This study was performed to determine the optimal dose of ozone for wound healing. Twenty Korean black goats were allocated to 4 groups. The skin wound with $1{\times}1cm$ was induced over the hoof. For 5 minutes per day, 0.1 ppm (Group I), 1 ppm (Group II) and 6 ppm (Group III) ozone gas was applied. In control group, 5 ml of normal saline was applied daily on the wound. Wound square was determined using color image analyzer at day 0, 4 and 14. Soft tissue depth of wound was determined using radiograph. Before the application of ozone gas, $1{\times}1cm$ of skin tissue was resected and observed for histopathologic findings. To determine the healing effect, $2{\times}1cm$ of wound tissue was resected and observed for histopathologic findings at day 14. In mean squares of wound, all experimental groups revealed more reduced mean squares than control group at day 4, and especially Group II (p<0.05) and Group III (p<0.05) revealed significant reduction. At day 14, Group I and Group II (p<0.05) revealed more reduced mean squares than control group, but Group III revealed less reduced mean squares than control group. In soft tissue depth of wound, all experimental groups and control group revealed swelling at day 4, compared with day 0. At day 14, all experimental groups and control group revealed reduction, compared with day 4, and especially Group II (p<0.05) revealed significant reduction. In histopathological findings, inflammation findings in epidermis and dermis, hemorrhagic finding in epidermis were revealed in control group. In Group I, inflammation finding was reduced, compared with control group. Formation of scab and many reepithelialization of epidermis were revealed. In Group II, inflammation findings in epidermis and dermis which were observed in control group were not found, normal skin tissue was revealed. In Group III, inflammation findings in epidermis and dermis were revealed, reepithelialization of epidermis was not founded. Slow healing process was observed, compared with Group I and II. It was concluded that Group II has excellent promotion effect of wound healing.
Purpose: Many topical agents had been used for contaminated wound treatment. Although antimicrobial ointments were widely used as topical agents, their comparative antibacterial and wound healing effects were largely unreported. The purpose of this study was to compare antibacterial effects and wound healing effects of common topical ointments on infected full thickness skin defect in the mouse. Methods: One full thickness skin defects in the mice (n=60) were developed on the back and left open for twenty-four hours. Sixty mice were divided into four groups: group B (dressing with Bactroban$^{(R)}$, n=15), group I (dressing with Iodosorb$^{(R)}$, n=15), group T (dressing with Terramycin$^{(R)}$, n=15), group G (control group, dressing with dry gauze, n=15). The size of wound defects and the grades of wound healing were evaluated at 4, 7, 10 days, and antibacterial effect was evaluated with restricted zone in Mueller Hinton agar by disk diffusion method. After the wound was left open for twenty-four hours, many Staphylococcus aureus was cultured. The wound defect size was decreased in order of Bactroban$^{(R)}$ (B), Iodosorb$^{(R)}$ (I), Terramycin$^{(R)}$ and gauze dressing group in all days, but difference among experimental groups was not statistically significant. The grade score of wound healing was increased in order of Bactroban$^{(R)}$, Iodosorb$^{(R)}$, Terramycin$^{(R)}$ and gauze dressing group, and the difference was statistically significant. Antibacterial effect for was increased in order of Bactroban$^{(R)}$, Iodosorb$^{(R)}$, Terramycin$^{(R)}$ and gauze dressing group, and the difference was statistically significant. Conclusion: Topical antimicrobial ointments application was effective against wound infections by S. aureus. Bactroban$^{(R)}$ may be an optimal topical treatment for infected wounds according to this study. However, further study is necessary to evaluate the clinical efficacy of antimicrobial ointments and to search for the mechanisms that explain their effects.
