  • Mass culture of microalgae as feed for mollusc, crustaceans and fish is an important components of the mariculture industry (Metting Jr., 1996) Growth activator for tetraselmis suecica were screened in methanol and water soluble extracts of several seaweed tissues. Also examined in this study were change in growth rate, biochemical composition, and digestion efficiency, of T.suecica cultured with and without the most effective of these extracts, the water soluble component extracted form M. nitidium. Effects of Co-Expression of Liver X Receptor β-Ligand Binding Domain with its Partner, Retinoid X Receptor α-Ligand Binding Domain, on their Solubility and Biological Activity in Escherichia coli

  • In this presentation, I describe the expression and purification of the recombinant liver X receptor β-ligand binding domain proteins in E. coli using a commercially available double cistronic vector, pACYCDuet-1, to express the receptor heterodimer in a single cell as the soluble form. I describe here the expression and characterization of a biologically active heterodimer composed of the liver X receptor β-ligand binding domain and retinoid X receptor α-ligand binding domain. Although many of these proteins were previously seen to be produced in E. coli as insoluble aggregates or "inclusion bodies", I show here that as a form of heterodimer they can be made in soluble forms that are biologically active. This suggests that co-expression of the liver X receptor β-ligand binding domain with its binding partner improves the solubility of the complex and probably assists in their correct folding, thereby functioning as a type of molecular chaperone.

Chemical Design of Highly Water-Soluble Ti, Nb and Ta Precursors for Multi-Component Oxides

  • Novel citric acid based Ti, Nb and Ta precursors that are highly stable in the presence of water were developed. No alkoxides of Ti, Nb and Ta were utilized in the preparation, instead much less moisture-sensitive metallic Ti, NbCl5 and TaCl5 were chosen as starting chemicals for Ti, Nb and Ta, respectively. The feasibility of these chemicals as precursors is demonstrated in the powder synthesis of BaTi4O9, Y3NbO7 and LiTaO3. The water-resistant Ti precursor was employed as a new source of water-soluble Ti in the amorphous citrate method, and phase pure BaTi4O9 in powdered form was successfully synthesized at 800 ?. The Pechini-type polymerizable complex method using the water-resistant Nb and Ta precursors was applied to the synthesis of Y3NbO7 and LiTaO3, and both the powder materials in their pure form were successfully synthesized at reduced tempera-tures, viz. 500-700 ?. The remarkable retardation of hydrolysis of these water-resistant precursors is explained in terms of the partial charge model theory.

대장균에서 초고온성 샤페로닌과 alginate lyase의 공발현 (Coexpression of Alginate Lyase with Hyperthermophilic Archaea Chaperonin in E. coli)

  • Pseudoalteromonas elyakovii 유래 alginate lyase 유전자(aly)를 대장균에서 발현시켰을 때, 발현된 대부분의 유전자 산물은 내포체라는 불용성 응집체 형태로 생산되었다. Alginate lyase를 가용성 및 활성형으로 생산하기 위해 Aeropyrum pernix K1 유래의 초고온성 샤페로닌 ApCpnA와 ApCpnB를 공발현 파트너로 도입하였다. aly와 ApCpnA와 ApCpnB 각각과의 공발현 결과, aly 단독발현 때의 alginate lyase 활성 10.1 unit/g-soluble protein에서 ApCpnA와의 공발현 때는 83.1 unit/g-soluble protein, ApCpnB와의 공발현 때는 100.3 unit/g-soluble protein으로 가용성 및 활성형으로의 alginate lyase 생산이 8~10배 크게 향상되었다. 다양한 배양 조건들의 조사를 통해 alginate lyase 최대 생산을 위한 조건은 다음과 같았다: 1.0 mM IPTG, $25^{\circ}C$ 배양 온도(IPTG 유도 후), ApCpnB 공발현 파트너. 이러한 공발현 시스템은 대장균에서 기능적으로 또한 의학적으로 중요한 재조합 단백질의 산업적 생산에 크게 유용하게 사용될 것이다.

