Effects of temperature, pH, and salt concentration on the preservation of the infective juvenile entolopathogenic nematodes, Steinernema carpocapsae, were analyzed. The effects of acclimation to sublethal low temperature and glycerol concentrations on rapid cold hardening of the nematodes were also evaluated to set up the conditions for their cryopreservation. All tested environmental factors could change the storage effectiveness of the nematodes. These analyses indicated that the optinal storage condition for the nematodes consisted of $15^{\circ}C$, pH 8.5, and 0.5-1.0% salt concentration. The nematodes showed a rapid could hardening in response to $5^{\circ}C$ for 2h. Glycerol induced the nematodes to increase the cold hardiness. Its high concentration over 10%, however, gave a harmful effect on the nematode survival.
During the course of study on plant parasitic nematodes associated with medicinal herbs in Korea, high population of different stages of citrus nematode, Tylenchulus semipentrans was found in association with Pyrola japonia Klenze in Imsil, Chunbuk province. The second stage larvae and males were isolated from rhizosphere soil ; immature and mature females were detached from the roots of infested plants of Pyrola japonica. Besides citrus it has been reported to parasitize on roots of Syringa vulgaris, Diospyros lotus, olive, grapevine, and pear in different parts of the world. This is the first record of T. semipenetrans parasitized on a medicinal plant, Pyrola japonica.
The present study described the isolation of 2-methylbutyric acid (2-MBA) produced from Bacillus pumilus L1, to subsequently investigate its nematicidal activity for the control of the root-knot nematode. The results showed that 2-MBA could be purified by chromatographic techniques and was identified using nuclear magnetic resonance and liquid chromatography-mass spectrometry. Crude extract and partially purified compounds had a significant effect on the inhibition of egg hatchability and second-stage juvenile (J2) mortality. A dose-dependent effect of 2-MBA was observed for J2 mortality and egg hatchability. Egg hatchability was 69.2%, 59.9%, 32.7%, and 0.0% at 125, 250, 500, and $1000{\mu}g\;mL^{-1}$ of 2-MBA after 4 d of incubation, respectively. Meanwhile, J2 mortality was in the range of 24.4%-100.0% after 2 d of incubation, depending on the concentrations of 2-MBA used. A pot experiment also demonstrated that treatment of B. pumilus L1 culture caused a significant reduction in the number of galls, egg masses, and J2 population than that of the tap water (TW) control. However, as the B. pumilus L1 culture concentration was decreased, the efficacy of nematode control by treatment of B. pumilus L1 culture was reduced compared to that of TW. B. pumilus L1 inoculation at different concentrations also promoted cucumber plant growth. Therefore, our study demonstrated the potential of 2-MBA from B. pumilus L1 as a biocontrol agent against the root-knot nematode and a plant growth promoter for cucumber plants.

An endemic entomopathogenic nematode, Heterorhabditis megidis, was evaluated by its control efficacies against housefly, Musca domestica, and flower beetle, Gametis jucunda. In Petri-dish assay, the pathogenicity of H. megidis showed 456.4 infective juveniles/larva (IJs/larva) in median lethality (LC$_{50}$) against the second instar larvae of M. domestica and 238.9 IJs/larva against the second instar larvae of G. jucunda. This was contrasted with those of the other well-known entomopathogenic nematode, Steinernema carpocapsae, which showed 115.9 IJs/larva against M. domestica and 388.6 IJs/larva against G. jucunda. In field experiment, H. megidis were applied per square meter of pork farm with 1,000,000 IJs of H. megidis or apple orchard with 370,000 IJs, which were infested with M. domestica or G. jucunda, respectively. H. megidis showed 56.9% and 57.3% of control efficacies against M. domestica and G. jucunda, respectively. These results suggest a promising control technique in the field using H. megidis against M. domestica and G. jucunda.a.

This study was conducted to get basic information on the occurrence of plant-parasitic nematodes for the establishment of nematode management strategy in major potato production areas in Korea. Nationwide soil collection was done in 11 areas of Cheju, Yesan, Gimchun, Goryoung, Hong chun, Pyungchang, Gimjae, Milyang, Namwon, Gangnung, and Inje in 2004-2005. Root-hot nematode juveniles(J2) were detected in 30 samples among the 50 samples. The average density was 12-69 J2/100cc soil. Pratylenchus sp., Helicotylenchus sp., Ditylenchus sp., Tylenchus sp., and Tylenchorhynchus sp. were also detected in various locations, however, their densities were very low. Root-knot nematode females were collected from tomato roots inoculated with the potato field soils for PCR-RFLP identification. The females from Cheju, Milyang, and Goryung showed PCR products of 500 bp. And the Dra I restriction enzyme digestions showing 290 bp and 230 bp fragments confirmed their identity as Meloidogyne hapla.