Background Acellular dermal matrix (ADM) helps wound healing by stimulating angiogenesis, acting as a chemoattractant for endothelial cells, providing growth factors, and permitting a substrate for fibroblasts to attach. The current standard for using paste-type ADM (CG Paste) in wound healing is direct application over the wounds. The major concerns regarding this method are unpredictable separation from the wounds and absorption into negative-pressure wound therapy devices. This study aimed to investigate the effects of subcutaneous injection of paste-type ADM on wound healing in rats. Methods Full-thickness skin defects were created on the dorsal skin of rats. Eighteen rats were randomly divided into three groups and treated using different wound coverage methods: group A, with a saline dressing; group B, standard application of CG Paste; and group C, injection of CG Paste. On postoperative days 3, 5, 7, 10, and 14, the wound areas were analyzed morphologically. Histological and immunohistochemical tissue analyses were performed on postoperative days 3 and 7. Results Groups B and C had significantly less raw surface than group A on postoperative days 10 and 14. Collagen fiber deposition and microvessel density were significantly higher in group C than in groups A and B on postoperative days 3 and 7. Conclusions This study showed comparable effectiveness between subcutaneous injection and the conventional dressing method of paste-type ADM. Moreover, the injection of CG Paste led to improved wound healing quality through the accumulation of collagen fibers and an increase in microvessel density.

Herbal medicines are popular in Korea communities and used for a variety of diseases, and most herbal medicines has been used an oral administration for treating diseases, but some herbal medicines included Jawoongo has been used by applying on the skin. Jawoongo, a traditional oriental medicine ointment, have been used for treating various dermatitis associated with skin diseases such as tinea manuum, eczema and chilblain, etc. by promoting blood flow(活血), dispersing wind(散風) and moistening(潤燥). Jawoongo used in this study is derived from Yungigo supplemented with Adeps Suillus(豚脂). The aim of this study is to investigate the effect of Jawoongo plus Rehmanniae Radix(生地黃) on the healing of incisional skin wound in rats. After inducing an inflammation on the rats, we observed the amount of Leukocyte, the level of Cortisol and CRP(C-reactive protein), the histological parts and have obtained the healing effect of wound by using Jawoongo plus Rehmanniae Radix(生地黃).
Centella asiatica is an herbal plant used on different continents by diverse ancient cultures and tribal groups. Historically. the extract has been used as a wound healing agent, The extract has three different triterpenoid ingredients: asiaticoside. asiatic acid, and madecassic acid. It has been reported that its wound healing activity is associated with the modulation of collagen synthesis in the skin dermis. The wound healing property of the extract has led to its commercial introduction under the trade name, Madecrlssol. As part of our program toward the development of new wound healing agents. structere activity relationship (SAR) studies have been performed by modifying asiatic acid. In this communication. the SAR study of esiatic acid for the development of an efficient woind healing agent is reported.
Burns are one of the most devastating forms of trauma and wound healing is a complex and multicellular process, which is executed and regulated by signaling networks involving numerous growth factors, cytokines, and chemokines. Recombinant human granulocyte macrophage colony-stimulating factor (rhGM-CSF) was specifically produced from rice cell culture through use of a recombinant technique in our laboratory. The effect of rhGM-CSF on promotion of deep second-degree burn wound healing on the back skin of a hamster model was evaluated through a randomized and double-blind trial. As macroscopic results, hamster skins of the experimental groups showed earlier recovery by new epidermis than the control groups. Immunohistochemical reactions of proliferating cell nuclear antigen and transforming growth factor-b1, which are indicators of cell proliferation, were more active in the experimental group, compared with the control group. On electron microscopy, basal cells in the epidermis of the experimental group showed oval nuclei, prominent nucleoli, numerous mitochondria and abundant free ribosomes. In addition, fibroblasts contained well-developed rough endoplasmic reticulum with dilated cisternae. Bundles of collagen fibrils filled the extracellular spaces. Particularly, ultrastructural features indicating active metabolism for regeneration of injured skin at 15 days after burn injury, including abundant euchromatin, plentiful free ribosomes, and numerous mitochondria, were observed. These findings suggest that use of rhGM-CSF could result in accelerated deep second-degree burn wound healing in animal models.
