Ultrastructure of the Gill of the Parrot fish, Oplegnathus fasciatus (돌돔 (Oplegnathus fasciatus) 아가미의 미세구조)

  • The ultrastructure of the gills of Oplegnathus fasciatus was examined by means of light and transmission electron microscopes. The gills have primary and secondary filaments (lamellae). The following cells are identified and described : epithelial cell, pillar cell, chloride cell and mucose cell. The simple epithelial layer consists of squamous epithelium containing a large nucleus and the surface is covered with some of microridges. The lamella pillar structures are characterized by axial microtubules and lateral membrane interdigitations. Chloride cells contain a lot of mitochondria and specifically developed tubular systems. The rough endoplasimic reticulum and golgi complex, and some of mucous granules were observed in immature mucous cells. The mature mucous cells were AB-PAS positive, globular in shape, and had mucous granules of similar size with various electron densities.

Effect of Buffalo Rat Liver Cell and Platelet Derived Growth Factor on the Development of In Vitro Matured/In Vitro Fertilized Bovine Oocytes (Buffalo Rat Cell과 Platelet Derived Growth Factor가 체외수정란의 체외발육에 미치는 효과)

  • The experiments reported here take advantage of the large number of in vitro matured and in vitro fertilized(IVM /IVF) bovine oocytes which can be produced, permitting the design of controlled experiments to establish a simple defined medium for the study of early embryo requirements. A total of 1,386 IVM /IVF oocytes were used to compare a simple defined medium(KSOM) with more complex culture conditions used successfully for culture of bovine embryos but do not permit study of specific requirements. All experiments were extensively replicated factorials. In Experiment 1, KSOM was superior to Menezo B$_2$ medium in producing morulae plus blastocysts from IVM /IVF oocytes(33 vs 20%, P<0.()5). The yield of morulae plus blastocysts with KSOM was 22% and with BRLC added was 30%. In Experiment 2, (a 2x2 factorial of KSOM with or without BRLC and 0, 1 ng /ml of platelet derived growth factor, PDGF) more morulae plus blastocysts (40%) were produced in KSOM-BRLC co-culture containing 1 ng /ml PDGF than in the control KSOM(12%). In Experiment 3, there was no dose response when 0, 1 and 5 ng /ml of PDGF were added. The results with simple defined KSOM medium are sufficiently promising to indicate that specific requirements of the embryo may be examined in future studies with KSOM as a base.

Investigation of the ASTM International frost heave testing method using a temperature-controllable cell

  • Frost heave can cause uneven ground uplift that may damage geo-infrastructure. To assist damage-prevention strategies, standard frost heave testing methods and frost susceptibility criteria have been established and used in various countries. ASTM International standard testing method is potentially the most useful standard, as abundant experimental data have been acquired through its use. ASTM International provides detailed recommendations, but the method is expensive and laborious because of the complex testing procedure requiring a freezing chamber. A simple frost heave testing method using a temperature-controllable cell has been proposed to overcome these difficulties, but it has not yet been established whether a temperature-controllable cell can adequately replace the ASTM International recommended apparatus. This paper reviews the applicability of the ASTM International testing method using the temperature-controllable cell. Freezing tests are compared using various soil mixtures with and without delivering blow to depress the freezing point (as recommended by ASTM International), and it is established that delivering blow does not affect heave rate, which is the key parameter in successful characterization of frost susceptibility. As the freezing temperature decreases, the duration of supercooling of pore water shortens or is eliminated; i.e., thermal shock with a sufficiently low freezing temperature can minimize or possibly eliminate supercooling.

  • The microanatomy and ultrastructure of the digestive diverticulum of Mytilus galloprovincialis were described using light and electron microscopy. The digestive diverticulum of tawny color was surrounded the stomach and connected to stomach by a primary duct. Digestive diverticulum is composed of numerous digestive tubules. The epithelial layer of a simple digestive tubule, which is simple, is composed of basophilic cells and digestive cells. Basophilic cells are columnar in shape, and has a well-developed endoplasmic reticula, tubular mitochondria, Golgi complex and membrane-bounded granules of high electron density in the cytoplasm. Whereas digestive cells are columnar in shape, with development of microvilli and cilia on the free surface. Pinocytic vasicles, active lysosomes and numerous mitochondria were observed in the apical cytoplasm of digestive cells. The results of this study suggest that basophilic cell and digestive cell of the digestive tubule are specialized in the extracellular and intracellular digestion, respectively.

  • This paper presents the technology for the design, fabrication, and characterization of a microfluidic system interface (MSI); the purpose of this technology is to enable the integration of complex microfluidic systems. The MSI technology can be applied in a simple manner for realizing complex arrangements of microfluidic interconnects, integrated microvalves for fluid control, and optical windows for on-chip optical processes. A microfluidic system for the preparation of genetic samples was used as the test vehicle to prove the effectiveness of the MSI technology for packaging complex microfluidic systems with multiple functionalities. The miniaturized genetic sample preparation system comprised several functional compartments, including compartments for cell purification, cell separation, cell lysis, solid-phase DNA extraction, polymerase chain reaction, and capillary electrophoresis. Additionally, the functional operation of the solid-phase extraction and PCR thermocycling compartments was demonstrated by using the MSI.

