Isolation and Identification of Korean type Streptococcus mutans (한국형 Streptococcus mutans의 분리 및 동정)

  • S. mutans known as a causative causative agent of dental caries was isolated from a carious lesion of Korean in the present study. The physiological, biochemical characteristics and polysaccharide pattern of these isolates were compated with those of four laboratory strains ; AHT(a), FA-1F(b), LM7(e), and OMZ65(g). One hundred strains of oral streptococci were isolated from dental caries sites of Korean (one male and one female). Among these, 3 strains were identified as S. mutans. These strains were able to grow on selective media MS, MST, MSP, MSP1, MSB, MSBT and were stained dark pink when sprayed with solutions of mannitol and TTC. So, these strains were called strain 108, 110, and 120, respectively. Strain 108, 110, and 120 were bacitracin resistnt. As these strains contained particularly hippurate hydrolysis enzyme, they were distinguished from laboratory strains. Apart from laboratory strains, the strain 108 was not capable of fermenting lactose, the strain 110 was not able to ferment sorbitol, inulin, melibiose, raffinose and the strain120 was incapable of fermenting inulin, raffinose. All fractions of extracellular and ecll bound polysaccharide of the strain108, 110, and 120 were consisted of more glucan than fructan. Aside from laboratory strains, the isolated strains were composed of more water-insoluble glucans related adherence on solid surface than water-soluble. According to these results, the strain108, 110, and 120 had native characteristecs of S. mutans, but they were different from laboratory strains in some characteristics. Therefore, each of them was given a name to S. mutans KHC108, KHC110, and KHC120, respcetively.

NIRS ANALYSIS OF MOLASSES AND EATS USED AT THE ANIMAL FEEDS INDUSTRY

  • Fats and molasses are used, at the present time, in a considerable proportion as ingredients for the animal feed industry. They are mainly used as energy sources, but also they provide other characteristics of technological and nutritional interest (dust reduction, increase in palatability, etc). Both semi-liquid ingredients have numerous aspects in common from the point of view of their use in livestock feeds, as well as of their analytical control. Feed manufacturers use several criteria to evaluate the quality of fat and molasses. Furthermore, the traditional methods currently used, for their evaluation (eg. fatty acids, sugars, etc) are expensive and more sophisticated that the traditionally used for solid ingredients. The objective of the present work is to carry out a viability study to evaluate the ability of NIRS technology for the quality control of fat and molasses. Samples of liquid molasses (n = 42) and liquid fat ( n = 61), provided by a feed manufacturer, were scanned in a FOSS-NIR Systems 6500 monochromator equipped with a spinning module. The samples were analysed by folded transmission, using a sample cup of 0.1mm pathlength and gold surface reflector. For molasses, calibration equations were developed for the prediction of moisture (SECV=1.69%; $r^2$=0, 42), gross protein (SECV=0, 14%; $r^2$=0, 99), ashy (SECV=0, 60%; $r^2$=0, 84), NaCl (SECV=0, 05%; $r^2$=0, 99) and sugars (SECV=1, 04%; $r^2$=0, 86). For animal fats calibrations were obtained for the prediction of moisture (SECV=0, 14%, $r^2$=0, 88), acidity index (SECV=0, 83%, $r^2$=0, 82), MIU (SECV=0, 38%, $r^2$=0, 94) and unsaponifiables (SECV=0, 45%, $r^2$=0, 87). High accuracy calibration equations were also obtained for the prediction of the fatty acid profile. The equations have $r^2$values around 0.9 or highest. The results showed that NIRS technology could provide rapid and accurate results and reduce analytical costs associated to the quality control of two Important feed ingredients of a well known chemical variability.

Drying Characteristics and Preferences for Steamed Chestnut-Sweet Potato Slab after Cold Air Drying (냉풍 건조 조건에 따른 증절간 밤고구마의 건조 특성과 기호도)

  • Cold air drying was conducted on steamed chestnut-sweet potato to improve its quality, convenience, and preference as snack. Steamed sweet potato was dried from 10 to $25^{\circ}C$ for 48 hours, and moisture contents, colors, texture, and taste were evaluated. The lowest moisture content was 4.53% at $25^{\circ}C$. Lightness decreased while other color values (a, b, and ${\Delta}$E) increased with increasing drying temperature and drying time. Reducing sugar and soluble solid contents ranged from 93.22~190.35 mg/g and 17~60 $^{\circ}Brix$, respectively. The highest hardness of chestnut-sweet potato was 25.13 kgf/$cm^2$, and springiness and cohesiveness were 88.60, and 94.87%, respectively, when dried at $25^{\circ}C$ for 48 hours. Sensory evaluation score was highest at $20^{\circ}C$ for 24 hours. The optimum drying conditions were determined to be $19^{\circ}C$ and 39.5 hours by RSM(response surface methodology).

