Although the number of protein-coding genes is not highly variable between plant taxa, the DNA content in their genomes is highly variable, by as much as 2,056-fold from a 1C amount of 0.0648 pg to 132.5 pg. The mean 1C-value in plants is 2.4 pg, and genome size expansion/contraction is lineage-specific in plant taxonomy. Transposable element fractions in plant genomes are also variable, as low as ~3% in small genomes and as high as ~85% in large genomes, indicating that genome size is a linear function of transposable element content. Of the 2 classes of transposable elements, the dynamics of class 1 long terminal repeat (LTR) retrotransposons is a major contributor to the 1C value differences among plants. The activity of LTR retrotransposons is under the control of epigenetic suppressing mechanisms. Also, genome-purging mechanisms have been adopted to counter-balance the genome size amplification. With a wealth of information on whole-genome sequences in plant genomes, it was revealed that several genome-purging mechanisms have been employed, depending on plant taxa. Two genera, Lilium and Fritillaria, are known to have large genomes in angiosperms. There were twice times of concerted genome size evolutions in the family Liliaceae during the divergence of the current genera in Liliaceae. In addition to the LTR retrotransposons, non-LTR retrotransposons and satellite DNAs contributed to the huge genomes in the two genera by possible failure of genome counter-balancing mechanisms.
Phytophthora root rot of Chniese cabbage and spinach is reported for the first time in Korea. The diseases ocurred at Yangju, Seosan and Yeocheon in Korea from 1995 through 1998, mainly in lowland and submerged areas. Symptoms consisted of stunt, yellows, wilt and eventual death due to root rot. Fourteen isolates collected from naturally infected plants were all identified as P. drechsleri based on mycological characteristics. PCR-RFLP analysis of rDNA of the isolates confirmed the above result, since the restriction band patterns of the small subunit and internal transcribed spacers were identical to P. drechsleri and P. cryptogea, but distinct from closely related species of P. erythroseptica, P. cambivora, P. sojae and P. megasperma. The pathogen showed strong pathogenicity to Chinese cabbage, moderate to spinach, radish, cabbage and tomato, and weak or none to brown mustard, kale, chicory and pepper in pathogenicity tests.
We established an efficient plant regeneration system for Catharanthus roseus L. (G.) Don through somatic embryogenesis. Embryogenic callus was induced from hypocotyl of seed germinated in vitro. Somatic embryogenesis in Catharanthus has been categorized into three distinct stages: (1) initiation and proliferation of embryo; (2) maturation, and; (3) germination or plantlet conversion. Beside plant growth regulators, various stages of embryogenesis were screened for their response to a wide variety of factors (pH, gelrite, light, sugar alcohols, polyethyleneglycol and amino acids), which affect embryogenesis. All of the tested factors had a small to marked influence on embryogeny and eventual conversion to plantlets. The plantlets were acclimatized successfully in a greenhouse. To our knowledge, this is the first report describing a detailed study of various cultural factors which regulate embryogenesis in C. roseus. The results discussed in this paper may be used in mass propagation to produce medicinal raw material, and the embryo precursor cells could be used in genetic modification programmes that aim to improve the alkaloid yield as well.
Germplasm characterization and evolutionary process in viable populations are important links between the conservation and utilization of plant genetic resources. Here, an investigation is made, based on molecular and biochemical techniques for assessing and exploiting the genetic variability in germplasm characterization of taro, which would be useful in plant breeding and ex situ conservation of taro plant genetic resources. Geographical differentiation and phylogenetic relationships of Indian taro, Colocasia esculenta (L.) Schott, were analyzed by random amplified polymorphic DNA (RAPD) and isozyme of seven enzyme systems with specific reference to the Muktakeshi accession, which has been to be proved resistant to taro leaf blight caused by P. colocasiae. The significant differentiations in Indian taro cultivars were clearly demonstrated by RAPD and isozyme analysis. RAPD markers showed higher values for genetic differentiation among taro cultivars and lower coefficient of variation than those obtained from isozymes. Genetic differentiation was evident in the taro accessions collected from different regions of India. It appears that when taro cultivation was introduced to a new area, only a small fraction of genetic variability in heterogeneous taro populations was transferred, possibly causing random differentiation among locally adapted taro populations. The selected primers will be useful for future genetic analysis and provide taro breeders with a genetic basis for selection of parents for crop improvement. Polymorphic markers identified in the DNA fingerprinting study will be useful for screening a segregating population, which is being generated in our laboratory aimed at developing a taro genetic linkage map.
