The herb, Cajanus indicus L, is well known for its hepatoprotective action. A 43 kD protein has been isolated, purified and partially sequenced from the leaves of this herb. A number of in vivo and in vitro studies carried out in our laboratory suggest that this protein might be a major component responsible for the hepatoprotective action of the herb. Our successive studies have been designed to evaluate the potential efficacy of this protein in protecting the hepatic as well as renal tissues from the sodium fluoride (NaF) induced oxidative stress. The experimental groups of mice were exposed to NaF at a dose of 600 ppm through drinking water for one week. This exposure significantly altered the activities of the antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase (GST), glutathione reductase (GR) and the cellular metabolites such as reduced glutathione (GSH), oxidized glutathione (GSSG), total thiols, lipid peroxidation end products in liver and kidney compared to the normal mice. Intraperitoneal administration of the protein at a dose of 2 mg/kg body weight for seven days followed by NaF treatment (600 ppm for next seven days) normalized the activities of the hepato-renal antioxidant enzymes, the level of cellular metabolites and lipid peroxidation end products. Post treatment with the protein for four days showed that it could help recovering the damages after NaF administration. Time-course study suggests that the protein could stimulate the recovery of both the organs faster than natural process. Effects of a known antioxidant, vitamin E, and a non-relevant protein, bovine serum albumin (BSA) have been included in the study to validate the experimental data. Combining all, result suggests that NaF could induce severe oxidative stress both in the liver and kidney tissues in mice and the protein possessed the ability to attenuate that hepato-renal toxic effect of NaF probably via its antioxidant activity.

The purpose of this study was to compare the growth-inhibiting effect of sodium fluoride and stannous fluoride on a cariogenic strain of Streptococcus mutans exposed to the different concentration of $SnF_2$ and NaF The result were as follows: 1. The growth rate of the Streptococcus mutans was unaffected by 75 ppmF, slowed by 150 ppmF, and immediately arrested by 300 or 600 ppmF where increments of NaF were added to actively growing 12-hour broth culture. 2. $SnF_2$ slowed the growth rate at 75 ppmF, was to bactericidal initially at 150 and 300 ppmF, and was to totally bactercidal at 600 ppmF. 3. $SnF_2$ has been shown to be more effective than NaF in inhibiting the growth activity of Streptococcus mutans. 4. The inhibitory effect of fluoride compounds on the growth of Streptococcus mutans was increased in the decreased pH at concentration of 600 ppmF.
The use of mouth-rinses containing sodium fluoride after tooth brushing has been proposed as a potentially useful measure for the prevention of dental caries in the practice of public health. About 700 middle school girls rinsed their mouths in the school once a week with sodium fluoride sodution after tooth-brushing. Tooth brushing was practiced everyday after meals and the rinses were carried out under the supervision of the teachers. The procedure consisted of gargling 10cc of fluoride solution for one minute and immediately followed by a second rinse with tap water. The rinsing was started in May 1976 and continued for one year. The results were as follows: 1) The inhibition of the 'D' Person rate was 76.04%, 'M' person rate was 80.79%, 'F' person rate was 4.06% and, 'D.M.F.' person rate was 76.86%. 2) the reduction of the 'D.T.' Index was 81.94%, 'M.T.' Index was 85.71%, 'F.T.' Index was 7.69% and, 'D.M.F.T.' Index was 76.83%. 3) Mouth rinsing with 0.3% NaF solution and tooth brushing are of value in the mass control of dental caries.
The kinetics of the loss of leaf fresh weight during incubation of barley and rice leaves in 9% or 15% NaCl solutions were biphasic, indicating the existence of a controlling mechanism for water transport. The first rapid phases reached their plateaus within 1 and 2 h in the case of rice and barley leaves, respectively. When barley leaves were fed with sodium fluoride, an inhibitor of phosphatase inhibitor, through their epicotyls for 3 h in darkness, prior to the treatment of NaCl, the biphasic pattern shown during NaCl treatment was disappeared resulting in linear decreases in the relative fresh weights. The results suggest that NaF accelerates salt-induced water efflux from plant cells, possibly by inhibiting the protection mechanism that may act in NaF-untreated leaves. The linear water loss can be explained in terms of phosphorylation of aquaporin by blocking its dephosphorylation in the presence of the phosphatase inhibitor to keep aquaporin in a phosphorylated form. However, the effect of NaF shown in barley leaves were not observed in rice. These results suggest that the regulation of water transport depends on plant species, and the mechanism for the controlling water transport in rice is different from that of barley.