  • A novel surface-attached solid dispersion is designed to improve the solubility and oral bioavailability of poorly water-soluble drugs without crystalline change. Accordingly, it draws increasing interest because of excellent stability and no pollution for accomplishing enhanced solubility and bioavailability, which have recently been highlighted in connection with a number of higher value-added poorly water-soluble drugs. In addition, excellent stability can be attained when the poorly water-soluble drugs are not dissolved but dispersed in water and provide no crystallinity change. This solid dispersion is given by means of attaching the dissolved carriers such as hydrophilic polymer and surfactant to the surface of dispersed drug particles followed by changing the hydrophobic drug to hydrophilic form. The aim of the present review is to outline the preparation, physicochemical property and bioavailability of novel surface-attached solid dispersion with improved solubility and bioavailability of poorly water-soluble drugs without crystalline change.

GroEL/ES Chaperone and Low Culture Temperature Synergistically Enhanced the Soluble Expression of CGTase in E. coli

  • The effect of culture temperature on the production of soluble form of B. macerans cyclodextrin glucanotransferase (CGTase) in recombinant E. coli was investigated. E. coli cell was cotransformed with two plasmids (pTCGT1 and pGroll) in which the cgt and groEL/ES genes are under the control of T7 promoter and pzt-1 promoter, respectively. When tetracycline (10 ng/ml) and IPTG (l mM) were added as inducers at the early-exponential phase (2 h) and mid-exponential phase (3h), respectively, the solubilization of the inclusion body CGTase was greatly dependent on the temperature of the culture. At low culture temperature of $25^\circ{C}$, 2- or 3-fold higher activity and specific activity were obtained over $37^\circ{C}$. SDS-PAGE analysis revealed that about 62% of CGTase in the total CGTase protein was found in the soluble fraction by applying overexpression of GroEL/ES chaperone and by cultivation of E. coli at $25^\circ{C}$, whereas 33% of CGTase was detected in the soluble fraction at $37^\circ{C}$. Therefore, the expression of GroEL/ES and cultivation at $25^\circ{C}$ greatly enhanced the soluble production of CGTase in E. coli.

  • The influences of sodium chloride, polyethylene glycol 4000 and 20000 on 5-fluorouracil release from disk type silicone polymer devices were examined in isotonic phosphate buffer. These water soluble cosolvent and sodium chloride caused devices to swell in aqueous media. Sodium chloride exerted the greatest influence on drug release. The addition of water soluble cosolvent or sodium chloride to silicone polymeric devices permitted controlled release of 5-fluorouracil, presumably due to the change of the physical microstructure of silicone network, and the solubility and diffusivity of 5-fluorouracil. It seemed that the water soluble drug was released through the hydrophilic pores or pathways formed in the device by the incorporation of a water soluble cosolvent or sodium chloride.

The Role of Ca Equilibrium on the Functional Properties of Cheese: A Review

  • The functional properties of cheese, such as texture and melt, are known to be controlled by several important parameters. Recently, the characteristics of Ca in cheeses, especially the form of Ca (insoluble (INSOL) or soluble) and the shift in Ca equilibrium (i.e., from INSOL to soluble Ca), during aging has received a lot of attention. The INSOL form of Ca, which is present as a structural form in casein, plays a critical role in determining the functional properties of cheese during the early period of ripening (~1 mon). It seems that there is always a reduction of INSOL Ca content in cheese during ripening and there are also factors that can affect the shift in Ca equilibrium. These factors may include the composition of cheese milk, cheese manufacturing pH, acid development during aging, adopting curd-washing in various methods, pre-acidification of milk, etc. There have been many studies showing that the rheological and melting properties of cheese during ripening were significantly (p<0.05) affected by the shift in Ca equilibrium. Therefore, for cheese makers, it is now possible to predict/manage the functional properties of cheese by monitoring and controlling Ca equilibrium in cheese during aging.
