Recently, single cell gel electrophoresis, also known as comet assay, is widely used for the detection and measurement of DNA strand breaks in vitro and in vivo in many toxicological fields such as radiation exposure, human monitoring and toxicity evaluation. As well defined, comet assay is a sensitive, rapid and visual method for the detection of DNA strand breaks in individual cells. Briefly, a small number of damaged cells suspended in a thin agarose gel on a microscope slide were lysed, unwinded, electrophoresed, and stained with a fluorescent DNA binding dye. The electric current pulled the charged DNA from the nucleus such that relaxed and broken DNA fragments migrated further. The resulting images which were subsequently named for their appearance as comets, were measured to determine the extent of DNA damages. However, some variations could be occurred in procedures, laboratories's conditions and kind of cells used. Hence, to overcome and to harmonize these matters in comet assay, International Workshop on Genotoxicity Test Procedure (IWGTP) was held with several topics including comet assay at Washington D.C. on March, 1999. In spite of some consensus in procedures and conditions in IWGTP, there are some problems still remained to be solved. In this respect, we attempted to set the practical optimal conditions in the experimental procedures such as lysis, unwinding, electrophoresis and neutralization conditions and so on. First of all, we determined optimal lysis and unwinding time by using 150 $\mu$M methyl methanesulfonate (MMS) which is usually used concentration. And then, we determined optimal positive control concentrations of benzo(a)pyrene (BaP) and MMS in the presence and absence of S9 metabolic activation system, respectively.

This study develops a seat with electric motor technology for a one-ton grade commercial vehicle. While applying electric motor technology, the FMVSS 210 seat frame strength test is also conducted to examine the product's weak parts. The seat frame strength test used the FMVSS 210 test standard and the ANSYS program was used to simulate the test and identify weak parts in the deformation and strain values. The test results showed that the cushion frame and slide rail connection bracket were fractured at loads of about 10,000 N. Similarly, the maximum stress and strain values in the bracket were obtained in the simulation results. On this basis, it was evaluated that the connection part bracket was a considerably weak part in the case of the first model, and changing the shape of the bracket and reinforcing the strength were required. In addition, the seat belt anchorage test results and simulation results were compared to assure their validity. In the comparison results, the error for each is about 5-10%. Therefore, the simulation performed in this study is considered to have produced reasonably accurate results.
Purpose: In order to ensure that vegetable seedlings (with a soil block around their roots) are planted in an upright orientation after metering in a vegetable transplanter, they need to be dropped freely from a certain height. The walk-behind hand-tractor-powered machines do not have sufficient space to drop the seedlings from that height. In the present work, a hopper-type planting device was developed for the walk-behind hand-tractor-powered vegetable transplanter to ensure that the soil block seedlings are planted in an upright orientation. Methods: Various dimensionless terms were developed based on the dimensional analysis approach, and their effect on the planting of soil block seedlings in an upright orientation (planting efficiency) was studied. The optimum design dimensions of the hopper-type planting device were identified by the Taguchi method of optimization. Results: The ratio of the height of free fall to the sliding distance of the seedling on the surface of the hopper had the highest influence on planting efficiency. The planting efficiency was highest for plants with a height $15{\pm}2cm$. The plant handling Froude number, in interaction with the design of the hopper-type planting device, also significantly affected the planting efficiency. Of the hopper design factors, the length of the slide of the seedlings on the surface of the hopper was most important, and induced sufficient velocity and rotation to cause the seedling to fall in an upright orientation. An evaluation of the performance of the planting device under actual field conditions revealed that the planting efficiency of the developed planting device was more than 97.5%. Conclusions: As the seedlings were fed to the metering device manually, an increase in planting rate increased missed plantings. The planting device can be adopted for any vegetable transplanter in which the seedlings are allowed to drop freely from the metering device.
