Effects of Hyunsamkabhangbang on the Hyperthyroidism Induced by Sodium Levothyroxine in Rats (현삼갑항방(玄參甲亢方)이 Sodium Levothyroxine 투여로 유발된 백서의 갑상선중독증에 미치는 영향)

  • Objective : This study was to elucidate the effects of Hyunsamkabhangbang (HSK) on the hyperthyroidism induced by sodium levothyroxine. Method : Sprague-Dawley rats were separated into five groups. Except the normal group, the other four groups were treated with sodium levothyroxine 160${\mu}g/kg/day$ for 5 days by oral administration. Among the four groups, all except one (as control) were treated with various concentrations of HSK (0.5, 1.0, 2.0 g/kg/days) for 3 days separately. T3-uptake, T3, T4, TSH, total cholesterol, free fatty acid, ${\beta}$-lipoprotein, glucose, ALP, AST, ALT change in the serum and body weight of rats were measured after medication of solid extract of HSK. Results : HSK decreased significantly serum T3, T4, T3-uptake, ALP, free fatty acid and glucose level in a dose-dependent manner compared with control. HSK increased significantly serum TSH and total cholesterol level and body weight in a dose-dependent manner compared with control. HSK also increased serum ${\beta}$-lipoprotein level compared with control, but not statistically meaningfully. HSK increased significantly serum AST and ALT level compared with control, but relationship to hyperthyroidism is not found. Conclusions : From these results, it could be concluded that HSK lowers excess thyroid hormone and increased metabolism, resulting in improvement of hyperthyroid state.

  • Objectives : The aim was to study experimentally the effects of KABHANG-BANG(갑항방) on the hyperthyroidism of rats induced by sodium levothyroxine. Methods : Spargue-Dawley rats were separated into four groups. Except normal group, the other three groups were treated with sodium levothyroxine $32{\mu}g$/200g/days for 5 days by oral administration. Among the three groups, two except one(as control) were treated KHB extract 225mg/200g/days(sample A), 450mg/200g/days(sample B) for 3 days separately. T3-uptake, T3, T4, r-TSH, total cholesterol, total protein change in the Serum of rats were measured after medication of solid extract of KHB. Results : As a result of this study, in the serum T3 and T4 level were decreased in sample A and B compared to controls. The serum r-TSH, total cholesterol and total protein were increase in sample A and B compared to controls. In the serum T3-uptake content, Smple B showed significant decrease in comparison with control group, but Sample A did not show significant decrease. Conclusion : According to the above results, it is suggested that KHB is effective to cure hyperthyroidism.

  • L-Thyroxine possessing a chiral center, the naturally occurring thyroid hormone has been used for the treatment of thyroid dysfunctions and marketed as levothyroxine (L-thyroxine) sodium salt. In this study, after extraction of levothyroxine tablet as a pre-treatment process, direct enantiomer separation of thyroxine on crown ether derived chiral columns for determination of optical purity was performed using reversed mobile phase with acid additive. The chromatographic method developed in this study was applied in the determination of optical purity of several current domestic and foreign commercialized levothyroxine tablets. Optical purity values of these commercialized L-thyroxine sodium tablets except one were higher than 99 percents.

The Effects of YUKWOOLTANG(六鬱湯) on the Hyperthyroidism of Rats (육울탕이 백서의 갑상선기능항진증에 미치는 영향)

  • Objectives : The aim was to study experimentally the effects of YUKWOOLTANG(li${\`{u}}$y${\`{u}}$-t${\={a}}$ng; YWT) induced by sodium levothyroxine. Methods : Spargue-Dawley rats were separated into four groups. Except normal group, the other three groups were treated with sodium levothyroxine $32{\mu}g$/200g for 5 days. Among the three groups, two except one(as control group) were treated YWT extract separately(sample A:75mg/200 g, sample B: 150mg/200 g ). T3-uptake, T3, T4, r-TSH, total cholesterol, total protein change in the serum of rats were measured after medication of solid extract of YWT. Results : As a result of this study, in the serum T3-uptake content, sample A showed significant decrease in comparison with control group, but sample B group did not show significant decrease. 2. In the serum T3 content, sample B showed significant decrease in comparison with control group. In the serum T4 content, YWT showed significant decrease in comparison with control group. The serum r-TSH was shown with valuable increase in sample B of YWT. The serum total cholesterol was not shown with valuable increase in YWT. The serum total protein was shown with valuable increase in YWT. Conclusion : These results suggest that YWT is effective to cure Hyperthyroidism.

