  • The Study was conducted to develope the low temperature tolerant methane-producing bacteria(LTTB) and to increase the efficiency of anaerobic fermentation for the treatment of agricultural and livestock wastes at low temperature. The samples were collected from muddy soil, water logged sediment, organic layer and anaerobic sludge at three latitudes, $34.8{\sim}37.4\;^{\circ}N(Korea)$, $41.4\;^{\circ}N(USA)$ and $54.5{\sim}56.9\;^{\circ}N(Canada)$. They were used for determination of the methanogenesis rates for isolation and identification of the LTTB. The methanogenesis rate of smaples at low temperature were higher in the cellulose medium than methanol medium. The methanogenesis rate in the samples of subarctic region were $15{\sim}19$ moles/ml during 30 days at low temperature($8\;^{\circ}C$), whereas not detected in the samples of temperate region. The methanogenesis rate in the enrichment culture of subarctic samples were inhibited by the $40\;{\mu}g/ml$ of streptomycin + vancomycin or ampicillin + oleandomycin which were not effect to the methanogens. An inhabitation of high temperature tolerant methane producing bacteria was identified in the samples of temperate region, whereas that of the LTTB growing at $8{\sim}13^{\circ}C$ was identified in the subarctic region.

Selection and Cultural Characteristics of Whole Chicken Feather-Degrading Bacterium, Bacillus sp. SMMJ-2 (Whole Chicken Feather-Degrading Keratinolytic Protease 생산균주의 분리 및 특성)

  • Feather, generated in large quantities as a byproduct of commercial poultry processing, is almost pure keratin, which is not easily degradable by common professes. Four strains, SMMJ-2, FL-3, NO-4 and RM-12 were isolated from soil for production of extracellular keratinolytic protease. They were identified as Bacillus sp. based on their morphological and physiological characteristics. They shown high protease activity on 5.0% skim milk agar medium and produced a substrate like mucoid on keratin agar medium. Bacillus sp. SMMJ-2 had a faster production time for producing keratinolytic protease than other strains. This strain did not completely degrade whole chicken feather for five days in basal medium but completely degraded whole chicken feather when supplied with nitrogen source for 40hours in keratinolytic producing medium ($0.7%\;K_{2}HPO_{4},\;0.2%\;KH_{2}PO_{4},\;0.1%$ fructose, 1.2% whole chicken feather, $0.01%\;Na_{2}CO_3$, pH 7.0). When supplied with chicken feather as nitrogen source, keratinolytic protease activity was 89 units/ml/min. When soybean meal was used as nitrogen source, the keratinolytic protease production reached a maximum of 106 units/ml/min after 48 hours under $30^{\circ}C$, 180 agitation. To isolate the keratinolytic protease, the culture filtrate was precipitated with $(NH_4)_{2}SO_4$ and acetone. The recovery rate of keratinolytic protease was about 96% after treatment with 50% acetone. The enzyme was stable in the range of $30{\sim}50^{\circ}C$ and pH $6.0{\sim}12.0$.

The Study on the Effects of the Economical Use of Irrigation Water by Different Irrgation Periods and Its Methods on the Growth, Yield and the Factors of Rice Plants. (절수의 시기 및 방법의 차이가 수도생육 수량과 기타 실용형태질 미치는 영향)

  • Higher yield in rice paddies is greatly dependent on adequately balanced and timely supply of water. A majority of rice paddy in Korea is generally irrigated by rainfall, but in many cases it has to be supplemented by artificial irrigation for optimum rice culture. Although the water requirement of rice plant is far higher than that of other crops, submerged condition of rice paddy is not necessarily required. The moisture requirement of rice plant varies with its growing stages, and it is possible to increase the irrigation efficiency through reduction of water loss due to percolation in rice paddies. An experiment was conducted on the effectiveness of economical use of water by different irrigation period and different method of cultivation. The experimental plots were set up by means of randomized block design with three duplications; (a) Alltime submerged (b) Economically controlled, and (c) Extremely controlled. Three different irrigation periods were (a) Initial stage (b) Inter-stage, and (c) last stage. The topsoil of the three plots were excavated to the depth of 30cm and then compacted with clay of 6 cm thickness. Thereafter, they were piled up with the excavated top soils, leveled and cored with clay of 6cm thickeness arround footpath in order to prevent leakage. The results obtained frome the experiments are as follows; (1) There is no difference among the three experiment plots in terms of physical and chemical contditions, soil properties, and other characteristics. (2) Colulm length and ear length are not affected by different irrigation methdos. (3) There is no difference in the mature rate and grain weight of rice for the three plots. (4) The control plot which was irrigated every three days shows an increased yield over the all the time submerged plot by 17 persent. (5) The clay lined plot whose water holding capacity was held days long, needs only to be irrigated every 7 days. (6) The clay lined plot showes an increased yield over the untreated plot; over all the time submerged plot by 18 percent, extremely controlled plot by 18 percent, and economically controled plot by 33 percent.

