Beneficial Effect of an Agar Mask against Skin Damage Induced by UV Exposure in SKH-1 Hairless Mice (UV조사에 의해 유발된 SKH-1 hairless 마우스의 피부노화에 미치는 한천마스크의 개선효과)

  • To investigate the beneficial effects of an agar gel mask (AGM) on UV-induced photoaging, SKH-1 hairless mice were treated with a topical application of AGM and an AGM dipped in essence (AGMdE). The mice were divided into an no radiation group, UV + AGM, UV + AGMdE, and UV + vehicle (PBS) treatment groups. Alterations in skin wrinkles, skin phenotype, histological structures, oxidative status, and toxicity were then evaluated during 4 weeks of exposure. The topical application of AGM and AGMdE inhibited wrinkle formation, suppressed the erythema index, prevented transepidermal water loss, and enhanced skin hydration. In addition, epidermal thickness recovered to a similar level as that in the no irradiation group in the UV + AGM and UV + AGMdE treatment groups compared with the UV + vehicle (distilled water) group. Furthermore, the expression levels of matrix metalloproteinase-1 (MMP-1) and tyrosinase were reduced in the UV + AGM and UV + AGMdE treatment groups, although the highest level varied. Moreover, superoxide dismutase (SOD) activity was significantly lower in the UV + AGM and UV + AGMdE treatment groups as compared with the UV + vehicle group. No significant alterations induced by most toxic compounds were measured in serum biochemical markers and liver and kidney histological features of the UV + AGM and UV + AGMdE treatment groups. These results suggest that AGM may protect against skin aging by regulating skin morphology, histopathological structures, and oxidative conditions.

Characteristics of BGsome-Coated Illite as a Face Powder (BGsome이 코팅된 일라이트 및 이를 함유한 페이스 파우더의 특성)

  • BGsome coating on the surface of illite powder was attempted to impart skin affinity and improve various characteristics of the powder for makeup cosmetics. BGsome is a type of hydrated liquid crystalline vesicles prepared by the hydration of 1,3-butylene glycol (BG) dissolved lecithin. BGsome droplets were coated through wet process and the coated powders were analyzed by TGA, SEM, and particle size measurements. Effect of BGsome coating on the physical properties, such as flow-ability, dispersibility, spreadability, and adhesion, were examined. The repose angle of BGsome-coated illite was decreased, which means that BGsome coating improves the flowability of the powder. Dispersibility, spreadability, and adhesion of the coated illite were also improved remarkably. The result of adhesion tests showed that the BGsome-coated illite was evenly adhered onto artificial leather surface with almost no lumps. The flowability, dispersibility, spreadability and adhesion of face powders containing the BGsome-coated illite were also improved.

Antidepressant effect of water extract of Taraxacum platycarpum through BDNF, ERK and CREB pathway (BDNF, ERK 및 CREB 경로를 통한 포공영 추출물의 항우울 효과)

  • Objectives : Taraxacum platycarpum H. Dahlstedt has been reported to have several biological properties such as skin hydration and antiinflammation. The purpose of this study was to examine the antidepressive effects of water extract of T. platycarpum (WTP) on an animal model of depression. Methods : In the present study, normal ICR mice (4 weeks) were used, and orally administered with WTP (25, 50 and 100 mg/kg). Depression-like behavior was monitored the forced swimming test (FST) and tail suspension test (TST) in mice. The locomotor activity was evaluated to eliminate the false-positive activity in the open field test (OFT). Fluoxetine, the selective serotonin reuptake inhibitor, as a positive control was intraperitoneally administered at a dose of 15 mg/kg at 30 min before starting the behavioral test. Moreover, we evaluated the effects of WTP on the expression of brain-derived neurotrophic factor (BDNF) and the extracellular signal-regulated kinase (ERK)/ cyclic AMP response-element binding protein (CREB) signaling pathway in the hippocampus using Western blot. Results : The administration of WTP (50 and 100 mg/kg) significantly (P < 0.05, respectively) reduced the immobility time during FST and TST without accompanying changes in locomotor activity by OFT. Furthermore, WTP at dose of 100 mg/kg increased the BDNF expression and the phosphorylation of ERK and CREB in the hippocampus region. Conclusions : These results suggest that WTP has a useful anti-depressant effect through the regulation of BDNF/ERK/CREB signaling pathway.

