Quantitative Evaluation of Radix Astragali through the Simultaneous Determination of Bioactive Isoflavonoids and Saponins by HPLC/UV and LC-ESI-MS/MS

  • The three major active isoflavonoids (calycosin-7-O-β -glucoside, isomucronulatol 7-O-β-glucoside, formononetin) and two main saponins (astragaloside I, astragaloside IV) in an extract of Radix Astragali were determined using rapid, sensitive, reliable HPLC/UV and LC-ESI-MS/MS methods. The separation conditions employed for HPLC/UV were optimized using a phenyl-hexyl column (4.6 × 150 mm, 5 μm) with the gradient elution of acetonitrile and water as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 230 nm. The specificity of the peaks was determined using a triple quadrupole tandem mass spectrometer equipped with an electrospray ionization (ESI) source that was operated in multiple reaction monitoring (MRM) in the positive mode. These methods were fully validated with respect to the linearity, accuracy, precision, recovery and robustness. The HPLC/UV method was applied successfully to the quantification of three major isoflavonoids in the extract of Radix Astragali. The results indicate that the established HPLC/UV and LC-ESI-MS/MS methods are suitable for the quantitative analysis and quality control of multi-components in Radix Astragali.

가상현실 환경에서 3D 가상객체 조작을 위한 인터페이스와 인터랙션 비교 연구 (Comparative Study on the Interface and Interaction for Manipulating 3D Virtual Objects in a Virtual Reality Environment)

  • Recently immersive virtual reality (VR) becomes popular due to the advanced development of I/O interfaces and related SWs for effectively constructing VR environments. In particular, natural and intuitive manipulation of 3D virtual objects is still considered as one of the most important user interaction issues. This paper presents a comparative study on the manipulation and interaction of 3D virtual objects using different interfaces and interactions in three VR environments. The comparative study includes both quantitative and qualitative aspects. Three different experimental setups are 1) typical desktop-based VR using mouse and keyboard, 2) hand gesture-supported desktop VR using a Leap Motion sensor, and 3) immersive VR by wearing an HMD with hand gesture interaction using a Leap Motion sensor. In the desktop VR with hand gestures, the Leap Motion sensor is put on the desk. On the other hand, in the immersive VR, the sensor is mounted on the HMD so that the user can manipulate virtual objects in the front of the HMD. For the quantitative analysis, a task completion time and success rate were measured. Experimental tasks require complex 3D transformation such as simultaneous 3D translation and 3D rotation. For the qualitative analysis, various factors relating to user experience such as ease of use, natural interaction, and stressfulness were evaluated. The qualitative and quantitative analyses show that the immersive VR with the natural hand gesture provides more intuitive and natural interactions, supports fast and effective performance on task completion, but causes stressful condition.

Development of a One-Step Duplex RT-PCR Method for the Simultaneous Detection of VP3/VP1 and VP1/P2B Regions of the Hepatitis A Virus

  • The simultaneous detection and accurate identification of hepatitis A virus (HAV) is critical in food safety and epidemiological studies to prevent the spread of HAV outbreaks. Towards this goal, a one-step duplex reverse-transcription (RT)-PCR method was developed targeting the VP1/P2B and VP3/VP1 regions of the HAV genome for the qualitative detection of HAV. An HAV RT-qPCR standard curve was produced for the quantification of HAV RNA. The detection limit of the duplex RT-PCR method was 2.8 × 101 copies of HAV. The PCR products enabled HAV genotyping analysis through DNA sequencing, which can be applied for epidemiological investigations. The ability of this duplex RT-PCR method to detect HAV was evaluated with HAV-spiked samples of fresh lettuce, frozen strawberries, and oysters. The limit of detection of the one-step duplex RT-PCR for each food model was 9.4 × 102 copies/20 g fresh lettuce, 9.7 × 103 copies/20 g frozen strawberries, and 4.1 × 103 copies/1.5 g oysters. Use of a one-step duplex RT-PCR method has advantages such as shorter time, decreased cost, and decreased labor owing to the single amplification reaction instead of four amplifications necessary for nested RT-PCR.

