A Comparative Study of the Anti-cancer Effects of Smilacis Chinae Rhizoma and Smilacis Glabrae Rhizoma on Stomach Cancer SNU-1 Cells (토복령(土茯苓)의 기원식물별 위암 세포 SNU-1에 대한 항암효과 비교연구)

  • Objectives : The purpose of this study was to compare the anticancer effects of Smilacis Chinae Rhizoma (SCR) and Smilacis Glabrae Rhizoma (SGR) on stomach cancer SNU-1 cells. Methods : The cell proliferation, apoptosis and cytokine level from the extracts were examined in order to compare the anticancer effects of SCR and SGR on SNU-1 cells. Results : SCR had greater anticancer effects than SGR in terms of the inhibition of cell proliferation and apoptosis in the SNU-1 cells. SCR decreased TGF-$\beta$ and SGR increased IL-2 in SNU-1 cells. SCR decreased more TGF-$\beta$ and increased more TNF-$\alpha$ compared to SGR. Conclusions : There were few significant differences according to the concentration and fraction, but a greater anticancer effect of SCR was shown as compared with SGR.

  • In the result of investigating traditional chinese medical literatures to understand definite immunopharmacologic effects of drugs for clearing away heat and detoxicating such as Sophorae tonkinensis Radix, Houttuyniae Herba, Smilacis glabrae Rhizoma, Belamcandae Rhizoma, Lasiosphaera seu Calvatia, Paridis Rhizoma, Lobeliae chinensis Herba, we could reach conclusions as follows: 1. Sophorae tonkinensis Radix, Houttuyniae Herba can increase voracity of leukocytes and activities of T cells. 2. Sophorae tonkinensis Radix, Smilacis glabrae Rhizoma can inhibit DTH. 3. Sophorae tonkinensis Radix, Houttuyniae Herba increase activities of B cells in low density but decrease activities of B cells in high density. 4. Sophorae tonkinensis Radix, Houttuyniae Herba, Smilacis glabrae Rhizoma, Belamcandae Rhizoma have anti-inflammatory effects. Studies about drugs for clearing away heat and detoxicating is on. but their range is wide and their effects are numerous. Therefore. we think that more deep and wide researches about them shoud be accomplished.

Studies on the Antioxidation and Antimicrobial Effect of Smilacis Glabrae Rhizoma (토복령의 항산화작용 및 항균 효과에 대한 연구)

  • Objectives : This study was performed to investigate the effects of Smilacis GlabraeRhizoma on antioxidation and antimicrobial activity. Methods : In this study, we investigated the effects of peroxide radicals on hydrogen donating activity and linoleic acid, and the MDA contents on the hepatic lipids of rats, via methanol extractions and subfractions of Smilacis Glabrae Rhizoma. Results : 1. Hydrogen donating activity was very great for the radical scavenging effects, depending on the additional concentration at the fraction level of chloroform and ethyl acetate. 2. The peroxide radicals in linoleic acid were lower depending on the additional concentration, at the fraction level of ethyl acetate, than the controls. We concluded that both had significant anti-oxidation effects. 3. MDA contents of the hepatic lipid had also inhibition effects on lipid radicals, in proportion to the concentration of n-hexane, chloroform, and ethyl acetate fraction level. 4. After extracting Smilads Glabrae Rhizoma with 80% methanol, we experimented with the extracts the antibiosis each concentration, for 5 bacilli, Bacilus subtilis, staphylococcus aureus, Escherichia coli, Salmonela typhimurium, and Alcaligenes faecalis. While the effects showed differentiations by concentration, they had usually the significant inhibition effect for the multiplication at 37.5~75ug/ml. To identify the effective constituents, we identified the antibiosis of the fractions assaying cyclically hexane, chloroform, ethyl acetate, and butanol. The result showed that antimicrobial activation of both Gram-positive and Gram-negative bacillus except for E. coli was measured highest at the fraction level of BuOH and water. Conclusions : This result suggest that the extractions of Smilacis Glabrae Rhizoma, at ethyl acetate fraction, had significant anti-oxidation effects and at BuOH and water fraction had relatively strong antimicrobial activity against Bacilus subtilis, staphylococcus aureus, Salmonela typhimurium, and Alcaligenes faecalis.