The Effects of Radix Scrophulariae (玄蔘) on the Hyperthyroidism of Rats (현삼이 갑상선기능항진 유발 백서에 미치는 영향)

  • Objective: The aim was to study experimentally the effects of Radix Scrophulariae (玄蔘) induced by Sodium levothyroxine. Methods: Sprague-Dawley rats Were separated into four groups. Except for the normal group, the other three were treated with sodium levothyroxine $160{\mu}g/kg$ for 5 days. Among the three groups, two (except the control) were treated with Radix Scrophulariae extract 0.5g/kg (HNS 1) or 1.0g/kg (HNS II). $T_3-uptake,\;T_3,\;T_4$, TSH, total cholesterol, glucose, ALP, AST, ALT, free fatty acid, $\beta-lipoprotein$, and change in the serum of rats were measured after medication of solid extract of Radix Scrophulariae. Results: As a result of this study, HNS II showed significant decrease in comparison with the control in the serum $T_3-uptake,\;T_3$ ALP and free fatty acid content. HNS I and HNS II showed significant decrease in comparison with the control in the serum of $T_4$ and glucose content. HNS I and HNS II showed significant increase in comparison with the control in the serum of TSH and total cholesterol content. In body weight, HNS I and HNS II showed significant increase in comparison with the control. Conclusion: These results suggest that Radix Scrophulariae is effective to cure hyperthyroidism.

The Effects of Dioscorea bulbifera L. on Hyperthyroidism of Rats (황약자가 갑상선기능항진 유발 백서에 미치는 영향)

  • Objective: The aim was to study experimentally the effects of Dioscorea bulbifera L. induced by sodium levothyroxine. Methods: Sprague-Dawley rats were separated into four groups. Except for the normal group, the other three were treated with sodium levothyroxine $160{\mu}g/kg$ for 5 days. Among the three groups, two (except the control) were treated with Dioscorea bulbifera L. extract of 0.75g/kg (sample A) or 1.5g/kg (sample B). $T_3-uptake,\;T_3,\;T_4$, TSH, total cholesterol, glucose, ALP, AST, ALT, tree fatty acids, $\beta-lipoprotein$, and change in the serum of rats were measured after medication with solid extract of Radix Scrophulariae. Results: $T_4$ concentration and $T_3-uptake$ level decreased in sample A and B compared to controls. $T_3$ concentration decreased but not statistically meaningful. Considering lipid metabolism, only free fatty acid level was significantly reduced glucose level also significantly decreased in sample B. Serum AST, ALT and ALP levels were slightly elevated but there was no meaning statistically, which doesn't mean Dioscorea bulbifera L. has liver toxicity because the quantity of the extract perhaps couldn't reach the minimal dose causing liver toxicity, which according to an article was 60g. Body weight significantly increased in comparison to controls. Conclusion: These results suggest that Dioscorea bulbiftra L. lowers excess thyroid hormone and increased metabolism, resulting in improvement of hyperthyroid state.

Gene Expression of Hyperthyroid Rats treated by Scrophularia buergeriana Miquel (현삼(玄蔘)이 갑상선기능항진증(甲狀腺機能亢進症) 유발(誘發) 흰쥐의 유전자(遺傳子) 발현(發顯)에 미치는 영향(影響))

  • Objective : The study was done to investigate Scrophularia buergeriana Miquel's effects in the genomic level, by measuring gene expression in hyperthyroid induced rats using cDNA micro array. Methods : Spargue-Dawley rats were separated into three groups(each with 10 rats). Except normal group, the other two groups were treated with Sodium levothyroxine $160{\mu}g$/kg/days for 5 days by oral administration. After 2 hours of the last intake of Sodium levothyroxine on two experimental group, one group was treated Scrophularia buergeriana Miquel extract 1.0g/kg/days for 3 days. The other groups(normal group, administration group) was treated normal saline 1.0g/kg/days for 3 days. Gene expression of hyperthyroid rats were measured after medication of solid extract of Scrophularia buergeriana Miquel with cDNA microarray. Results : In thyroid tissue, the numbers of the genes showed over twice increase and decrease in the control group compared to the normal group were 296, in the Scrophularia buergeriana Miquel administration group compared to the normal group were 859 and in the hypothalamus, the numbers of the genes showed over twice increase and decrease in the control group compared to the normal group were 416, in the Scrophularia buergeriana Miquel administration group compared to the normal group were 391. Conclusion : According to the above results, it is suggested that Scrophulari buergeriana Miquel suppress hyperthyroidism effectively and regulate the gene expression in the thyroid and brain.

