  • These surveys were carried out to investigate of damage of agricultural chemicals, tabacco, and factory smoke on silkworm rearing Autumn in 1975 1. Damaging rate in chemicals, tabacco and factory smoke was 1.4, 0.6 and 0.1 percent each other. 2. In damage by agricultural chemicals, it appeared to be chemicals for rice (49.4%) orchard (21.2%) and vegetable (12.3%) in order. 3. Status of damaging rate in tabacco showed 60 and 18 percent in tabacco field and tobacco drying place near mulberry field. and showed 14 and 8 percent in rabacco drying place and tabacco field near rearing house. 4. The damage of factory smoke is largely stone powder of cement.

  • Several polyhydroxylated alkaloids were isolated from the extracts of freeze-dried silkworm powder, and purified by ion exchange chromatographic analysis. Through the HPLC analysis, we could identify 1-deoxynojirimycin (DNJ) and a kind of calystegin $B_2$ (HS-58) as well as a noble compound (HS-74) from the purified polyhydroxylated alkaloids. In order to know the characteristics of these isolated alkaloids as enzyme inhibitors, glycosidase inhibition activities of these identified alkaloids including other two non-purified alkaloids (SWP 3-1 and SWP 3-2) were investigated.

  • This experiment was conducted to investigate the effects of $\alpha$-amylase, added to artificial diet, on the larval growth and on the amount of blood sugar of the silkworm Bombyx mori L.. The artificial diet containing 20% mulberry leaf powder was fed on the 5th instar silkworm. The results obtained were summarized follows: 1. The digestibility in both sexes was increased by feeding artificial diet with $\alpha$-amylase and increase of the digestibility was greater in the female than in the male. 2. Larval body weight was increased in the female but was the male, compared with those in each sex of the control, respectively. 3. There was an increase of the single cocoon weight and cocoon shell weight, as compared with that of the control. 4. It was evident that the artificial diet containing $\alpha$-amylase was more effective than the control, and the effect of the diet was considerable higher in the female than in the male. 5. The amount of the blood sugar on the 7th day of 5th instar was decreased as compared with that of the control.

Biological Activity and Chemical Characteristics of Fermented Silkworm Powder by Mold (유용곰팡이 균주에 의한 발효 누에분말의 이화학적 특성 및 생리활성)

  • Five mold strains - Mucor racemousus, Rhizopus oryzae, Aspergillus oryzae, Aspergillus kawachii, and Monascus purpureus - were used for the fermentation of both freeze-dried and air-dried silkworm powders. The concentrations of proteins and minerals, electrophoretical protein patterns, fatty acid composition and the activities of fibrinolytic and antioxidation with freeze-dried silkworm (FDSW) or air-dried silkworm powders (ADSW) fermented by the five molds were investigated. The concentrations of major minerals in fermented FDSW and ADSW powders were K by 72.0-76.3 and 77.1-78.9 ppm, Mg by 29.6-49.7 and 44.3-58.7 ppm, Ca by 1.9-14.9 and 9.8-21.6 ppm and Zn by 0.64-0.70 and 4.17-4.52 ppm, respectively. Major fatty acids in fermented FDSW and ADSW powders were linolenic acid, oleic acid, and palmitic acid. There were slightly varietal differences in electrophoretical protein patterns when total protein patterns of fermented FDSW and ADSW powders were analyzed by native-and SDS-polyacrylamide gel electrophoresis (PAGE). DPPH radical scavenging activity was slightly stronger in fermented ADSW powders than that in fermented FDSW. Fibriolytic activity was only detected in the FDSW fermented by Aspergillus kawachi and Monascus purpureus. These results may provide the basic data to understand the biological activities and chemical characteristics of silkworm fermented by mold, which can be used for the development of functional foods.