We compared the diagnostic accuracy of liquid-based cervicovaginal cytology using $MonoPrep2^{TM}$ system (Monogen, Herndon, Virginia, USA), a manual system based on membrane filtration method, with conventional Pap smear. Study population included 92 patients visiting the gynecologic department under the suspicion of uterine cervical disease. In thirty of them, surgical biopsy was performed. $MonoPrep2^{TM}$ system provided well-preserved monolayer specimen with good nuclear morphology. However, about 19% of specimens were inadequate to interpret due to low cellularity. The detection rate of abnormal cells more than ASCUS (atypical squamous cells of unknown significance) was 23.9% and higher than 19.4 % of conventional Pap smear. Diagnostic concordance rate with conventional Pap smear was 81%, and severe discordance rate influencing on the management of patient was 7.6 %. Among these seven cases, $MonoPrep2^{TM}$ system was more diagnostic only in four. In comparison with histology, the sensitivity of diagnosis of $MonoPrep2^{TM}$ system was 78.9% and slightly higher than 73.5% of conventional Pap smear. However, the specificity was 81.1% and lower than 90.9% of Pap smear. In conclusion, $MonoPrep2^{TM}$ system provided diagnostic accuracies similar to the conventional Pap smear. The inexpertness of slide preparation and the low cellularity were considered to endow a limitation in more accurate evaluation.
Surface energy is the principal factor of anti-condensation. High surface energy appears hydrophilic itself and low surface energy represents hydrophobic itself. The contact angle is widely being used for measurement of surface energy of materials, evaluation of coating performances, measurement of wettability, and so on. However, the existing contact angle measuring system is so expensive for purchasing and complicated, so it takes a lot of time and money to use. This study was conducted to develop the algorithm for evaluating hydrophilic performance through measuring the contact angle of water droplet automatically, and fabricate relatively simple measuring system using a low-cost monochrome camera and image processing. A constant amount of water was firstly allocated on a slide by a micropipette, and then the image of water droplet was captured by monochrome digital camera and sent to a computer. The image was binarized and then reduced noises by labeling. Finally, the contact angle of water droplet was computed by using three points (left, right, and top coordinates), simple linear mathematics, and trigonometric function. The experimental results demonstrated the accuracy and reproducibility of the developed system showing less deviations and deviation ratio.
The cytology of sputum specimens, at a medical facility in Korea, is using the liquid-based cytology and conventional smear method. It confirms the diagnosis using conventional smear methods and the liquid-based cytology. In addition, the diagnosis uses two kinds of each method, compares the suitability, sensitivity and specificity for each test and tries to understand the efficient method needed. It divided sputum specimens in half and liquid-based cytology. A conventional smear method was conducted and the Papanicolaou's staining was conducted using Autostainer. Diagnosed each slide of staining produced smear slides specimens by using an optical microscope. The result of the liquid-based cytology and conventional smear method was that the liquid-based cytology was of a higher-grade than the conventional smear method. It was 36.8%, and 62.8% in the same condition. But only one case was of a lower grade. Liquid-based cytology showed 86.2% of adequacy, 31.0% of sensitivity and 97.5% of specificity. The conventional smear method showed 54.4% of adequacy, 19.6% of sensitivity and 100% of specificity. If it was conducted with two methods at the same time, the results would be similar to the liquid-based cytology. In conclusion, liquid-based cytology has a more superior adequacy and sensitivity than the conventional smear method. The combination of the two methods is similar to the result of only conducting a liquid-based cytology. Conducting a liquid-based cytology rather than the combination of the two ways of sputum cytoscopy, is to be considered as an efficient method to achieve diagnostic accuracy, reduce labor of clinical technologist and pathologist, and to reduce the expense of patients.
