피부 소양 마우스 모델의 최근 연구 동향 -Scratching behavior model을 중심으로 (The latest study tendency in mouse model of skin pruritus - Mainly on scratching behavior model)

  • Objectives : This study was carried out to investigate the latest study tendency in mouse model of skin pruritus published since 2005 and to arrange various experimental methods. Methods : We examined the journal(such as Experimental dermatology and British journal of dermatology) and in Pubmed since 2005, regarded pruritus and scratching behavior model as key words. Results and Conclusion : 1. BALB/c, hairless, NC/Nga and ICR mice were the most used in scratching behavior model. According to scratching-induced agents. we need to select experimental mice. 2. There are various methods inducing skin pruritus; by cohabitation with NC/Nga mice having severe skin lesions, by injection intradermally(or subcutaneously) with agent inducing inflammation, by inducing contact dermatitis with TNCB, TNFB, destruction of skin barrier and by transgenic mice. 3. Injection intradermally(or subcutaneously) with agent inducing inflammation is the most used out of methods inducing skin pruritus. Compound 48/80, histamine, substance P and others(chloroquine, serotonin, carrageenin, TPA etc.) were included in agents inducing inflammation and pruritus in skin pruritus model. 4. Video camera, SCLABA system, MicroAct and acoustic evaluation system were included in evaluation methods of scratching behavior mouse model.

Inhibition of Collagenase by Naturally-Occurring Flavonoids

  • We examined the inhibitory activities of various flavonoids, including the flavanones, flavones/isoflavones and flavonols, on collagenase from Clostridium histolyticum to establish their therapeutic potential against skin inflammation and photoaging. In general, the flavonols were stronger inhibitors than the flavones/isoflavones, and this indicated the importance of the C-3 hydroxyl substitution. Quercetin was the most active flavonoid among those tested, and it showed an $IC_{50}$ of 286 ${\mu}M$. These novel results suggest that certain flavonoids, particularly the flavonols, may prevent collagen breakdown by inhibiting collagenase in inflamed skin as well as photoaged skin.

  • There have been various inflammatory skin disorders in humans including atopic dermatitis, eczema and psoriasis. Although some drugs have been used for these disorders, there is an urgent need for safer and more effective topical anti-inflammatory agents. Plant flavonoids possess anti-inflammatory activity and some of them have multiple pharmacological mechanisms, inhibition of eicosanoid metabolizing enzymes, histamine release and/or down-regulation of pro inflammatory gene expression. These properties of flavonoids may be suitable for treating chronic skin inflammatory disorders. Especially, wogonin, some prenylated flavonoids and biflavonoids have a strong potential as new anti-inflammatory agents by topical application.

Nitric Oxide Signal Transduction and Its Role in Skin Sensitization

  • Nitric oxide (NO) is a signaling molecule that plays a crucial role in numerous cellular physiological processes. In the skin, NO is produced by keratinocytes, fibroblasts, endothelial cells, and immune cells and is involved in skin functions such as vasodilation, pigmentation, hair growth, wound healing, and immune responses. NO modulates both innate and adaptive immune responses. As a signaling molecule and cytotoxic effector, NO influences the function of immune cells and production of cytokines. NO is a key mediator that protects against or contributes to skin inflammation. Moreover, NO has been implicated in skin sensitization, a process underlying contact dermatitis. It modulates the function of dendritic cells and T cells, thereby affecting the immune response to allergens. NO also plays a role in contact dermatitis by inducing inflammation and tissue damage. NO-related chemicals, such as nitrofatty acids and nitric oxide synthase (NOS) inhibitors, have potential therapeutic applications in skin conditions, including allergic contact dermatitis (ACD) and irritant contact dermatitis (ICD). Further research is required to fully elucidate the therapeutic potential of NO-related chemicals and develop personalized treatment strategies for skin conditions.