Analysis of Consciousness and Model on Land for the Another use After Quarrying (채석장의 부지 활용에 대한 의식 및 모델 분석)

  • The study was conducted to develop an effective forest resources use models for an alternate use of abandoned quarry by an attitude survey. According to the result of survey, a pessimistic view due to dust, noise pollution, and forest damage was 5% higher than an affirmative view by economic benefits from the development of quarry. The 42% of the respondents preferred the alternate use of abandoned quarry and the 25% of the respondents wanted an art and cultural space. The optimum size of alternate use was 5-10 ha (43%) with the requirement of nearby residents (32%). According to the SWOT analysis for abandoned quarry, the strength factors were an effective use of land, the content development of modern industrial inheritance + cultural and art fusion, attraction for nearby city and visitors, a harmony of beauty landscape and clean environment, and a sustainable increase of domestic and foreign visitors with the 5-day-work week. The opportunity factors were the improvement of traffic networks through KTX and local highway, the creation of the new growth engines with the establishment of artistic creation belts, the providing of unique cultural and art space through grafting of tour and education, the creation of local income through stone processed goods, and the vitalization of local development through eco-city. The weakness factors were a psychological remoteness and backwardness, and the weakness of staying tour infra. The threat factors were a poor financial support for sustainable development in nearby quarry and a modify of legal and institutional system for the alternated use of abandoned quarry. The developed restoration models for the alternate use in abandoned quarry are classified to a sculpture park, a waterfall and lake park, a rock-climbing, a sports park + forest park, a native botanical garden, a culture and art park, a complex park, a water storage site, a water storage site to extinguish forest fire, a geriatric hospital, an agricultural facility, and a school site types etc. The results suggest that the alternate use in the abandoned soil and stone quarry is needed to establish facility use models with consideration of user's preference.

Biological Control of Anthracnose (Colletotrichum gloeosporioides) in Red Pepper by Bacillus sp. CS-52 (Bacillus sp. CS-52를 이용한 고추 탄저병 (Colletotrichum gloeosporioides) 방제 특성)

  • This study was carried out in order to develop a biological control of anthracnose of red pepper caused by fungal pathogens. In particular, this study focuses on the Colletotrichum species, which includes important fungal pathogens causing a great deal of damage to red pepper. Antagonistic bacteria were isolated from the soil of pepper fields, which were then tested for biocontrol activity against the Colletotrichum gloeosporioides anthracnose pathogen of pepper. Based on the 16S rRNA sequence analysis, the isolated bacterial strain CS-52 was identical to Bacillus sp. The culture broth of Bacillus sp. CS-52 had antifungal activity toward the hyphae and spores of C. gloeosporioides. Moreover, the substances with antifungal activity were optimized when Bacillus sp. CS-52 was grown aerobically in a medium composed of 0.5% glucose, 0.7% $K_2HPO_4$, 0.2% $KH_2PO_4$, 0.3% $NH_4NO_3$, 0.01% $MnSO_4{\cdot}7H_2O$, and 0.15% yeast extract at $30^{\circ}C$. The inhibition of spore formation resulting from cellulase, siderophores, and indole-3-acetic acid (IAA), were produced at 24 h, 48 h, and 72 h, respectively. Bacillus sp. CS-52 also exhibited its potent fungicidal activity against anthracnose in an in vivo test, at a level of 70% when compared to chemical fungicides. These results identified substances with antifungal activity produced by Bacillus sp. CS-52 for the biological control of major plant pathogens in red pepper. Further studies will investigate the synergistic effect promoting better growth and antifungal activity by the formulation of substances with antifungal activity.

