The anti-histamine effect of water soluble alkaloids extracted from solanum nigrum L. (용규에서 추출된 수용성 알칼로이드성분의 항히스타민 효과)

  • The whole herbs of solanum nigrum L were extracted in boiling water (SNL-W), and the extracts were separated with butyl alcohol fraction (SNL-W/B) and aqueous fraction (SNL-W/W) by the solvent extraction method. The total alkaloid and total saponin content mensuration were used to identify the alkaloid composition of methanol fraction extracted from the aqueous fraction. The venom of honey bee was used to induce the rat peritoneal mast cell to secreting the histamine. The results show that the water soluble alkaloid composition of solanum nigrum L (SNL-W/W/M) has a significant inhibitory effect on the histamine release.

  • The leaves of eggplant (Solanum melongena L.) were extracted with 80 % aqueous MeOH, and the concentrated extract was partitioned with n-hexane, EtOAc, n-BuOH, and water fractions. From the n-BuOH fraction, five compounds were isolated through the repeated silica gel, octadecyl silica gel, and Sephadex LH-20 column chromatographies. On the basis of physic-chemical and spectroscopic data including mass spectrometry, infrared spectroscopy, and nuclear magnetic resonance, they were identified to be caffeic acid (1), chlorogenic acid (2), cryptochlorogenic acid (3), panasenoside (4), and (6R,7E,9R)-4,7-megastigmadien-3-one-9-${\beta}$-${\small{D}}$-glucopyranoside (5). Compounds 3 and 4 were isolated for the first time from the leaves of S. melongena L. in this study.

  • The experiments were conducted to identify several factors affecting isolation and culture of mesophyll protoplasts in Solanum melongena var. fructualbo. Higher viable plotoplasts were obtained, when isolated in 1.5% macerozyme, 0.2% macerozyme, 0.6M mannitol, 0.01M MES, 0.2% BSA containing solution adjusted to pH 6.3 for 4 hours. One hour plasmolysis of the material before digestion of leaf tissue was effective for protoplast yield and viability. The method of washing and purification of crude protoplasts, ESS process with 0.7M mannitol and 0.6M sucrose solution. was the best way to get purified protoplasts with viability. As isolated protoplasts were cultured in 8P-KM medium at a density of $2.5{\times}10^4/ml$, the cells were enlarged after 3 to 5 days from culture, subsequently the cells were divided and resulted in colonies.

  • 21plants, which collected from Korea and Japan, were applied to antitumor and cytotoxic screening tests against sarcoma 180 a ascitec in mice. The results are summariged as follows : 1) The total packed cell volume method has been used for the antimeoplastic screening for from natural higher plants in Korea. By this method, we have found out that Selaginella involves, Patrinia hispida, Archyranthes japonica and Solanum nigrum having significant activity and also Cydonia sinensis and Rubia akane showed slight activity to antitumor 2) The total packed cell volune method has been used for the antineoplastic screening for from natural higher plants in Japan. Among the 21 tested plants, lsodon japonicus having strong antitumor activity and also Torilis japonica, Aralia elata, Leonurus sibiricus and Rubia cordifolia showed significant activity to anticancer tumor while Forsythia spp and Solanum nigrum showed slight activity to antitumor. 3) Among the 21 tested Korea plants, lsodon excisus and Forsythia Koreana showed strong antitumor activity by the V79 cytotoxic cell screening test.

  • The study aimed to confirm transformation, morphology, steroid alkaloidal TLC pattern and growth rate of hairy roots. The Solanum nigrum plantlets were inoculated with Agrobacterium rhizogenes strain 15834. Hairy roots were induced by plasmid. Agropine and mannopine were detected in the hairy roots. The organization of hairy roots on the transactional morphology was undifferentiated. Culture on the medium containing hormone(IBA 2, kinetin 0.1mg/l) altered hairy roots into callus. The growth rate of hairy roots on the NN30 liquid medium was 52 times heavier than in the original state and this was higher than on the other media. The results of TLC analysis indicated that the hairy roots produced steroidal alkaloids resembling those of normal roots.

Purification and Characterization of Tyrosinase from Solanum melongena

  • Tyrosinase was purified from Solanum melongena by ammonium sulfate precipitation, Sephadex G-150 and DEAE-Sephacel column chromatography. The molecular weight of the purified tyrosinase was approximately 88,600 daltons with 805 amino acid residues. The amino acid composition showed the characteristic high contents of glycine, glutamic acid and serine residues. The enzyme had high substrate specificity towards (+)-catechin. The $K_m$, value for L-DOPA was 20.8 mM. L-ascorbic acid, ${\beta}-mercapto-ethanol$, sodium diethyldithiocabamate, KCN and $NaN_3$ had strong inhibitory effects on enzyme activity. Sodium diethyldithiocabamate was a competitive inhibitor of the enzyme with a $K_i$ value of $5.2{\times}10^{-2}\;mM$. The optimum pH of the enzyme was 9.0 and the optimum temperature was $65^{\circ}C$ with L-DOPA as a substrate. In addition, the activity was enhanced by addition of $Ca^{+2}$ or $Cu^{+2}$, but decreased in the presence of $Fe^{2+},Fe^{3+}$ and $Zn^{2+}$ ions.