Suppression of Cyclooxygenase-2 Expression of Skin Fibroblasts by Wogonin, a Plant Flavone from Scutellaria Radix

  • Previously, wogonin (5,7-dihydroxy-8-methoxyflavone) was found to suppress proinflammatory enzyme expression including cyclooxygenase-2 (COX-2), contributing to in vivo anti-inflammatory activity against skin inflammation. However, the detailed effect on each skin cell type has not been understood. Therefore, present investigation was carried out to find the effect of wogonin on inflammation-associated gene expression from skin fibroblasts in culture using reverse transcriptase-polymerase chain reaction. As a result, it was found that wogonin (10 - 100 ${\mu}$M) clearly down-regulated COX -2 expression from NIH/3T3 cells treated with 12-O-tetradecanoylphorbol 13-acetate, interleukin-1${\beta}$ or tumor necrosis factor-a. But, the expression levels of COX-1, interleukin-1${\beta}$ and fibronectin were not significantly affected. This finding was well correlated with significant reduction of prostaglandin E$_2$(PGE$_2$) production by wogonin. As a comparison, NS-398 (selective cyclooxygenase-2 inhibitor) did not suppress COX -2 expression and other gene levels, while PGE$_2$production was potently reduced at 0.1 - 10 ${\mu}$M. All these results suggest that COX -2 down-regulation of skin fibroblasts may be, at least in part, one of anti-inflammatory mechanisms of wogonin against skin inflammation.

In Vitro Anti-Inflammatory and Skin Protective Effects of Codium fragile Extract on Macrophages and Human Keratinocytes in Atopic Dermatitis

  • Codium fragile has been traditionally used in oriental medicine to treat enterobiasis, dropsy, and dysuria, and it has been shown to possess many biological properties. Atopic dermatitis (AD) is one of the types of skin inflammation and barrier disruption, which leads to chronic inflammatory skin diseases. In the current investigation, the protective effects of C. fragile extract (CFE) on anti-inflammation and skin barrier improvement were investigated. In LPS-stimulated RAW 264.7 cells, nitric oxide generation and the expression levels of interleukin (IL)-1β, IL-4, IL-6, iNOS, COX-2, and tumor necrosis factor-alpha (TNF)-α were reduced by CFE. CFE also inhibited the phosphorylation of NF-κB-p65, ERK, p-38, and JNK. Additionally, CFE showed inhibitory activity on TSLP and IL-4 expression in HaCaT cells stimulated with TNF-α/interferon- gamma (IFN-γ). Enhanced expression of factors related to skin barrier function, FLG, IVL, and LOR, was confirmed. These findings implied that CFE may be used as a therapeutic agent against AD due to its skin barrier-strengthening and anti-inflammatory activities, which are derived from natural marine products.

백작약 열수추출물의 피부노화 억제 효과(I) -염증 및 피부장벽 손상 완화 효과를 중심으로- (Inhibitory Effects of Peonia japonica Water Extract on Skin Aging (I) - Focussed on Alleviative Effects of Inflammation and Skin Barrier Damage -)

  • To investigate the alleviative effects of Peonia japonica water extract(PJWE) on inflammation and skin barrier damage, both the irradiation of UVB and the application of squalene monohydroperoxide (Sq-OOH) to the backs of hairless mice were performed for 4 weeks. And at the same time experimental materials were applied topically. The skin erythema indices for the positive control (PC, 0.01% retinoic acid) and experimental (E, PJWE) groups were lower than that of the control (C) group. Whereas both the lipid and water capacities for the PC and E groups were higher than those of the C group. Epidermis and dermis of the C group were remarkably thickened in comparison with the PC and E groups. Relatively much less number of inflammatory cells, including lymphocytes, neutrophils and macrophages were found in dermis of the PC and E groups compared with the C group. Lipid lamellae of the C group were broken severely showing an irregular arrangement and lipid content was much reduced. Whereas those of the PC and E groups were almost intact with a regular arrangement, which were similar to that of the N group. Taken the results all together, it was confirmed that PJWE could be effective natural herbal material for the alleviation of inflammation and skin barrier damage in hairless mice skin which were induced by UVB and Sq-OOH.