  • Objective : Inflammatory cytokines have a close relationship to insulin dependent diabetes mellitus (IDDM). The inhibitory effect of Smilacis Glabrae Rhizoma (SGR) were examined on production of nitric oxide (NO), prostaglandin $E_2$ $(PGE_2)$, synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and NF-${\kappa}$B activation in Raw 264.7 cells. Methods: Raw 264.7 cells were pretreated with SGR(20, 50, 100 ${\mu}g$/ml), and then cultured with lipopolysaccharides (LPS). Cell viability was measured by MTT assay; inhibition of NO, $PGE_2$, and TNF-${\alpha}$ production were measured by Griess reagent and enzyme-linked immunosorbent assay(ELISA). Induction of COX-2 and iNOS were determined by western blotting analysis. Inhibition of NF-${\kappa}$B was measured by immunofluorescence assay (IFA). Results: SGR inactivated NF-${\kappa}$B, and inhibited the production of NO, iNOS, and $PGE_2$. Inhibition of COX-2 and TNF-${\alpha}$ could not be confirmed. Conclusions: From the above result. SGR was found to have an anti-inflammatory effect of inhibition of NO, iNOS, and $PGE_2$ production via inhibition of NF-${\kappa}$B.

Isolation of a Calcium-Binding Fraction from a Hot-Water Extract of Smilax rhizoma (청미래덩굴 뿌리 열수 추출물로부터 칼슘 결합 물질의 분리)

  • We isolated a calcium-binding substance from Smilacis rhizoma hot-water extract using ion exchange, normal phase HPLC, and gel filtration chromatography; fractions were analyzed for calcium-binding activity. Fractions (F6) with the highest calcium-binding activity from the resource Q coulmn were pooled and further purified on an $NH_2$ column. Two major peaks were separated and the fraction (F61) with the higher calcium-binding activity was then loaded onto a $Superdex^{TM}$ column. A single peak (F611) with calcium-binding activity was finally obtained. These results suggest that the isolated calcium-binding fraction could be used as a functional food additive, similar to a calcium supplement, in the food industry.

The Effect of Smilacis glabrae Rhizoma aqua-acupuncture on injured kidneys Induced by $HgCl_2$ Intoxication in Rats (토복령약침자극(土茯笭藥鍼刺戟)이 $HgCl_2$중독(中毒) 흰쥐의 손상신(損傷腎)에 미치는 영향(影響))

  • Purpose: In order to study the therapeutic effect of TBR(Smilacis glabrae Rhizoma) aqua-acupuncture on the $HgCI_2$(mercury bichloride) intoxicated kidneys of rats. Methods: 10% TBR extract solution was aqua-punctured into the bilateral loci of Shin-su(BL23), every other day for 18 days, and the amount of leucocytes, erythrocytes, thrombocytes, hematocrit, hemoglobin, serum BUN and creatinine levels were measured and renal mercury level and histopathological findings were studied as well. The experimental groups were into nonmaI group, control group, sample I group, sample II group and sample III group. Results: 1. There were not any significant changes of leucocytes, erythrocytes and serum creatinine level in the TBR aqua-acupuncture group, saline aqua-acupuncture group and TBR aqua-acupuncture group respectively as compared with control group. 2. It showed a significant increase of hematocrit and hemoglobin level in the TBR aqua-acupuncture group on the 8th experimental day as compared with the control group. 3. It showed a significant increase of platelet in the TBR aqua-acupuncture group on the 8th experimental day, whereas significant decreases on the 18th experimental day as compared with the control group. 4. It showed a significant decrease of serum BUN level in the TBR aqua-acupuncture group on the 8th and 18th experimental day as compared with the control group. 5. It showed a significant decrease of renal mercury level in the TBR aqua-acupuncture group on compared with the control group. 6. In the histopathological findings, TBR aqua-acupuncture group showed slight atrophy of the renal tubules, mitosis and regeneration of tubular lining cells with visible nuclei, whereas the control group showed acute tubular necrosis, especially severe necrosis of tubular epithelial cells. Conclusions: Smilacis glabrae Rhizoma aqua-acupuncture on injured kidneys Induced by HgCl2 Intoxication in Rats increases hematocrit, hemoglobin and platelet level, and decreases BUN, renal mercury level. This suggests that Smilacis glabrae Rhizoma aqua-acupuncture helps to recover kidney from HgCl2 intoxication.

