Anti-skin Aging Properties of Different Solvent Extracts from Ishige foliacea (넓패(Ishige foliacea) 추출물의 피부노화 억제활성)

  • In this study, the antioxidant and anti-skin aging properties of the Korean marine algae Ishige foliacea were investigated. Solvent extracts from I. foliacea were prepared with 70% ethanol, 80% methanol, and water. The extraction yields of various solvent extracts ranged from 9.55% to 35.12%. In terms of antioxidant activity, the ethanol extract showed the highest ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging activity, nitrite oxide scavenging activity, reducing power, and FRAP (ferric reducing antioxidant power). Regarding anti-skin aging activity, evaluation of the skin whitening and anti-wrinkle activities revealed that the methanol, water, and ethanol extracts possessed the highest tyrosinase (IC50=0.98 mg/mL), elastase (IC50=0.15 mg/mL), and collagenase (IC50=0.06 mg/mL) inhibitory activities, respectively. These results suggest that I. foliacea holds potential as an antioxidant and anti-skin aging substance in food and cosmetic materials.

Development of Pre-assessment Indicator for Skin Aging and Anti-aging Effect of Inner Skin by the Cream Product Containing Gynostemma pentaphyllum Gypenoside Fraction (피부 노화의 선평가 지표 개발 및 그에 따른 칠엽담 지페노사이드 분획물 함유 크림의 피부 내부 노화 개선 효과)

  • The human skins had a lot of changes as aging. The synthesis of collagens and elastic fibers were reduced and the expression of matrix metalloproteinase (MMPs) was increased in the dermis. These changes have been known to be responsible for skin wrinkle formation. In this study, we tried to develop pre-indicator of inner skin aging, and investigated the effect of the cream product containing Gynostemma pentaphyllum gypenoside fraction (Gynostemma pentaphyllum gypenoside cream). 21 ~ 34 years old 52 Korean female volunteers without crow's feet were chosen for this clinical study. The starting point of inner skin aging was confirmed through dermal density measurement, even though outside skin wrinkles were not observed. In addition, 4 and 8 weeks treatment of G. pentaphyllum gypenoside cream significantly increased age-related reduction of dermal density. These results suggest that the evaluation of dermal density can be used as pre-indicator of skin aging. Furthermore, G. pentaphyllum gypenoside fraction is a potential anti-wrinkle agent for prevention and treatment of skin aging.

A novel nicotinoyl peptide, nicotinoyl-LVH, for collagen synthesis enhancement in skin cells

  • A novel Nicotinoyl fused peptide, Nicotinoyl-LVH, was synthesized by solid phase peptide synthesis method, purified, and tested in cultured skin cells. Nicotinoyl-LVH enhanced the expression level of collagen mRNA and its fragments in fibroblasts. These data show that this novel Nicotinoyl peptide is a promising biomaterial in the anti-aging functional cosmetic market.

Inhibition Effects of Frankincense Oil on Skin Aging (II): Focussed on Histological Observation (프랑킨센스 오일의 피부노화 억제 효과 (II) - 조직학적 관찰을 중심으로 -)

  • The purpose of this study was to investigate the effects of frankincense oil in skin aging animal model. Skin aging was induced by both the irradiation of UVB and the application of squalene monohydroperoxide (Sq-OOH) to the back of experimental animals for 4 weeks. And at the same time experimental materials were applied topically. Six to seven weeks female SHR-1 hairless mice were divided into five groups including normal (N: saline), control (C: UVB+Sq-OOH+saline), vehicle control (VC: UVB+Sq-OOH+jojoba oil), positive control (PC: UVB+Sq-OOH+0.01% retinoic acid) and experimental (E: UVB+Sq-OOH+3% Frankincense oil) groups, five animals each group. Lipid lamella and lipid content in stratum corneum of the E group were almost intact with a regular arrangement which were similar to the N group. Collagen fibers in dermis of the E group were almost intact with a regular arrangement which were similar to the N group. Relatively much less number of mast cells and inflammatory cells were found in the E group compared to the C group. The activities of XO, SOD and CAT were no significant difference between the E and N groups. In conclusion, the application of frankincense oil to the skin aging animal model reduced both the generation of free radicals and the damage of skin tissues. Therefore, frankincense oil can be used practically for the prevention or improvement of skin aging in terms of health promotion and beauty for the people.

Aging and UV Irradiation Related Changes of Gene Expression in Primary Human Keratinocytes

  • The epidermis is a physiological barrier to protect organisms against environment. During the aging process, skin tissues undergo various changes including morphological and functional changes. The transcriptional regulation of genes is part of cellular reaction of aging process. In order to examine the changes of gene expression during the aging process, we used the primary cell culture system of human keratinocytes. Since UV radiation is the most important environmental skin aggressor, causing skin cancer and other problems including premature skin aging, we examined the changes of gene expression in human keratinocytes after UV irradiation using oligonucleotide microarray containing over 10,000 genes. We also compared the gene expression patterns of the senescent and UV treated cells. Expression of the variety of genes related to transcription factors, cell cycle regulation, immune response was altered in human keratinocytes. Some of down-regulated genes are represented in both senescent and UV treated cells. The results may provide a new view of gene expression following UVB exposure and aging process in human keratinocytes.