Report on the sediment types, environmental parameters, density and biometry of Manila clam Ruditapes philippinarum in Gyeonggi Bay off the west coast of Korea (경기만 바지락양식장의 저질조성 서식밀도 및 바지락성장의 특성에 관한 보고)

  • From January to December 2007, we have investigated sediment types, environmental factors and density of clam Ruditapes philippinarum inhabited at Oeri, Naeri, Jonghyeon and Bangmiri tidal flats in Gyeonggi Bay off the west coast of Korea. Sediment types of Oeri and Jonghyeon were characterized with well sorted muddy sand. In contrast, Bangmiri tidal flat was mainly composed of poorly sorted gravelly muddy silt and Naeri was characterized with very poorly sorted muddy-sandy gravel. During the course of study, the surface water temperature, salinity, dissolved oxygen and pH ranged 3.2 to $27.0^{\circ}C$, 21.7 to 33.3 ‰, 5.6 to 12.7 mg/l and 7.36 to 8.82, respectively. The suspended solid (SS) ranged from 3.2 to 1,266.0 mg/l and chlorophyll-a level in the water column ranged $0.3-36.1{\mu}g/l$, respectively. The density of clam in the study areas was mainly determined by the harvesting activities rather than the types of substrate. Relatively higher density of clam was observed in Oeri and Bangmiri where the clam seeds were sowed by the villagers. Condition factor of clams were found to be higher in Oeri where the substrate sorting was better with higher level of chlorophyll a. It was believed that sediment types and the level of available food the two governing factors controlling the density and growth of clam in Gyeonggi Bay.

A Brazing Defect Detection Using an Ultrasonic Infrared Imaging Inspection (초음파 열 영상 검사를 이용한 브레이징 접합 결함 검출)

  • When a high-energy ultrasound propagates through a solid body that contains a crack or a delamination, the two faces of the defect do not ordinarily vibrate in unison, and dissipative phenomena such as friction, rubbing and clapping between the faces will convert some of the vibrational energy to heat. By combining this heating effect with infrared imaging, one can detect a subsurface defect in material in real time. In this paper a realtime detection of the brazing defect of thin Inconel plates using the UIR (ultrasonic infrared imaging) technology is described. A low frequency (23 kHz) ultrasonic transducer was used to infuse the welded Inconel plates with a short pulse of sound for 280 ms. The ultrasonic source has a maximum power of 2 kW. The surface temperature of the area under inspection is imaged by an infrared camera that is coupled to a fast frame grabber in a computer. The hot spots, which are a small area around the bound between the two faces of the Inconel plates near the defective brazing point and heated up highly, are observed. And the weak thermal signal is observed at the defect position of brazed plate also. Using the image processing technology such as background subtraction average and image enhancement using histogram equalization, the position of defective brazing regions in the thin Inconel plates can be located certainly.

The Durability of Geotextiles for Outdoor Exposure during the Construction of Solid Waste Landfill (폐기물 매립지 건설시 옥외 노출된 Geotextiles의 내구성에 관한 연구)

  • Recently, many landfills are constructed by using geosynthetics with earth materials. Geotextiles in geosynthetics are applied for the various purposes such as filters of the leachate collection and removal system and protectors of geomembrane liner. However geotextiles can be exposed to direct sunlight during the construction of landfill for several months. As you know the exposure of polymers to sunlight can be a major source of degradation of them. This study is to suggest a criteria on the installation of heavy weight geotextiles at the landfill. For this purpose, several different geotextiles were evaluated by outdoor exposure at two different locations and under the different seasons for one and half years. As the results of outdoor exposure, polypropylene geotextiles with 500g/$m^2$ and 700g/$\textrm{m}^2$were maintained around 40% of the initial strength during for one and half years. However, the polypropylene geotextile of 1000g/$\textrm{m}^2$showed 65% of the retained strength for the same duration of exposure. The retained strength of all polypropylene geotextile reduced dramatically with the cumulative sunshine radiation on a horizontal surface of around 1500MJ/$\textrm{m}^2$.

Changes in Physicaochemical Properties of Soil, Yield, and Milling Quality of Rice Grown under the Long-term No-till Rice System (장기 무경운 벼 생산체계에서 토양의 이화학성, 쌀 수량 및 도정형질의 변화)

  • This experiment was conducted to evaluate the differences in physicochemical properties of soil, grain yield and milling recovery ratio and grain appearance of rice grown in long-term no-till and ordinary till systems. The paddy in no-till rice system was unploughed but straw-mulched for 15 years from 1988 to 2002 at the Experimental Paddy of Gyeongnam Agricultural Research and Extension Services, while the paddy in ordinary till system was ploughed and puddled every crop year, A 5cm organic layer was formed in 11-year no-till rice system, in which exchangeable cation and phosphate were accumulated. In no-till paddy organic matter, bulk density and solid phase of surface soil significantly increased, while pH, exchangeable cation, phosphate, liquid and gaseous phase decreased. Tillage made the very top soil soft, but made deep soil below 5cm significantly hard. In the 1st year of no-till, topsoil showed hard, but in the process of the time in no-till system, the top- and sub-soil showed softer, Yield and yield components of rice showed no differences between till- and no-till rice systems. Milling recovery ratio and grain appearance were not significantly different between no-till and till-rice systems.

