Isolation and Characterization of Eukaryotic Translation Initiation Factor 5A (eIF-5A) from Potato (감자로부터 Eukaryotic Translation Initiation Factor 5A (elF-5A) 유전자의 동정 및 발현 분석)

  • Differential display based on PCR was employed to identify genes expressed during tuber-developing stage of potato (Solanum tuberosum L. cv. Irish Cobbler). An eukaryotic initiation factor 5A (eIF-5A) clone isolated from a cDNA library constructed with developing micro-tuber using a probe of PCR fragment. We isolated three positive clones and ore of them contained open reading frame. This clone revealed high sequence similarity to tomato eIF 5A cDNA. At the DNA level, there is 94.8% identity with the tomato eIF-5A4, whereas at the protein level there is a high identity with 97.5%. The potato eIF 5A clone is 716 bp in length and contains a single open reading frame from 57 to 539 bp, a 56 bp 5'-untranslated region and a 177 bp 3'-untranslated region. The deduced protein composed of 160 amino acid residues, with a predicted molecular mass of 17.4 kD and an estimated pl of 5.5. The sequence of 12 (STSKTGKHGHAK) amino acids among eIF-5A proteins is perfectly conserved from yeast to human. That sequence in potato eIF-5A protein is also conserved at position 46 to 57 amino acid. This region embeds the post-translational modification site of the lysine residue (at the seventh K) to hypusine that is crucial to eIF-5A activity. The northern blot analysis of eIF5A has shown abundant expression, mainly in flower organs (stamen, ovary, petal, sepal), fruit and stolen.

Effects of Tuber Size on Field Growth and Yield of 'Dejima' Potatoes Produced through Wick-based Hydroponics (심지양액재배 '대지'의 씨감자 크기에 따른 포장 생육과 수량)

  • Two field experiments were carried out in 2002 to evaluate effects of the sizes of tubers produced through a wick-based hydroponics on growth and yield of seed potatoes (Solanum tuberosum L. 'Dejima'). Nine sizes of seed tubers (<1, 1~3, 3~5, 5~7, 7~10, 10~20, 20~30, 30~50, and 50~80 g) were used in spring crop. In fall crop, however, tubers except for the 1 g and 50~80 g wereused with tubers 7 and 15 g produced through aeroponics. Emergence rate greatly increased as tuber size was increased up to 10~20 g and then leveled off in both croppings. The number of days from planting to emergence in spring crop was about 34 days regardless of tuber size, while that in fall crop decreased from 34 to 16 days as tuber size increased from 1~3 to 30~50 g. The number of stems per plant, stem length and diameter, and leaf size tended to increase with tuber size. As the seed tuber size increased up to 10~20 g, the number of tubers per plant, average tuber weight, total and seed tuber yields linearly increased and then slowly increased with seed tuber size. Total and seed potato yields were not significantly different between tubers produced through a wick-based hydroponics and an aeroponics when similar sized tubers were planted in the fall cropping.

Effects of Seed Storage Methods and GA3 Application on Seed Germination and Seedling Growth of Solanum lyratum Thunb. (종자저장방법 및 GA3처리가 배풍등 종자 발아와 유묘 생육에 미치는 영향)

  • This study was conducted to determine the effects of seed storage methods ($-20^{\circ}C$, stratification, $2^{\circ}C$ dry, $2^{\circ}C$ wet and room temperature) and $GA_3$ application (control, $dH_2O$, 10, 100, 1000 ppm) on seed germination and seedling growth of S. lyratum. As a result seed germination rate of S. lyratum was the highest at 91% when seeds were stored at room temperature and then soaked for 24 hours in $GA_3$ 10 ppm. And seedlings of S. lyratum showed the best quality when seed were stored at $2^{\circ}C$ dry and then soaked for 24 hours in $GA_3$ 1000 ppm, with the growth characteristics of plant height (47 mm), number of leaves (8), leaf width (12 mm), leaf length (19 mm), fresh weight (aerial/root part; 471/476 mg), dry weight (aerial/root; 106/41 mg) and seedling quality indices (106). Therefore, S. lyratum seed were stored at $2^{\circ}C$ dry, and then soaked for 24 hours in $GA_3$ 1000 ppm, seed germination rate was more than 80% and production of superior quality container seedlings.

