용규(龍葵) 추출물이 백혈병 세포의 Apoptosis 유도에 미치는 영향 (Induction of Mitochondria-mediated Apoptosis by Solanum Nigrum in Leukemia Cells)

  • Objectives In human myeloid leukemia cells, there are no specific features of apoptosis compared with apoptosis in other cell types. Solanum nigrum L.(SNL) is a deciduous tree, which is widely distributed in Korea with reported anti-tumor, anti-inflammatory and non-specific immune-enhancing properties. Although the plant has been clinically used for treating a variety of diseases, its bioactive ingredients are unknown and its mode of action potential has never been investigated. Thus anti-tumor property of methanol extract was investigated. Methods In this study, anti-tumor property of methanol extract was investigated by determining its in vitro growth-inhibitory effects on human myeloid leukemia cells. XTT proliferation assay, DNA fragmentation, immunoblot analysis, densitometric analysis were used. Results 1. The methanol fraction of the extracts of SNL induced mitochondria-mediated apoptosis in human myeloid leukemia cells. 2. The methanol fraction exhibited relatively higher cytotoxic activity in a dose-dependent manner than chloroform, and hexane fraction. 3. Typical ladder profile of Oligonucleosomal fragments were appeared. 4. The secreted cytosolic cytochrome C level was increased by treatment of methanol fraction. Conclusions Methanol fraction of SNL is capable of inducing apoptosis in human myeloid leukemia cells.

백모등 엑스의 간기능개선 효과 (Effect of Solanum lyratum Extract on the Hepatotoxicity of Carbon Tetrachloride in Rats)

  • The studies were attempted to investigate the therapeutic effect of Solanum lyratum extract(SLE) on the liver damage induced by carbon tetrachloride in rats. Male rats (Sprangue-Dawley) were treated with subchronic concentration of $CCl_4$, which was 1 mg/kg of $CCl_4$ via per oral administration daily for three days. The level of serum cholesterol, transaminase, bilirubin and BSP were measured. Treatment of SLE significantly decreased the increment in serum transaminase activities, total cholesterol and enlargement of liver but serum bilirubin was not reduced. The protective effect was observed when SLE was given daily for 4 weeks after $CCl_4$ treated for 3 days. SLE administration to rats prevented the development of fatty change, necrosis, and fibrosis induced by $CCl_4$. The results suggest that SLE has appreciable therapeutic effect on $CCl_4$ induced hepatotoxicity.

Molecular Cloning and Expression of Dihydroflavonol 4-reductase Gene in Tuber Organs of Purple-fleshed Potatoes

  • A full-length cDNA encoding dihydroflavonol 4-reductase (st-dfr) of potato was isolated by rapid amplification of cDNA ends, and their expression was investigated from purple-fleshed potato (Solanum tuberosum L. cv. Jashim). The st-dfr exists as a member of a small gene family and its transcripts was abundant in the order of tuber flesh, stem, leaf, and root. The expressions of st-dfr gene were light inducible and cultivar dependant. Transgenic potato plants harboring antisense st-dfr (AS-DFR) sequences were analyzed. The accumulation of mRNA was nearly completely inhibited as a result of introducing an AS-DFR gene under the control of the 35S CaMV promoter into the red tuber skin Solanum tuberosum L. cv. Desiree. The anthocyanin content of the tuber peels of the transgenic lines was dramatically decreased by up to 70%. The possible production of flavonols in the peels of AS-DFR transgenic potatoes was discussed.

