Growth Response and Changes of Nitrate and Sucrose Content in Tomato under Salt Stress Condition (염스트레스에 의한 토마토 생장반응과 식물체내 Nitrate 및 Sucrose 변화)

  • This experiment was carried out to find the growth response and changes of nitrate and soluble sugar content in tomato leaves with salt stress. Tomato (Solanum lycopericum) seedlings were grown under different electrical conductivity (EC) levels adjusted with $CaCl_2$ as 1, 2, and $6dS\;m^{-1}$. The growth response and contents of nitrate and soluble sugar in tomato plants were examined at 7 and 14 days after salt treatment. Leaf area and dry weight ratio of shoot to root of tomato plants were decreased as EC level increased. Photosynthetic rate of leaves was reduced under high EC level due to the stomatal closure and the reduction of transpiration rate. The soluble sugar and starch content were lower in the tomato leaves grown under high EC level. Total nitrogen and nitrate contents were decreased in high EC level, whereas the ammonium content was increased. High-salt stress induced the accumulation of salt crystal in mesophyll cells of tomato leaf.

Application of Slow-Freezing Cryopreservation Method for the Conservation of Diverse Potato (Solanum tuberosum L.) Genotypes

  • Cryopreservation has been recognized as a practical and efficient tool for the long-term storage of vegetatively propagated plants. This study was conducted to investigate the effects of slow-freezing techniques on the cryopreservation of potato. In vitro plantlets of the potato genotypes of 'Atlantic', 'Superior’, 'Namseo', 'J138', and 'CTO5-5' were cold acclimated, and the excised axillary buds were precultured, osmoprotected, exposed to plant vitrification solution, frozen slowly to $-40^{\circ}C$ and then rapidly plunged into liquid nitrogen, thawed and finally plated on the regeneration medium. It was found that the higher the sucrose concentrations in the subculture medium of donor plantlets, the higher the survival rates of shoot tips after cryopreservation, and the highest survival (20%) was observed in the medium added with 0.25 M sucrose. As for the effect of cooling, $0.3^{\circ}C/min$ cooling speed showed the highest survival (25%). Different varieties showed different responses over different cryopreservation treatments. Survival rate was increased by slow-freezing technique method as compared with that of the basic cryopreservation method of vitrification alone in the diverse potato genotypes. Leaf and tuber morphologies of potatoes regenerated after cryopreservation using slow freezing technique were similar to those derived from the in vitro stock plantlets.

Growth, quality, and yield characteristics of transgenic potato (Solanum tuberosum L.) overexpressing StMyb1R-1 under water deficit

  • This study was conducted to evaluate agronomic characteristics such as growth, quality, and yields of StMyb1R-1 transgenic potato and also to obtain the basic data for establishing assessment guidelines of transgenic potato. Three transgenic lines (Myb 1, Myb 2, and Myb 8) were cultivated under conventional irrigation, drought condition, and severe drought condition and were analyzed by comparing with wild type, non-transgenic cv. Superior. Myb 2 showed a different flower color from wild type and Myb 1 had much bigger secondary leaflets than wild type. Myb 1 and Myb 2 showed higher $P_2O_5$ content in both top and root zone and longer shaped tubers than wild type. In yield factors, transgenic lines had more tubers than wild type, however their yield decreases were severe because of the poor enlargement of tuber under water deficit condition. This tendency was noticeable in Myb 1 and Myb 2. In TR ratio, chlorophyll content, dry matter rate, and relative water content, there were no big differences between transgenic lines and wild type. Meanwhile, in phenotype, growth, quality, and yield factors, substantial equivalent was confirmed between Myb 8 and wild type. Then, Myb 8 showed the highest marketable tuber yield under conventional irrigation, while showed lower level than wild type under water deficit. Judged by this result, the enhancing droughttolerance by StMyb1R-1 gene might actually not mean the enhancement of photosynthesis or starch accumulation in tuber and, furthermore, not the yield improvement. More detailed research will be required to accurately understand the relationship between StMyb1R-1 and yield factors.

  • 1) Optimum temperature was $15^{\circ}C$ for C. bursa-pastoris, $20^{\circ}C$ for C. album, $30^{\circ}C$ for P. oleracea, A. lividus, E. crus-galli, D. sanguinalis, and 4 showed wide range of germination temperature. 2) Emergence of C. bursa-pastoris, and C. album was best at $14.8^{\circ}C$ of soil temperature, E. crus-galli at $23^{\circ}C$, E. indica, A. lividis and P. oleracea at $27.1^{\circ}C$ and A. retroflexus and D. sanguinalis at $31.1^{\circ}C$. 3) A. retroflexus and P. oleracea started to germinate at 30% water absorption stage and A. lividus, C. album, S. viridis, and D. sanguinalis at 40% and E. indica at 70%. 4) Germination of weed species was decreased as PEG 6000 induced osmotic potential lowered. C. album, P. oleracea, D. sanguinalis, A. lividus, and Solanum nigrum were germinated at -5.0 bar osmotic potential and C. album and P. oleracea were germinated at -7.0 bar.

