Establishment of the Optimum Culture Conditions for Mozzarella Cheese manufacturing by Streptococcus macedonicus LC743 with Immunomodulating Activity (면역 활성능을 가진 Streptococcus macedonicus LC743을 이용한 모짜렐라 치즈 제조의 최적 배양조건 확립)

  • This study aimed to establish the optimum culture conditions for Mozzarella cheese by using Streptococcus macedonicus LC743, a strain selected for its immunomodulatory activity. For process optimization, 1.0% and 2.0% strain was inoculated and incubated at $32^{\circ}C$ and $35^{\circ}C$, respectively. The general components, bacterial count, total solids, yields, and immunomodulatory activity of the Mozzarella cheese were investigated. When the strain was inoculated at 2.0% and incubated at $32^{\circ}C$, the product quality and immunomodulatory activity was optimal and required minimal processing time. Therefore, 2.0% of S. macedonicus LC743 starter culture was added to milk at $32^{\circ}C$, after pasteurization at $63^{\circ}C$ for 30 min, and agitated for 4~5 min after addition of $230{\mu}L/kg$ of rennet. Curd was made by setting the milk 25~35 min after addition of 0.01~0.02% calcium chloride. The curd was cut at 0.1~0.12% acidity (81 min later) and after heating the cheese to up to $43^{\circ}C$. Whey was removed at an acidity of 0.17~0.18% by agitation for 53 min. Next, cheddaring for 210 min up to an acidity of 0.6~0.65%, stretching at $72{\sim}75^{\circ}C$, and molding at $65{\sim}70^{\circ}C$ were performed, and the product was allowed to cool down to $5^{\circ}C$. Salting was done with a solution of $18{\sim}20^{\circ}B{\acute{e}}$ at $12^{\circ}C$ for 20 min and drying occurred at 80~90% relative humidity at $10^{\circ}C$ for 2~3 days.

Effect of Cultural Regions or Methods on Postharvest Physiological Characteristics and Qualities of Paprika Fruits (재배지역과 재배방법이 파프리카 과실의 수확 후 생리 및 품질에 미치는 영향)

  • This study was conducted to compare the postharvest physiological characteristics of 2 paprika cultivars; 'Special', 'Fiesta' grown in 2 regions: Chuncheon(plain region) and Hwacheon(semi-high land region), and by 2 cultural methods; soil and hydroponic(rock wool culture). Respiratory rate of paprika fruit was higher in soil culture, and ethylene production was higher in Chuncheon region of both cultivars. As the thickness of fruit flesh showed thicker in Hwacheon region and hydroponic culture, the firmness of fruit was in proportion to thickness. Vitamin C content was higher in red-cultivar, 'Special'. and in Chuncheon region. Soluble solids was higher in 'Special', but there was no different between cultural regions. The fruit of both cultivars was grown smaller in Chuncheon region, and the fruit shape of 'Fiesta' progressed faster in soil culture. The surface color of fruit was higher in Hwacheon, and although there was no significantly different between cultural method, both cultivars grown by hydroponics was shown vivid color. As above results, for the long term circulation and high quality of fruit, paprika may be cultivated by hydroponics in high-altitude region.

Favorable Irrigation Timing with Timer and Fruiting Position Focused on the Fruit Quality and Harvesting Time in Perlite Culture of Muskmelon (멜론의 펄라이트 재배시 타이머 제어에 의한 급액 시간과 착과절위가 과실의 품질과 수확시기에 미치는 영향)

  • The efficient timer-controlled irrigation and the favorable fruiting position were investigated far highly quality melon fruits from Feb.18 to July Si 1999. The nutrient solution was supplied either at every hour from 6:00 to 18:00 (T-1) or at 6:00, 8:00, 10:00, 11:00, 12:00, 12:30, 13:00, 13:30, 14:00, 14:30, IS:00,16:00, and 17:00 (T-2). A fruit was set at the first node of the fruit bearing branch from the 10, 12, or 13th node of the main stem. Pot weight was maintained at almost n constant level, regardless of the daily integrated solar radiation in T-2. Soluble solids content (SSC) and fresh weight of fruit were not significantly different among the irrigation treatments at each harvesting time. At the first harvest, SSC and fresh weight of fruit were not significantly different between the fruiting positions within the irrigation treatment. At the second harvest, SSC was higher in T-2 than T-1. The SSC was low in the fruit of the loth node in T-1, while it was not significantly different between fruiting positions in T-2. Fruit fresh weight was the highest at the 12 and 13th nodes in T-1, and the 13th node in T-2. Fresh and dry weights of leaf except petiole, regardless of harvesting time, increased as the node position was higher, The higher the fruiting position was, the lower the leaf weight was. Therefore, it is recommended to irrigate more frequently during the mid-noon. Fruits can be harvested earlier at the lower nodes in the spring crop production.