  • To understand the properties of the cauliflower mosaic virus (CaMV) 35S promoter and the effect of the deleted maize alcohol dehydrogenase I-S (Adhl-S) intron 1 on the expression of the CaMV $35S{\beta}-glucuronidase$ (GUS) gene in potato (Solanum tuberosum L. cv. Superior), we constructed a chimeric gene and transferred it into potato with Agrobacterium tumefaciens mediated method. The pLS201, a gene transfer vector of 17.7 kilobase pairs, was composed of the CaMV 35S promoter, the 249 base pairs of deleted maize Adhl-S intron 1, the GUS reporter gene, and the kanamycin resistance gene as a selectable marker for transformation. The GUS activity was examined by histochemical and spectrophotometric assay in transformed potato plants. The GUS activity was found primarily around the vascular tissue cells in stem and root. In the spectorophotometric assay, the level of GUS activity of transgenic potato transformed with CaMV 35S/249 bp of intron 1 fragment-GUS (pLS201) was compared with that of potato transformed with CaMV 35S-GUS (pBI121). The quantitative spectrophotometric assay showed that the level of GUS activity in potato transformed with pLS201 was higher in leaf, stem and root by 30-, 34- and 42-fold, respectively than those in potato transformed with pBI121. This results indicate that the inclusion of the deleted maize Adhl-S intron 1 resulted in increament of the GUS gene expression in transgenic potato.potato.

Selection of Herbicide Resistant Potatoes Transformed with Phosphinothricin Acetyltransferase Gene (Phosphinothricin Acetyltransferase 유전자 도입에 의한 제초제 저항성 감자의 선발)

  • This experiment was conducted to introduce PAT (phosphinothricin acetyltransferase, non-selective herbicide bialaphos resistant gene) gene into potato (Solanum tuberosum. cv. Desiree). Optimal shoot regeneration from leaf discs and stem segments was obtained in MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L BA, and the frequency of shoot regeneration was 54% in left discs and 46% in stem segments. In this condition, leaf discs and stem segments of potato were co-cultivated with A. tumefaciens MP90 which contained binary vector with GUS: :NPTII gene and PAT gene. Transgenic shoots were regenerated from leaf and stem-derived calli on selection medium with 100mg/L kanamycin. The 100${\mu}M$ acetosyringone treatment during the co-cultivation highly enhanced(4 times than the control) the shoot regeneration on selection medium. When the putative transgenic plants were transferred to medium with 10mg/L basta, all of them were survived. After PCR. GUS test, and Southern blot analysis of the survived plant, we confirmed that the gene was stably integrated into the potato genome and expressed. After the transgenic plants were transplanted in soil, and the transgenic plants were sprayed with the herbicide basta (300ml/10a), the transgenic plants remained green but control plants were died.

The Skin Protecting Effects of Ethanolic Extracts of Eggplant Peels (가지 외피 에탄올 추출물의 피부보호효과)

  • We investigated the in vitro antioxidant and antimelanogenesis effects of ethanol extracts from eggplant (Solanum melongena L.) peels. The total phenolics and chlorogenic acid in ethanol extracts were 2,465 mg/100 g and 2.08 mg/100 g, respectively. ABTS radical scavenging activity, ferric reducing/antioxidant power assay, and malondialdehyde (MDA) inhibitory effect of the extracts increased in a dose-dependent manner. In addition, the extracts generally showed strong UV absorption in the range of UV-B (290-320 nm). The $IC_{50}$ of mushroom tyrosinase inhibitory activity of ethanol extracts from eggplant peels was 870 ${\mu}g/mL$. Importantly, the melanin syntheses of B16/F10 melanoma cells were decreased by extracts in a concentration-dependent manner. Overall, these results suggest that eggplant peels can be potentially applied as a anti-melanogenic agent as well as an antioxidant resource.

Processing Quality of Potato (Solanum tuberosum L.) Tubers as Influenced by Soil and Climatic Conditions (감자의 가공품질에 영향을 미치는 토양 및 기상조건)

  • In order to examine the difference in processing quality of potato tubers among localities, chemical properties of soils were analyzed and climatic conditions were investigated. Potatoes (Solanum tuberosum L.) were grown at seven localities of Korea during two years from 1994 to 1995. Soil samples and tubers were obtained from 2 to 3 commercial farms per locality with 10 days interval from 70 days before harvesting. As the result of that, higher correlation in processing quality was found with organic material content among soil conditions. On the climatic conditions, minimum temperature and sunshine hours during the period from 30 to 11 days before harvesting exhibited highly significant negative correlations with all quality parameters except reducing sugar content. Additionally, regression equations based on the observed level of these factors showed the relatively high coefficients of determination for dry matter content and chip color. To produce higher quality potatoes for processing, therefore, climatic conditions such as minimum temperature and sunshine hour and soil condition such as organic matter content have to be considered before the selection of areas or fields.

