By cytotoxicity screening of the 65 Korean medicinal plants against leukemia L1210 and P388D$_{1}$ cell line using the MTT assay in vitro, Albizzia julibrissin was studied. This plant was extracted with MeOH and MeOH extract was solvent-fractionated with CHCl$_{3}$, EtOAc and n-BuOH in sequence. Each fraction by various solvents system was purified by column chromatography and preparative TLC, and four compounds were isolated. The structure of each compound was deduced from UV, IR, $^{1}$H-HMR, $^{13}$ C-NMR and CI-MS spectral data. The cytotoxic activity ($IC_{50}$/_ of the compounds, quercetin-3-rhamnoside, 4',5,7-trihydroxyfla-van-3-glucoside, spinasterol-3-glucoside and acacic acid lactone, were evaluated as 5 $\mu\textrm{g}$/ml, 2$\mu\textrm{g}$/ml, 1 $\mu\textrm{g}$/ml against L1210 and as 9$\mu\textrm{g}$/ml, 10$\mu\textrm{g}$/ml, 1.5 $\mu\textrm{g}$/ml, 1.5 $\mu\textrm{g}$/ml and 0.9 $\mu\textrm{g}$/ml against P388D$_{1}$, respectively.

Microarray technology has contributed Significantly to the understanding of bacterial genetics and transcriptional regulation. One neglected aspect of this technology has been optimization of microarray-generated signals and quality of generated information. Full genome microarrays were developed for Clostridium acetobutylicum through spotting of PCR products that were designed with minimal homology with all other genes within the genome. Using statistical analyses it is demonstrated that Signal quality is significantly improved by increasing the hybridization volume. possibly increasing the effective number of transcripts available to bind to a given spot, while changes in labeled probe amounts were found to be less sensitive to improving signal quality. In addition to Q-RT-PCR, array validation was tested by examining the transcriptional program of a mutant (M5) strain lacking the pSOL1 178-gene megaplasmid relative to the wildtype (WT) strain. Under optimal conditions, it is demonstrated that the fraction of false positive genes is 1% when considering differentially expressed genes and 7% when considering all genes with signal above background. To enhance genomic-scale understanding of organismal physiology, using data from these microarrays we estimated that $40{\sim}55%$ of the C. acetobutylicum genome is expressed at any time during batch culture, similar to estimates made for Bacillus subtilis.
Whole plants of Zostera asiatica were extracted twice with acetone/methylene chloride (A+M) and methanol (MeOH) in turn. The combined crude extracts were evaporated in vacuo and then the residue was partitioned between water and methylene chloride. The aqueous layer was fractionated into $H_2O$ and n-butanol and then the organic layer was also fractionated into 85% aq. MeOH and n-hexane, successively. The crude extracts and their solvent fractions were evaluated for their inhibitory effect on growth of human cancer cells AGS, HT-29, MCF-7, and HT-1080 cells by MTT reduction assay. Among samples tested, 85% aq. MeOH and n-hexane fractions showed strong cytotoxic effect against AGS, HT-29, and MCF-7 cells. On the other hand, for HT-1080 cell, 85% aq. MeOH fraction exhibited the strongest cytotoxic effect.
Natural antimicrobial substance from peanut (Arachis hypogaea) shells was isolated and structurally elucidated. Peanut shells were extracted with methanol (MeOH) and concentrated in vacuo, MeOH extract was solvent-fractionated with ethyl acetate (EtOAc) and various buffer to obtain EtOAc acidic, neutral, and phenolic fractions. EtOAc neutral fraction, which showed antimicrobial activity, was purified through silica gel adsorption column, Sephadex LH-20 column, ODS column, and high performance liquid chromatographies, and its active substance was isolated and identified as pratensein (3',5,7-trihydroxy-4'-methoxyisoflavone) by spectroscopic methods of proton-nuclear magnetic resonance, mass spectrometry, and nuclear overhauser enhancement spectroscopy.