  • The pig slurry leachate was dark brown-colored solution that leaches out of woodchip trickling filter. The purpose of this research was to investigate the effect of pig slurry leachate and byproduct on growth characteristics of leaf lettuce in hydroponics culture. The effects of addition of fish meal, bone meal and crab shell for the growth of leaf lettuce were investigated. Leaf lettuce were grown in each of six combination treatment solutions; slurry leachate, slurry leachate + fish meal, slurry leachate+bone meal, slurry leachate + crab shell and chemical hydroponic solution for lettuce based on EC content. The chemical nutrient solution was the solution of National Horticulture Research Station for the growth of lettuce. The all of nutrient solution was adjusted 1.5 mS/cm in EC in hydroponics culture. 1. The pH level of leachate of trickling filter was increased and EC decreased gradually during treatment. Pig slurry leachate was low in suspended solids (SS), phosphorus (P), but rich in potassium (K). 2. The plot of slurry leachate (SL) was lowest in the growth characteristics of lettuce. The leaf length and width of lettuce treated with mixture plot of slurry leachate and fish meal (SL + FM) was higher compared with plot in slurry leachate. The chlorophyll reading was reduced in plot treated with slurry leachate, but that in plot of SL+FM was similar compared with control plot. 3. The fresh weight of lettuce showed lowest in the plot treated with slurry leachate. The addition of fish meal increased the yield of comparing plot of slurry leachate, but plots of bone meal and crab shell addition were not significantly difference. The fresh weight of leaf lettuce in plot of SL+FM was 87% as 400.0g compared with control. In conclusion, the mixture solution of pig slurry leachate and fish meal could be used as a nutrition solution of organic lettuce hydroponics.

Breeding of Strawberry 'Okmae' for Forcing Culture with Less Labor Requirement for Fruit Thinning (적과 노력 절감용 촉성 딸기 '옥매' 품종 육성)

  • A new strawberry cultivar (Fragaria ${\times}$ ananassa Duch.) 'Okmae' was developed from a cross between 'Toyonoka' and 'Maehyang'. This cultivar has a good fruit quality and suitable number of flowers per cluster for labor saving in fruit thinning. 'Gyoengnam No. 1' was selected as an elite line with vigorous plant growth, higher soluble solids content and fruit firmness after examining its characteristics and productivity in forcing cultures from 2007 to 2009. Farmer's field trial of 'Gyoengnam No. 1' was conducted in 2010 and it was registered as 'Okmae' thereafter. The general characteristics of 'Okmae' are vigorous growth habit, erect plant type, less number of leaf and elliptic leaf shape. 'Okmae' has long peduncle and 9-10 flowers per flower cluster which need less labor for fruit thinning. Fruits of 'Okmae' are conical having a bright red and glossy skin color. Although 'Okmae' has a smaller average fruit number per plant of 21.9 than that of the control cultivars, it's possible to produce high yield because of greater fruit weight of 26.0 g. 'Okmae' showed a high soluble solids content of $11.6^{\circ}Brix$, low acidity of 0.37%, and high firmness of $14.5g{\cdot}mm^{-2}$. It is sensitive to anthracnose and powery mildews, but high-quality strawberry can be harvested by using effective control measure.

Characteristics of New Ever-bearing Strawberry 'Bokha' Bred for Tropical and Subtropical zone Cultivation (열대 및 아열대 지역 재배용 사계성 딸기 '복하' 육성)

  • 'Bokha' is a new strawberry (Fragaria x ananassa Duch.) cultivar, which was released by the Highland Agriculture Research Institute in 2016. The 'Bokha' cultivar originated from a cross between 'Goha' and 'Saebong No. 3' that showed excellent ever-bearing characteristics, including continuous flowering habit and high soluble-solids contents under long-day and high temperature conditions in 2011. This cultivar was initially named 'Saebong No. 8' after examining its characteristics and productivity in summer culture from 2013 to 2015. After regional adaptability tests in 2016, 'Bokha' was selected from Saebong No. 8 as an elite cultivar. The general characteristics of 'Bokha' include semispreading type, elliptical leaves, and moderately vigorous growth. The fruits are conical in shape, and red in color. 'Bokha' plants have 32.7 leaves, 9.9 more than 'Goha' plants. The soluble-solids content of 'Bokha' was 9.2%, which was 0.2% higher than that of 'Goha'. The average fruit weight of 'Bokha' was about 9.5g and the marketable yield was $27,701kg{\cdot}ha^{-1}$, 72% higher than 'Goha'. 'Bokha' is suitable for tropical and subtropical zone cultivation as a high soluble solids contents cultivar, because it shows continuous flowering habit under long-day and high temperature conditions.

