Phylogenetic Analysis of Pectobacterium Species Using the 16S-23S rRNA Intergenic Spacer Regions

  • For the taxonomic evaluaition, 15 strains of the genus Pectobacterium and Erwinia were analyzed for 16S-23S rDNA intergenic spacer regions (ISRs). These species contained two types of ISRs, large and small ISRs. Large ISRs were on the range of 474-569 bp size, and coding transfer $\textrm{RNA}^{11e}$($\textrm{tRNA}^{11e}$) and $\textrm{tRNA}^{Ala}$. Small ISRs were 354-459 bp in length and coding $\textrm{tRNA}^{Glu}$. The sequence variations of two ISRs among species and strains were very high as compared with 16S rRNA gene sequences. By phylogenetic trees on the basis of two ISRs, Pectobacterium ere differentiated into P. carotovorum-P. cactiaidum group and P. chrysanthemi group. However, the taxonomic position of E. cypripedii and E. rhapontici, which were not clear on taxonomic delineation between Pectobacterium and Erwinia, were not clearly resolved on the basis of ISRs.

Reliable Identification of Bacillus cereus Group Species Using Low Mass Biomarkers by MALDI-TOF MS

  • Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS)-based pathogen identification relies on the ribosomal protein spectra provided in the proprietary database. Although these mass spectra can discern various pathogens at species level, the spectra-based method still has limitations in identifying closely-related microbial species. In this study, to overcome the limits of the current MALDI-TOF MS identification method using ribosomal protein spectra, we applied MALDI-TOF MS of low-mass profiling to the identification of two genetically related Bacillus species, the food-borne pathogen Bacillus cereus, and the insect pathogen Bacillus thuringiensis. The mass spectra of small molecules from 17 type strains of two bacilli were compared to the morphological, biochemical, and genetic identification methods of pathogens. The specific mass peaks in the low-mass range (m/z 500-3,000) successfully identified various closely-related strains belonging to these two reference species. The intensity profiles of the MALDI-TOF mass spectra clearly revealed the differences between the two genetically-related species at strain level. We suggest that small molecules with low molecular weight, 714.2 and 906.5 m/z can be potential mass biomarkers used for reliable identification of B. cereus and B. thuringiensis.

어류 이동용 여수로의 자유수면 수치해석 (Numerical Modeling of Free Surface at the Fish Migration Spillways)

  • 두가지 어류이동용 여수로 설계안에 대하여 레블�V 유한요소법을 이용하여 자유수면을 포함한 흐름을 모의하였다. 댐 상류부에서 여수로 입구 방향으로 직각형 유입부를 갖는 여수로와 곡선형 유입부의 여수로에 대하여 흐름의 형상을 모의, 비교하였으며, 또한, 수치해석 결과를 여수로 바닥면의 압력분포와 자유 수면형상에 대하여 수리실험 결과를 통하여 검증하였다. 직각형 여수로가 여수로 유입부에서 자유수면 형상과 압력분포에서 다소 큰 내부 변형율을 보이는 반면, 곡선형 유입부 여수로에서는 작은 변형율을 관찰 할 수 있었다. 여수로 정상부의 대기압보다 작은 부압의 존재 형태는 두가지 형상 모두 유사함을 알 수 있었지만, 전반적으로는 곡선형 유입부 여수로가 어류 이동에 유체역학적으로 더 우호적인 환경을 제공함을 알 수 있었다.

