Absorption Study of Genistein Using Solid Lipid Microparticles and Nanoparticles: Control of Oral Bioavailability by Particle Sizes

  • In this study, the effect of particle size of genistein-loaded solid lipid particulate systems on drug dissolution behavior and oral bioavailability was investigated. Genistein-loaded solid lipid microparticles and nanoparticles were prepared with glyceryl palmitostearate. Except for the particle size, other properties of genistein-loaded solid lipid microparticles and nanoparticles such as particle composition and drug loading efficiency and amount were similarly controlled to mainly evaluate the effect of different particle sizes of the solid lipid particulate systems on drug dissolution behavior and oral bioavailability. The results showed that genistein-loaded solid lipid microparticles and nanoparticles exhibited a considerably increased drug dissolution rate compared to that of genistein bulk powder and suspension. The microparticles gradually released genistein as a function of time while the nanoparticles exhibited a biphasic drug release pattern, showing an initial burst drug release, followed by a sustained release. The oral bioavailability of genistein loaded in solid lipid microparticles and nanoparticles in rats was also significantly enhanced compared to that in bulk powders and the suspension. However, the bioavailability from the microparticles increased more than that from the nanoparticles mainly because the rapid drug dissolution rate and rapid absorption of genistein because of the large surface area of the genistein-solid lipid nanoparticles cleared the drug to a greater extent than the genistein-solid lipid microparticles did. Therefore, the findings of this study suggest that controlling the particle size of solid-lipid particulate systems at a micro-scale would be a promising strategy to increase the oral bioavailability of genistein.

Potential Dioxin and Furan Sources from Hospital Solid Waste Streams : A Pilot Study

  • This Pilot study identifies potential dioxin and furan sources and medical plastic wastes produced from hospital solid waste streams. In this study, air emissions of dioxins and furans from sources in the U.S., which were estimated by the U.S. Environmental Protection Agency (EPA), were summarized. Potential loading routes of dioxins and furans to the U.S.-Canada Great Lakes region have also been identified in trcent surveys. In addition, medical and hospital solid wastes produced in typical large city hospitals were characterized as important potential sources of dioxins and furans. Plastic contents in medical Plastics Characterization Survey (MMPCS), plastics composition data were obtained from a survey of five Massachusetts Hospitals and Medical Centers. By identifying plastic wastes as a percentage of total hospital wastes, we were able to use data from a preliminary study that analyzed the waste stream of 16 major New York City hospitals (NYCMWS) characterizing the plastic content of the wasters. This study determined the types of plastic wastes included in each medical waste stream (regulated medical waste or non regulated medical waste) and it discussed the potential for recyclibility of hospital plastic wastes. The combination of the NYCMWS and the MMPCS surveys provides for the first overview of the size of the recycling task of hospital plastic wastes and the potential of dioxin elimination if dioxin generating plastics were to be eliminated from hospital use.

Effect of Overlayer Thickness of Hole Transport Material on Photovoltaic Performance in Solid-Sate Dye-Sensitized Solar Cell

  • The photovoltaic performance of solid-state dye-sensitized solar cells employing hole transport material (HTM), 2,2',7,7'-tetrakis-(N,N-di-p-methoxyphenyl-amine)-9,9'-spirobifluorene (spiro-MeOTAD), has been investigated in terms of HTM overlayer thickness. Two important parameters, soak time and spin-coating rate, are varied to control the HTM thickness. Decrease in the period of loading the spiro-MeOTAD solution on $TiO_2$ layer (soak time) leads to decrease in the HTM overlayer thickness, whereas decrease in spin-coating rate increases the HTM overlayer thickness. Photocurrent density and fill factor increase with decreasing the overlayer thickness, whereas open-circuit voltage remains almost unchanged. The improved photocurrent density is mainly ascribed to the enhanced charge transport rate, associated with the improved charge collection efficiency. Among the studied HTM overlayer thicknesses, ca. 230 nm-thick HTM overlayer demonstrates best efficiency of 4.5% at AM 1.5G one sun light intensity.

A Study on the Product Design and Prototype Manufacturing of a Plastic V-Belt Pulley (플라스틱 V-벨트 풀리 설계 및 시작품 제작에 관한 연구)

  • In this study, product design and prototype manufacturing of a plastic water-pump pulley has been tried. The designed model is supposed to be made of 33 % glass reinforced resin of which the tensile strength is 180 MPa, and has 24 ribs on each side to increase its structural strength. Structural analysis under a static load of 300 kgf acting on both edges of the belt has been carried out using a commercial finite element code, MARC. The analysis result showed the maximum effective stress near a rib of designed model would be at most 35 MPa (less than 20% of the tensile strength), therefore, the plastic product would be sufficiently safe under that loading condition. On the basis of the structural analysis, a prototype of the designed model has been manufactured by using the fused deposition modelling (FDM) method which is one of the rapid prototyping (RP) methods, using ABS resin and support materials. The CAD data exported to the RP system in STL format was prepared by a commercial solid modeling software, SolidWorks. It has been proved that the plastic pulley can successfully replace the existing flow-formed steel product.

  • A development of a composite solid propellant is carried out for an application to gas generators as an energy source of rocket system. With HTPB as a propellant binder which has 80% of particle loading ratio, a favorable rheology, and moderate curing properties at the range of $-50^{\circ}C{\sim}70^{\circ}C$, AN is selected as the first kind of oxidizer having the characteristics of a low flame temperature, minimal particle residual as well as nontoxic products. AP is the second oxidant for ballistic property control. A series of experiments for the improvement of physical properties were conducted and resulted in the propellant formulation having 30% of strain rate at 8 bar of max. stress.

