Fermentation Characteristics of Honey Wine by Saccharomyces bayanus (Saccharomyces bayanus를 이용한 벌꿀 발효주의 양조 특성)

  • Fermentation characteristics for the production of honey wine (mead and melomel) was investigated. Among the yeast strains tested, Saccharomyces bayanus showed higher alcohol production and better fermentability than the other strains at low temperature. Optimum pH and temperature for the production of honey wine by Saccharomyces bayanus were pH 4.0 and $20^{\circ}C$, respectively, and optimum soluble solid content of diluted honey solution for the fermentation was between 24 and $27^{\circ}Brix$. Total acidity and pH of honey wine (mead) did not change considerably during the whole period of fermentation, but those of Tangerine and Japanese apricot honey wine (melomel) changed during the fermentation. As the fermentation progressed, reducing sugar decreased continuously until the late period of fermentation, while alcohol content increased continuously during the same period. After fermentation of 21 days, honey wine consisted of about $8.5{\sim}9.1^{\circ}Brix$ of soluble solid, $1.90{\sim}2.32%$ of reducing sugar with the conversion rate of $90{\sim}92%$. After 21 days of fermentation, alcohol contents of mead fermented with polyflower and acasia flower were 13.3 and 13.7%, respectively. Final alcohol content was not affected significantly by the source of honey. While pH of the osmotically extracted fruit honey juice decreased rapidly to pH $2.92{\sim}2.97$ after 13 days of fermentation, total titratable acidity of Tangerine and Japanese apricot honey wine were 0.30 and 0.53%, respectively. After 13 days of fermentation, reducing sugar of fruit honey wines were reduced to $2.03{\sim}2.87%$, alcohol content were reached up to 13.1 and 12.5% for Tangerine and Japanese apricot honey wine, respectively. Generally, honey extracted fruit juices were fermented more efficiently than diluted honey juice.

The changes of pectic substances of $emph{Kimchis}$ with different garlic contents during the fermentation periods (마늘첨가량을 달리한 김치의 펙틴질의 변화)

  • The changes of acidity, the content of alcohol-insoluble solid (AIS) and pectic substances of Kimchis which were prepared with various garlic contents and fermentation at $21^{\circ}C$ were investigated. Acidity and the content of water soluble pectin of Kimchis increased but the content of AIS and protopectin decreased during the fermentation. As the Kimchis contained high garlic contents compared with the Kimchis deleted garlic, acidity of those rapidly increased but the content of AIS of those slightly decreased with fermentation. In the case of Kimcltis contained high garlic contents, protopectin decreased and water soluble pectin increased with fermentation, but the amount of changes was small.

  • Soybean meal (SBM), a commonly used protein source for animal feed, contains anti-nutritional factors such as trypsin inhibitor, phytate, oligosaccharides among others, which limit its utilization. Microbial fermentation using bacteria or fungi has the capability to improve nutritional value of SBM by altering the native composition. Both submerged and solid state fermentation processes can be used for this purpose. Bacterial and fungal fermentations result in degradation of various anti-nutritional factors, an increase in amount of small-sized peptides and improved content of both essential and non-essential amino acids. However, the resulting fermented products vary in levels of nutritional components as the two species used for fermentation differ in their metabolic activities. Compared to SBM, feeding non-ruminants with fermented SBM has several beneficial effects including increased average daily gain, improved growth performance, better protein digestibility, decreased immunological reactivity and undesirable morphological changes like absence of granulated pinocytotic vacuoles.

Mevinolin Production by Monascus pilosus IFO 480 in Solid State Fermentation of Soymeal

  • Mevinolin, a fungal metabolite, is a potent inhibitor of 3-hydroxy-methyl-3-glutaryl-coenzyme A (HMG-CoA) reductase, the rate-controlling enzyme in cholesterol biosynthesis. In this investigation, the optimum factors for mevinolin production by Monascus pilosus IFO 480 in soymeal fermentation were studied. The highest yield of mevinolin, 2.82 mg mevinolin per g dry weight, without citrinin (a toxic fungal secondary metabolite) was obtained after 21 days of fermentation at $30^{\circ}C$ at 65% moisture content, particle size 0.6-0.9 mm, and initial substrate pH of 6.0. Mevinolin was present in the fermentation substrate predominantly in the hydroxycarboxylate form (open lactone, 92.1-97.3%), which is currently being used as a hypocholesterolemic agent.

The Changes of Pectic Substances and Enzyme Activity, Texture, Microstructure of Anchovy Added Kimchi (멸치 첨가 김치의 숙성 중 펙틴 함량, 효소 활성, 조직감과 미세구조의 변화)

  • This study was intended to observe the changes of pectic substances and enzyme activities, texture, microstructure of anchovy added kimchi during fermentation for 4 weeks at 4$^{\circ}C$. Content of alcohol insoluble solid(AIS) and HCl soluble pectin(HClSP) were decreased, whereas content of hot water soluble pectin(HWSP) was increased during fermentation. Content of HClSP was higher and HWSP was lower in anchovy added kimchi than control. Activity of pectinesterase(PE) was decreased, whereas activity of polygalacturonase(PG) was increased during fermentation. In anchovy added kimchi, PG activity was lower than control. Changes in microstructure of Chinese cabbage and kimchi during fermentation was lower than control. Changes in microstructure of Chinese cabbage and kimchi during fermentation was observed ; in the raw cabbage, parenchyma cells, intercellular space and middle lamella were clearly shown. But in salted cabbage, middle lamella became separated. In the late stage of fermentation, parenchyma cell walls were wrinkled and collapsed. Puncture forces of kimchi were decreased, whereas cutting forces of kimchi were increased as fermentation proceeded. The firmness was slightly higher in anchovy added kimchi than control at the late stage fermentation, which may be explained by the PG activity.