The Roles of Two hfq Genes in the Virulence and Stress Resistance of Burkholderia glumae

  • The Hfq protein is a global small RNA chaperone that interacts with regulatory bacterial small RNAs (sRNA) and plays a role in the post-transcriptional regulation of gene expression. The roles of Hfq in the virulence and pathogenicity of several infectious bacteria have been reported. This study was conducted to elucidate the functions of two hfq genes in Burkholderia glumae, a causal agent of rice grain rot. Therefore, mutant strains of the rice-pathogenic B. glumae BGR1, targeting each of the two hfq genes, as well as the double defective mutant were constructed and tested for several phenotypic characteristics. Bacterial swarming motility, toxoflavin production, virulence in rice, siderophore production, sensitivity to $H_2O_2$, and lipase production assays were conducted to compare the mutant strains with the wild-type B. glumae BGR1 and complementation strains. The hfq1 gene showed more influence on bacterial motility and toxoflavin production than the hfq2 gene. Both genes were involved in the full virulence of B. glumae in rice plants. Other biochemical characteristics such as siderophore production and sensitivity to $H_2O_2$ induced oxidative stress were also found to be regulated by the hfq1 gene. However, lipase activity was shown to be unassociated with both tested genes. To the best of our knowledge, this is the first study to elucidate the functions of two hfq genes in B. glumae. Identification of virulence-related factors in B. glumae will facilitate the development of efficient control measures.

Molecular characteristics of Budgerigar fledgling disease polyomavirus detected from parrots in South Korea

  • Background: Budgerigar fledgling disease polyomavirus (BFDV) is the pathogen that causes budgerigar fledgling disease in psittacine species. The clinical signs of PBFV infection include ascites, hepatitis, and crop stasis. BFDV is associated with a high mortality rate in nestling birds. In contrast, adult birds only have mild symptoms such as feather dystrophy. Objectives: This study aimed to determine the prevalence, genetic characteristics, and phylogenetic analysis of BFDV in pet parrots in Korea. Methods: Fecal and tissue samples were collected from 217 pet parrots from 10 veterinary hospitals including Chungbuk National University Veterinary Hospital. The molecular screening was performed using polymerase chain reaction (PCR) analysis of the small t/large T antigen gene segment. Full-length genome sequencing with the Sanger and phylogenetic analysis were performed on BFDV-positive samples. Results: The PCR results based on the small t/large T antigen gene marker indicated that BFDV DNA was present in 10 out of 217 screened samples. A whole-genome sequence was obtained from six strains and phylogenetic analysis revealed no significant relationship existed between the species and geographical locations amongst them. Conclusions: The prevalence of BFDV infection in South Korea is not high when compared to the prevalence of BFDV in other parts of the world, however, it has been reported sporadically in various species and geographic locations. The whole-genome analysis revealed 0.2%-0.3% variation in intragenomic homogeneity among the six strains analyzed. Korean strains are separately on the phylogenetic tree from their counterparts from China and Japan which might reflect the substantial genetic variation.

사과와 고추에서 분리한 Colletotrichum gloeosporioides의 색소형성형 및 유성세대형 계통의 배양적 특징 (Cultural Characteristics of Chromogenic and Teleomorphic Strains of Collectotricum gloeosporioides Isolated from Apple and Red pepper)

  • 사과와 고추에서 분리된 Colletotrichum gloeosporioides의 색소형성형 및 유성세대형 계통균주들의 배양적 및 분생포자의 특성을 비교하였다. 유성세대 형성 계통들의 균사생장은 감자한천배지와 V-8배지에서 색소형성형계통에 비하여 빨랐다. 사과와 고추에서 분리된 색소형성형계통은 감자한천배지와 V-8배지에서 회백${\sim}$회록색의 균사가 환문을 이루면서 군데 군데에 황색${\sim}$연분홍색의 포자덩어리를 생성하였고 자낭포자세대를 형성하는 것은 없었다. 고추에서 분리된 계통은 사과와 감자한천배지에 한쪽 끝이 좁은 포자를 생성하였으나 V-8배지와 물한천잎배지의 것은 방추형이고 더 작았다. 사과에서 분리된 색소형성계통은 공시된 모든 배지에서 방추형의 분생포자를 생성하였다. 사과와 고추에서 분리된 유성세대 형성 계통들은 원통형의 포자를 형성하였는데 대부분은 양끝이 둥글었으며 감자한천배지와 V-8배지에서는 소흑점이 산재한 회백${\sim}$흑록색의 환문균층을 이루면서 자낭포자세대를 형성하였다.