ICR mouse에 있어 UVB조사로 유도된 접촉 과민반응에 대한 EGb 761의 억제 효과 (Prevention of Ultraviolet B Radiation (280-320 nm) Induced Contact Hypersensitivity by EGb 761)

  • Exposure of skin to UVB radiation can cause the induction of inflammation and impairment of contact hypersensitivity(CHS) response. Several studies have shown that polyphenolic compounds isolated from EGb 761 afford protection against UVB. In this study, we demonstrated that topical application of EGb 761, before 1MED(1.4 KJ/$m^2$), 1.5MED (2.1 KJ/$m^2$), 2MED (2.8 KJ/$m^2$) of UVB exposure to ICR mice prevented UVB-induced inflammation and inhibition of the contact hypersensitivity response. The skin-fold swelling from 1MED, 1.5MED, 2MED of UVB exposure highly significantly increased after twice irradiation. Topical application of EGb 761(0.1%, 1%, 4%), 5 days prior to UVB exposure reduced skin thickness compared to non-treated mice. Exposure of shaved abdominal skin of mice to 1MED, 1.5MED and 2MED of UVB radiation resulted in suppression of contact sensitization through the skin to 56.23%, 65.12%, 74.02%, compared to normal unirradiated skin. Topical application of EGb 761(0.1%, 1%, 4%), 5 days prior to or 5 days after exposure to 1MED and 2MED of UVB resulted in protection against suppression of contact hypersensitivity in mouse dorsal skin. These protective effects were dependent on the dose of EGb 761 employed. The present study show that EGb 761 protect UVB-induced inflammation and immune suppression. Also, we suggest that EGb 761 can provide protection from photoimmunosuppression.

Investigation of Antimicrobial Activity of Rutaceae Fruit Ethanol Extracts Against Microorganisms-induced Skin Inflammation

  • 피부 염증 개선용 화장품 천연소재를 탐색하고자 피부 염증 유발 미생물 6종에 운향과 과실류 에탄올 추출물을 농도별로 처리하여 항균력을 비교 조사하였다. 운향과 과실류는 전과와 과피로 나누어 70% 에탄올로 추출하여 파우더 형태로 만들어 사용하였다. 그 결과 Staphylococcus aureus 균주를 제외한 5종의 균주에서는 항균활성을 확인할 수 있었고, 특히, Staphylococcus epidermidis 균주에 대한 항균활성이 생 레몬 전과에서 가장 우수하였다. Escherichia coli 균주에서는 생 레몬 전과와 생 탱자 과피에서만, Propionibacterium acnes 균주에서는 건조 레몬 전과에서만 항균활성이 확인되었다. Pityrosporum ovale 균주에 대한 항균활성은 생과의 경우 유자 전과, 레몬 전과, 밀감 껍질 순으로 항균활성을 확인할 수 있었고 건과 중에서는 오렌지 껍질, 탱자 껍질, 밀감 껍질, 레몬 껍질 순으로 항균활성을 확인할 수 있었다. 본 연구의 결과 운향과 과실류 중에서 피부 염증 유발과 관련된 피부 상재균에 대해 전반적으로 높은 항균활성을 나타낸 것으로 확인된 레몬이 피부 염증 개선용 화장품 천연소재로서 사용 가능성이 높을 것으로 사료된다.

Hijikia fusiforme Attenuate the Mast Cell-mediated Inflammatory Response through NF-kB Pathway Regulation

  • Hijikia fusiforme (HF), a member of brown algae family, exerts various pharmacological effects, including preventing arteriosclerosis and menopausal disorders. This study aimed to elucidate the ameliorative effect of HF on skin inflammation. We investigated the antioxidant and anti-inflammatory effect of HF extract to evaluate its potential as a functional materials. The antioxidant activity of HF was determined using 2,2-diphenyl-1-picrylhydrazy (DPPH) scavenging and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+) scavenging. To determine the pharmacological mechanism of HF in inflammatory reaction, we evaluated the effects of HF on interleukin (IL)-8, IL-6 and tumor necrosis factor (TNF)-α production and nuclear factor-κB (NF-κB) activation in activated- human mast cells (HMC)-1. Results showed that HF had the high DPPH and ABTS+ radical scavenging activity, and it suppressed IL-8, IL-6 and TNF-α production in a concentration-dependent manner. Moreover, HF significantly ameliorated NF-kB activation in activated-HMC-1. Hence, these results provide evidence that HF's potential for skin inflammation therapy.