To enhance our understanding of immunological stimulating effects through the pathway by which nitric oxide (NO) activity and alkaline phosphatase(ALP) enzyme activity from the marine algae, Ulva lactuca, we have investigated NO activity by using mouse RAW264.7 cell line. And ALP enzyme activity performed by spleen of ICR mouse. The results showed that NO activity of the $H_2O$ fraction is the most effective than activities of other solvent fractions.
The cytotoxic activity of methanol extracts of 25 leguminous seeds in vitro was evaluated by sulforhodamine B assay, using the five human solid A549 lung, SK-OV-2 ovarian, SK-MEL-2 melanoma, XF-498 CNS and HCT-15 colon tumor cell lines. The responses varied with both cell line arid leguminous seed used. Extracts of Canavalia lineata and Glycine soja revealed potent cytotoxic activity against A549 arid SK-MEL-2 cell lines. Moderate activity was observed in the extracts of Cassia obtusifolia and Glyeine max var. chungtae, and C. lineata and Vigna angulasis against SK-MEL-2 and HCT-15 cell lines, respectively. The other seed extracts were ineffective against model tumor cell lines. Because of their potent cytotoxic activities, the activity of each solvent fraction from C. lineata and G. soja was determined and the potent activity was produced from their chloroform fractions. As a naturally occurring therapeutic agent, leguminous seeds described could be useful for developing new types of anti-tumor agents.
The antioxidant activities of a methanolic extract of Eisenia bicyclis and its organic solvent fractions, including dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc), n-butanol (n-BuOH), and water ($H_2O$) fractions, were investigated. Scavenging activities against DPPH, hydroxyl, superoxide anion, and peroxynitrite radicals were evaluated using electron spin resonance spectrometry; intracellular reactive oxygen species (ROS) were evaluated by a 2',7'-dichlorofluorescein diacetate assay using RAW264.7 mouse macrophages. The antioxidant activities of the individual fractions were: EtOAc>n-BuOH>$CH_2Cl_2$ >$H_2O$. The EtOAc fraction exhibited strong radical scavenging activity and a significantly reduced ROS level in RAW264.7 cells. Moreover, the phenolic contents of the extract and fractions followed the same order as their radical scavenging activities. Our results indicate that E. bicyclis is a valuable natural source of antioxidants that may be applicable to the functional food industry.
The effect of an extract of Dalbergiae Lignum and four components that were isolated from the extract on the anticarcinogenic phase II marker enzyme, quinone reductase (QR), was investi-gated. Of the solvent extracts of Dalbergiae Lignum, the CH$_2$CI$_2$ fraction was the most potent in inducing QR activity, with a CD value (the concentration required to double the QR activity) of 29.5 $\mu$/mL. The CH$_2$CI$_2$ extract was further separated into six compounds, four of which were identified as 4-methoxydalbergione, latifolin, 4',6-dihydroxy-7-methoxyflavanone, and obtusafu-ran. Obtusafuran [CD = 1.1 $\mu$M; chemopreventive index (CI) = 101.9] and latifolin (CD = 1.7 $\mu$M; CI = 154.6) displayed potent QR inducing activity and high chemopreventive indices. Lati-folin and 4-methoxydalbergione were identified as strong DPPH-scavengers with half-maximal free radical scavenging concentrations of 15.9 and 17.2 $\mu$M, respectively.

The major aim of this work is the research of secondary metabolites isolated from the aerial parts of Petasites japonicus. The plant material is extracted with a polar solvent, which is 95% by volume methanol at room temperature. The concentrated extract was partitioned as EtOAc, n-BuOH, and $H_2O$ fractions. From the EtOAC and n-BuOH fraction, two bakkenolides and two caffoylquinic acid were isolated using the Diaion HP-20, silica gel, ODS-A, and Sephadex LH-20 column chromatographies. According to the results of the results of physico-chemical and spectroscopic data including NMR, MS and UV. The chemical structures of the compounds were respectively determined as bakkenolide B (1), bakkenolide D (2), 1,5-dicaffeoylquinic acid (3), and 5-O-caffeoylquinic acid (4). These results suggest that the compounds isolated from the aerial parts of this plant were almost identical with known components of Petasites japonicus. However, it is necessary to investigate more about the difference of amounts of constituents according to harvest area and time.
Emission of hazardous and volatile organic chemicals from solid waste landfill site was become to important issue because of environmental pollution and health risk by such chemicals. Laboratory batch and continuous experiments were conducted respectively to elucidate isothermal sorption behaviors and transport phenomena(by gas through unsaturated solid waste layer) in wet solid waste-gas system. Source separated and size reduced refuse(bulky waste) and incinerated ash were used after controlling water content, and trichloroethylene(TCE) was chosen among many such chemicals because of it's generality among those man-created pollutants. Isothermal TCE sorption equilibria wet solid waste-gas system can be described in linear equation and partition coefficient in this system can be estimated approximately by the simple equation derived from schematic structure of the system. Transport equation modified by instantaneous equilibrium sorption fraction and kinetic sorption rate(overall mass transfer capacity coefficient) simulated well the column experiment results.
