Effect of Single Nucleotide Polymorphisms of Acetyl-CoA Carboxylase α(ACACA) Gene on Carcass Traits in Hanwoo (Korean Cattle)

  • Meat production and quality traits in beef cattle are largely affected by genetic factors. Acetyl-Coenzyme A carboxylase-${\alpha}$ (ACACA) plays a key role in the regulation and metabolism of fatty acid biosynthesis in mammalian animals. The gene encoding ACACA enzyme was chosen as a candidate gene for carcass and meat traits. In this study, we investigated effects of single nucleotide polymorphisms (SNPs) in the ACACA gene on beef carcass and meat traits in Hanwoo (Korean cattle) populations. We have sequenced a fragment of intron I region of the Hanwoo ACACA gene and identified two SNPs. Genotyping of the two SNP markers (g.2344T>C and g.2447C>A) was carried out using PCR-SSCP analysis in 309 Hanwoo steers to evaluate their association with carcass and meat production traits. The g.2344C SNP marker showed a significant increasing effect on LW (p = 0.009) and CW (p = 0.017). Animals with the CC genotype had higher CW and LW compared with TT and TC genotypes (p<0.05). The g.2447A SNP marker was associated with higher MC (p = 0.019). Animals with the AA genotype had higher MC than animals with CC and CA genotypes (p<0.05). Although the degree of linkage disequilibrium (LD) was not strong between g.2344T>C and g.2447C>A in the LD analysis, four major haplotype classes were formed with two SNP information within the ACACA gene. We constructed haplotypes using the HaploView software package program and analyzed association between haplotypes and carcass traits. The haplotype of ACACA gene significantly affected the LW (p = 0.027), CW (p = 0.041) and MC (p = 0.036). The effect of h1 haplotype on LW and CW was larger than that of h3 haplotype. Animals with the h1 haplotype also had greater MC than did animals with h2 haplotype. Consequently, the ACACA gene could be useful as a DNA marker for meat production traits such as carcass yield and meat contents in Hanwoo.

노인요양시설 입소자들의 의료적 요구에 대한 간호직 종사자들의 관리 경험 (Nursing Staffs' Experiences of Managing Medical Needs of the Elderly in Korean Long-term Care Facilities)

  • Purpose: This qualitative study was designed to explore and describe the nursing staff reported experiences of managing the medical needs of nursing home residents in South Korea. Methods: Using a focus group interview method, qualitative data were collected from March to May 2012. Twenty five registered nurses and nurse aids working in nursing homes were recruited through a convenient sampling method. Participants participated in one of four focus group interviews lasted up to two hours in which their day-to-day experiences of taking care of the medical needs of the elderly residents. All interviews were recorded and transcribed in verbatim, and analyzed according to the qualitative thematic analysis method using MAXQDA software. Results: The overriding theme was 'experiencing differences between the requirements of the regulatory system and the actual reality of the work place.' The findings of differences between what were true and what is required was based in four subcategories. The nursing home residents had far more medical needs than what were reported. Another finding was that the family members were seen as non-supportive and negligent towards the residents. Conclusion: The findings of the study support the need for changes in the regulatory system. The regulatory limitations of current system with a shortage of resources pushed the participants to experience discrepancies between the required regulation and the reality. Additional research could contribute more exemplars to support changes.

고수두 1,000 MW 석탄화력발전소 냉각수계통 수격방지장치의 성능특성 (The Performance Characteristics of Anti-Surge Devices for High Head Cooling Water Systems in 1,000 MW Thermal Power plants)

  • 최근 환경 규제 정책 강화에 따른 입지 제약발생으로 발전소 부지가 높아지고 냉각수 관로의 길이가 증가되어 냉각수 계통 내 수격현상이 심화된다. 이는 발전소 안정성에 심각한 영향을 주게 된다. 본 연구에서는 수두가 높은 1,000 MW급 대용량 석탄화력발전소 냉각수계통에 대한 안정성을 확보하고자 비정상상태 1차원 해석 상용 소프트웨어인 LIQT 7.2을 사용하여 과도현상을 분석하고, 수격현상을 저감하기 위해 수격방지장치를 개별 및 조합 적용하여 성능특성에 대한 효과를 예측하였다. 수격방지 장치를 설치하지 않고 펌프가 불시 정지되었을 경우 발생하는 냉각수 서지압력은 펌프 출구 측에서 가장 크게 나타났다. 이러한 냉각수계통의 서지압력을 저감시키는 가장 효과적이고 간단한 방법은 펌프 보호를 위해 필수적 장치인 유압구동역류방지밸브에 진공파괴밸브를 조합한 것이다.

