage of time and holding methods after making foods, by means of evaluating time, temperature and microbiological quality during various phases in product flow of cold soybean noodles serviced by an industry feeding operation for 500 persons a day, measuring pH & Aw and analyzing factors affecting microbiological growth conditions. The results were as follows: 1. According to phases in product flow of cold soybean noodles, it showed 15.6 hours of mean of needed time, $24.2^{\circ}C$ of room temperature, 5.1~7.6 of pH value & 0.95~0.98 of Aw except dry noodles. These conditions were good for multipling of microbe, and the phases with potential sanitary danger were ingredient, pre-preparation, holding before assembly and service and assembly & service. 2. As for holding methods and passage of time, holding at cold table was more effective than holding at room temperature as time past. 3. As for equipments using for making food, dipper, basket, kitchen board & kitchen towel showed high microbiological value in total aerobic plate counts and kitchen towel and stainless-steel were showed high microbioloical value in coliform counts. 4. E. coli, food poisoning bacteria was detected from barrel filled with soybean soup and also soybean itself.

$\beta$-아밀라아제의 정제에 관한 연구 (Purification of Soybean $\beta$-Amylase)

  • Soybean $\beta$-amylase was purified by DEAE-cellulose ion exchange chromatography, Sephadex G-100 gel chromatography, CM Sephadex C-50 ion exchange chromatography and CM Sephadex C-50 ion exchange rechromatography The purified enzyme showed 1, 020 unit/mg of specific activity. The purified enzyme was identified as homogenious by disc PAGE and analysis of reaction product.

두류식품의 지역 이름 브랜드화의 효과: 한국 소비자의 종적 데이터 분석을 중심으로 (The Effects of Regional Branding on Soybean Products: Evidence from Consumer Longitudinal Data in Korea)

  • Purpose - This study investigates the purchase pattern relating to soybean products in Korea. Specifically, the effect of branding based on a regional name was analyzed in terms of consumer purchase frequencies. The primary purpose of this study is to understand why family characteristics affect product selection for a regional brand in the soybean food category. Research design, data, and methodology - We used data collected by the Rural Development Administration (RDA) of Korea. The RDA has monitored agricultural food consumers for years in order to obtain purchase records. Panel participants live in regions near the capital city of Seoul, Korea. Examining data from January 2010 to May 2016, 667 families were selected for analysis. The final data set was 1,335,402. Each purchase item by each individual family was aggregated to a countable weekly observation. To analyze the data set quantitatively, zero-inflation regression was adopted, which was appropriate to avoid biases from overly dispersed observations. Results - We hypothesized the effects of regional branding from the viewpoint of the family characteristics. The first hypothesis was that the number of children would be positively associated with the purchase of a regional brand of soybean products. The result strongly supported this hypothesis. The second hypothesis was that the number of family members would be negatively associated with the purchase of the soybean products of a regional brand. Based on empirical analysis, we concluded that this hypothesis was partially supported. The third hypothesis was the presence of an interaction effect between the number of children and the family size, which was supported by the results. As a supplementary analysis, we also tested mean-variance differences in terms of categories and regional branding with corporate branding. Conclusion - The results of this study provide insights for regional branding strategies in agricultural food management. This study appears to be one of the seminal studies trying to analyze purchase patterns from longitudinal observations. In addition, this study adopted variables characterizing family lifestyle. This study confirmed that children and family size should be considered when soybean product brands are introduced.