Aluminum Powder Metallurgy Current Status, Recent Research and Future Directions

  • The increasing interest in light weight materials coupled to the need for cost -effective processing have combined to create a significant opportunity for aluminum P/M. particularly in the automotive industry in order to reduce fuel emissions and improve fuel economy at affordable prices. Additional potential markets for Al PIM parts include hand tools. Where moving parts against gravity represents a challenge; and office machinery, where reciprocating forces are important. Aluminum PIM adds light weight, high compressibility. low sintering temperatures. easy machinability and good corrosion resistance to all advantages of conventional iron bm;ed P/rv1. Current commercial alloys are pre-mixed of either the AI-Si-Mg or AL-Cu-Mg-Si type and contain 1.5% ethylene bis-stearamide as an internal lubricant. The powder is compacted in closed dies at pressure of 200-500Mpa and sintered in nitrogen at temperatures between $580~630^{\circ}C$ in continuous muffle furnace. For some applications no further processing is required. although most applications require one or more secondary operations such as sizing and finishing. These sccondary operations improve the dimension. properties or appearance of the finished part. Aluminum is often considered difficult to sinter because of the presence of a stable surface oxide film. Removal of the oxide in iron and copper based is usually achieved through the use of reducing atmospheres. such as hydrogen or dissociated ammonia. In aluminum. this occurs in the solid st,lte through the partial reduction of the aluminum by magncsium to form spinel. This exposcs the underlying metal and facilitates sintering. It has recently been shown that < 0.2% Mg is all that is required. It is noteworthy that most aluminum pre-mixes contain at least 0.5% Mg. The sintering of aluminum alloys can be further enhanced by selective microalloying. Just 100ppm pf tin chnnges the liquid phase sintering kinetics of the 2xxx alloys to produce a tensile strength of 375Mpa. an increilse of nearly 20% over the unmodified alloy. The ductility is unnffected. A similar but different effect occurs by the addition of 100 ppm of Pb to 7xxx alloys. The lend changes the wetting characteristics of the sintering liquid which serves to increase the tensile strength to 440 Mpa. a 40% increase over unmodified aIloys. Current research is predominantly aimed at the development of metal matrix composites. which have a high specific modulus. good wear resistance and a tailorable coefficient of thermal expnnsion. By controlling particle clustering and by engineering the ceramic/matrix interface in order to enhance sintering. very attractive properties can be achicved in the ns-sintered state. I\t an ils-sintered density ilpproaching 99%. these new experimental alloys hnve a modulus of 130 Gpa and an ultimate tensile strength of 212 Mpa in the T4 temper. In contest. unreinforcecl aluminum has a modulus of just 70 Gpa.

Radioactivity Analysis of $^{55}Fe\;and\;^{63}Ni$ in Dismantled Concrete (해체 콘크리트 폐기물에 포함된 $^{55}Fe$$^{63}Ni$ 방사능 분석)

  • Combined method of extraction chromatography and liquid scintillation analysis was established for determinating $^{55}Fe\;and\;^{63}Ni$ radioactivity in solid samples. Activated concrete wastes generated from dismantling KRR-2 were analysed. The sequential separation including precipitation and extraction chromatography resulted in the above 90% chemical recoveries of Fe. Above 62% recoveries of Ni were obtained by this procedure exception to 43.6 and 46.5% recoveries. The seperation and counting procedure was also confirmed with spiked samples of known quantity. The measured and spiked quantity were agreed with the 3.7% and 0.7% variations in the $^{55}Fe\;and\;^{63}Ni$ experiments, respectively. The radioactivities of $^{55}Fe$ in the dismantled concretes are shown from below MDA to maximum 362 Bq/g. The radioactivities of $^{63}Ni$ in all concrete samples are below MDA. The $^{63}Ni$ doesn't exist in dismantled concretes from KRR-2. The radioactivity of $^{55}Fe$ is decreased rapidly as the sampling depth is increased from the concrete surface.

A Study on the Characteristic of Indoor Green-Wall Design - With Focused on Preference of Emotional Image Language - (실내 벽면녹화디자인 특성 연구 - 감성이미지 언어에 따른 선호도를 중심으로 -)

  • This study examines potential growth and different perspectives on green-walls, which are being re-evaluated recently. The aim is to identify the viewer preferences concerning the walls by collecting a wide spectrum of information to create an interior design related comprehensive and intellectual database. Also based on this proposition, choosing a green surface of indoor wall as a specific example stimulus, identifying a relationship betweeationsviewer's preference factors. The relationship will formulate detailed and absolute qualities, which will reach potential areas that the green walls can be widely applied in. As a result of a factor analysis, the viewers classified into the 3 factors as is pleasant, gorgeous and rich about the stimulus. Preferred key factors, which are closely related to emotional image language, were; cool, tidy, comfortable and beautiful. The common factors in preferred design stimulus, in order of importance are color>elements>image/form/plants. Specific levels of design factors according to relevance are contrast>furniture> modern>central/creeper foliage plants. In the meantime, this study is leading the process of quantitative measurement of green-walls to a new design direction and it is critical to consistently experimenting to back up the theory with solid evidence.