In this paper, we develop the image transfer remote maintenance monitoring system for hand held device which can compensate defects of human mistake. The human mistakes always happen when the worker communicate information each other to check and maintenance the equipment of the power plant under bad circumstance such as small place and long distance in power plant. A worker couldn't converse with other when in noisy place like Power plant. So, we make some hand device for handy size and able to converse in noisy place. The developed system can have improvement of productivity through increasing plant operation time. And developed system is composed of advanced H/W(hard ware) system and S/W(soft ware)system. The H/W system consist of media server unit, communication equipment with hand held device, portable camera, mike and head set. The advanced s/w system consist of data base system, client pc(personal computer) real time monitoring system which has server GUI(graphic user interface) program, wireless monitoring program and wire ethernet communication program. The client GUI program is composed of total solution program as pc camera program, and phonetic conversation program etc.. We analyzed the required items and investigated applicable part in the image transfer remote maintenance monitoring system with hand held device. Also we investigated linkage of communication protocol for developed prototype, developed software tool of two-way communication and realtime recording skill of voice with image. We confirmed the efficiency by the field test in preventive maintenance of plant power.
This study is to analyse the feasibility for providing Co-generation plant in Apartment Complex for 4 typical Apartment Complexes located Seoul metropolitan area, The selected complexes are three midium-large size Apartment[nearby 35pyoug of floor area] and one complex of small size Apartment[below 25 pyoung of floor area] for comparison. The necessary data for the study were collected with visitation of each site. The study showed very positive result for the three medium-large size Apartment Complexes of which the average floor area is more than 25 pyoungs, while negative result for the Complex of which average floor area is less than 25 pyoungs. Other than floor size it was found that the electric consumption density also influence the economic feasibility. In study the unit fixed cost of the energy produced from Co-generation plant is one third of the unit variable cost[fuel cost] and it seems better to select high thermal efficiency machine for Co-generation plant even with some higher cost of the plant.
Acrobeles ciliatus (von Linstow, 1877) and Plectus parietinus (Bastian, 1865) are newly collected from Korea. Acrobeles is similar to those nematode belongs to Rhabditidae and Panagrolaimidae by having terminal bulb without medium bulb but different by having complex, vines shaped labial attachment. A. ciliatus differed from other Acrobeles species by lateral fields with two incisures, primary axils U-shaped, and secondary axils V-shaped. Head region set off with the neck, three high labial probolae present, each having 5 tines at inner margins and 5-7 at outer margins, and two elongate apical tines. Also, it differs from the longer female body. Plectus parietinus is distinguished from all other species of Plectus by the prominent hypodermal glands, the relatively small amphid, the distinct and well set-off lips and by the subdorsal distal caudal seta on the left side of the tail. The cooperation with other work types and designing procedure in plant industry and engineering showed clear differences from general construction industries. Current problems are that design's consistency is reduced because of the false system of interface and it ultimately causes frequency changes of design and delayed submission of final book. To solve the problems, the improvements of designing procedure were studied by focusing on the field of architectural designing and the range was limited from power plant construction industry to engineering designing field. If combining GA with the field of architectural designing and receiving data previously, it's possible to conduct expectable designing on small changes in the future, so it's expected to improve greatly cooperation with interface among the work types of power plant engineering received previously from the aspect of consistency. For the application of designing tool, a checklist, interim check will be conducted if progressed by list's division in the designing procedure, not book's completing period. In the interface among the designing work types, direct confirmation and revision must be conducted. In case of 3D modeling, the reflection of input data must be conducted from the basic designing so as to solve interferences intensively.
A full-length cDNA encoding dihydroflavonol 4-reductase (st-dfr) of potato was isolated by rapid amplification of cDNA ends, and their expression was investigated from purple-fleshed potato (Solanum tuberosum L. cv. Jashim). The st-dfr exists as a member of a small gene family and its transcripts was abundant in the order of tuber flesh, stem, leaf, and root. The expressions of st-dfr gene were light inducible and cultivar dependant. Transgenic potato plants harboring antisense st-dfr (AS-DFR) sequences were analyzed. The accumulation of mRNA was nearly completely inhibited as a result of introducing an AS-DFR gene under the control of the 35S CaMV promoter into the red tuber skin Solanum tuberosum L. cv. Desiree. The anthocyanin content of the tuber peels of the transgenic lines was dramatically decreased by up to 70%. The possible production of flavonols in the peels of AS-DFR transgenic potatoes was discussed.
This study was conducted to investigate the relationship between yearly variations of climatic elements and yearly variations of productivity in perilla. In addition, correlation coefficients among yield and yield components were estimated. The data of yield and yield components were investigated for 10 years from 1991 to 2000. The meteorological data gathered at the Yeosu Weather Station for the same period were used to find out the relationships between climatic elements and productivity. Yearly variation of the amount of precipitation in September was large with coefficients of variation(c. v.) of 11.1%, but the coefficient of variance(c. v.) in July and August were relative small with 1.8, 2.1%, respectively. Number of cluster per hill and weight of 1,000 grains were greatly with c. v. of 76.1, 79.3%, respectively, but the coefficients of variance(c. v.) of plant height and seed yield were more less with 9.58, 10.60%, respectively. Correlation coefficients between precipitation of September and seed yield were positively significant correlation at the level of 5.1%, respectively, but the duration of sunshine in September and seed yield were negatively significant at the level of 5.1%, respectively. Correlation coefficients of these, the plant height, number of branches per plant, cluster length, number of cluster per hill, weight of 1,000 grains and seed yield were positively significant at the level of 5.1% respectively.
