Study on Iodine Labelling (I) Influence of Reducing Agent and Iodate-$^{131}I$ in Sodium iodide-$^{131}I$ solution on Labelling

  • In iodine-131 labelling of iodocompounds such as tetrachloro-P-tetraiodo R-fluorescein, sodium orthoiodohippurate and a non-iodocompound, human serum albumin (HSA), the labelling rates and yields are accurately compared with each other. The reaction systems conducted for each compounds were different conditions: sodium iodide-$^{131}$ I containing reducing agent, sodium iodide-$^{131}$ I free from reducing agent, and sodium iodide-$^{131}$ I free from reducing agent but containing considerable amount of iodide-$^{131}$ I etc. The labelling yields were generally poor; 10% in the case of using sodium iodide-$^{131}$ I containing redoing agent, and 50~60% in the case of using sodium iodide-$^{131}$ I free from reducing agent but containing considerable amount of iodide-$^{131}$ I. However, fair yields were obtained in the case of using sodium iodide-$^{131}$ I free from reducing agent and mostly in the form of iodide-$^{131}$ I. The reaction entities involved in these reactions are also briefly discussed.

Determination of Trace Iodide in Sodium Bisulfite Aqueous Solution by Ion Chromatography with UV Detection (이온크로마토그래피를 이용한 Sodium bisulfite 수용액 중의 미량 요오드 정량)

  • The iodide was recovered from a simulated spent fuel to the sodium bisulfite aqueous solution. It was discussed that the trace iodide (below 1 ppm) was determined without the matrix effect of 0.1 M sodium bisulfite and 1 mM $HNO_3$ in aqueous solution by ion chromatography with UV detection. AS4A-SC (DIONEX) column and UV-absorption spectrophotometer were used. The UV-absorption spectra of sodium bisulfite, nitric acid and iodide were obtained, and then 230 nm was selected as an absorption wavelength for iodide determination. 0.1 M NaCl eluent was optimum condition. In this condition the calibration curve of iodide was obtained on the range of about 0-1,000 ppb. The linear coefficient was 0.99993 and the detection limit was 5 ppb. The relative standard deviation was 1.26%.

  • A stepwise gradient elution with two wavelengths detection was performed for the separation and determination of some anions in saline water. The eight anions such as iodate, bromate, nitrite, bromide, nitrate, chromate, iodide and thiocyanate were successfully separated using AS-7 column and sodium chloride/sodium phosphate buffer solution as an eluant within 40 min. The separation behaviors of anions were studied at various sodium chloride concentrations. The peak shapes of anions of bromate, nitrite, bromide and nitrate gradually broadened as the concentration of sodium chloride increased until 1.0 M in the sample solutions. However, no effect was observed in the peak shapes of chromate, iodide and thiocynate. A good linearity was obtained at the range of ppm(mg/L). The detection limit was proved to be $10-720{\mu}g/L$ for the eight anions with $50{\mu}L$ injection volume. This method was applied to the determination of $Br^-$, ${NO_3}^-$ and $I^-$ in sea water.

  • Ion chromatography was applied to determine iodide remained in sulfuric acid aqueous solution after adsorption procedure. Iodide was determined in 0.25 M, 0.5 M and 1 M sulfuric acid solution on time variation. Because sulfuric acid in solution plays as an oxidizer, the concentration of iodide decreased with increasing concentration or sulfuric acid. Thereafter, sulfuric acid was neutralized with sodium hydroxide. Good linearity(r=0.99999) was obtained at the range of 0-20 mg/L 1 in 0.5 M sodium sulfate matrix.

  • Sodium periodate ($NaIO_4$) and potassium iodide (KI) in aqueous ammonia has been used for the one-pot synthesis of nitriles from the corresponding aldehydes and alcohols in moderate to good yield. This transformation, proceeds via an in situ oxidation- imination-aldimine oxidation sequence.