Recovery of Paclitaxel from Suspension Culture Medium with Hydrophobic Resin (흡착제를 이용한 택서스속 식물세포 배양액으로부터 Paclitaxel 회수)

  • The soluble paclitaxel was found in the supernatant of the plant cell cultures of Taxus chinensis, The percentage of soluble paclitaxel depends on paclitaxel concentration in bioreactor. As paclitaxel concentration decreases the percentage of soulbe paclitaxel increases. it is therefore important to develop a new process for the recovery of soluble paclitaxel. The use of hydrophobic resin HP20 gives nearly perfect recovery of paclitaxel in supernatant. The resin was more effective in treatment of th cell and debris free filtrate probably because of the reduced solids content In this case 3 g.l resin and 1 day reaction were enough for recovery the soluble paclitaxel in medium.

  • This experiment has investigated the effect of growth, yield and fruit quality of fig plant by different concentration of nutrient solution. Nutrient solution for pig plant were three concentrations of the balanced nutrient formula development by Japanese Horticultural Experiment Station. Plant height, number of leaves, stem diameter and number of fruit per plant were the best at 1/2 concentration. However, leaf length and leaf width did not show any difference in other treatment. Fruit length, fruit diameter and soluble solids did not differ from the different concentration of nutrient solutions. However, the fruit weight of fig plant was heavier by hydroponics than by soil culture in 2nd experiment. Early stage growth of fig plant was better at low concentration of nutrient solution and yield was better at high concentration. The result of this experiment will be utilized in the new application for fig plant hydroponics.

Influence of Environmental Factors on the Prevalence of the Ovarian Parasite, Marteilioides chungmuensis, in Crassostrea gigas, Cultured in Pukman Bay, Tongyeong (양식환경이 통영 북만의 참굴, Crassostrea gigas에 기생하는 난소기생충, Marteilioides chungmuensis 감염에 미치는 영향)

  • Occurrence and prevalence of Marteilioides chungmueasis have been reported in several waters around Tongyeong but no report has been made for Pukman bay. Therefore, we investigated that the prevalence and infection intensities in Pukman Bay at the inside and the outside areas which are hydrographically divided by tidal current. Furthermore, various environmental parameters were investigated in order to elucidate effective parameter for parasitic infection. Infection rates of Marteilioides chungmuensis in adult oysters were ranged 3.3-20.0% at the inside area during September 2002 through January 2003, and 3.3-30.9% at the outside area during August 2002 through January 2003. External manifestation of infected oyster consisted of abnormal egg masses with nodular appearance in the soft tissue. Histopathological symptoms included massive hemocytic infiltration within or around the follicle wall and atrophic epithelium of digestive diverticula. For the environmental parameters, comparative study made differences between two side of the Bay during the infection period: inside > outside for SS while inside < outside for chlorophyll-a. A positive relationship was observed between chlorophyll-a and infection period, which might indicate the difference in food availability between two areas. The prevalence of ovarian parasite Marteilioides chungmuensis, therefore, was highly associated with food availability. Pearson's correlation analysis was made between environmental parameters and infection prevalence. Significance was observed in water temperature (p < 0.05), suspended solids (p < 0.01) and chlorophyll-a (p < 0.05). A principle component analysis showed that infection of the ovarian parasite, Marteilioides chungmuensis, exhibited effects of seasonality (component I = 55.2%) and chemical/physical environmental factors (component II = 24.4%). These results clearly indicate that the infection of ovarian parasite, M. chungmuensis in the Pacific oyster Crassostrea gigas is closely associated with seasonality and food availability.

Effect of Fish Farm Fecal Solid Diets on the Growth and Energy Budget of Juvenile Sea Cucumber Apostichopus japonicus (육상어류양식장 고형오물을 공급한 해삼(Apostichopus japonicus)의 성장과 에너지 수지)

  • We conducted a 90-day feeding experiment to evaluate the growth and energy budget of the sea cucumber Apostichopus japonicus. Sea cucumbers with a mean initial wet body weight of 3.03±0.06 g were fed one of the following three diets: an eel fecal solid diet, rainbow trout fecal solid diet, or a commercial diet at a water temperature of 17±1℃ and salinity of 32±1 psu. The results suggested that the diets affected the final body weight, specific growth rate (SGR), food ingestion, feces production ratio, and hence the growth and energy budget of the sea cucumbers. Sea cucumbers fed the eel and rainbow trout fecal solid diets showed poorer energy absorption, assimilation, and growth than those fed the commercial diet. The sea cucumbers fed the commercial diet had a significantly higher SGR and proportion of energy used for growth than those fed the other two diets. Sea cucumbers fed the rainbow trout fecal solid diet showed a comparatively higher ingestion rate and feces production ratio than those fed the commercial diet. The eel and rainbow trout fecal solid diets, therefore, were not suitable for sea cucumbers in intensive cultivation. Our findings will facilitate further development of more appropriate diets for culture of sea cucumber.