온도차를 갖는 경사진 평행평판 내의 혼합대류 열전달 (Mixed Convection between Inclined Parallel Plates with different Temperatures)

  • Experiments are performed to study the mixed convection flow and heat transfer in an inclined parallel plates with the upper part cooled and the lower part heated uniformly. The Reynolds number ranges from $4.0{\times}10^{-3}\;to\;6.2{\times}10^{-2}$, the angle of inclination, ${\theta}$, from 0 to 45 degree from the horizontal line, and Pr of the high viscosity fluid is 909. In this paper, the PIV(Particle image velocimetry) with TLC(Thermo-sensitive liquid crystal) tracers is used for visualizing and analysis. This method allows simultaneous measurement of velocity and temperature field at a given instant of time. Quantitative data of the temperature and velocity are obtained by applying the color-image processing to a visualized image, and neural network is applied to the color-to-temperature calibration. This paper describes the methods, and presents the quantitative visualization of mixed convection. From this study, it is found that the flow pattern can be classified into three patterns which are affected by Reynolds number and the angle of inclination.

A Generic Quantitative Relationship to Assess Interdependency of QoE and QoS

  • Researchers as well as network providers are increasingly interested in understanding how a network user's experience changes in relation to different quality of service (QoS) parameters, with many attempts being made to identify the general relationship between quality of experience (QoE) and QoS. Several relations in different function forms have been proposed. There are instances when this variety of proposed functions potentially results in one problem having completely dissimilar solutions at the same time. The question then is which function form is the one that can better explain this relationship. In this paper, three major existing quantitative relationships between QoE and QoS are investigated. After conducting a comprehensive comparative analysis between them, a new definition of QoE-QoS dependency appears. It then results in identifying a new mutual relationship between QoE and QoS. The newly proposed relationship improves the deficiency of previous QoE-QoS relationships along with solving the problem of diversity of simultaneous solutions for one problem.

Simultaneous Determination of Polycyclic Aromatic Hydrocarbons by Near Infrared Spectroscopy using a Partial Least Squares Regression

  • Polycyclic aromatic hydrocarbons(PAHs) are widely distributed in the environment and are often implicated as potential carcinogens. The chromatographic methods of detection and quantitative determination of PAHs in environmental samples are costly, time consuming, and do not account for all kinds of PAHs. This work describes a quantitative spectroscopic method for the analysis of mixtures of eight PAHs using multivariate calibration models for Fourier transform near infrared(FT-NIR) spectral data. The NIR spectra of mixtures of PAHs (anthracene, pyrene, 1,2-benzanthracene, perylene, chrysene, benzo(a)pyrene, 1-methylanthracene and benzo(ghi)perylene) were measured in the wavelength range from 1100 nm to 2500 nm. The spectral data were processed using a partial least squares regression. We have studied the spectral characteristics of NIR spectra of mixtures of PAHs. It was possible to determine each PAM used in this study at the environmental level(mg L-1) in the laboratory samples. Further development may lead to the rapid determination of more PAHs in typical environmental samples.

Simultaneous Determination of Four Compounds from Artemisia capillaris using High Performance Liquid Chromatography-Ultraviolet Detector (HPLC-UVD) and Their Quantitative Study in Artemisia Genus

  • Artemisia capillaris Thunb. (Compositae) is a native herb of East Asian countries and has used for the treatment of jaundice, high liver fever, and digestive diseases for a long time, as well as being developed as the source of herbal preparations until now. The major components from A. capillaris were chlorogenic acid (1) and its derivatives substituted with caffeoyl moieties, such as 3,5-dicaffeoylquinic acid (2) and 4,5-dicaffeoylquinic acid (3), and coumarins, such as scoparone. In the study, four compounds, chlorogenic acid, 3,5-dicaffeoylquinic acid, 4,5-dicaffeoylquinic acid and scoparone (4) in the 70% ethanolic extract of A. capillaris were simultaneously determined by using HPLC-UVD system. This method was validated with the terms of linearity, precious and accuracy according to ICH guidelines. The developed method was successfully applied for the quantitative analysis of Artemisia genus, A. capillaris, A. iwayomogi, A. princeps, and A. argyi, distributed in Korea.