  • This study was conducted to investigate the biochemical properties of isolated bacteria, low temperature tolerant methanogens which were selected for use as inoculum for anaerobic fermentation of agricultural and livestock wasted at low temperature. The results, obtained were summarized as follows: Low temperature tolerant methanogens were isolated from the samples which showed the high methanogenesis rate by enrichment culture at low temperature in methanol medium. These methanogens, Methanobacterium M-251 and Methanobacterium M-253 were isolated from swampy sediment at latitude $56.9^{\circ}$, Methanosarcina mazei M-372 from lake sediment IV at latitude $55.0^{\circ}$ N, and Methanobacterium formicicum M-375 from tidal land soil at latitude $37.0^{\circ}N$, respectively. The isolated anaerobic bacteria could not use sugars as carbon sources. The optimum pH value for the growth of M-251 and M-375 was 6.8, but those for M-253 and M-372 6.5 and 7.0, respectively. The minimum growth temperature of isolated, M-251 and M-253 were $8^{\circ}C$ and the optimum temperature $30^{\circ}C$, while the minimum of M-392 and M-395 were $13^{\circ}C$ and the optimum $37^{\circ}C$. The growth rate of isolates at $17.5^{\circ}C$ were lower by 32-50% than that of $30^{\circ}C$. The isolated Methanobacterium strains such as M-251, M-253, and M-375 have lower cell yield, 0.38-1.21g/1M $CH_4$ than 1.14-1.51g/1M $CH_4$ of Methanosarcina mazei M-372.

Effect of Chlorella vulgaris CHK0008 Fertilization on Enhancement of Storage and Freshness in Organic Strawberry and Leaf Vegetables (Chlorella vulgaris CHK0008 시비가 유기농 딸기와 엽채소의 저장성과 신선도 향상에 미치는 영향)

  • This study aimed to enhance storage and freshness of strawberry fruits and foliage vegetables by spray treatment with Chlorella vulgaris as a bio-fertilizer. The tested strain, C. vulgaris CHK0008, was isolated from an organically cultivated rice paddy and identified as C. vulgaris by its morphology and 18S rDNA and 23S rDNA sequence homology. We successfully cultured C. vulgaris CHK0008 in BG11 modified medium (BG11MM) and adjusted $2.15{\times}10^6cell/mL$ C. vulgaris CHK0008 to one OD value by measuring the optical density at 680 nm using a UV-vis spectrophotometer. The soluble solid content of 'Seolhyang' and 'Yukbo' strawberry fruits treated by spray application with C. vulgaris CHK0008 was enhanced by 22.2% and 11.5% respectively, compared to untreated controls. Additionally, the decay rates of treated 'Seolhyang' and 'Yukbo' strawberry fruits decreased 63.8% and 74.4% respectively, compared to untreated control. Surface color changes and chlorosis of leaves in leaf vegetables such as lettuce, kale, red ornamental kale, white ornamental kale and beet were observed in samples treated with water spray for 10 days after cold storage. However, the decay rate of leafy vegetables treated with foliar application of 25% C. vulgaris CHK0008 liquid culture was significantly decreased compared to that of the untreated control during storage at $4^{\circ}C$.

Isolation and Characterization of Bacillus Strain as a Potential Biocontrol Agent (환경친화적 미생물농약으로서의 잠재성을 가진 세균의 분리 및 특성)