지의류 추출물의 구강세균에 대한 항균효과 (Antimicrobial activities of oral bacteria by lichen extracts)

  • Objectives : By investigating antimicrobial activity of natural extracts, identifying its usefulness as antibiotic material to oral bacteria. Methods : Antimicrobial activity tests of 25 natural extracts were implemented on 6 type strains alleged to cause dental caries and 10 clinical strains isolated and identified from dental caries. Results : Among medicinal plants, Coptis japonica, Scutellaria baicalensis, Dictamnus dasycarpus showed antimicrobial activity. Among lichens, 3 methanol extracts and 6 acetone extracts showed antimicrobial activity. Minimum Inhibitory Concentration(MIC) test of Usnea aurantiacoatra KoLRI 004184, an acetone extract of lichen with the highest antimicrobial activity, on 6 type strains involved in dental caries and 10 strains isolated from dental caries resulted as follows; Actinomyces oris 1041 $12{\mu}g/ml$, for Corynebacterium durum 3151 $13{\mu}g/ml$, for Rothia dentocariosa KCTC $3204^T$ $14{\mu}g/ml$, for R. dentocariosa 911 $15{\mu}g/ml$, for R. dentocariosa 1822 $12{\mu}g/ml$, for Lacto-bacillus casei KACC $12413^T$ $12{\mu}g/ml$, for L. acidophilus KACC $12419^T$ $16{\mu}g/ml$, for L. rhamnosus 2421 $15{\mu}g/ml$, for Streptococcus mutans KCTC $3065^T$ $15{\mu}g/ml$, for S. mutans 121 $13{\mu}g/ml$, for Streptococcus oralis 2221 $18{\mu}g/ml$, for S. salivarius KCTC 5512 $14{\mu}g/ml$, for S. salivarius 122 $12{\mu}g/ml$, for S. sanguinis KCTC $3284^T$ $14{\mu}g/ml$, for S. sanguinis 912 $14{\mu}g/ml$, for Neisseria sp. KEM232 $12{\mu}g/ml$. Conclusions : Even a small amount of extract from lichen Usnea aurantiacoatra KoLRI 004184 showed very high level of antimicrobial activation against all 16 bacterial strains involved in dental caries.

Mannose-resistant Hemagglutination(MRHA) 및 Colonization Factor Antigen I(CFA I)을 표현하는 대장균의 장관내 존재와 설사증 발현과의 관계 (Gastrointestinal Carriage of Escherichia coli with Hemagglutination Activity and Colonization Factor Antigen I and its Relation to Diarrhea)

  • Colonization factor antigen I(CFA I) has been shown to be one of several virulence factors that promote attachment of enterotoxigenic E. coli(ETEC) to small intestinal epithelial cells of humans. The ability of ETEC to produce mannose-resistant hemagglutination(MRHA) of human blood group A has been used to detect CFA I. To determine gastrointestinal carriage in Korean children of E. coli with MRHA and CFA I, 116 strains of E. coli from diarrheal children admitted to Hanyang University Hospital were examined for MRHA of human erythrocytes and the presence of CFA I. Of 45 ETEC strains, 18(40%) gave a positive MRHA($MRHA^+$) and eight(18%) were positive for CFA I(CFA $I^+$). ETEC with CFA I were all heat-stable enterotoxin(ST) producers and two of these strains were of serogroups $O_{25}$. Of 17 classic enteropathogenic E. coli(EPEC), 7(41%) were $MRHA^+$ but all were negative for CFA I(CFA $I^-$). Of 30 enteroadherent E. coli(EAEC) strains, 11(37%) were $MRHA^+$ and one was CFA $I^+$. Of 24 nonpathogenic E. coli, 4(17%) were $MRHA^+$ but all were CFA $I^-$. It was shown that MRHA was common in all strains of E. coli, CFA I was limited only to ST producing ETEC and EAEC; although MRHA is a useful screening procedure, serologic tests seem to be necessary to comfirm CFA I production. CFA I was associated with a lower proportion of ETEC isolates in Korea than has been reported for other locations.