The Methodological Study on Atopic Dermatitis in the latest clinical study (최신(最新) 임상연구(臨床硏究)를 중심으로 한 아토피 피부염에 관한 방법론적(方法論的) 연구(硏究))

  • Objective : The objective of this study is to analyze an increasing rate, difference of attack rate in age, relationship between atopic dermatitis and breast-feeding, the relationship between atopic dermatitis and the Sasang constitution, and various treatments of atopic dermatitis. Methods : This clinical study was carried out with 22 theses which are related with diagnosis, treatments, prognosis and control of atopic dermatitis. The 22 theses are carried on J Korean Oriental Pediatrics, J Korean Oriental Ophthalmology, J Korean Oriental Med, J Korean Academy of Pediatric Allergy and Respiratory Disease, J Korean Acad Fam Med and Korean J Food & Nutr. Results : The prevalence rate, attack rate and diagnosis rate of atopic dermatitis are increased in the year of 2000, compared with those of 1995. Comparing age of patient between the year of 1992 and 2002, the attack rate of atopic dermatitis is increased quickly over 7 years old. Specific immunoglobulin E(IgE) antibodies detected in patients under 1 year old was exclusively caused by food. But for the age over 7 years old, food and inhalant allergen are detected in the year of 2002 in compare with that of 1992. Because of breast-feeding, intemperate diet adjustment during the period of maternity of family history of atopic dermatitis, the attack rate of atopic dermatitis is increased in infant. The types of patients are categorized according to Sasang constitution and Soeumin group was largest. The Oriental medicine treatments of atopic dermatitis are bath & skin hydration, avoidance from antigen, dietetic treatment, external treatment, medication and phototherapy. Conclusion : The atopic dermatitis is associated with breast-feeding, Sasang constitution types. More active approach for the treatment and prevention of Atopic dermatitis in children are needed.

Establishment of reference intake of water for Korean adults in 2015 (2015 한국 성인의 수분 섭취기준 설정)

  • Purpose: This review was performed to establish a reference intake of water for Korean (KDRI-water) adults in 2015 by examining current knowledge of the relationship between water intake and health and the general status of water intake and loss. We expect that this study will be utilized for further refinement of KDRI-water. Methods: Documents were searched using RISS, NDSL, DBPIA, CINAHL, and Pubmed with the keywords 'water intake, water supply, water ingestion, hydration, dehydration, water balance, and fluid balance'. Results: Water balance is essential for the maintenance of health. Based on this assumption, numerous studies have been performed to investigate the association of water intake with several diseases such as urolithasis, obesity, diabetes, and cancer as well as other health problems, including constipation, cognition, and fetal weight. Effects of water intake for prevention or relief of these health problems vary. Water is supplied to the body by eating foods and drinking liquids such as plain water and beverages. Metabolic water is another source of water input. Water is lost through urine, skin, respiration, and feces. KDRI-water 2015 was set by adequate intake (AI) based on water intake volume, which was the sum of water intakes from foods and fluids reported by the Korea National Health and Nutrition Examination Survey, with extra milk intake of 200 mL. AIs in some age groups were modified considering their estimated energy requirements. Conclusion: Accurate data of water intake is critically important for the establishment of KDRI-water. Therefore, improvement of systems investigating water intake is required, and more studies on the status of water intake and loss in Korean people are needed for definite KDRI-water establishment.