Direct design of partially prestressed concrete solid beams

  • Tests were conducted on two partially pre-stressed concrete solid beams subjected to combined loading of bending, shear and torsion. The beams were designed using the Direct Design Method which is based on the Lower Bound Theorem of the Theory of Plasticity. Both beams were of $300{\times}300mm$ cross-section and 3.8 m length. The two main variables studied were the ratio of the maximum shear stress due to the twisting moment, to the shear stress arising from the shear force, which was varied between 0.69 and 3.04, and the ratio of the maximum twisting moment to the maximum bending moment which was varied between 0.26 and 1.19. The required reinforcement from the Direct Design Method was compared with requirements from the ACI and the BSI codes. It was found that, in the case of bending dominance, the required longitudinal reinforcements from all methods were close to each other while the BSI required much larger transverse reinforcement. In the case of torsion dominance, the BSI method required much larger longitudinal and transverse reinforcement than the both the ACI and the DDM methods. The difference in the transverse reinforcement is more pronounce. Experimental investigation showed good agreement between design and experimental failure loads of the beams designed using the Direct Design Method. Both beams failed within an acceptable range of the design loads and underwent ductile behaviour up to failure. The results indicate that the Direct Design Method can be successfully used to design partially prestressed concrete solid beams which cater for the combined effect of bending, shear and torsion loads.

Two dimensional time-dependent creep analysis of a thick-walled FG cylinder based on first order shear deformation theory

  • In this paper the time-dependent creep analysis of a thick-walled FG cylinder with finite length subjected to axisymmetric mechanical and thermal loads are presented. First order shear deformation theory (FSDT) is used for description of displacement components. Inner and outer temperatures and outer pressure are considered as thermo-mechanical loadings. Both thermal and mechanical loadings are assumed variable along the axial direction using the sinusoidal distribution. To find temperature distribution, two dimensional heat transfer equation is solved using the required boundary conditions. The energy method and Euler equations are employed to reach final governing equations of the cylinder. After determination of elastic stresses and strains, the creep analysis can be performed based on the Yang method. The results of this research indicate that the boundaries have important effects on the responses of the cylinder. The effect of important parameters of this analysis such as variable loading, non-homogeneous index of functionally graded materials and time of creep is studied on the behaviors of the cylinder.

Enhanced Liver Targeting by Synthesis of $N_{\b{1}}-Stearyl-5-Fu$ and Incorporation into Solid lipid Nanoparticles

  • To enhance the liver targeting and reduce the side effects of 5-fluorouracil (5-Fu), it was acylated by stearyl chloride to obtain .$\b{N}_{\b{1}}$stearyl-5-Fu (5-FuS). The chemical structure of the prodrug was confirmed by Nuclear Magnetic Resonance and Infrared Spectrometry. 5-FuS was incorporated into solid lipid nanoparticles (SLN), which were prepared by the physical agglomeration method. The mean diameter of 5-FuS-SLN was 240.19 nm and the drug loading was 20.53%. The release characteristics in vitro of 5-FuS-SLN were fitted to the first-order pharmacokinetic model. Compared with 5-Fu injection, a study on the distribution of 5-FuS-SLN in mice showed that 5-FuS-SLN could double 5-Fu concentration in mice livers. The main pharmacokinetic parameters of 5-FuS-SLN in rabbits is shown as follows: $V_d$=0.04336L/kg, $T_{1/2} \beta$=1.2834h, CL=0.1632 L/h. In conclusion, 5-FuS-SLN has significant liver targeting properties. The employment of a prodrug to enhance drug liposoluble properties and the preparation method presented in this paper, seem to be an alternative strategy to the traditional colloidal delivery system.

Hydrogenation of Ethyl Acetate to Ethanol over Bimetallic Cu-Zn/SiO2 Catalysts Prepared by Means of Coprecipitation

  • A series of bimetallic Cu-Zn/$SiO_2$ catalysts were prepared via thermal decomposition of the as-synthesized $CuZn(OH)_4(H_2SiO_3)_2{\cdot}nH_2O$ hydroxides precursors. This highly dispersed Cu-solid base catalyst is extremely effective for hydrogenation of ethyl acetate to ethanol. The reduction and oxidation features of the precursors prepared by coprecipitation method and catalysts were extensively investigated by TGA, XRD, TPR and $N_2$-adsorption techniques. Catalytic activity by ethyl acetate hydrogenation of reaction temperatures between 120 and $300^{\circ}C$, different catalyst calcination and reduction temperatures, different Cu/Zn loadings have been examined extensively. The relation between the performance for hydrogenation of ethyl acetate and the structure of the Cu-solid base catalysts with Zn loading were discussed. The detected conversion of ethyl acetate reached 81.6% with a 93.8% selectivity of ethanol. This investigation of the Cu-Zn/$SiO_2$ catalyst provides a recently proposed pathway for ethyl acetate hydrogenation reaction to produce ethanol over Cu-solid base catalysts.

The Measurement Technique of Burning Rate in Solid Proplellant at High Pressure (고체추진제의 고압 연소속도 측정기법)

  • The burning characteristics of HTPB/AP solid propellants were measured by closed bomb of internal volume of 200 cc and 700 cc up to 30,000 psia. The burning rates of closed bomb method showed good agreement with those of strand burner method between 1,000 psia and 5,000 psia, and the sharp increment of pressure exponent(n) around 6,000 psia as a result of testing in accordance with loading densities. The burning rate measured in 200 cc and 700 cc of internal volume of closed bomb agreed well without the relation of internal volume size.