한국산 Rotifer, Brachinus plicatilis와 B. calyciflorus의 크기 및 내구란 형성 (Size and Resting Egg Formation of Korean Rotifer, Brachionus plicatilis and B. calyciflorus)

  • 어류 종묘생산시 중요한 먹이생물로 사용되는 한국산 rotifer를 순수 분리하기 위하여 염전, 기수호, 하구 및 어류 양식장을 중심으로 채집하였다. 해수산 rotifer, B. plicatilis는 13 지역에서 L-type rotifer 2 strains과 S-type rotifer 14 strains을 순수 분리하였다. 담수산 rotifer, B. calyciflorus는 4곳의 양식장에서 4 strains을 순수 분리하였다. 순수 분리된 해수산 rotifer, B. plicatilis. L-type과 S-type rotifer strain의 피갑장과 내구란 장경의 분포는 각각 $244.3\~255.3\;{\mu}m,\;142.4\~145.5\;{\mu}m$$131.0\~165.8\;{\mu}m,\;93.7\~116.4\;{\mu}m$로 나타났다. 또 담수산 rotifer, B. calyciflorus strains의 피갑장과 내구란 장경의 분포는 각각 $211.8\~229.9\;{mu}m$$126.8\~140.2\;{\mu}m$로 나타났다. B. plicatilis는 성장과 내구란 생산이 매우 다양하게 나타났다. L-type과 S-type의 rotifer의 최고 밀도는 각각 O-L이 200개체/ml였고, H-S가 753.3개체/ml로 가장 높게 나타났다. 생산된 내구란은 S-type에서는 YY-S가 86.7개/ml로 가장 높았고, L-type에서는 O-L이 45.8개/ml로 높았다.

개와 고양이에서 분리된 메티실린 내성 포도상구균의 내성인자 분포조사 (Study on the Methicillin-resistant Gene Distribution of Staphylococci Isolated from Dogs and Cats)

  • 인의학에서 메티실린 내성 포도상균주는 병원감염의 주요 원인균으로 보고되고 있지만 소동물에서 이에 대한 연구는 거의 없다. 본 연구에서는 2002년 8월부터 2003년 7월까지 개와 고양이에서 분리된 136개의 포도상구균 분리주 (coagulase 양성 87주, coagulase 음성 49주)에 대하여 항생제 감수성 검사와 이들 분리주에서 메티실린 내성 유전자인 mecA 분포상황을 조사하였다. 136개 분리주중 43주 (31.6%)가 mecA 유전자를 가지고 있었고, 유전자의 분포율은 균주에 따라 상당한 차이를 보였다. 43주의 mecA 양성균주 중 31주 (72.1%)가 oxacillin 내성을 보여 mecA 양성균주가 반드시 oxacillin 내성과 일치하는 것은 아님을 시사하였다. 그러나 mecA 양성균주일수록 oxacillin 내성율이 높았는데 S. intermedius의 71.4% (p<0.001), coagulase 음성균주의 경우 72.4%가 내성을 보였다 (p<0.001). 분리주의 94주(69%) 적어도 하나 이상의 항생제에 내성을 보였고 특히 31주(22.8%)는 4가지 이상의 항생제에 동시에 내성을 보였다. Penicillin 항생제에 내성율이 71.7%로 가장 높았다. 본 연구는 국내 소동물에서 mcA 양성균주가 존재하며, 이러한 균에 의해 유도된 감염증을 치료할 때 다제내성의 특성 때문에 항생제 선택의 폭이 매우 제한될 수 있음을 시사한다.