First Report of Pink Rot of Potato (Solanum tuberosum) Caused by Phytophthora erythroseptica in Korea (Phytophthora erythroseptica에 의한 감자 홍색부패병 발생)

  • Pink rot of potato (Solanum tuberosum L.) occurred at Pyeongchang in Gangwon and at Bosung in Junnam province since 1999. The disease incidence in the surveyed areas was about 5% of harvested potatoes in 2002. Affected tubers showed a dull brown appearance and the lenticels and eyes on tubers turned dark brown. The cut surface of the diseased tuber colored faint pink and the entire surface of the diseased tuber becomes deep salmon pink within 30 min. The pathogen isolated from the diseased tubers was identified as Phytophthora erythroseptica based on morphological and cultural characteristics. Mycelial mat was fairly fluffy, rosette or stellate patterns and rounded or angular hyphal swellings were farmed in water, Temperature for mycelial growth was ranged from 5 to 3$0^{\circ}C$ and optimal temperature was $25^{\circ}C$. Non-papillate sporangia were persistent on stalk and ellipsoid, ovoid, obpyriform or distorted in shape, often with a constriction distal in the middle. Size of sporangia was 41.3~69.6$\times$26.8~47.4 (av, 55.5$\times$37.1) ${\mu}{\textrm}{m}$. Sexuality of Phytophthora erythroseptica was homothallic. Oogonia were 30~46 ${\mu}{\textrm}{m}$ in diameter and oospores were 28~35 ${\mu}{\textrm}{m}$ in diameter, Elongated or cyclindrical antheridia were all amphigynous. This is the first report on potato pink rot caused by Phytophthora erythroseptica in Korea.

Effects of Foliar Application of Chitosan and Seedling Types on Increasing the Production of Fall-growing Potatoes(Solanum tuberosum) in Jeju Island (Chitosan 처리와 종묘종류가 가을감자의 생산성 향상에 미치는 영향)

  • This study was conducted to investigate the effect of foliar application of Chitosan and seedling types on increasing the production of fall-growing potatoes(Solanum tuberosum) at the height of 250m from the sea in Jeju Island. The experiment main-plots consisted of four Chitosan applications, and subplots had three seedling types as cutting plug seedlings, mine-tuber plug seedling planting plot and seed potatoes planting plot. The foliar application of Chitosan of 2000ppm was carried out 0, 1, 3, 5 times at intervals of ten days after ten days of transplanting. Plant height and top dry weight were significantly more vigorous as three to five application of Chitosan. In the small tubers(under 80g), the number of tubers and tuber yield were relatively increased in the seed potatoes planting plot and mine-tuber plug seedling plot, but the large tubers(over 80g) was higher in the cutting plug seedling planting plot. T-N content in leaves was increased as the number of application of Chitosan was increased. A similar tendency was shown in K, P Ca, Mg and Na. The growth and tuber yield in cutting plug seedlings, mine-tuber plug seedling planting plot and seed potatoes planting plot were effectively increased as three to five foliar applications of Chitosan was increased.

Suppression of reactive oxygen species generation as a part of antioxidative effect of plant extracts (식물추출물 항산화효능 기전의 일부로서의 활성산소 발생 억제 효과)

  • Chemical scavenging of reactive oxygen species (ROS) is considered a major mechanism of antioxidant effects, but preventing ROS generation can be more efficient in attenuating oxidative damage. In this study, the extracts of plants, Solanum lycopersicum, Ailanthus altissima, Equisetum arvense, and Oenothera biennis, were tested to determine whether their antioxidative effects are driven by the prevention of superoxide generation from mitochondria, a major ROS generator. While all the extracts efficiently attenuated the elevation of ROS levels in human fibroblasts and inflammation-induced mice, those of S. lycopersicum, A. altissima, and O. laciniata only suppressed mitochondrial ROS generation and reduced levels of lipofuscin and lipid peroxidation. Furthermore, the extracts of A. altissima and O. laciniata extended the lifespan of fruit flies. Our results suggest that plant extracts with anti-oxidative effects differ in their ability to prevent ROS generation, which may be associated with the attenuation of oxidative damage in cells and animal tissues.