A Study on the Quality Characteristics of Doenjang Prepared with Paecilomyces japonica, from Silkworm (누에 동충하초(Paecilomyces japonica)를 첨가하여 제조한 된장의 품질특성 변화에 관한 연구)

  • Change in Quality Properties of Doenjang Prepared with the Powder of Paecilomyces japonica and extract of P. japonica by different solvents were investigated during 90 days of fermentation at 20$^{\circ}C$. The moisture content was not significantly different, pH of Doenjang made with P. japonica was lower than control group and decreased continuously according to the fermentation time. Amino nitrogen continuously increased till 60 days and decreased slightly on 90 days. Value of L, a, b in Color decreased with proportion to fermentation period and L, a, b value of Doenjang made with P. japonica was lower and in Particular that of Doenjang made with P. japonica powder was lowest. From the result of sensory evaluation test, the color of control group was similar to “yellow”but that of Doenjang made with powder of P. japonica was close to“dark brown”and that of Doenjang made with extract of P. japonica was darker than that of control group and the preference of dark color was low. Texture was“glossy and smooth”in all and preference was high. In salty taste, Doenjang by P. japonica addition was stronger and Doenjang made with extract was stronger than that made with powder. Doenjang made with powder of P. japonica was weaker than other groups in sweet taste. In flavor and overall Preference, Doenjang made with P. japonica was scored lower slightly in than control group and Doenjang made with powder of P. japonica was the lowest in score.

Anti-Diabetic Effect of Sericultural Product in High Fat Diet-Fed Mice (고지방식이급여 마우스에서 잠상산물의 항당뇨 효능)

  • The objective of this study was to identify and compare the anti-diabetic effects of mulberry leave (ML), silkworm (SK), mulberry fruit (MF), and Cudrania tricuspidata BUREAU (CT) extracts in high fat diet (HFD)-induced obese and diabetic mice. C57BL/6N mice were assigned to six groups: normal diet (ND, n=7), HFD (n=10), HFD with 5% ML powder (ML, n=10), HFD with 2% SK powder (SK, n=10), HFD with 5% MF powder (MF, n=10), and HFD with 5% CT powder (CT, n=10). Mice were fed their assigned diet for 14 weeks. ML group showed significant reduction in levels of plasma glucose and insulin compared with the HFD group. Plasma total cholesterol (T-C) was significantly reduced by ML and SK compared with the HFD group. Plasma high-density lipoprotein cholesterol (HDL-C) and HTR (HDL-C to T-C ratio) levels of the ML, SK, MF, and CT groups were significantly elevated compared to the HFD group. Moreover, concentrations of hepatic T-C and triglycerides in the ML, SK, MF, and CT groups were significantly reduced in comparison to the HFD group. Levels of pAKT, pS6K, and pAMPK significantly increased in the ML group compared with the HFD group. Taken together, ML appears to be the most potent anti-diabetic and anti-dyslipidemic substance among sericultural products. ML could be developed as a potential agent for diabetes and its complication management.

  • To obtain basic data for mass production of Isaria japonica, cultural characteristics of japonica were investigated by using liquid, solid media and silkworms pupa. Mycelia grew favorably at the temperature of $23{\sim}28^{\circ}C$ on MYG medium with pH 7.0. The fruiting-body of I. japonica was induced below $20^{\circ}C$ in MYG liquid medium (Malt yeast glucose) under fluorescent light. In MYG basal medium mixed with pupal powder of silkworms, the fresh weight of fruiting-bodies was increased with increasing concentration of pupal powder. The highest yield of fruiting bodies was obtained in carbon-rich medium supplemented with pupal powder of silkworm. Also, fruiting-bodies of I. japonica were produced massively on the silkworm pupa placed on the stainless tray in the shortest time. The structure and shape of fruiting-bodies were coral-like, many-branched types with numerous conidiospores.

Fatty Acid Composition of Adipose Tissues in Obese Mice and SD Rats Fed with Isaria sinclairii Powder

  • Isaria sinclairii (Cicada Dongchunghacho) was studied as a potential crude natural food in powdered form. The role of tissue fatty acids in relation to the anti-obesity effects of I. sinclairii (IS) was examined by feeding the powder to SD rats ad libitum at 0, 1.25, 2.5, 5 and 10% (calculated about 8 g/kg) of the feed for a period of 3 months and 6 months. The fatty acid composition profile as indicated GC-MS, showed significantly slight dose-dependent increases in the levels of unsaturated fatty acids, particularly, arachidonic acid (C20: 4n6), oleic acid, linoleic acid, eicosadienoic acid, eicosapentaenoic acid (EPA) (C20: 5) concentration in the the ad libitum IS-fed groups compared to the control group in SD abdominal fat over 6 month period. Over viewing of the SD and Ob mice treated Isaria sinclairii powder; there were increases in the single (mono) unsaturated fatty acids ratio but decreases in polyunsaturated fatty acid. In IS-fed groups in proportion to the treatment period, this Dongchunghacho also induced an increase in the level of same result of unsaturated fatty acid in C57BL/6 obese (ob/ob) mice over a 6-month period treatment compared to those given 10% dry mulberry leaf powder (ML) or silkworm powder mixed with the standard diet.