Antioxidant, anti-inflammatory, antibacterial and ovoprotective effects of mixture of Ulmi cortex and Smilacis rhizoma extracts (유백피, 토복령 추출물 혼합물의 항산화, 항염, 항균 및 난소세포 보호효과)

  • Objectives : US extract is a mixture of each extract of Ulmi cortex and Smilacis rhizoma. In this study, we investigated the antioxidant, anti-inflammatory, antibacterial, and ovoprotective effects of US extract in in vitro model to identify potential candidates for improving female reproductive function. Methods : The antioxidant activity of US extract was measured using 1,1-diphenyl- 2-picrylhydrazyl free radical and superoxide anion radical scavenging assays. The anti-inflammatory effect of US extract on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells were determined with a nitric oxide (NO) assay, enzyme linked immunosorbent assays, and western blots analysis. The antibacterial activity of US extract against vaginitis infection microorganisms were determined with disc diffusion and minimum inhibitory concentration assays. The ovoprotective effect of US extract on 4-vinylcyclohexene diepoxide (VCD)-induced ovotoxicity in CHO-K1 cells were evaluated with a cell viability assay. Result : US extract showed good antioxidant capacity and inhibited LPS-induced NO production as well as iNOS and COX-2 expression and secretion of pro-inflammatory cytokine IL-6 without affecting the cell viability. It showed significant clear zones for Staphylococcus aureus and Candida albicans but did not indicate the clear zones for Escherichia coli and Enterococcus faecium. VCD-induced ovotoxicity in CHO-K1 cells was significantly reduced by US extract pre-treatment. Conclusions : These results demonstrate that US extract has antioxidant activity, anti-inflammatory effects on the LPS-stimulated macrophages, antibacterial activity against vaginitis infection microorganisms, and protective effects on the ovarian cells against VCD-induced ovotoxicity. These findings suggest that the US extract can be used as new prescriptions, supplements, functional foods, and cosmetics for improving female reproductive function.

The Effect of Smilacis Glabrae Rhizoma on Rats with Gout induced by Monosodium Urate (토복령(土茯苓)이 Monosodium Urate로 유발(誘發)된 백서(白鼠)의 통풍(痛風)에 미치는 영향(影響))

  • This study was designed to investigate the Effects of Smilacis Glabrae Rhizoma(SGR) on the gout. After pretreatment of SGR(50, 500mg/kg) for 5days, the Effects of SGR was evaluated on changes Joint value increase ratio, Serum albumin, Serum globulin, glutamate dxalacetate transminase(AST), glutanate pyruvate transminase(ALT), blood urea nitrogen(BUN), Serum creatitine, Serum uric acid, xanthine oxidase activity, WBC, Erythrocyte Sedimentation Rate(ESR), platelet. The results were obtained as follows ; Joint valueincrease ratio was decreased in 50mg/kg, 500mg/kg SGR taken group, but changes were not significantly different with the control group. AST, ALT were not significantly different in all SGR taken groups compared with the control group. Serum BUN, creatinine were significantly decreased in 500mg/kg SGR taken group compared with control group. ESR was significantly decreased in all SGR taken groups compared with the control group. WBC, platelet were significantly decreased in 500mg/kg SGR taken group compared with control group. Serum uric acid was not significantly different in all SGR taken groups compared with the control group. Xanthine oxidase activity was significantly decreased in 500mg/kg SGR taken group compared with control group. From above results, it may be concluded that SGR can be used for treatment and prevention of gout.