A Study on functional cosmetic ingredients of the invasive plant Spartina anglica (생태교란종 영국갯끈풀의 기능성화장품 원료로서 효능 연구)

  • The Spartina anglica is recognized as a highly invasive plant and active eradication methods are required. In this study, we aimed to determine the physiologic activities of Spartina anglica extracts as a cosmetic ingredient. Antioxidant properties were investigated by different chemical methods including radical quenching (DPPH and ABTS), reducing power(FRAP) assay and aerial part of S. anglica(SAA) extract presented the strongest antioxidant activities. The significant cytotoxicity was not observed up to a concentration of 0.5 mg/mL in RAW264.7 cells and NHDF cells. The anti-inflammatory activity of S. anglica belowground(SAB) extract had strong effects on cell-based systems, including LPS-induced NO and cytokines(TNF-a and IL-6) production in RAW264.7 cells. Collagen synthesis and skin hydration gene expression of S. anglica extract showed the highest anti-wrinkle and moisturizing effect in NHDF cells. Results presented in this study tend to show that the ethanol extracts of S. anglica could be exploited as useful-bio-resource for bioactive substances in functional cosmetics.

The Enhancing Effect of Atopic Dermatitis by cosmetics containing estern medical herbs (한방소재 화장품의 아토피 피부염 개선 효과)

  • Sixteen moderate atopic dermatitis patients were treated with applying toner and cream using estern medical herbs[Estern medical complex(Radix Glycyrrhizae, Radix Angelicae Dahuricae, Rhizoma Ligustici Chuanxiong, Fructus ponciri Seu Aurantii Immaturus, Radix Rehmanniae Praeparata, Radix Puerariae, Rhizoma Corydalis), Cypress, Green tea, Radix Astragali, Red ginseng, Syrup ferment, Apple ferment filtrate] on the atopic area 2-3 times per day for 6 weeks. After 6 weeks of study, the result was a decrease in SCORAD index($26.9{\pm}11.8$ to $14.9{\pm}9.7$(p<0.05)), IgE($641.9{\pm}1294.6$ to $565.8{\pm}1076.8$), Eosinophil count($246.7{\pm}203.5$ to $203.3{\pm}130.7$(p<0.05)) and TEWL($16.7{\pm}5.1$ to $15.4{\pm}8.7$(p<0.05)). And skin hydration was increased($36.6{\pm}10.0$ to $44.0{\pm}10.3$(p<0.01)) maintaining skin pH level. The patient;s and physician's global assessment also improved. Therefore, estern medical herbs may play a role in treatment of atopic dermatitis.

Efficacy and Safety of Topical Application of Epidermal Growth Factor (EGF) for Korean Acne Patient (한국인 여드름 환자에서 표피성장인자가 함유된 외용제의 피부 적용에 대한 유효성 및 안전성 평가)

  • Acne vulgaris is a chronic inflammatory condition characterized by comedo, papule, cyst, nodule and postinflammatory hyperpigmentation. Meanwhile, it is also induced by adverse event of drugs. Among them, acneiform folliculitis is a side effect of epidermal growth factor receptor (EGFR) inhibitor, which is an anticancer agent, and its incidence may occur in upward of 75 ~ 100% of cases. The main method of acne vulgaris treatment is oral antibiotics, retinoids, topical medication and so on. However, it is limitation that teratogenicity caused by retinoids and antibiotic resistance increased by using antibiotics. In this study, we aimed to evaluate the clinical efficacy and safety of topical recombinant human (rh) EGF in treating facial acne vulgaris. Twenty three Koreans (age: 10 ~ 29 years) with mild to moderate acne vulgar participated in the study and applied topical rhEGF cream (trouble control EGF) with 3 products (trouble control clarifying cleansing foam, trouble control all-clear filling toner, redroll calming moisture) on their face twice daily for four weeks. Several assessment methods were applied: Acne lesion counts score by investigator's global assessment, efficacy and satisfaction score by subjects. Skin sebum output level, hydration level and redness level were also measured at each visit. At the final visit, skin sebum level, transepidermal water loss, skin redness statistically decreased and acne lesions (comedone, papule) were statistically reduced. No severe side effects were observed during the study. In conclusion, topical rhEGF seems to be an effective and safe adjuvant treatment option for mild acne vulgaris.