  • This study was conducted to investigate the effects of Cordyceps ochraceostromat, silkworm cocoon, and conjugated linoleic acid (CLA) on the quality and storage properties of pork sausage manufactured with protein recovered from breast of spent laying hen during 4 wks of storage at $4^{\circ}C$. Pork sausages were prepared using 100% ham (control) and 40% recovered protein from breast of spent laying hen to replace pork (T1), and with added different sources to final concentrations of 0.1% Cordyceps ochraceostromat powder (T2), 0.1% silkworm cocoon powder (T3), 0.1% CLA (T4), 0.05% Cordyceps ochraceostromat + 0.05% silkworm cocoon (T5), 0.05% Cordyceps ochraceostromat + 0.05% CLA (T6), and 0.05% silkworm cocoon + 0.05% CLA (T7). The treatments T5 and T7 had higher (p<0.05) protein content than control, but control had lower fat content than other samples during 4 wks of storage at $4^{\circ}C$. Lightness was significantly lower in the treatment samples than control. However, there was no significant difference in water holding capacity between the sausage samples, whereas, cohesiveness and chewiness were significantly higher (p<0.05) in the control than other treatments. All sausage samples showed a significant increase in volatile basic nitrogen (VBN) and total plate counts with extending storage time (p<0.05), and VBN values of treatments were lower than the control. However, the treatment samples showed a significant decrease (p<0.05) in thiobarbituric acid reactive substances over the increasing storage time. Therefore, our results suggested that the 40% recovered protein to replace pork and with added different sources decreased lipid oxidation and protein denaturation of pork sausages, thereby enhancing self-life, compared to normal pork sausage (control).

Anti-obesity Effect of Ethanol Extracts from Silkworm (Bombyx mori) Pupae Powder Fermented with Cordyceps militaris in the Primary Adipocytes and High Fat Diet-induced Obesity Model Mice (일차 지방세포와 고지방식이로 유발한 비만모델동물에서 Cordyceps militaris로 발효시킨 누에 (Bombyx mori) 가루 에탄올 추출물의 항비만 효과)

  • Silkworm (Bombyx mori) pupae have been widely worked in wound dressing, hepatoprotective activity, antigenotoxicity, control of glucose level and anti-cancer activity. To investigate the anti-obesity activity of ethanol extract of silkworm pupae powder fermented with Cordyceps militaris (ESfC), the free glycerol release and cAMP concentration as well as fat accumulation were measured in the primary adipocytes of SD (Sprague Dawley) rats and high fat diet (HFD)-treated C57BL/6 mice treated with 12 weeks. Firstly, the presence of the cordycepin with lipid lowering effect was confirmed in ESfC using HPLC analysis. The level of free glycerol and cAMP concentration was significantly increased in the primary adipocytes treated with high dose of ESfC ($400{\mu}g/ml$) although these levels were consistently maintained in other dose ESfC treated groups. In HFD-induced obesity model, the increased fat weight and size of adipocytes in HFD+Vehicle treated group was recovered in HFD+ESfC treated group. Also, the liver weight and the number of lipid droplets were higher in HFD+Vehicle treated group than No treated group. But, this level was significantly decreased in HFD+ESfC treated group compared with HFD+Vehicle treated group. Furthermore, a similar recovery was detected on the phosphorylation of periliphin and HSL, and ATGL expression. Overall, the results of the present study provide some scientific evidences that ESfC can stimulate lipolysis in primary adipocytes and prevent fat accumulation in HFD-treated obesity model, and therefore have the potential for use as anti-obesity agents to treat obese patient.