In vitro maturation using αMEM with reduced NaCl enhances maturation and developmental competence of pig oocytes after somatic cell nuclear transfer

  • Background: Compared to medium containing 108 mM sodium chloride (NaCl), in vitro maturation (IVM) using a simple medium with reduced (61.6 mM) NaCl increases the cytoplasmic maturation and embryonic development of pig oocytes. Objectives: This study determines the effect of a complex medium containing reduced NaCl on the IVM and embryonic development of pig oocytes. Methods: Pig oocytes were matured in Minimum Essential Medium Eagle-alpha modification (αMEM) supplemented with 61.6 (61αMEM) or 108 (108αMEM) mM NaCl, and containing polyvinyl alcohol (PVA) (αMEMP) or pig follicular fluid (PFF) (αMEMF). Medium-199 (M199) served as the control for conventional IVM. Cumulus cell expansion, nuclear maturation, intra-oocyte glutathione (GSH) contents, size of perivitelline space (PVS), and embryonic development after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) were evaluated after IVM. Results: Regardless of PVA or PFF supplementation, oocytes matured in 61αMEM showed increased intra-oocyte GSH contents and width of PVS (p < 0.05), as well as increased blastocyst formation (p < 0.05) after PA and SCNT, as compared to oocytes matured in 108αMEMP and M199. Under conditions of PFF-enriched αMEM, SCNT oocytes matured in 61αMEMF showed higher blastocyst formation (p < 0.05), compared to maturation in 108αMEMF and M199, whereas PA cultured oocytes showed no significant difference. Conclusions: IVM in αMEM supplemented with reduced NaCl (61.6 mM) enhances the embryonic developmental competence subsequent to PA and SCNT, which attributes toward improved oocyte maturation.

Corrosion of Quartz Crystal Marine Sensors in Sea Water (항만센서용 수정진동자의 해수에 의한 부식)

  • A quartz crystal analyzer is utilized to monitor the corrosion process of an aluminum surface of a quartz crystal for marine sensor by sea water. A quartz crystal having 2000 $\AA$ of aluminum layer is installed in a specially designed cell and is in contact with sea water imitated electrolyte solution. While a constant potential is applied to the cell, the resonant frequency and resonant resistance are simultaneously measured using the quartz crystal analyzer. In addition, surface topographs are taken with an atomic force microscope(AFM) and the element analysis of the surface is conducted using an energy dispersive X-ray spectrometer(EDX). The simultaneous measurement of resonant frequency and resonant resistance during the corrosion process explains the change of surface structure caused by the corrosion. The variation of resonant frequency addresses the amount surface metal dissolution. As a conclusion, it is found that a simple measurement using the quartz crystal analyzer can replace the complex monitoring employing large equipments in the investigation of a corrosion process of sensor surface.

Proteomic Application in Cell Biology (세포생물학과 Proteomics 응용)

  • As the complete genomic sequences accumulate, the use of global techniques became possible. DNA microarray is a powerful technology for measuring global mRNA levels. This method, however, does not provide information on post-translational modifications of proteins. In addition, mRNA levels do not strictly correlate with protein concentrations, especially for lower-abundance proteins. Therefore, studies at the level of transcription are not sufficient to understand cellular activity. Proteomic techniques to analyze protein expression and function at the large-scale have been developed and used. This review introduces a simple explanation for proteomic analysis and examples of how proteomics is applied in cell biology.

Molecular Identification of the Toxic Alexandrium tamiyavanichii (Dinophyceae) by the Whole-cell FISH Method

  • The dinoflagellate Alexandrium tamiyavanichii Balech, a producer of toxins causing paralytic shellfish poisoning (PSP), has recently been considered as one of main organisms responsible for toxication of shellfish in Japan. In this study, A. tamiyavanichii was subjected to a molecular phylogenetic analysis inferred from 28S rDNA D1-D2 sequences and a species-specific LSU rRNA-targeted oligonucleotide DNA probe was designed to identify A. tamiyavanichii using the whole cell-FISH (fluorescence in situ hybridization). The sequences of the 28S rDNA D1-D2 region of A. tamiyavanichii showed no difference from A. cohorticular AF1746l4 (present name A. tamiyavanichii) and formed a distinct clade from the 'tamarensis species complex'. The probe, TAMID2, reacted specifically with A. tamiyavanichii cultured cells, without any cross-reaction with other species belonging to the same genus, including A. tamarense, A. catenella, A. affine, A. fraterculus, A. insuetum and A. pseudogonyaulax. In a test of cross-reactivity with a field sample, TAMID2 reacted consistently with only A. tamiyavanichii, indicating that the present protocol involving the TAMID2 probe might be useful for detecting toxic A. tamiyavanichii in a simple and rapid manner.

  • In the newly developed control method, the current flowing into SPE cell is the only one considerable factor. So, the structure of control circuit becomes simple and the manufacturing cost of the control device decreases. In conventional power comparison MPPT control method however, a voltage and current coming out from PV cell should be feedbacked to chase maximum power point at every moment. Then, the structure of control circuit becomes so complex and the risk of control failure is much higher than the novel MPPT control method. Therefore, PV generation system by a novel MPPT control method is especially operated much more safely in case of a huge system, because the voltage coming out from PV-cell is not needed to be feedbacked. In this paper, the PV-SPR system was actually manufactured based on the simulation model of PSCAD/EMTDC program and the results tested were shown. Authors are sure that it is the most useful method to maximize power from PV to SPE with only a feedback of SPE input current.