Skin Volume Augmentation and Anti-wrinkle Effects of Tribulus terrestris Fruit Extract (질려자 추출물의 피부 볼륨 증진 및 주름개선 효과)

  • Aging of the face is mainly related to the features that are sagging or loss of elasticity of the skin by reducing the volume around the eyes or cheek. Intrinsic aging can be seen to cause thinner dermis, reduction of extracellular matrix and subcutaneous fat. This study was carried out to investigate the skin volume augmentation and anti-wrinkle effects of Tribulus terrestris fruit extract. Skin anti-aging effect of Tribulus terrestris fruit extract was evaluated by using lipid accumulation, expressin of type I procollagen and elastin in preadipocytes and human dermal fibroblasts. Tribulus terrestris fruit extract augmented preadipocytes differentiation about 56% at 100 µg/mL. The type I procollagen and elastin were increased about 35% and 25% by treatment 20% Tribulus terrestris fruit extract, respectively. The clinical study also showed that skin sagging, skin elasticity, and dermal density improved without adverse effect following 4 week application of cream containing 2% Tribulus terrestris fruit extract. We suggest that Tribulus terrestris fruit extract can have the good possibility as skin volume augmenting, skin elasticity and wrinkle improving agent.

Clinical Study of Cream Containing Pueraria mirifia for Skin Elasticity (Pueraria mirifica 추출물 함유 화장품의 피부 탄력 임상 효능 평가)

  • Recently, along with the remarkable increase in interest in natural products, the application of phytoestrogens in anti-aging products has become very important. In the focus, we developed Pueraria mirifica (PM), in this in vivo study on skin elasticity and wrinkles, and investigated the anti-aging properties. Thirty healthy women volunteers, between the ages of 30 and 60, applied the cream formula with $4\% Pueraria mirifica or placebo, on the face area twice a day for 16 weeks. The evaluation was made by silicon replica analysis, Cutometer SEM 575 and Comeometer followed by statistical treatment using the student's t-test. Results of skin elasticity measurement showed that topical application on the skin increased the total surface of skin elasticity in comparison with placebo group (p<0.05). However, PM cream does not have prominent effect on the depth of wrinkles and skin moisturization. This in vivo study demonstrates that, PM exhibits a significant effect on skin elasticity therefore, is of great interest in anti-aging and firmness skin care products.

Suppression of Cellular Senescence by Cordycepin in Replicative Aged Human Dermal Fibroblasts (Cordycepin에 의한 피부 섬유아세포 세포노쇠화 개선효과)

  • Cell senescence can be identified by cellular changes that occur as a result of intrinsic aging and/or diseases. In case of skin cells, aging and cell senescence caused by external factors results in cessation of cell proliferation and cellular malfunction, which, in turn, accelerates skin aging. In this study, inhibition of cell senescence and enhancement of cell function were studied using cordycepin to evaluate the potential for skin anti-aging agent. By comparing with the number of senescence associated with ${\beta}$-galactosidase (SA-${\beta}$-gal) positive cells in young and replicative aged human fibroblasts, it was found that replicative aged cells showed higher expression of ${\beta}$-galactosidase. Treatment of cordycepin - known as an anti-oxidative and anti-inflammatory agent - reduced ${\beta}$-galactosidase expression in senescent cells and enhanced cell survival in serum-free culture condition. Cordycepin also showed superb inhibition of ROS, which is another indicator of cell senescence. The results of this study proved the anti-aging effect of cordycepin on human fibroblasts and also proposed a possibility of its use as an anti-aging cosmetic ingredient.

Anti-aging Effects Saccharomyces Fermented Modified Kyungohkgo Extract on Skin (경옥고(瓊玉膏) 가미방(加味方) 효모(酵母) 발효물(醱酵物)이 피부(皮膚) 노화(老化)에 미치는 영향(影響))

  • Objectives : We investigated the anti-aging effects on skin with Saccharomyces fermented modified Kyungohkgo extract (SFKE). Methods : Fermented modified Kyungohkgo extracts by 14 different kinds of microorganism were prepared and most effective Saccharomyces fermented modified Kyungohkgo extract among these was selected for anti-aging agent. We measured various effects related to skin such as scavenging activity against free radical, cell proliferation, collagen synthesis and toxicity of SFKE were evaluated and compared with modified Kyungohkgo extract (KE). Results : Free-radical scavenging activity and cell proliferating ratio of SFKE were 2 and 8 times higher than those of KE. The SFKE could significantly increase the collagen synthetic ratio compared with KE treated group. SFKE showed no toxicity at all tested concentrations. Conclusions : The results of our study propose that SFKE have good anti-aging effects on skin.

In vitro screening of anti-skin aging and antioxidant properties of aqueous/solvent extracts from distinctive stages of silkworm (Bombyx mori L.) pupae

  • Silkworm pupae, a by-product of the silk industry are known to be valuable resource of nutrients for humans as well as animals besides encompassing diverse bioactive constituents. However, there is a paucity of knowledge on their role in amelioration of oxidative stress and anti-skin aging properties. In the present study, we evaluated the inhibitory effect of aqueous as well as ethanolic (30% and 70%) extracts from distinct stages of male and female silkworm pupae belonging to two silkworm varieties on skin aging-related enzymes. The activities of collagenase, elastase and tyrosinase were effectively inhibited by 70% ethanolic silkworm pupal extracts (SPE), followed by 30% with aqueous extracts exhibiting meager inhibitory potential. SPE were also investigated for their antioxidant activity in oxidative-stressed murine fibroblasts (L929). The intracellular ROS and lipid peroxidation induced by tert-butyl hydroperoxide (t-BHP) in fibroblasts was better attenuated by pre treatment with ethanolic (30%) and aqueous extracts, respectively. The safety of the extracts was determined by studying their effect on fibroblast cell viability and it was found that none of the extracts were cytotoxic. Our findings indicate the potential utility of SPE as anti-aging components in cosmeceuticals.