Experiments were carried out to know the effects of emulsifiable DBCP, (Dibromo Chloropropane) for control of the citrus-root nematode, Tylenchulus semipenetrans in Orchards of chinese citron and mandarine orange for four years of 1967-1970. Nematode population density decreased as the amount of DBCP increased. In mandarine orange orchard, the nematode population density decreased after second treatment mainly because of bad drainage of the soil, while in Chinese citron orchard, it fell off sharply after just one treatment since the soil was well drained. Yields of Chinese citron increased as much as $26\%$ in 4.4l/10a injection, 1$18\%$ in 4.4l/10a drench; yields of mandarine orange increased as much as $11\%$ in 4.41/10a injection and $13\%$ in 4.4l/10a drench.
To investigate the effects of Brassicaceae cover crops on clover cyst nematode, Heterodera trifolii, 10 cultivars consisting of six of oil radish and four of white mustard were planted in the nematode infected field at Jeongseon city. Two months after planting, the cover crops were plow down and incorporated into the soil using rotavator, decomposed for 1 month, then transplanted kimchi-cabbages. After 70 days, the density of eggs inside of the cyst and the number of females in the soil were examined. As a result, the reproduction rates of eggs in each plots of Adios and Anaconda cultivars, which were 0.04 and 0.02, respectively, were greatly reduced. The number of females in the plots of above two cultivars showed means at 2.5 and 3.5 per 300 ㎤ soil, which were lower than those of other plots. In addition, fresh weights of three plants in the two plots, which were 7.67 and 7.35 kg, were significantly higher than that of the control plot. Collectively, these results suggest that the two cultivars of Brassicaceae cover crops, Adios and Anaconda, could be used for reducing the cyst nematode density.
Fifty-two nematode predatory fungi were isolated from 37 soil samples collected from eight provinces in Korea. Isolated fungi were tested their predacity against Rhabditis sp. and Meloidogyne incognita in petri dish, and against M. incognita in greenhouse pot experiments. Fifty isolates had trapping organ of adhesive networks and two isolates had adhesive column or adhesive knob. In petri dish experiments, 5 1 isolates against Rhubditis sp. and 26 isolates against M. incognita showed over 91 % of predacity; in greenhouse experiments, however, only three isolates showed over 81% of predacity. These results imply that the results from the laboratory experiments are not consistent with those from the greenhouse experiments. Therefore, to select a promising biocontrol predatory fungi for plant-parasitic nematodes, the screening experiment should be conducted in conditions close to nature.
For the biological control of the root-lesion nematodes, Pratylenchus spp., which damage directly and indirectly to the leaf perilla, the nematical effect of three nematode-trapping fungi, Arthrobotrys oligospora, A. conoides and A. dactyloides was evaluated in the field. Three species of Arthrobotrys were isolated from the culture soil of leaf perilla in 1998 and were observed the capture of the root-lesion nematodes, Pratylenchus spp. by adhesive hyphal networks or constricting rings on agar. At 40 days after treatment, the plant-parasitic nematodes and root-lesion nematode populations were approximately increased 3.5 fold in untreated control plot, while the nematode population in fungi treatment plots was similar to initial population. In the A. dactyloides plot, however, the population of plant-parasitic nematodes and Pratylenchus spp. was approximately reduced 65% and 53%, respectively. Thus, the fungus A. dachyloides should provide as biological agent for the control of Pratylenchus spp.
Clover cyst nematode, Heterodera trifolii are one of the major pests that damage Chinese cabbage in high land cultivation area and have recently been distributed in Korea. This study was conducted to investigate the effect of temperature and inoculation density on the proliferation of clover cyst nematode. The clover cyst nematode was proliferated at $15-30^{\circ}C$ and the egg, female adult and cyst number was the highest at $20^{\circ}C$. As the inoculation density increased, the number of female cyst nematodes increased with the inoculation density and when 16 eggs were inoculated per gram of soil, the amount of growth, number of egg and cyst was the highest. Number of females and eggs were formed more in cabbage than in kale regardless of inoculation density. Chinese cabbage was a favorable host for clover cyst nematode growth than kale.