Conservation Management Methods and Vascular Plants of Major Sites in Changwon-si (창원시 주요지역의 식물상 및 보전관리방안)

  • The vascular plants of major sites in Changwon-si were listed 456 taxa (9.3% of all 4,881 taxa of vascular plants); 112 families, 293 genera, 371 species, 2 subspecies, 66 varieties and 17 forms. The vascular plants of Changwon-cheon were listed 133 taxa, Junam reservoir were listed 90 taxa, Seongju-sa were listed 293 taxa, and Yungji park were listed 164 taxa. Divided into 456 taxa; woody plants were 160 taxa (35.1%) and herbaceous plants were 296 taxa (64.9%). Furthermore, therophytes (Th) were 111 taxa (24.3%), hemicryptophytes (H) were 75 taxa (16.4%), megaphanerophytes (MM) were 63 taxa (13.8%) showed high proportional ratio in life form. Based on the list of rare plants by the Korea Forest Service and Korea National Arboretum, 10 taxa (1.8% of all 571 taxa of rare plants); Aristolochia contorta (LC), Euryale ferox (VU), Melothrua japonica (LC), Utricularia pilosa (CR), Hydrocharis dubia (LC), Carex idzuroei (DD), Acorus calamus var. angustatus (LC), etc. Based on the list of endemic plants by the Korea National Arboretum, 10 taxa (3.0% of all 328 taxa of endemic plants); Salix pseudolasiogyne, Philadelphus incanus, Indigofera koreana, Lespedeza ${\times}$ maritima, Stewartia pseudocamellia, Weigela subsessilis, Carex okamotoi, etc. Based on the list of specific plants by floral region were total 43 taxa (4.0% of all 1,071 taxa of specific plants); 4 taxa (Euryale ferox, Hydrocharis dubia, Carex idzuroei, etc.) in class IV, 9 taxa (Ilex crenata, Acer palmatum, Stewartia pseudocamellia, Melothria japonica, Fatsia japonica, etc.) in class III, 3 taxa (Caltha palustris var. membranacea, Nymphoides indica, etc.) in class II, 26 taxa (Cyrtomium fortunei, Chloranthus japonicus, Quercus variabilis, Ulmus parvifolia, Aphananthe aspera, etc.) in class I. Based on the list of naturalized plants, 48 taxa (Rumex crispus, Chenopodium album var. album, Amaranthus patulus, Phytolacca american, Brassica juncea var. integrifolia, Potentilla paradoxa, Robinia pseudoacacia, Euphorbia maculata, Oenothera odorata, Cuscuta pentagona, Veronica persica, Plantago lanceolata, Diodia teres, Helianthus tuberosus, Dactylis glomerata, etc.), naturalization rate was 10.5% of all 456 taxa of vascular plants and urbanization index was 15.0% of all 321 taxa of naturalized plants. Ecosystem disturbing wild plants were 3 taxa (Rumex acetocella, Solanum carolinense, Ambrosia artemisiifolia).

Effects of $\textrm{CO}_2$ concentration and air current speed on the growth and development of plug seedlings under artificial lighting (인공광하에서 $\textrm{CO}_2$ 농도와 기류속도 제어가 플러그묘의 생육에 미치는 효과)

  • This study was conducted to investigate the effects of $CO_2$ concentration(310 or 950$\mu$ mol.mol$^{-1}$ ) and air current speed(0.3, 0.5, 0.7 or 0.9m.s$^{-1}$ ) on the growth and development of eggp1ant Plug seedlings (Solanum melongena L.) under artificial 1ighting. For the treatment of $CO_2$ enrichment, stem length and diameter, the ratio of stem length to stem diameter, plant height, leaf area, net photosynthetic rate, top dried weight were significantly different at 1% level. Stem length of plug seedlings decreased at the condition under enriched $CO_2$ and high air current speed above plug stand. Stem diameter of plug seedlings increased and plant height decreased with the increasing $CO_2$ concentration. Plug seedlings had maximum net photosynthetic rate at the air current speed of 0.7m.s$^{-1}$ . Net photosynthetic rate at $CO_2$ concentration of 950$\mu$mol.mol$^{-1}$ increased by 46% than those at 310$\mu$mol.mol$^{-1}$ . Thus $CO_2$ enrichment would be effective for the production of plug seedlings with high quality.