  • 무기체계 SW 개발 전반에 대한 규정인 방위사업청의 무기체계 SW 개발 및 관리 매뉴얼이 개정될 때마다 SW 신뢰성 및 품질향상을 위해 SW 신뢰성 시험의 수준과 범위를 상향하는 방향으로 변경됨에 따라 SW 신뢰성 시험은 SW 개발자에게 큰 부담이 되고 있다. 특히 동적시험은 무기체계 SW를 구현하는데 필요한 일정과 인력에 육박할 정도의 비용이 소요되고 있으며, 1회성으로 그치지 않고 소스코드 형상이 변경될 때마다 수행해야 한다. 본 논문에서는 동적시험 도구인 VectorCast를 이용하여 동적시험을 최초 1회 수행한 후 소스코드 형상변경으로 인한 동적시험 회귀시험 시 사람의 개입을 최소화하여 자동으로 회귀시험을 수행하고 결과 보고서를 생성해주는 회귀시험 자동화 프로그램인 VectorCast Environment Manager 개발에 대해서 기술한다.

지속가능한 순천만을 위한 보호지역 확대와 정책적 활용을 위한 생태계 서비스 지불제(PES)의 적용 (Establishment of Additional Protected Areas and Applying Payment for Ecosystem Services(PES) for Sustainability of Suncheonman-Bay)

  • Suncheonman-Bay and its surrounding areas play important roles as habitats for migratory birds. However, sustainable management of these areas is difficult because of the development pressure of private lands. Therefore, the areas surrounding Suncheonman-Bay must be classified as additional protected areas; for this, it is necessary to gather concrete and objective evidence and ensure protected area management. Further, compensation measures must be considered when acquiring a private property as an additional protected area. In this study, we distinguish protected areas, such as core, buffer, and transition areas, within a private area by using data from the Winter Waterbird Census of Korea and MARXAN software, a spatial conservation prioritization tool. We applied ecosystem services to apply Payment for Ecosystem services (PES) as compensation measures. Watershed conservation (supply), climate control (regulation), supporting habitats (support), and recreation (culture) etc. were evaluated by calculating the economic value of these ecosystem services. Eastern, western, and northern forests and rice fields of Suncheonman-Bay were shown to have a number of core areas for the preservation of endangered species. The ecosystem service value of the additional protected areas was estimated at 17.5 million KRW/ha/year. We believe that our study result could be used to establish protected areas to preserve major habitats, as well as include areas adjacent to such major habitats that play a vital role in endangered species conservation. In addition, through this study, we highlight the need for an objective basis to establish protected areas.

Lin28 regulates the expression of neuropeptide Y receptors and oocyte-specific homeobox genes in mouse embryonic stem cells

  • Objective: Lin28 has been known to control the proliferation and pluripotency of embryonic stem cells. The purpose of this study was to determine the downstream effectors of Lin28 in mouse embryonic stem cells (mESCs) by RNA interference and microarray analysis. Methods: The control siRNA and Lin28 siRNA (Dharmacon) were transfected into mESCs. Total RNA was prepared from each type of transfected mESC and subjected to reverse transcription-polymerase chain reaction (RT-PCR) analysis to confirm the downregulation of Lin28. The RNAs were labeled and hybridized with an Affymetrix Gene-Chip Mouse Genome 430 2.0 array. The data analysis was accomplished by GenPlex 3.0 software. The expression levels of selected genes were confirmed by quantitative real-time RT-PCR. Results: According to the statistical analysis of the cDNA microarray, a total of 500 genes were altered in Lin28-downregulated mESCs (up-regulated, 384; down-regulated, 116). After differentially expressed gene filtering, 31 genes were selected as candidate genes regulated by Lin28 downregulation. Among them, neuropeptide Y5 receptor and oocyte-specific homeobox 5 genes were significantly upregulated in Lin28-downregulated mESCs. We also showed that the families of neuropeptide Y receptor (Npyr) and oocyte-specific homeobox (Obox) genes were upregulated by downregulation of Lin28. Conclusion: Based on the results of this study, we suggest that Lin28 controls the characteristics of mESCs through the regulation of effectors such as the Npyr and Obox families.

Loss of Heterozygosity at the Calcium Regulation Gene Locus on Chromosome 10q in Human Pancreatic Cancer

  • Background: Loss of heterozygosity (LOH) on chromosomal regions is crucial in tumor progression and this study aimed to identify genome-wide LOH in pancreatic cancer. Materials and Methods: Single-nucleotide polymorphism (SNP) profiling data GSE32682 of human pancreatic samples snap-frozen during surgery were downloaded from Gene Expression Omnibus database. Genotype console software was used to perform data processing. Candidate genes with LOH were screened based on the genotype calls, SNP loci of LOH and dbSNP database. Gene annotation was performed to identify the functions of candidate genes using NCBI (the National Center for Biotechnology Information) database, followed by Gene Ontology, INTERPRO, PFAM and SMART annotation and UCSC Genome Browser track to the unannotated genes using DAVID (the Database for Annotation, Visualization and Integration Discovery). Results: The candidate genes with LOH identified in this study were MCU, MICU1 and OIT3 on chromosome 10. MCU was found to encode a calcium transporter and MICU1 could encode an essential regulator of mitochondrial $Ca^{2+}$ uptake. OIT3 possibly correlated with calcium binding revealed by the annotation analyses and was regulated by a large number of transcription factors including STAT, SOX9, CREB, NF-kB, PPARG and p53. Conclusions: Global genomic analysis of SNPs identified MICU1, MCU and OIT3 with LOH on chromosome 10, implying involvement of these genes in progression of pancreatic cancer.

