Iontophoretic Transdermal Delivery of Alendronate in Hairless Mouse Skin (In-vitro에서 전기이온영동법을 이용한 알렌드로네이트의 경피약물전달)

  • This study examined the transdermal delivery of alendronate across hairless mouse skin. The effects of iontophoresis, perforation with a microneedle, and a combination of a microneedle pretreatment and iontophoresis were evaluated in vitro test. Hydrogel patches were polymerized by UN polymerization to supply a hydrogel patch to the iontophoretic transdermal drug delivery system. The alendronate content in the iontophoretic delivery patch was $5.0\;mg/cm^3$. The amounts of alendronate that permeated across the hairless mouse skin when current densities of 0.25 and $0.50\;mA/cm^2$ were supplied to the iontophoretic alendronate patch were $0.80{\pm}0.03$ and $2.00{\pm}0.02{\mu}g$, respectively. After pretreatment with a microneedle, the amounts of alendronate that permeated across the hairless mouse skin increased to $70.65{\pm}0.37$ and $162.23{\pm}0.40{\mu}g$, respectively. The biocompatibility of the iontophoretic alendronate patch was examined according to the international standardization organization 10993.

Characteristics and Hair Growth Efficacy of Water Dissolving Micro-needles Containing Minoxidil Regarding Length of Micro-needles (미녹시딜을 내포한 용융형 고분자 마이크로 니들의 길이에 따른 특성 및 발모 효능 관찰)

  • Two types of polymeric micro-needles with 250 and $750{\mu}m$ lengths were prepared by a micromolding process. The geometry of dissolving micro-needles with minoxidil was investigated and the mechanical property of polymer micro-needles was also evaluated. Water dissolving micro-needles containing minoxidil were successfully inserted into skin and delivered minoxidil to the skin layer after 30 min of insertion. The observed hair growth was less when the minoxidil treatment was not applied or used without the micro-needles than that the minoxidil was applied by micro-needles. Histologically, these hair follicles became bigger, and the hair papillae were totally encircled by the hair bulbs. In this feasibility test, we show the successful delivery of minoxidil, and effective treatment of hair loss can be possible by using dissolving micro-needles with minoxidil.

Cosmeceutical Properties of Fructan (Levan) Produced by Zymomonas mobilis

  • Fructan, a polysaccharide existing in plants or produced by microorganisms, is a sugar polymer of fructose with $\beta$-2,6 linkages. In this study, we investigated some cosmeceutical properties of Fructan such as moisturizing effect, cell proliferation effect, anti-inflammation effect and cell cytotoxicity. Zymomonas mobilis, a microorganism producing Fructan, was cultured in a medium containing 10% sucrose and 2% yeast extract as main components for 24 hours at 37$^{\circ}C$ and pH 7. Fructan was obtained by precipitation from the cultured medium by adding alcohol (alcohol ratio of 1:3) after removing the enzyme by centrifuging. Fructan exhibited almost same moisturizing effect as hyaluronic acid and cell proliferation effect on human fibroblast and keratinocyte as well. Moreover, on cell proliferation test on bio-artificial skin constructed by 3-dimensional(3-D) culture after inducing primary skin inflammation with 0.5% sodium lauryl sulfate (SLS), the 3-D artificial skin treated with 0.01 mg/ml, 0.05mg/ml of Fructan exhibited higher cell proliferation than the 3-D artificial skin treated with SLS only. On anti-inflammation test on 3-D artificial skin evaluated by measuring secreted quantity of interleukin-1$\alpha$ (IL-1$\alpha$) which is a pre-inflammatory mediator induced by SLS, the quantity of IL-1$\alpha$on the 3-D artificial skin treated with 0.01 mg/ml, 0.05mg/ml of Fructan was less than the one on the 3-D artificial skin treated with SLS only. As a result of these studies, Fructan has anti-inflammation effect against inflammatory reaction by a skin irritant as well as cell proliferation effect in bio-artificial skin. Fructan was also evaluated as a safe material without any toxicity in safety tests using fibroblasts and animals.

Effect of Mori Follium Extract on the Melanogenesis and Skin Fibril Matrix (상엽(桑葉) 추출물의 미백활성 및 피부섬유구조 형성에 미치는 효과)

  • Objectives : The Skin is composed of multiple layers, including the epidermis, dermis, and hypodermis. It provides a vital barrier structure that protects vertebrates from external environmental antigens, solvents, ultraviolet light, microorganisms, toxins, and weather conditions. Although several biological effects of Mori Follium have been reported, beneficial effects of Mori Follium in skin health remain unclear. In this study, we prepared water extract of Mori Follium (MLE) and evaluated the effects on melanin accumulation and expression levels of skin fibril-related proteins.Methods : The cytotoxicities of MLE in B16F10 melanoma and human skin fibroblasts (HSF) were examined by MTT assay. Inhibitory effect of MLE on the α-MSH- and IBMX-induced melanosis in B16F10 melanoma was examined. The expression levels of fibronectin, collagen 1α2, and CCN2 in MLE-treated HSF were analyzed by reverse transcription-polymer chain reaction (RT-PCR) and western blotting.Results : The MLE treatment for 24 h did not affect to the B16F10 and HSF at concentrations of 1, 10, 50, 100, 200, 400 and 800 ㎍/ml. The MLE treatment for 72 h significantly and dose dependently suppressed melanin accumulation in B16F10 melanoma. In addition, the MLE treatment up-regulated expression levels of skin fibril-related genes such as fibronectin, collagen 1α2, and CCN2 in HSF. Our western blot analysis revealed MLE-induced up-regulation of skin fibril-related genes required the activation of CCN2 protein.Conclusions : In conclusion, these findings suggest that the MLE could be used in development of cosmetic natural material of maintaining healthy skin.