Proliposomal Clenbuterol Patch for Transdermal Delivery (프로리포솜을 이용한 클렌부테롤의 경피흡수 제제화)

  • Proliposomal patch of clenbuterol, ${\beta}_2-agonist$ bronchodilator, was prepared and its feasibility as a novel transdermal drug delivery system was examined. Proliposomal granules containing clenbuterol was prepared by a standard method using sorbitol and lecithin with (Rx 2) or without cholesterol (Rx 1). The porous structure of sorbitol in the proliposomes was maintained allowing tree flowability of the granules. Following contact with water, the granules were converted probably to liposomes almost completely within several minutes. It indicates that proliposomes may be hydrated, when they are applied on the skin under occlusive condition in vivo, by the sweat to form liposomes. Clenbuterol release from Rx 1 and Rx 2 proliposomes to pH 7.4 isotonic phospate buffer (PBS) across cellulose membrane (mol. wt. cut-off of 12000-14000) was retarded significantly compared with that from the mixture of clenbuterol powder and blank proliposomes. Interestingly, proliposomes prepared with lecithin and cholesterol (i.e., Rx 2 proliposomes) showed much more retarded release of clenbuterol than proliposomes prepared only with lecithin (i.e.. Rx 1 proliposomes), indicating that clenbuterol release from proliposomes can be controlled by the addition of cholesterol to the proliposomes. Proliposomal patches were prepared using PVC film as an occlusive backing sheet, two sides adhesive tape (urethane, 1.45 mm thickness) as a reservoir for proliposome granules and Millipore MF-membrane (0.45 mm pore size) as a drug release-controlling membrane. Rx 1 or Rx 2 proliposomes containing 4.6 mg of clenbuterol were loaded into the reservoir of the patch. Clenbuterol release from the patches to pH 7.4 PBS was determined using USP paddle (50 rpm)-over-disc release method. Clenbuterol release from the proliposomal patches was much more retarded even than from a matrix type clenbuterol patch (Boehringer Ingelheim ltd). Being consistent with clenbuterol release from the proliposomal granules, the release from the patches was highly dependent on the presence of cholesterol in the proliposomes : Patches containing Rx 2 proliposomes showed several fold slower drug release than patches containing Rx 1 proliposomes. When the patch containing Rx 1 proliposomes was applied on to the back of a hair-removed rat, clenbuterol concentration in the rat blood was maintained during 6-72 hrs. Transdermal absorption of clenbuterol from the patch was accelerated when the patch was prehydrated with 50 ml of pH 7.4 PBS before topical application. Above results indicate that sustained transdermal delivery of clenbuterol is feasible using proliposomal patches if the cholesterol content and pore size of the release rate-controlling membrane of patches, for example, are appropriately controlled.

Moisturizing and Anti-oxidation Effect of Astragalus membranaceus Root Extract (황기추출물의 보습 및 항산화 효과)

  • We investigated the effect of moisturizing and anti-oxidation of Astragalus membranaceus (Astragali Radix) root extract with respective to growing districts and extract methods for the purpose of development of cosmetic ingredients. Astragalii Radix was collected in Jecheon, Jeongseon, Taebaek, and Yeongju in Korea and China as growing districts. Formononetin was determinated by HPLC method as one of the various active agents in Astragalus membranaceus root extract. The 75% ethanol extract demonstrated to be more effective than $H_2O$-extracted one for a scavenging activities to DPPH radicals and reactive oxygens. The 75% ethanol extract showed $IC_{50}$ (50% scavenging concentration) of 2.162 mg/mL and 2.981 mg/mL in case of free radical scavenging activity test and reactive oxygen scavenging activity test, respectively Especially, free radical scavenging activity of isoflavonoids isolated from ethylacetate fraction was similar to scavenging activity of genistein. Astragalus membranaceus root extract of Jecheon district by sonicating extraction method was more effective in skin hydration compared with others.
