Ozone has been a problem in big cities. That is secondary air pollutant produced by nitrogen oxide and VOCs in the atmosphere. In order to solve this, the first to be the analysis of the $NO_x$ and VOCs. The main source of nitrogen oxide is the road mobile. Industrial sources in Seoul are particularly low, and mobile traffics on roads are large, so 45% of total $NO_x$ are estimated that road mobile emissions in Seoul. Thus, it is necessary to clarify the relation with the activity of road mobile source and $NO_x$ concentration. In this study, we analyzed the 4 locations with roadside automatic monitoring systems in their center. The V.K.T. calculating areas are set in circles with 50 meter spacing, 50 meter to 500 meter from their center. We assumed the total V.K.T. in the set radius affect the $NO_x$ concentration in the center. We used the hourly $NO_x$ concentrations data for the 4 observation points in July for the interference of the other sources are minimized. We used the intersection traffic survey data of all direction for construction of the V.K.T. data, the mobile activities on the roads. ArcGIS application was used for calculating the length of roads in the set radius. The V.K.T. data are multiplied by segment traffic volume and length of roads. As a result, the $NO_x$ concentration can be expressed as linear function formula for V.K.T. with high predictive power. Moreover we separated background concentration and concentrations due to road mobile source. These results can be used for forecasting the effect of traffic demand management plan.
Fusarium wilt caused by Fusarium oxysporum has become a serious problem world-wide and relies heavily on chemical fungicides. We selected Pseudomonas sp. NJ134 to develop an effective biocontrol strategy. This strain shows strong antagonistic activity against F. oxysporum. Biochemical analyses of ethyl-acetate extracts of NJ134 culture filtrates showed that 2,4-diacetylphloroglucinol (DAPG) was the major compound inhibiting in vitro growth of F. oxysporum. DAPG production was greatly enhanced in the NJ134 strain by adding mannitol to the growth media, and in vitro antagonistic activity against F. oxysporum increased. Bioformulations developed from growth of NJ134 in sterile bean media with mannitol as the carbon source under plastic bags resulted in effective biocontrol efficacy against Fusarium wilt. The efficacy of the bioformulated product depended on the carbon source and dose. Mannitol amendment in the bean-based formulation showed strong effective biocontrol against tomato Fusarium wilt through increased DAPG levels and a higher cell density compared to that in a glucose-amended formulation. These results suggest that this bioformulated product could be a new effective biocontrol system to control Fusarium wilt in the field.
Satellite data have an intrinsic problem due to a number of various physical parameters, which can have a similar effect on measured radiance. Most evaluations of satellite performance have relied on comparisons with limited spatial and temporal resolution of ground-based measurements such as soundings and in-situ measurements. In order to overcome this problem, a new way of satellite data evaluation is suggested with statistical tools such as empirical orthogonal function(EOF), and singular value decomposition(SVD). The EOF analyses with OMI and OMI HCHO over northeast Asia show that the spatial pattern show high correlation with population density. This suggests that human activity is a major source of as well as HCHO over this region. However, this analysis is contradictory to the previous finding with GOME HCHO that biogenic activity is the main driving mechanism(Fu et al., 2007). To verify the source of HCHO over this region, we performed the EOF analyses with vegetation and HCHO distribution. The results showed no coherence in the spatial and temporal pattern between two factors. Rather, the additional SVD analysis between $NO_2$ and HCHO shows consistency in spatial and temporal coherence. This outcome suggests that the anthropogenic emission is the main source of HCHO over the region. We speculate that the previous study appears to be due to low temporal and spatial resolution of GOME measurements or uncertainty in model input data.
A strong fibrin-specific fibrinolytic enzyme was produced from Bacillus subtilis KH-4 isolated from Deonjang, a Korean fermented soybean paste similar to Japanese miso. The addition of glucose as a carbon source resulted in the highest levels of caseinolytic and fibrinolytic activities. Likewise, the addition of yeast extract as the nitrogen source resulted in the highest caseinolytic and fibrinolytic activities (3473.2 unit and 47.4 munit, respectively), It was observed that out of all metal ion sources only calcium (chloride) enhanced caseinolytic and fibrinolytic activities, with increases of 4949.3 unit and 58.2 unit/mg, respectively. The optimal temperature for the production of the enzyme was found to be 4$0^{\circ}C$ in the optimal medium (glucose 20 g, yeast extract 5 g, CaCl$_2$l g, and NaCl 2 g). The maximum fibrinolytic activity was observed at the late stationary phase. B. subtilis KH-4 produced a fibrinolytic enzyme at 4$0^{\circ}C$, after 30 h growth, which increased up to 54 h and then remained constant. These results suggest that Deonjang has potential as a source of physiologically active anti-thromotic enzymes.
The behavior of Penicillium camembertii and Geotrichum candidum growing in submerged pure cultures on simple (glutamate) or complex (peptones) substrates as nitrogen and carbon sources and lactate as a second carbon source was examined. Similar to the behavior previously recorded on a simple substrate (glutamate), a clear differentiation between the carbon source and the energy source was also shown on peptones and lactate during P. camembertii growth, since throughout growth, lactate was only dissimilated, viz., used for energy supply by oxidation into $CO_2$, whereas peptides and amino acids from peptones were used for carbon (and nitrogen) assimilation. Because of its deaminating activity, G candidum preferred peptides and amino acids to lactate as energy sources, in addition to being assimilated as carbon and nitrogen sources. From this, on peptones and lactate, G candidum grew faster than P. camembertii (0.19 and 0.08 g/l/h, respectively) by assimilating the most readily utilizable peptides and amino acids; however, owing to its lower proteolytic activity, the maximum biomass was lower than that of P. camembertii (3.7 and 5.5 g/l, respectively), for which continuous proteolysis and assimilation of peptides were shown.