Changes of Vitamin C and Fermentation Characteristics of Kimchi on Different Cabbage Variety and Fermentation Temperature (배추품종 및 숙성온도를 달리한 김치의 발효 및 비타민 C의 변화양상)

  • This study was conducted to determine the changes in the contents of vitamin C and fermentation characteristics of kimchi on different Chinese cabbage variety(Koreangji Yureum: KY, Kalak Shin Il Ho: KS) and fermentation temperature(5oC, 15oC). Kimchi were fermented at 5oC and 15oC for 140 days and 25 days respectively. In all samples, total acidity increased, and the pH, total sugar and reducing sugar content decreased as the fermentation proceeded. Also, the number of lactic acid bacteria reached the maximum at the optimum fermenting stage. On the other hand, total ascorbic acid, dehydroascorbic acid and ascorbic acid from individual kimchi solid tissue significantly decreased with fermentation periods, however, increased kimchi liquid at the 7day in KS 15(kimchi prepared with Kalak Shin Il Ho at 15oC), KY 15(kimchi prepared with Korangji Yureum at 15oC) and at the 65day in KY 5(kimchi prepared with Korangji Yureum at 5oC) respectively. Therefore, different fermentation temperature as well as variety affected ascorbic acid contents and chemical characteristics of kimchi.

  • In order to use the glutamic acid fermentation residuum in agriculture effectively, comparison experiments of the residua liquid and dried solid, and compost on radish (Raphanus Sativus) and chinese cabbage (Brassica chinesis) were carried out, together with an investigation on some physical property changes of soil brought about by the use of the residua and compost. 1. An equal or somewhat superior effect of the glutamic acid fermentation residua solid and liquid, to urea was observed. 2. Both residua liquid and dried solid, tended to acidify soil and the tendency was observed to be somewhat servere in the latter. 3. The liquid fermentation residuum compounded with minor elements such as iron, manganese, zinc, copper and boron increased the yield of chinese cabbage compared with the liquid residuum alone. 4. Not only as a N, K fertilizer, the liquid residuum of glutamic acid fermentation could also be used as a source for liquid or solid compound fertilizer.

  • Microbial production of cellulolytic enzymes by Aspergillus niger in solid state fermentation (SSF) and submerged state fermentation (SF) in laboratory scale was compared. Czapek Dox liquid broth amended with cellulose (0.5%) was used for cultivation in SF, whereas rice bran was used as a solid support, moistened with cellulose, amended Czapek Dox broth for growth in SSF. The production of Carboxymethyl cellulase, Filter paperase and ${\beta}$-Glucosidase was monitored at regular intervals. The peak production of the enzymes occurred within 3 days of incubation in SSF as against $\geq$ 7 days in SF. SSF gave higher yields of enzymes in comparison to SF. Maximum titres of 0.40, 0.62 and 0.013 U/ml in respect of FPase, CMCase and ${\beta}$-glucosidase in SSF were recovered as against 0.13, 0.06 and 0.0013 U/ml in SF respectively, at their respective peak time intervals. Hence, SSF appeared to be a better choice for production of cellulolytic enzymes by Aspergillus niger.

Physicochemical Properties of Dongchimi Added with Gatt (Brassica juncea)

  • To improve Dongchimi (watery radish kimchi) quality and preservation, 0, 3, 5, 10, and 15% of gatt (Brassica juncea; leaf mustard) per radish was added. Chemical characteristics were determined during fermentation at $10^{\circ}C$ for 45 days. Total acidity increased slowly by addition of gatt during initial fermentation period, and 15% treatment showed lowest total acidity. Total vitamin C content increased initially in all treatments depending on gatt content, and decreased thereafter. Period for reaching maximum value was delayed by addition of gatt. Highest total vitamin C was found in 15% treatments. In the case of reducing sugar, 5% treatment showed highest contents. Lactic, succinic, and tartaric acid contents consistently increased during fermentation, while those of malic and citric decreased. Turbidity and total solid contents of Dongchimi liquid increased in all treatments as fermentation proceeded, although the extent was rather suppressed by addition of gatt. Colorimetric lightness values decreased, while the initial increased and then decreased in redness and yellowness. Addition of gatt at above 15% weight level per radish accelerated fermentation at the later fermentation stage thus it should be avoided. Most changes in typical characteristics of fermentation were similar depending on treatments. More acceptable Dongchimi could be prepared by fermenting at 5% gatt concentration under given conditions.

Bioprocess Strategies and Recovery Processes in Gibberellic Acid Fermentation

  • Gibberellic acid (GA$_3$) is a commercially important plant growth hormone, which is gaining much more attention all over the world due to its effective use in agriculture and brewing industry. Industrially it is produced by submerged fermentation technique using Ascomycetous fungus Gibberella fujikuroi. Solid state and immobilized cell fermentation techniques had also been developed as an alternative to obtain higher yield of GA$_3$. This review summarizes the problems of GA$_3$ fermentation such as production of co-secondary metabolites along with GA$_3$, substrate inhibition and degradation of GA$_3$ to biologically inert compound gibberellenic acid, which limits the yield of GA$_3$ in the fermentation medium. These problems can be overcome by various bioprocessing strategies e.g. two - stage and fed batch cultivation processes. Further research on bioreactor operation strategies such as continuous and / or extractive fermentation with or without cell recycle / retention system need to be investigated for improvement in yield and productivity. Down stream processing for GA$_3$ isolation is also a challenge and procedures available for the same have been critically evaluated.