  • Mammalian tyrosinase plays an important role in the process of melanin polymer biosynthesis by catalyzing the hydroxylation of tyrosine to 3,4-dihydroxyphenylalanine(DOPA) and the oxidation of DOPA to dopaquinone. These processes are major determinant of human skin color and involved in localized hyperpigmentation. Therefore, the enzyme inhibitors have been of great concern as skin-whitening cosmetics. Methanol extracts of 174 oriental herbs were screened for the mushroom tyrosinase inhibitory activity.

Preparation of Lip Balm Utilizing Functionalities of Colorants Extracted from Marine Algae (해조류 추출색소의 기능성을 활용한 립밤 제조)

  • The aim of this study is to utilize the algae extracts with functionalities such as antioxidant and antimicrobial properties for preparing lip balm. Functional properties of extracts from Dictyota coriacea and Ulva pertusa were evaluated in terms of DPPH radical scavenging activity and antimicrobial activity. Lip balms contained the algae extracts were tested its efficacy by measuring the moisture retention of lip skin after applying human subjects. According to the results of skin moisture retention measurement, lip balm was effective to increase the moisture retention of lip skin. It is considered that the utilization of colorants extracted from algae which are verified of antioxidant effect and antimicrobial activity is valuable and useful in developing the cosmetic products.

Development of Artificial Skin from Chitosan Derivatives

  • Chitosan derivitives, a sulfated N-acetylchitosan was synthesized, and artificial skin of sulfated N-acetylchitosan and N-carboxyl butyl chitosan were investigated. Sulfated derivatives of chitosan were analyzed by {TEX}${13}^C${/TEX}-NMR and the structure on N-acetyl chitosan 3,6-O-disulfate were confirmed. Rabbits underwent a midline laparotomy followed either by a bilateral peritoneal sidewall abraison(3.0×1.5cm). The injured surface was then covered with 0.2mm thick sulfated N-acetyl chitosan membrane. Sulfated N-acetyl chitosan membrane was found to reduce postsurgical bleeding after abraison of peritoneal surface treated with sulfated N-acetyl chitosan membrane. Sulfated N-acetyl chitosan implanted rabbit showed quick wound healing than N-carboxybutyl chitosan. With a sterilization procedure of chemical sterilization, sulfated N-acetyl chitosan seem to be better substitutes than N-carboxybutyl chitosan.

  • Trimethylglycine, commonly named betaine, is the most simple amphoteric molecule. It is completely vegetal (1,2), as it is produced in the sugar industry by industrial chromatography of molasses. While abundantly used in foods and diet supplements, many interesting applications in cosmetics have recently been investigated, like its capability to increase the volume and stability of foams in surfactant solutions. For its special chemical structure (it is the internal salt of a weak acid and a strong alkali) trimethylglycine is a solvent and buffering agent for strong acids and Lewis' acids. It allows to improve the efficiency of $\alpha$- and $\beta$-hydroxy acids in increasing the physiological rate of epidermal cell renewal, while keeping a low skin-irritation level. In oral care cosmetics, it acts as a mucous membrane protectant (3). For its special water co-ordination capability, its solubilising power, polymer swelling capability, after-feel improvement in hair products, skin moisturization and elasticity enhancing properties, trimethylglycine provides unusual characteristics to many products intended for skin maintenance (4).(omitted)

3-Dimensional Coating Polymer Microneedles for Economical and Efficient Transdermal Drug Delivery (경제적이고 효과적인 경피 약물전달을 위한 3차원 구조의 코팅 고분자 마이크로니들)

  • Polymer microneedles can be fabricated by a micromolding method, an easy and cost-effective method. However, it is not easy to achieve uniform coating with an aqueous coating solution due to hydrophobic surface of polymer microneedles. 3-Dimensional coating polymer microneedles could deliver more than twice as much dose as in-plane metal microneedles by increasing coating area and the number of microneedles per unit area. A uniform coating was not obtained by addition of coating additives in the coating solution. The satisfied coating was achieved by treatment of surface of polymer microneedle with metal deposition and UV/ozone, and UV/ozone treatment was an ultimate surface treatment method based on biological safety. Calcein coating polymer microneedles were prepared by using UV/ozone treatment and followed dip-coating, and they delivered calcein in porcine skin successfully after 15 min of insertion.