Background: Dental caries is one of several prevalent oral diseases caused by dental plaque biofilms. This study evaluated the anti-cariogenic effects of a bamboo salt (BS) and sodium fluoride (NaF) mixture on oral bacteria. Methods: The effects of several mixtures of NaF and BS on acid production, growth, and adhesion to glass beads of Streptococcus mutans, and their anti-cariogenic properties were investigated. The growth of S. mutans was measured according to optical density at 3, 6, 9, 12, 15, 18, and 24 hours after treatment using spectrophotometry at a wavelength of 600 nm, while pH was measured using a pH meter. Adhesion of S. mutans was measured according to the weight of glass beads from each group before and after incubation. Gene expression was measured using real-time polymerase chain reaction. Acid production and growth patterns of S. mutans were compared using repeated measures analysis of variance, followed by Scheffe's post-hoc test. The Kruskal-Wallis test was used to compare adhesion, followed by the Mann-Whitney test. Gene expression in the experimental and control samples was compared using the Student's t-test. Results: Growth, acid production, and adhesion of S. mutans were inhibited in all experimental groups. Expression of gft and fructosyltransferase in S. mutans was inhibited in all groups. A mixture of NaF and BS significantly reduced growth, acid production, adhesion, and gene expression of S. mutans compared with the other groups. Conclusion: Results of the present study demonstrated that a mixture of NaF and BS was useful as a mouth rinse in preventing dental caries.
이 연구의 목적은 silver diamine fluoride(SDF)를 도포할 때 불화나트륨(sodium fluoride, NaF) 바니쉬와 요오드화 칼륨(potassium iodide, KI)이 SDF의 재광화 효과에 미치는 영향을 유치의 법랑질 인공 우식 병소에서 미세경도의 변화를 통해 측정하고, 도포 전과 후의 표면 형태를 주사전자현미경(scanning electron microscopy, SEM)을 통해 관찰하여 비교하는 것이다. 발치된 유구치의 인접면에 법랑질 인공 우식병소를 형성하고 비커스 미세경도를 측정한 후, 4개의 군으로 나누어 I군에는 SDF, II군에는 SDF와 NaF 바니쉬, III군에는 SDF와 KI, 그리고 IV군에는 증류수를 처리하였다. 8일 간 pH cycling을 시행한 후 비커스 미세경도를 다시 한번 측정하여 미세경도의 변화량을 계산하였다. 또한 치아 연마 후, 인공 우식 유발 후, 그리고 pH cycling 이후 각 군에 대하여 각각 2개의 시편을 이용하여 SEM를 촬영하여 표면의 형태를 관찰하였다. 연구결과 미세경도는 III군에서 가장 크게 증가했다. I군과 II군 모두 미세경도의 증가를 보였으나 두 군 사이에는 통계학적으로 유의한 차이가 없었다. IV군에서도 미세경도의 증가를 보였으나 그 변화 정도는 가장 적게 나타났다. SEM 관찰 결과 I, II, III군이 우식 유발 직후나 IV군과 비교하여 표면이 더 균일해지고 불규칙성이 감소된 양상을 보였으며, III군에서 무정형의 피막 형태의 침착이 나타났다. 이러한 결과를 종합해 보면 SDF만을 도포한 군과 SDF와 NaF, SDF와 KI를 함께 도포한 경우 모두 표면 형태가 균일해지고 미세경도가 증가하였으며 이와 같은 결과를 기반으로 구강 내의 환경에서도 재광화 효과를 가질 수 있을 것으로 사료되었다. 또한 이 연구의 제한된 조건 하에서 NaF 바니쉬를 사용하였을 때에는 SDF의 재광화 효과를 증가시키지 못했으며, SDF와 KI를 함께 도포한 경우 SDF의 재광화가 효과가 증가되는 결과를 보였으나 추후 실제 구강 내에서의 다양한 상황을 고려한 추가적인 연구가 필요할 것으로 생각된다.