Background: This study was conducted to analyze the impact of having a usual source of care on health behaviors of the middle aged and the elderly, in order to investigate the potential effect of enhancing primary health care in a Korean context. Methods: This study constructed a balanced panel of middle-aged and elderly samples using the Korea Health Panel 2016-2018, and fixed-effect models were used to analyze the data. Results: Among three sets of dependent variables (physical activity, smoking, drinking), statistically significant results were found only in physical activity. Subgroup analysis showed that this effect was not observed in the late elderly (aged 75 and older) and those without chronic diseases. Conclusion: Results of the study implied that enhancing primary health care among middle age and the elderly may have an effect on improving health behaviors. Moving forward to person-centered primary health care from disease-focused primary health care should be considered in high-risk groups such as the middle aged and the elderly with chronic diseases.
Purpose: This is a descriptive research to examine the levels of ADL, self efficacy, and life satisfaction of elderly institution residents, and the relations among the factors. Method: Data were collected through a questionnaire survey of elders aged over 65 at nursing homes in 9 areas throughout the country. Measures were the physical function scale, the general self efficacy scale, and LSI Z. Collected data were analyzed using SAS/PC11.0. Bacillus subtilis B22, a chemotrophic and aerobic bacterial strain was isolated from homemade kimchi, identified by 16S rRNA gene sequencing. B22 was primarily screened by biochemical, carbon source utilization tests. B22 was used to produce pectinase and ${\beta}$-glucosidase by submerged fermentation under different light sources. B22 was incubated in pectin media and basal media (pH 7.0) under blue, green, red and white light-emitting diodes (LEDs), fluorescent white light, and in darkness at $37^{\circ}C$, orbital shaker 150 rpm for 24 hours. Fermentation under blue LEDs maximized pectinase production ($71.59{\pm}1.6U/mL$ at 24 h) and ${\beta}$-glucosidase production ($56.31{\pm}1.6U/mL$ at 24 h). Further, the production of enzyme increased to pectinase ($156{\pm}1.28U/mL$) and ${\beta}$-glucosidase ($172{\pm}1.28U/mL$) with 3% glucose as a carbon source. Activity and stability of the partially purified enzymes were higher at pH 6.0 to 8.0 and $25-55^{\circ}C$. The effect on the metal ions $Na^+$ and $K^+$ and (moderateactivity) $Mn^{2+}$ and $Ni^{2+}$ increased activity, while $Hg^{2+}$, $Cu^{2+}$, $Fe^{2+}$, and $Fe^{2+}$ inhibited activity. EDTA, phenylmethylsulfonyl fluoride and 5,5-dithiobis (2-nitrobenzoicacid) reduced activity, while tetrafluoroethylene and 1,10-phenanthroline inhibited activity. The amylase was highly tolerant of the surfactants TritonX-100, Tween-20, Tween-80 and compatible with organic solvents methanol, ethanol, isoamylalcohol, isopropanol, t-butylalcohol and the oxidizing agents hydrogen peroxide, sodium perborate and sodium hypochlorite, although potassium iodide and ammonium persulfate reduced activity. These properties suggest utility of pectinase and ${\beta}$-glucosidase produced by B. subtilis B22 under blue LED-mediated fermentation for industrial applications.
Nitrate reductase deficient (NR) mutant lines were selected indirectly by their resistance to 100mM chlorate in cell cultures of P. parviflora. A total of 585 chlorate resistant lines were confirmed by a second passage on a high concentration of chlorate. Frequency of spontaneous mutation was $9.7{\times}10^{-7}$ in 3 month old suspension-cultured cells, and in non-selective media containing amino acids as sole nitrogen source. The frequency of mutation could be increased up to 11-fold by culture for 12 months. Out of 40 randomly selected calli, 22 were fully deficient in NR. The rest of the clones contained a decreased level of NR activity. Further characterization was carried out in 13 mutant lines which were fully deficient in NR and in 5 mutant lines containing residual (0-7.0%) NR activity, as compared to wild-type cells cultured on the same medium. The $NR^-$ mutants were tentatively classified as defective in the NR apoenzyme (nia-type; 11 mutant lines including the 5 with residual NR activity) or in the molybdenum cofactor (cnx-type; 7 mutant lines) by the XDH activity. The cnx-type could be further classified into two groups. In one group (5 mutant lines) of these, the NR activity could be partially restored by nonphysiologically high (1.0mM) molybdate in the culture medium. Both types of $NR^-$ mutants were unable to grow on minimal medium containing nitrate as sole nitrogen source, but grew well on amino acids. They also proved to be extremely sensitive to the standard medium ($MSP_1$) containing nitrate and ammonium. Shoot regeneration was obtained only in the $NR^-$ mutants, which contained residual NR activity, but they so far have failed to grow into plants.