  • Appreciable radiation exposures certainly occur in the workers who handle radioiodine in biochemical research, nuclear medicine diagnostics with the development of nuclear industries. But in the case of occurring the nuclear accidents, the early medical treatment of radiation injury should be necessary but little was reported in korea till now. Accordingly, to achieve of the basic data for protective roles and medical treatment of radiation injury, the present studies were carried out to evaluate the decontamination of radioiodine by the administration of the antithyroid drugs. The results observed are summarized as follows: 1. The administration of sodium iodide and potassium iodide results in rapid excretion of radioiodine and reduction of the whole body retention than the saline-only group. 2. Reguarding to thyroid protective effects, sodium iodide, potassium iodide and saline were effected significant in order. 3. In the control(saline) group, if administered with enough fluids, the whole body retention of radioiodine is reduced temporary shifts. But as far as radioprotective effects is concerned, saline was not more in the protective effects than the other groups. In conclusion, in case of nuclear accidents, if being administered sodium iodide and saline as quickly as possible, the radioprotective effects against the radiation hazard might be markedly increased in the internal contamination of radioiodine.

Expression of Sodium-Iodide Symporter (NIS) in Thyroid Nodules: Comparison of RT-PCR and Immunohistochemical Staining Methods (갑상선 결절에서 Sodium Iodide Symporter (NIS)의 발현: RT-PCR방법과 면역조직화학염색법의 비교)

  • Purpose: The sodium-iodide symporter (NIS) expression is an important factor in determining the sensitivity of radioiodine therapy in well-differentiated thyroid cancers. Several previous studies for the expression of NIS in thyroid tissues show diverse results. To investigate whether there is difference between methods in determining the expression of NIS in thyroid tissues of patients with thyroid nodules, we measured the expression ot NIS using two different methods (RT-PCR and immunoshistochemical staining) and compared the results. Materials & Methods: We measured the expression of NIS by reverse transcriptase-polymerase chain reaction (RT-PCR) and also by immunohistochemical staining using anti-NIS antibody in thyroid cancers and other benign thyroid diseases. We compared the results of each method. We included 19 papillary carcinomas, 1 follicular carcinoma, 7 medullary carcinoma, 4 adenomas and 7 nodular hyperplasias. Results: By RT-PCR analysis, 10 of 19 papillary carcinomas expressed NIS, but 1 follicular cancer didn't express NIS. By immunohistochemical staining, 15 of 19 papaillary carcinomas express NIS, but 1 follicular lancer didn't express NIS. There was a significant correlation between the semiquautitative results of RT-PCR and immunohistochemical staining of NIS expression. (p<0.01) Conclusion: Our data demonstrated that the expression of NIS in thyroid cancers and other benign diseases investigated by RT-PCR and immunohistochemical staining correlated well each other. However, by immunohistochemical staining, more NIS expression was found.

Enhancing the Physicochemical Properties of Sodium Iodide-based Root Canal Filling Material with Lanolin Incorporation

  • This study aimed to enhance the physicochemical properties of sodium iodide-based root filling materials, particularly solubility. In earlier developmental stages, the iodoform-containing paste exhibited high antibacterial efficacy but failed to meet only the solubility requirement among the ISO 6876 criteria. Therefore, this study focused on enhancing the physicochemical properties of the paste under development, particularly centering on reducing its solubility. Four experimental groups were established, including three control group. The previously developed D30 paste was named the Oil 33 group, and the control group was named the Vitapex® group. The Oil 50 group, in which the oil content was increased, and the Oil 45L group, in which lanolin was incorporated. The physical properties (solubility, pH, flowability, and film thickness) of the four pastes were evaluated according to the ISO 6876 standards. No significant differences were observed between the Oil 45L and Vitapex® groups in any of the physical property evaluations. While the Oil 33 and Oil 50 groups met the ISO 6876 standards for flowability and film thickness, the Oil 45L group met all the physical properties. However, reducing the overall oil content may be necessary to enhance the antimicrobial properties. The result of the physicochemical experiments showed that the Oil 45L group with the newly formulated composition and incorporated lanolin exhibited low solubility meeting the ISO 6876 standard of ≤ 3%. We were able to develop a paste with more stable solubility than previous iodide-based root-filling materials. Therefore, the oil content must be further adjusted to improve its antimicrobial properties. If other physical properties also meet the ISO 6876 standards and demonstrate excellent results in cytotoxicity tests, this root filling material could potentially replace existing options.