뇌-컴퓨터-인터페이스를 이용한 암환자들의 전전두엽 뇌파 분석 (Patterns Analysis of Prefrontal Brain Waves of Cancer Patients using Brain-Computer-Interface)

  • 암환자들은 암의 진행과 항암화학요법 등의 치료로 인해 심신의 불안정과 항상성의 저하로 큰 고통을 겪고 있다. 간편하면서 인체에 아무 해를 주지 않는 뇌파를 기반으로 하는 뇌-컴퓨터-인터페이스(BCI) 기술로서 암 환자의 상태를 모니터링하여 적절한 처치를 취할 수 있다는 것은 매우 중요한 일이다. 암환자들의 전전두엽에 헤드밴드 형태의 건성전극단자를 부착하고, 컴퓨터와 연결된 휴대용 뇌파측정 장치로 전전두엽 뇌파(Fp1, Fp2)를 측정하였다. 컴퓨터를 통하여 파장대 별로 얻어진 뇌파를 상호 연관성에 따라 뇌지수로 구분한 후 통계 처리하여 유의성을 검증하였다. 암환자군과 정상대조군을 비교한 결과 암환자군에 비하여 정상대조군이 기초율동지수, 주의지수, 정서지수, 항스트레지수와 좌우뇌균형지수에서 유의하게 높은 차이를 나타냈다. 따라서 뇌파 측정이 환자의 상태를 모니터링하는 중요한 도구로서의 가능성을 보였다.

PERT/CPM을 이용한 최적화된 협업 프로세스 수립 모형 (Establishing Model of Optimized Collaboration Procedure using PERT/CPM)

  • 다자간 협업이 요구되는 신규 사업 시 협업 절차를 수립하거나 이미 다자간 상호 연계 형태로 진행되는 협업 절차에서 문제점을 분석하여 객관적으로 증빙하고 제도를 개정하는 일은 매우 어려운 작업이다. 본 연구에서는 PERT/CPM 네트워크 다이어그램을 활용하여 협업 주체 간 업무 처리 절차를 수립하고 처리 소요 기간을 산출할 수 있는 협업 프로세스를 위한 최적화 수립 모형을 제안하였다. 또한 본 연구에서 개발한 협업 프로세스 최적화 수립 모형의 실효성과 유용성을 검증하기 위하여 고용노동부에서 실시하고 있는 학위연계형 일학습병행제 학습근로자 선발 절차의 협업 절차 최적화 작업에 적용하였다. 본 연구는 협업 절차를 새로이 수립하거나 재구성해야 하는 경우 뿐만 아니라 정보시스템 구축을 위해 협업 주체 간 정보시스템 간 연계 업무 절차를 표준화하기 위하여 유용하게 활용할 수 있다.

유아의 놀이성, 정서통제, 정서불안정이 또래놀이행동에 미치는 상대적 영향 비교 (A Comparison of Effects of Playfulness, Emotional Control, Emotional Instability on Young Children's Peer Play Behavior)

  • 본 연구에서는 유아의 놀이성, 정서통제, 정서불안정이 또래놀이행동에 미치는 상대적 영향을 비교하기 위해 어린이집에 다니는 만 4세 및 5세 유아 209명을 대상으로 연구를 실시하였다. 본 연구에서는 유아의 놀이성을 측정하기 위해 '놀이성척도'를, 정서통제, 정서불안정을 측정하기 위해 '정서규제체크리스트'를, 또래놀이행동을 측정하기 위해 '또래놀이행동척도'를 사용하였다. 수집된 자료는 t-검증, 피어슨의 적률상관계수, 단계적 회귀분석을 통해 분석되었다. 본 연구의 결과는 다음과 같다. 첫째, 연령에 따라 유아의 또래놀이행동, 놀이성, 정서통제, 정서불안정에 차이가 있는 것으로 나타났다. 둘째, 유아의 놀이상호작용은 정서통제와 놀이성에 의해 영향을 받는 것으로 나타난 반면, 유아의 놀이방해와 놀이단절은 정서불안정과 놀이성에 의해 영향을 받는 것으로 나타났다. 이와 같은 연구결과에 기초하여 유아의 또래놀이행동에 영향을 미치는 정서 관련 변인의 중요성에 대해 논의한 후 향후 과제와 제언을 제시하였다.