In order to study the effects of the low power density laser (AsGa) on the wound healing of tongue and skin, thirty two healthy rats were anesthetized with pentothal. The tongue wound, approximately 1 mm in depth and 2mm in diameter, was created on the anterior of the tongue and the skin wound, approximately, 2mm in depth and 6-8mm in diameter, was created on the back of rat with the tip of small rongeur forcep. Wounds of experimental groups were irradiated with AsGa laser (Stomalaser, SEDATELEC Co., France) every other day by ninth day. The areas of wounds were measured at 1, 3, 5, 7, 9 days after wounding and the specimens were sectioned, stained, and observed with light microscope. The results were as follows : 1. In the AsGa laser irradiated wound of tongue, the epithelial and fibroblastic regeneration were accelerated when compared with controls. 2. In the AsGa laser irradiated wound of skin, the epithelial and fibroblastic regeneration were accelerated when compared with controls. 3. When the wound areas were compared, there was a significant difference between control group and experimental group (P < 0.01: 2-Way NOVA).

Staphylococcus epidermidis is well-known not only as an innocuous normal flora species commonly isolated from human skin, but also as an important bacterial species to keep skin healthy, because this species can protect the human skin from pathogenic microorganisms. However, S. epidermidis turns into a potential pathogen in damaged skin, because these bacteria can easily form a biofilm on the wound area and provide antimicrobial resistance to other microorganisms embedded in the biofilm. Thus, it is important to kill S. epidermidis in the early stage of wound treatment and block the formation of biofilms in advance. In the present study, hydrogel wound dressings were fabricated using polyvinyl alcohol/polyethylene glycol containing silver citrate nanorods, which have been proven to have strong antimicrobial activity, especially against S. epidermidis, and their wound-healing efficacy was investigated in vivo using a rat experiment.

Aim: The aim of this study was to investigate the mechanism of promoted skin wound healing in skin defects with primary cultured oral mucosal keratinocytes. Materials and methods: Thirty adult female nude mice weighing $20{\pm}2g$ were used for the experiment. Primary cultured and suspended oral mucosal keratinocytes, labeled with BrdU, were scattered onto $1.5cm{\times}1.5cm$ sized full thickness skin defects in the experimental group(N=15), and no grafts were placed the control group(N=15). They were sacrificed at 3 days, 1 week and 2 weeks after the treatment respectively. Histological examination of each wounds were performed to review the healing progress on measuring the length from the wound margin to regenerating epithelial front. The role of keratinocytes were assessed by double immunohistochemical staining with Anti-BrdU and Anti-cytokeratin AE1/3. Results: In the experimental group the wound was completely covered with regenerating epithelia in 2 weeks, but partially regenerated in the control group. The immunohistochemical studies unexpectedly reveal that most of regenerating epithelial cells were induced from marginal epithelium of the margin, not from the scattered keratinocytes. Conclusion: We could successfully confirm that graft of primary cultured oral mucosal keratinocytes promotes the regeneration of skin defects.
Purpose : To compare the skin damage recovery efficacy of natural Angelicae Gigantis Radix extract (N-AGR) and cultivated A. Gigantis Radix extract (C-AGR). Through this, we confirmed whether the quality standards of herbal medicines recorded in the classic books make a difference in the experimental efficacy using epithelial cells. Methods : The quality standards of medicinal herbs in the classic books and the cultivation and processing conditions of two types of A. gigas were compared. After inducing oxidative stress with H2O2, cytoprotective property of N-AGR and C-AGR were evaluated through cell viability. Additionally, after wound formation of epithelial JB6 cells, N-AGR and C-AGR were treated to evaluate wound healing efficacy. Result : The natural A, gigas met the excellent quality standards of the classic books. N-AGR inhibited cell death by oxidative stress induced by H2O2, and was superior to C-AGR. Both N-AGR and C-AGR showed dose-dependent wound healing efficacy, but N-AGR was significantly superior to C-AGR. Conclusions : Through the oxidative stress of skin and skin wound healing efficacy experiments using epithelial cells, natural A. gigas showed superior efficacy compared to cultivated A. gigas.