Protective effects of Smilacis Glabrae Rhizoma on acute pancreatitis (토복령(土茯苓) 물 추출물의 급성 췌장염 보호 효과)

  • Objectives : Smilacis Glabrae Rhizoma (SGR) has been used in traditional medicine for treating diseases related to inflammation and cancer. However, the protective effect of SGR has not been reported in the study of acute pancreatitis (AP). Therefore, this study was performed to evaluate the protective effects of SGR water extract on cerulein-induced AP. Methods : AP was induced by intraperitoneal injection of supramaximal concentrations of stable cholecystokinin analogue cerulein ($50{\mu}g/kg$) 6 times at intervals of one hour in mice. SGR water extract (0.01, 0.05, or 0.1 g/kg) was administrated intraperitoneally 1 h prior to the first injection of cerulein. The mice were sacrificed at 6 h after the final injection of cerulein. Pancreas and lung were rapidly removed for histochemical examination and myeloperoxidase (MPO) assay. In addition, polymerase chain reaction (PCR) was performed to examine mRNA levels of proinflammatory cytokines such as interleukin(IL)-6, $IL-1{\beta}$ and Tumor necrosis $factor(TNF)-{\alpha}$. Results : Administration of SGR water extract significantly inhibited pancreatic weight to body weight ratio. In addition, SGR treatment inhibited the histological damages and increase of MPO activity in both pancreas and lung during AP. Also, mRNA levels of IL-6 but not $IL-1{\beta}$and $TNF-{\alpha}$ were inhibited by SGR water extract against AP. Conclusion : Our results revealed that pre-treatment of SGR water extract reduces the severity of cerulein-induced AP. Therefore, this study suggest that SGR could be used as a drug or agent to prevent AP.

Anti-oxidative and Anti-inflammatory Effect of Fractionated Extracts of Smilacis Glabrae Rhizoma in Human Umbilical Vein Endothelial Cell (혈관내피세포에서 토복령(土茯苓)의 항산화 및 항염증 효과)

  • Objectives : Smilacis glabrae rhizoma (SG) has been traditionally used as a herbal medication of musculoskeletal disorders like arthritis, pain, convulsions, and syphilis in traditional Korean medicine. This study was investigated anti-oxidative and anti-inflammatory effect of fractionated extracts of Smilacis Glabrae Rhizoma in Human Umbilical Vein Endothelial Cell (HUVEC). Methods : SG extract prepared with methanol, and then fractionated with hexane, dichloromethane, ethylacetate, n-butanol and water. Inhibitory effect of SG onto free radical generation was determined by measuring DPPH, superoxide anions and nitric oxide scavenging activities in vitro. Cytotoxic activity of extracts on RAW 264.7 cells was measured using 5-(3-caroboxymethoxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. Intracelluar oxidation was analysed by DCF-DA assay. The nitric oxide (NO) production was measured by Griess reagent system. The levels of ICAM-1 and VCAM-1 expression were confirmed by western blot. And proinflammatory cytokines were measured by ELISA kit. Results : Our results indicated that fractionated extracts, especially ethyl acetate (EA) extract, significantly inhibited free radical generation, the TNF-$\alpha$-induced intracellular oxidation. Furthermore, the EA extract protected TNF-$\alpha$-induced adhesion to THP-1, expression of adhesion molecules accompanied by an attenuation of IL-6 and IL-8 formation in HUVEC. Conclusions : These results indicate that EA extract of SG have potential as an agent of atherosclerosis and other chronic inflammatory diseases including diabetes, hypertension, and arthritis.