In this study, we investigated about cell toxicity and oxidative stress of HepG2 cell by treatment of sodium fluoride (NaF) and fluoride extracts from krill Euphausia superba meat, shell, whole body and krill meal. The cell toxicity showed significant at 300 and $500{\mu}g/mL$ NaF treatment group. But krill (Euphausia superba) fluoride extract (KFE) treatment in all groups were not toxic. The superoxide radical production increased significantly in NaF treated group, but there was no significant change in KFE treated group. The superoxide dismutase activity was a significant increase 21.5% at $100{\mu}g/mL$ and 24.7% at $300{\mu}g/mL$ treatment group of fluoride extracts from krill meat, and 8.7% at $300{\mu}g/mL$ in krill meals, compared to the control group. However, hydroxy radical flux and catalase and glutathione peroxidase activity of fluoride extracts from krill meat did not change. As a result, for a short period of time, NaF treatment in HepG2 cells affect the cell toxicity and oxidative stress, but in the case of KFE, these were not recognized. Thus, depending on the type of food ingested with fluoride, cell toxicity and oxidative stress was found to be different.
관상동맥 석회화 검사에 컴퓨터단층촬영(Comtuted tomography, CT)을 사용하지만 고위험군인 미세석회화 감별에는 어려움이 있다. $^{18}F$-sodium fluoride ($^{18}F-NaF$)가 미세석회화를 진단하는데 매우 유용하다는 연구가 해외에서 보고되고 있다. 본 논문에서는 $^{18}F-NaF$ PET 영상으로 석회화를 정량평가 하고 그 유용성을 알아보고자 한다. 환자는 총 45명($67.1{\pm}6.9$세)으로 $^{18}F-NaF$을 250 MBq 주입하고 1시간 뒤 30분간 영상을 획득하였다. 장비는 Discovery 710 (GE Healthcare, MI, USA)을 사용하였으며 모든 환자는 PET 검사 전 혈관조영CT (CTAngiography, CTA)를 촬영 하였다. 석회화의 SUVmax를 측정하고 좌심방의 배후방사능을 측정하여 Target to Background (TBR) 구한 뒤 정량 분석하였다. ROC 곡선(Receiver Operating Characteristic Curve)을 통하여 고위험군을 분류하였다. 융합영상에서 관상동맥 석회화는 226개로 나왔으며 SUVmax는 $1.15{\pm}0.39$으로 나왔다. 28명(62%)에서 58개가 고위험군으로(TBR>1.25)분류 되었다. 나머지 168개는 $TBR{\leq}1.25$로 나왔다. $^{18}F-NaF$ PET 영상으로 미세석회화의 정량평가가 가능하였고 고위험군을 분류할 수 있었다. 혈관조영CT와 $^{18}F-NaF$ PET을 병행한다면 위험도가 높은 미세석회화를 조기 진단하는 새로운 영상 진단법이 될 수 있을 것으로 사료된다.
Objectives : The aim of this investigation was to evaluate the remineralization effect of Bamboo salt and NaF+Bamboo salt solutions on bovine enamel formed incipient artificial enamel caries by microcomputed tomography (micro CT). Methods : Experimental solutions were distilled water(negative control), 2% sodium fluoride solution (2% NaF group), 3% bamboo salt solution (3% BS group) and the solution mixed 2% sodium fluoride solution and 3% bamboo salt solution (2% NaF+3% BS group). Specimens were prepared from extracted bovine teeth and divided into 4 groups of 10 specimens each by randomized blocks according to density. Then the specimens surface were divided equally into three parts to observe sound enamel area, incipient enamel carious area and remineralized enamel area. Only one-third of specimen surface was coated with nail varnish and these were exposed to a lactate carbopol buffer system for 72 hours. Then one-half of the demineralized enamel area was coated with nail varnish. The specimens were carried out under pH cycling model for 14 days as follows; samples were immersed in each experimental solution for 2 mins 3 times per day, demineralized for 4 hours and in mixed saliva for the remaining hours. After pH cycling, density was measured using micro CT. Results : All experimental groups showed remineralization effects except for negative control group(p<0.05). The differences of density after experimental solution treatment were statistically significant difference among 4 groups (p<0.01). The density difference values of groups were $0.04{\pm}0.01$ in negative control group, $0.19{\pm}0.01$ in 2% NaF group, $0.14{\pm}0.01$ in 3% BS group, and $0.21{\pm}0.01$ in 2% NaF+3% BS group. Conclusions : The bamboo salt solution showed remineralization effects on incipient artificial enamel caries and the solution mixed sodium fluoride solution and bamboo salt solution showed more remineralization effects than the bamboo salt solution. Thus, it is suggested that bamboo salt can be used as remineralization agent in incipient enamel caries lesion.