$k_0$-표준화방법에 의한 기기중성자방사화 분석법의 고찰 (The Review of Instrumental Neutron Activation Analysis by $k_0$-standardization method)

  • 기기중성자방사화 분석법은 핵 분석기술의 대표적인 방법으로 비파괴-동시 다원소분석의 장점과 함께 절대측정값에 의해 정량 할 수 있다는 특징을 갖고 있다. 최근에는 정확도와 편의성을 만족할 수 있는 $k_0$-정량법을 사용한 기기중성자방사화 분석법이 세계적으로 일반화되고 있다. 본 연구에서는 $k_0$-법의 적용을 위하여 이 방법의 전체적인 개념의 소개와 함께 $k_0$-파라미터를 측정하고자 하였다. 이를 위하여 $k_0$-법의 개념이해와 정량에 필요한 인자들의 정의 및 파라미터인 $Q_0$(${\alpha}$)와 f 값을 결정하기 위한 수식과 실험적 측정방법 등을 요약하였고 중성자조사공에 따라 특성 값을 갖는 ${\alpha}$와 f 값을 하나로 연구용원자로의 방사화분석용 조사공(NAA#1)에서 측정하여 $k_0$-법의 도입을 위한 기반을 마련하였다.

LC-ESI-MS/MS를 이용한 용담사간탕의 주요 성분 분석 (Quantitative Analysis of the Marker Constituents in Yongdamsagan-Tang using Liquid Chromatography-Electrospray Ionization-Tandem Mass Spectrometry)

  • Yongdamsagan-tang has been used to treat the urinary disorders, acute- and chronic-urethritis, and cystitis in Korea. In this study, an ultra-performance liquid chromatography-electrospray ionization-mass spectrometry (UPLC-ESI-MS/MS) method was established for simultaneous analysis of the 20 bioactive marker compounds, geniposidic acid, chlorogenic acid, geniposide, liquiritin apioside, acteoside, calceolarioside B, liquiritin, nodakenin, baicalin, liquiritigenin, wogonoside, baicalein, glycyrrhizin, wogonin, glycyrrhizin, wogonin, saikosaponin A, decursin, decursinol angelate, alisol B, alisol B acetate, and pachymic acid in traditional herbal formula, Yongdamsagan-tang. Chromatographic separations of all marker compounds were conducted using a Waters Acquity UPLC BEH $C_{18}$ analytical column ($2.1{\times}100mm$, $1.7{\mu}m$) at $45^{\circ}C$ using a mobile phase of 0.1% (v/v) formic acid in water and acetonitrile with gradient elution. The MS analysis was performed using a Waters ACQUITY TQD LC-MS/MS coupled with an electrospray ionization source in the positive and negative modes. The flow rate was 0.3 mL/min and injection volume was $2.0{\mu}L$. The correlation coefficient of 20 marker compounds in the test ranges was 0.9943-1.0000. The limits of detection and quantification values of the all marker components were 0.11-6.66 and 0.34-19.99 ng/mL, respectively. As a result of the analysis using the optimized LC-ESI-MS/MS method, three compounds, geniposidic acid (from Plantaginis Semen), alisol B (from Alismatis Rhizoma), and pachymic acid (from Poria Sclerotium), were not detected in this sample. While the amounts of the 17 compounds except for the geniposidic acid, alisol B, and pachymic acid were $0.04-548.13{\mu}g/g$ in Yongdamsagan-tang sample. Among these compounds, baicalin, bioactive marker compound of Scutellariae Radix, was detected at the highest amount as a $548.13{\mu}g/g$.

페놀성 화합물을 이용한 현지초의 UPLC 다성분 동시분석 개발 (Simultaneous Analysis of Phenolic Compounds in Geranium thunbergii Using UPLC)

  • The aim of this study was to develop a UPLC method for simultaneous analysis of 8 phenolic compounds including gallic aicd (1), protocatechuic acid (2), methyl gallate (3), ellagic acid (4), kaempferol-3-arabinofranosyl-7-rhamnoside (5), kaempferitrin (6), afzelin (7) and kaempferol-7-rhamnoside (8) isolated from Geranium thunbergii which has been traditionally used as anti-diarrheal agent. The UPLC method was optimized and validated using Halo C18 column ($4.6{\times}100mm$, $2.7{\mu}m$) consisting of MeOH and 0.1% formic acid at 260 nm in 25 minutes. In quantitative analysis of 8 compounds in MeOH extract of G. thunbergii, contents of 4-6 were 12.39, 20.52 and 21.45 mg/g, respectively. These compounds were measured as major phenolic compounds in G. thunbergii and can be useful as marker compounds for its quality control. These results suggest that the UPLC method can be contributed as basic data for quality evaluation of herbal preparations.