Appropriate Root-zone Temperature Control in Perlite Bag Culture of Tomato during Winter Season (저온기 토마토 펄라이트 자루재배시 최적 근권온도 조절 방법)

  • The effective method for heating root-zone during winter season was studied in the aspects of growth, yield and economics for tomato ($Solanum$ $lycopersicum$) in perlite bag culture. There were four root-zone heating treatments: two hours heating from one hour before to one hour after sunrise, four hours from two hours before to two hours after sunrise, 15 hours after sunset, and no heating. The growth characteristics of the upper parts of plants were not significantly different among the treatments, but root volume increased with longer heating of the root zone. The Plant Development Index, using stem diameter and the length between growing tip and the upper flowering truss, showed relation between yield per cluster and growth pattern. The treatment heating for four hours was the most economic in terms of growth and yield of tomato.

Analysis of Energy Balance in Closed Transplants Production System (폐쇄형 묘생산 시스템의 에너지 수지 분석)

  • This study was conducted to analyze the energy balance in closed transplants production system (CTPS) for the production of high quality transplants. Potato (Solanum tuberosum L. cv. Dejima) plug seedlings were grown for 15 days at air temperature of 20$^{\circ}C$, relative humidity of 70%, photoperiod of 16/8 h, and photosynthetic photon flux (PPF) of 200 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ following 5 days of tooting in CTPS. Electric energy consumption was 46% for lighting, 35% for cooling, 16% for heating, 2% for air circulation, and 1% for humidifying. The electric energy utilization efficiency and the short-wave energy utilization efficiency were 0.5% and 5.4%, respectively. These results suggest that CTPS has the feasibility for the commercial production of potato plug seedlings if the electric energy consumption in CTPS is reduced and the electric energy utilization efficiency is increased.

Plant Growth-Promoting Activity of Endophytic Fungi Isolated from the Roots of Native Plants in Dokdo Islands (독도 자생식물의 뿌리로부터 분리된 내생균의 식물생장촉진 활성)

  • Endophytic fungal strains were isolated from the roots of six species plants in the Dokdo islands. Native plant samples, such as Artemisia japonica, Chenopodium album and Solanum nigrum were isolated from Dongdo, and those such as Cyrtomium falcatum, Dianthus longicalyx and Tetragonia tetragonoides were isolated from Seodo. In total, thirty two fungal strains were isolated from these native plants. To identify the fungal strains, polymerase chain reaction (PCR) amplification of internal transcribed spacer (ITS: containing ITS1, 5.8s and ITS2 region) regions was done with universal primers ITS1 and ITS4. Endophytic fungi of four species were isolated from A. japonica, eight species from C. album, three species from S. nigrum, three species from C. falcatum, three species from D. longicalyx and eleven species from T. tetragonoides. Culture filtrates (CF) of isolated endophytic fungi were used to treatwaito-c rice seedlings to test plant growth-promoting activity. As a result of bioassay, Ca-5-2-2 strain isolated from C. album expressed highest plant growth-promotion activity. Of all the endophytic fungi isolated, Penicillium sp., Fusarium sp. and Aspergillus sp. were the most abundantly distributed fungal strains in the six plants used in this study.

Characterictics and Stability of Anthocyanin Pigment Extracted from Purple-Fleshed Potato (자색감자 Anthocyanin 색소의 특성 및 안정성)

  • Effects of pH, sugars, organic acids, metal ions, ascorbic acid and light on the stability of anthocyanin pigment extracted from purple-fleshed potato (Solanum tuberosum L.) were studied. The pH had marked influences on the color of the potato anthocyanin pigment: i.e., the lower the pH of the anthocyanin solution was, the more stable and intenser of the pigment was. It showed characteristic bathochromic shift as the pH of the solution increased. Generally, the addition of sugars into the purple-fleshed potato anthocyanin solution caused decrease in color stability of the pigment. Among the sugars tested, maltose was the most deleterious followed by sucrose, galactose, fructose and glucose. The addition of organic acids greatly increased the stability of the pigment. Malic acid was found to be the most effective in stabilizing the pigment followed by tartaric, citric and succinic acids, while malonic acid was found to be deteriorative to the stability of the pigment. Most metal ions except $Cu^{2+}$ increased stability of the pigment-especially, $Cd^{2+}\;and\;Al^{3+}$ were more effective than the others. Ascorbic acid degraded the pigment considerably, but the adverse effect was diminished by adding thiourea. Light gave an adverse effect to the stability of the purple-fleshed potato anthocyanin pigment, which could be minimized by shielding the light from the pigment.