To clarify whether inhibitory effect of estrogen on liver tumor is associated with cell proliferation, we investigated its role in diethylnitrosamine (DEN)-induced rat preneoplastic lesions, with time sequenced manners. F344 male rats (n = 90) were divided into three groups at 5 weeks of age. The mini-osmotic pumps providing a continuous infusion of DEN was implanted into the abdominal cavity of each animal in group 1, 2 and 3 at 6 weeks of age. To see the effect of estrogen, pellet containing 1 or 10 ${\mu}g$ of estradiol-3-benzoate (EB) was implanted subcutaneously in the animals of groups 2 or 3, respectively, one week prior to DEN treatment. Ten animals of each group were euthanized at 10, 14 and 18 weeks after DEN treatment. Liver tissues at each time point were fixed in 10% phosphate-buffered formalin and were processed and embedded in paraffin and 5 ${\mu}g$ sections mounted on a silanized slide. Glutathione S-transferase placental form (GST-P) positive foci and 5-bromo-2-deoxyuridine (BrdU) labeling cells were detected at each time point. Area of GST-P positive foci in DEN+EB 1 or 10 ${\mu}g$ group was significantly decreased compared to DEN alone at 14 weeks (p < 0.01 or p < 0.05, respectively) an at 18 weeks (p < 0.05 or p < 0.01, respectively). BrdU index in DEN+EB 1 or 10 ${\mu}g$ groups was significantly decreased compared to DEN alone at 14 weeks and at 18 weeks (p < 0.01). Taken together, we conclude that EB treatment decrease the DEN-induced liver preneoplastic lesions and this may be associated with decrease of cellular proliferation.

The purpose of this paper is to provide the statistical models for landslide prediction through quantification and AHP methods. Quantification method is a statistical method of providing quantity to qualitative variables by analyzing the observed data. In this paper, we suggest the quantification process based on the results of cannonical correlation analysis. In contrast with the quantification method which is based on given data the AHP(Analytic Hierarchy Process) technique is a kind of method based on questionaire data which is usually taken from professionals. We analyze both the real data(provided from KIGAM) and questionaire data collected from professionals of various related area. We developed two kinds of evaluation table which provide the scores of land slide possibility and the logistic model providing the probability of occurring landslide. Finally we compare the performance and evaluate the stability of the suggested two models.
Previous artificial latent fingerprint solution has shown unsatisfactory results. Therefore, in this study, we developed an artificial latent fingerprint solution close to the actual fingerprint composition by improving the lipid composition. We mixed lipid solution with amino acid solution at v/v ratios as follows: 2:3, 1:4, 1:5, 1:8, 1:10, 1:20. We then dropped the same amount of each proportion of artificial latent fingerprint solution on porous paper and non-porous slide glass. Subsequently, each sample was treated with Oil red O, Cyanoacrylate fuming and Basic yellow 40 staining. As the concentration of lipids decreased, the output also decreased. Both types of surfaces and all concentrations were visually confirmed very well. In addition, the reactivity to lipids was significantly higher compared to the previous artificial latent fingerprint solution. Furthermore, for the quantitative evaluation, it is necessary to conduct additional research on the printing of the artificial latent fingerprint solution.
This study carried out evaluation of vulnerability in accessability and functionality using road network that was extracted from Intelligent Transportation System(ITS) and digital map. It was built in order to figure out accessability that locational data which include community center, public facilities, medical facilities and highway IC. The method for grasping functionality are Digital Elevation Model(DEM) and land slide hazard map provided by Korea Forest Service. The evaluation criteria for figure out accessability was set to related comparison of average time in urban area. Functionality value was calculated by the possibility of backing the vehicle possibility of snowfall and landslides. At last, this research computed weighting value through Analytic Hierarchy Process (AHP), calculated a vulnerable score. As the result, the accessability of rural village came out that would spend more time by 1.4 to 3.2 times in comparison with urban area. Even though, vulnerability of the road by a snowfall was estimated that more than 50% satisfies the first class, however, it show up that the road were still vulnerable due snowing because over the 14% of the road being evaluated the fifth class. The functionality has been satisfied most of the road, however, It was vulnerable around Lake Daechung and Piban-ryung, Yumti-jae, Suriti-jae where on the way Boeun. Also, the fifth class road are about 35 km away from the city hall on distance, take an hour to an hour and a half. The fourth class road are about 25 km away from the city hall on distance, take 25 min to an hour. The other class of the road take in 30 min from the city hall or aren't affected of weather and have been analyzed that a density of road is high. In A result that compare between distribution and a housing density came out different the southern and the eastern area, so this result could be suggested quantitative data for possibility of development.
