Development of a Multiplex Polymerase Chain Reaction Assay for Detecting Five Previously Unreported Papaya Viruses for Quarantine Purposes in Korea

  • There are concerns about the introduction and spread of plant pests and pathogens with globalization and climate change. As commercial control agents have not been developed for plant viruses, it is important to prevent virus spread. In this study, we developed a multiplex polymerase chain reaction (PCR) detection method to rapidly diagnose and control three DNA (papaya golden mosaic virus, Lindernia anagallis yellow vein virus, and melon chlorotic leaf curl virus) and two RNA (papaya leaf distortion mosaic virus and lettuce chlorosis virus) viruses that infect papaya. Specific primer sets were designed for the virus coat protein. Performing PCR, clear bands were observed with no non-specific reaction. Our multiplex PCR method can simultaneously detect small amounts of DNA/RNA to diagnose five viruses infecting papaya and prevent the spread of the virus.

  • Occurrence of white scab of tea (CAmellia sinensis) showing numerous, small, circular, reddish or yellowish brown spots on young tea leaves was observed at a tea plantation of Boseung, Chonnam, from May to June in 1992-1994. At the late growth stages, the center of the spot became light gray. The causal fungus of the white scab was identified as Sphaceloma theae Kurosawa. Symptoms were produced 5∼6 days after inoculation by the artificial inoculation of s. theae conidia, which was similar to those on naturally infected leaves.

Bacterial Soft Rot of Radishby Erwinia chrysanthemi (Erwinia chrysanthemi에 의한 무 세균성 무름병)

  • Bacterial soft rot was observed on radish grown in Hongcheon, Kanwon-Do, Korea. The soft rot symptoms began as small water-soaked lesions. The water-soaked lesions enlarged rapidly in roots and produced a foul odor. When roots were affected in the field, the shoots also became infected and watery, causing infected plants to wilt, disorganize, and die. The causal organism was isolated from the lesions, and the identified as Erwinia chrysanthemi based on the morphological, physiological and biochemical characteristics. E. chrysanthemi is first described bacterium which causes bacterial soft rot on radish in Korea.

  • Black leg occurred in potato (Solanum tuberosum L.) grown in Pyungchang, Kangwon-Do, Korea. The symptoms began as small water-soaked lesions on stem, and the affected stems discolored black to brown. It became yellow under dry condition. When lower parts of potato were affected in the field, wilting of leaves and desiccation of the stem were developed. The causal organism was isolated from lesions and identified as Erwinia carotovora subsp. atroseptica based on the morphological, physiological and biochemical characteristics. E. carotovora subsp. atroseptica is the first described bacterium which causes black leg in potato in Korea.

  • In this paper, we design the H$\infty$ optimal controller satisfying robust stability and performance in spite of the plant uncertainty for an electro-mechanical actuator system and analyze the controller in frequency domain. H$\infty$ optimal controller K was designed using iteration algorithm suggested by DOYLE. Using the controller in an electro-mechanical actuator system, the joint with very small coupling rigidity coefficient was used to vary the control parameter. The plant unstructured uncertainty was assumed to be a multiplicative type.

  • Four sterigmata of papillaform E. violaceobrunneum are developed from crator-shaped basidium. The apex of sterigma is swollen to a small globeform. And then it is swollen to a clubform. The clubform is again swollen to a ellipticalform, and then more than six spots of spore surfaces are ramdomly depressed with hilum axes. When the depression of surface of a elliptical spore is over, the spore is a heterodiametrical spore of multi-angular.

  • The results of the relationship between standing crop of Miscanthus sinensis grassland and soil morphological characteristics are as follows. The Miscanthus sinensis grassland seems to grow well in volcainc ash soil. The depth of A layer was closely related to the standing crop of the Miscanthus sinensis grassland. The root systems of Miscanthus sinensis reached to its maximum in A layer. The root systems of Miscanthus sinensis showed its maximum at 20mm and below(soil hardness). The soil texture of A layer showed SL-SiL. The soil structure of A layer contained Massive-Small Granular. The soil colors of A layer expressed Dark Yellowish Orange-Brownish Black.

Root Nodule Specific Proteins of Alnus hirsuta (물오리나무(Alnus hirsuta)의 뿌리혹 특이 단백질)

  • Root nodule specific proteins of Alnus hirsuta were examined. SDS-PAGE pattern of the Alnus root nodule was simpler than that of soybean, showing five nodule specific proteins whose molecular weights were 48, 40, 36, 26 and 19 kD, respectively. Among them, 48 kD protein existed most abundantly and were composed of two subunits whose pI value were 4.0 and 4.3, respectively. The 48 kD protein seemed to be a heme containing protein based on reaction with diaminobenzidine. Although 19 kD protein was present in small amount, it was most similar to leghemoglobin in terms of its molecular weight.