Cell Surface Display of Four Types of Solanum nigrum Metallothionein on Saccharomyces cerevisiae for Biosorption of Cadmium

  • We displayed four types of Solanum nigrum metallothionein (SMT) for the first time on the surface of Saccharomyces cerevisiae using an α-agglutinin-based display system. The SMT genes were amplified by RT-PCR. The plasmid pYES2 was used to construct the expression vector. Transformed yeast strains were confirmed by PCR amplification and custom sequencing. Surface-expressed metallothioneins were indirectly indicated by the enhanced cadmium sorption capacity. Flame atomic absorption spectrophotometry was used to examine the concentration of Cd2+ in this study. The transformed yeast strains showed much higher resistance ability to Cd2+ compared with the control. Strikingly, their Cd2+ accumulation was almost twice as much as that of the wild-type yeast cells. Furthermore, surface-engineered yeast strains could effectively adsorb ultra-trace cadmium and accumulate Cd2+ under a wide range of pH levels, from 3 to 7, without disturbing the Cu2+ and Hg2+. Four types of surfaceengineered Saccharomyces cerevisiae strains were constructed and they could be used to purify Cd2+-contaminated water and adsorb ultra-trace cadmium effectively. The surface-engineered Saccharomyces cerevisiae strains would be useful tools for the bioremediation and biosorption of environmental cadmium contaminants.

Effects of Solanum lyratum on The $CCl_4$-indused$ Liver Damage in mice (배풍등(排風藤)이 $CCl_4$ 유발된 mouse의 간손상(肝損傷)에 미치는 영향)

  • This study was carried out to investigate the effects of Solanum lyratum Extract (SLE) on the recovery of liver in $CCI_{4}-intoxicated$ mice. In this study, ICR-Mice were divided into 3 experimental groups; Normal group was fed basal diet and was fed bassal diet for 2 Weeks, sample group was injected $CCl_{4}\;(0.6m{\ell}/kg)$ and was fed basal diet for 2 weeks, sample group was injected $CCl_{4}\;(0.6m{\ell}/kg)$ and was fed the SLE 500mg/kg fod 2 Weeks. In sample group, the level of serum AST, ALT, ALP activity decreased significantly compared with control group. This results suggest that SLE has appreciable therapeutic effect on $CCI_4$ induced hepatotoxicity. In sample group, the level of LPO in liver homogenates decreased significantly compared with control group. In sample group. the level of GSH, catalase in liver homogenates increased significantly compared with control group. This results suggest that SLE has appreciable antioxidant effect on $CCI_4induced hepatotoxicity. In conclusion the results suggest that SLE has appreciable therapeutic effect and antioxidant effect on $CCI_4$ induced hepatotoxicity.

Soft Rot of Eggplant (Solanum melongena) Caused by Choanephora cucurbitarum in Korea

  • In April 2002 and 2003, soft rot on fruit of eggplant (Solanum melongena) caused by Choanephora cucurbitarum was observed in the experimental fields at Gyeongnam Agricultural Research and Extension Services in Korea. The disease began with water-soaking and dark-green lesions, and then the infected tissues were rapidly rotten. Sporangium was subglobose in shape and sized $40{\sim}130\;{\mu}m$. Monosporous sporangiola were elliptic, fusiform or ovoid, brown in color, and measured as $12{\sim}20\;{\times}\;6{\sim}14\;{\mu}m$. Sporangiospores having three or more appendages were elliptic, fusiform or ovoid in shape, dark brown or brown in color, and sized $14{\sim}20\;{\times}7{\sim}16\;{\mu}m$. The fungus grew well on potato dextrose agar between 15 and $40^{\circ}C$ and its optimum growth temperature was $30^{\circ}C$. Based on morphological characteristics, the causal fungus of the fruit soft rot of eggplant was identified as C. cucurbitarum. This is the first report on the soft rot of S. melongena caused by C. cucurbitarum in Korea.

The Extracts of Solanum nigrum L. for Inhibitory Effects on HIV-1 and Its Essential Enzymes (Human Immunodeficiency Virus Type I에 대한 용규(龍葵) 추출물의 억제활성)

  • For the purpose of developing new anti-HIV agents from natural sources, the extracts of Solanum nigrum L. were tested for their inhibitory effects on HIV-1 replication and its essential enzymes as the reverse transcriptase (RT), protease and ${\alpha}$-glucosidase. In the assay of HIV-1-infected human T-cell line, water extracts inhibited the HIV- 1 -induced cytopathic effects with IC (inhibitory concentration) of 100 ug/ml. Moreover water extracts (100ug/ml) of aerial parts showed strong activity of 32.6% on anti-HIV-1 PR using the activity of the enzyme to cleave an oligopeptide. In the HIV-1 reverse transcriptase inhibition assay, aqueous extract a inhibited 17.4%, but no glucosidase inhibitory activities. We found out this result, for these samples it is possible that the inhibition of the viral replication in vitro is due to the inhibition at least one of PR and RT. It would be of great interest to identify the compounds which are responsible for this inhibition, since all therapeutically useful agent up to date are PR, RT and ${\alpha}$-glucosidase inhibitors.