  • Serous effusions have been most commonly associated with ascites, pericardial effusion, and heart failure. But, they have been considered to be an unusual form of complication in hypothyroidism and pleural effusion, which has been observed as an isolated finding in hypothyroidism is apparently rare and complete analysis of these types of hypothyroid-associated pleural effusions has yet not been described. We report a case of hypothyroidism associated with unilateral pleural effusion in a 77 year-old male patient who was improved through levothyroxine sodium with brief review of the literature.

A Comparative Study on the Etfects of 『Palmisoyo-San gamibang』, 『Gyebu-Tang』 and 『Boganyangje』 on the Thyrotoxicosis of Rats (갑상선중독유발 흰쥐에 미치는 팔미소요산가미방(八味消遙散加味方)과 계부탕(桂附湯)및 보간양제(補肝陽劑)의 영향 비교 연구)

  • In order to study comparatively the effects of "Palmisoyo-San gamibang(八味消遙散加味方)"(AP), "Gyebu-Tang(桂附湯)"(GT) and "Boganyangje(補肝陽劑)" (BG) on the thyrotoxicosis rats, we have made thyrotoxicosis rat model by administration sodium levothyroxine p.o. during 4 days(Control group), and have administered solid extract of AP(236.8mg/200g/day : Sample I group), GT(80mg/200g/day : Sample II group) and BG(70.4mg/200g/day : Sample III group) p.o. to thyrotoxicosis rats during 3 days from 3rd day. We measured the body weight(BW), body temperature(BT), levels of Serum $T_3{\cdot}T_4{\cdot}free\;T_3{\cdot}free\;T_4$ and TSH after administrating solid extract of AP, GT and BG. The results are summarized as follows: 1. Sample I group showed significantly high increasing rate compared with Control, Sample II and Sample III group, but it showed significantly lower levels of BT, $T_4$ and free $T_4$ than those of Control, Sample II and Sample III. group. 2. We could know that AP suppress the alteration of $T_4$ to $T_3$. 3. Sample II and Sample III group showed opposite results to Sample I group in the BW., BT, $T_4$ and free $T_4$ Especially Sample III was significantly higher than Control, Sample I and Sample II, so we could guess that BG stimulates the formation of thyroid hormones. According to the above experimental results, "Palmisoyo-San\;gamibang(八味消遙散加味方)" is assumed to have a curative effect against the thyrotoxicosis rat induced by sodium levothyroxine, and to suppress the alteration of $T_4$ to $T_3$. And we can suppose that "Boganyangje(補肝陽劑)" has a curative effect against the hypothyroidism.

Effects of Ukgan-san on the L-thyroxine-induced Rat Hyperthyroidism (억간산이 갑상선기능항진 유발 백서에 미치는 영향)

  • This study was to elucidate the effects of Ukgan-san on the hyperthyroidism induced by sodium levothyroxine. Hyperthyroidisms were induced by continuous subcutaneous treatment of LT4, 0.3 mg/kg, once a day for 27days, and 1,500, 1,000 and 500 mg/kg of Ulkansan extracts were orally administered, once a day for 15 days from 12th LT4 treatment, and the changes on the body, thyroid gland, liver and epididymal fat pad weights, serum triiodothyronine(T3), thyroxine(T4), thyroid-stimulating hormone(TSH), asparte aminotransferase (AST) and alanine aminotransferase (ALT) concentrations, hepatic lipid peroxidation(LPO), glutathione(GSH), superoxide dismutase(SOD) and catalase(CAT) activities were investigated with throid gland, liver and epididymal fat pad histopathological changes. The effects of Ukan-san extracts were compared with that of propyl thiouracil(PTU), a standard antithyroidic drug 10 mg/kg(intraperitoneally). 1,500 and 1,000 mg/kg of Ukan-san extracts reversed all LT4-induced hyperthyroidisms and these effects indicating their potential in the regulation of hyperthyroidism. Further, the Ukan-san extract normalized LT4-induced liver oxidative stresses, and also reduced liver damages and epididymal fat pad reducements suggesting its antioxidative and relative organ protective nature. However, nor favorable effects on LT4-induced hyperthyroidisms were detected in Ulkansan 500 mg/kg treated rats as compared with LT4 control rats in the present study. These effects of Ukan-san may help improvement of hyperthyroidisms and accompanied various organ damages.