  • Phytochemical study on the aerial parts of Solanum lyratum (Solanaceae) was carried out. On the basis of phytochemical and spectroscopic evidences, compound I was identified as mixtures of hexadecanoic acid methyl ester, 2,6,10,15-tetramethyl heptadecane, tricosane, tetracosane, pentacosane, docosanoic acid methyl ester, docosane, tricosanoic acid methyl ester, 8-hexyl pentadecane, tetracosanoic acid methyl ester, pentatriacontane, hexatriacontane, eicosane, hexacosane, hentriacontane and stigmasta-5,23-dien $3-{\beta}-ol$, and compound II, III, IV and V were identified as hexacosanoic acid methyl ester, ${\beta}-sitosterol-{\beta}-{_D}-glucoside$, $3-O-{\alpha}-{_L}-rhamnopyranosyl-(1->2)-{\beta}-{_D}-glucuronopyranosyl$ diosgenin and $3-O-{\alpha}-{_L}-rhamnopyranosyl-(1->6)-{\beta}-{_D}-glucopyranosyl$ quercetin (rutin), respectively. Rutin was identified as one of the active principles having antioxidative effect from S. lyratum.

Production of glycoalkaloids from callus cultures of Solanum hainanense Hance

  • Leaf explants of the Solanum hainanense plant, grown in vitro, were cultured in basal Murashige and Skoog (MS) media supplemented with 0.5 mg/L kinetin and 1 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) for callus initiation. For maintenance and proliferation, the callus was cultured on MS medium supplemented with 1 mg/L 6-benzylaminopurine (BAP) and 0.5 mg/L 2,4-D. The glycoalkaloid content in the callus was at its maximum after ten weeks of culture (188.65 mg/g), whereas that of the one-year-old control was 22.22 mg/g in the root and 5.99 mg/g in the stem. The glycoalkaloid extracted from the callus inhibited the activity of collagenase on collagen gel. High performance liquid chromatography (HPLC) analysis showed that biotransformation occurred when a callus was grown on medium supplemented with various carbon sources. These results suggest that callus of S. hainanense is a good material for production of glycoalkaloid.

Characterization of Polyphenoloxidase extracted from Solanum tuberosum Jasim

  • Polyphenoloxidase (PPO) was extracted from Solanum tuberosum Jasim by various chromatographic methods and was subsequently purified and characterized. PPO was purified upto 78-fold from the crude extract. SDS-PAGE profile of the enzyme showed a major subunit of PPO with molecular weight of 40 kDa. The optimum pH and temperature for the maximum activity of PPO was 6.5 and $25^{\circ}C$, respectively. The enzyme was found to be quite stable between 10 and $40^{\circ}C$, whereas it was almost inactivated at $70^{\circ}C$ when incubated for 30 min. Substrate specificity study indicated that catechol was the most suitable substrate for PPO isolated from purple-fleshed potato with a $K_m$ value of 21.1 mM. The most effective inhibitor was ascorbic acid, followed by L-cysteine, citric acid, EDTA, and boric acid. Studies on the effect of metal ion on PPO activity showed that magnesium and copper were inhibitory, while iron and zinc ions increased the activity of PPO.

Solanum nigrum L. Extract Inhibits Inflammation in Lipopolysaccharide-stimulated Raw 264.7 and BV2 Cells

  • Solanum nigrum L. (SNL), generally known as black nightshade, is traditionally used as medicine to reduce inflammation caused by several diseases like asthma, chronic bronchitis and liver cirrhosis. In this study, anti-inflammatory effects of SNL extract were examined and possible molecular mechanisms of the anti-inflammatory effects were investigated. The inhibitory effects of SNL extract on nitric oxide (NO), pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6) and Matrix metallopeptidase 9 (MMP-9) productions were dissected using lipopolysaccharide (LPS) stimulated murine macrophage-like cell line Raw264.7 cells and human microglial cell line BV2 cells. We further investigated whether SNL extract could suppress the phosphorylation of ERK1/2, JNK, and p38 and the nuclear expression of nuclear factor $NF-{\kappa}B$ p65 in LPS-stimulated Raw264.7 cells and BV2 cells. As a result, we showed that the SNL extract significantly decreased the production of pro-inflammatory cytokines, NO, and MMP-9. In addition, the SNL strongly inhibited the phosphorylation of ERK1/2, JNK, p38 and nuclear translocation of $NF-{\kappa}B$ p65 in activated cells. We confirmed that the extracts of SNL effectively inhibits the anti-inflammatory and may be used as a therapeutic to various inflammatory diseases.