  • In this study, to retain a stable bacterial inoculant, Bacillus strains showing antifungal activity were screened. The improved production, antifungal mechanism, and stability of the antifungal metabolite by a selected strain, AF4, a potent antagonist against phytopathogenic Botrytis cinerea, were also investigated. The AF4 strain was isolated from rhizospheric soil of hot pepper and identified as Bacillus subtilis by phenotypic characters and 16S rRNA gene analysis. Strain AF4 did not produce antifungal activity in the absence of a nitrogen source and produced antifungal activity at a broad range of temperatures (25-40℃) and pH (7-10). Optimal carbon and nitrogen sources for the production of antifungal activity were glycerol and casein, respectively. Under improved conditions, the maximum antifungal activity was 140±3 AU/ml, which was higher than in the basal medium. Photomicrographs of strain AF4-treated B. cinerea showed morphological abnormalities of fungal mycelia, demonstrating the role of the antifungal metabolite. The B. subtilis AF4 culture exhibited broad antifungal activity against several phytopathogenic fungi. The antifungal activity was heat-, pH-, solvent-, and protease-stable, indicating its nonproteinous nature. These results suggest that B. subtilis AF4 is a potential candidate for the control of phytopathogenic fungi-derived plant diseases.

Biodegradation of Phenol by Comamonas testosteroni DWB-1-8 Isolated from the Activated Sludge of Textile Wastewater (섬유 폐수 활성 슬러지에서 분리한 Comamonas testosteroni의 생물학적 페놀 분해)

  • Since industrialization, the production and utilization of various chemicals has contributed to improving the quality of our lives, but the subsequent discharge of massive waste is inevitable, and environmental pollution is becoming more serious every day. Exposure to chemicals as a result of environmental pollution is having a negative effect on human health and the ecosystem, and cleaning up the polluted environment that can affect our lives is a very important issue. Toxic aromatic compounds have been detected frequently in soil, groundwater, and wastewater because of the extensive use of oil products, and phenol, which is used to produce synthetic resins, textiles, and dyes, is one of the major pollutants, along with insecticides and preservatives. Phenol can cause dyspnea, headache, vomiting, mutation, and carcinogenesis. Phenol-degrading bacterium DWB-1-8 was isolated from the activated sludge of textile wastewater; this strain was identified as Comamonas testosteroni by 16S rRNA gene sequencing. The optimal culture conditions for the cell growth and degradation of phenol were 0.7% K2HPO4, 0.6% NaH2PO4, 0.1% NH4NO3, 0.015% MgSO4·7H2O, 0.001% FeSO4·7H2O, an initial pH of 7, and a temperature of 30℃. The strain was also able to grow by using other toxic compounds, such as benzene, toluene, or xylene (BTX), as the sole source of carbon.

Influences of the Plant Growth under Beta-Rays Irradiation at Low Dose (저 선량 베타선의 조사에 의한 식물의 생장에 미치는 영향)

  • This study is to analyze effects of the growth of Chunhyang Young Radish (CYR) and Altari Radish (AR) according to the exposure for 31 days at low dose ${\beta}$-rays. This test has one contrast sample and eleven test samples each as to AR and CYR. The seeds from contrast and test sample were planted in the culture soil after 8 seeds were chosen from each with identical condition. The accumulated dose of test samples has been measured at consistent time on a daily basis for 31 days. The growing process and germination have been measured twice at consistent time in each week. The number of leaves, length of first leave and weight have been acquired average value by measuring for 20 and 25 days, respectively after being planted. The result of test sample in case of 25 days shows that 5% increase in length and 36% increase in weight for AR each at accumulated dose 0.01 Gy compared to the contrast sample. And the length of CYR has increased by 13~17% and 1% at accumulated dose 0.01~0.08 Gy and 0.3 Gy compared to the contrast sample. For the weight at accumulated dose 0.05 Gy and 0.23 Gy has increased by 36% and 2% compared to contrast sample. As to the number of leaves, AR has increased by 0~50% at accumulated dose 0.01-0.32 Gy compared to contrast sample. It also shows that the CYR has increased to 0~67% at accumulated dose 0.01-0.62 Gy compared to contrast sample. As a result of this study, it indicates that both AR and CYR has generally increased in their length, weight, and the number of leaves at low level accumulated dose part 0.01~0.2 Gy. The size of cell, area of nucleus and density of cell for test sample has been observed quite similar to the ones from contrast sample through microscope. In conclusion, AR and CYR irradiated by ${\beta}$-rays have estimated that they are achieved a rapid growth at low level accumulated dose region corresponding to its radiation hormesis theory. Further studies need to confirm the correlation between the radiation hormesis and the growth of the plants.