가지(Solanum melongena L.) 활성물질의 라디칼 소거능과 산화적 스트레스에 대한 세포 보호 효과 (Free radical scavenging activity and protective effect from cellular oxidative stress of active compound from eggplant (Solanum melongena L.))

  • To investigate the protective effect of eggplant (Solanum melongena L.) and its active compound, delphinidin, we used in vitro and cellular system. The active fraction from eggplant, BuOH fraction, showed protective effect from hydrogen peroxide-induced oxidative stress in WI-38 fibroblast cells. It suggests that eggplant would have the protective activity from radical-induced oxidative damage and its BuOH fraction would play the crucial role with antioxidative activity. In addition, delphinidin, the active compound from eggplant, exerted the strong 1,1-diphenyl-2-picrylhydrazyl scavenging effect with $IC_{50}$ value of 6.59 ${\mu}g/mL$. Furthermore, the cellular oxidative stress was induced by 2,2'-azobis (2-amidinopropane) dihydrochloride (AAPH) in LLC-$PK_1$ cells, while treatment of delphinidin atteunated AAPH-induced oxidative stress as dose-dependent manner. The present study suggests the antioxidative activity of eggplant and delphinidin against free radical-induced oxidative stress.

감자(Solanum tuberosum)의 기내 소괴경 형성 단계에 따른 Peroxidase와 Esterase 동위효소의 양상 변화 (Changes in Isozyme Patterns of Peroxidase and Esterase during the Microtuberization of Potato(Solanum tuberosum))

  • The microtuber was efficiently formed on SH medium containing 9% sucrose from the in vitro propagated shoot of potato (Solanum tuberosum cv. Sumi). In order to investigate gene expression depending on the development stage of microtuber, we examined the changes of peroxidase and esterase activities, and their isozyme patterns as well. Peroxidase and esterase activities were the highest at the 7 day-culture of the microtuber and subsequently decreased on the stage of microtuberization, whereas esterase activity increased at the stage of 60 day-culture. However, their activities in the ordinary tuber were higher than those of 60 day-cultured microtuber. In addition, in the peroxidase isozyme pattern two new bands of pI 7.05 and pI 4.65 were appeared at the 15- day and 60 day-cultures, respectively, as shown by isoelectric focusing. Various bands in the sterase isozyme pattern were shown at the 7 day-culture, and the band patterns were a large difference, comparing those of shoot and tuber. New bands in the esterase isozyme pattern also appeared at the 15 day- (pI4.52) and 60 day-cultures (pI 4.48). These results suggest that the changes of peroxidase and esterase activities and isozyme patterns are an important factor in the differentiation and development of potato.

Suppressive Effects of Potato (Solanum tuberlosum) on Type II Collagen-Induced Arthritis in DBA/1J Mice

  • Collagen-induced arthritis (CIA) is a model for some types of human autoimmune rheumatoid arthritis (RA). In this study, we examined whether ethanol extract of potato (Solanum tuberosum) is efficacious against CIA in mice. Potato extracts (100 and 200 mg/kg) were orally administered to DBA/1J mice once daily for 49 day after initial immunization with type II collagen. Clinical assessment of disease and measurement of paw edema were conducted throughout the study. The production of CIA-related rheumatoid factor, anti-type II collagen antibody, and cytokines were examined in DBA/1J mice. Serum levels of AST, ALT, creatinine, and lipids were measured, and antioxidant enzyme activity in the spleen was also determined. The arthritis score and paw edema were markedly suppressed in the groups treated with potato extract. Levels of rheumatoid factor, anti-type II collagen antibody, interleukin (IL)-1, IL-6, LDL-cholesterol, and malondialdehyde in sera were also reduced by potato extract treatment. The activities of glutathione peroxidase and glutathione reductase were increased in the spleens of CIA mice treated with potato extract. These findings suggest that potato extract has suppressive effects on type II collagen-induced arthritis, an animal model for human RA.
