Functional Silk Proteins: Molecular Structure and Application to Biomaterials

  • Silk proteins consist of two major proteins, fibroin and sericin. There is currently an enormous reawakening of interest in these silk proteins as a biomaterial due to their mechanical and biological properties based on the detailed findings. Novel method for determination of the crystalline structure of silk proteins in an atomic level using nuclear magnetic resonance (NMR) was reviewed. Recent application of silks to biomaterials and prospects for future were discussed.

Effect of Processing Conditions on the Homogeneity of Partially Degummed Silk Evaluated by FTIR Spectroscopy

  • The partial degumming of silk has recently attracted researchers' attention because of its ability to produce silk textiles with new tactile properties, intermediate between the softness of fully degummed silk and the hardness of raw silk. However, it is difficult to obtain partially degummed silk in a homogenously degummed state due to the heterogeneous character of sericin removal. It is also difficult to examine the homogeneity of degumming. In the present study, Fourier transform infrared (FTIR) spectroscopy with attenuated total reflection (ATR) geometry was used to evaluate the effect of processing conditions on the degumming of silk yarns. The crystallinity index, calculated from FTIR spectra, showed an increase with the degumming ratio. Therefore, the homogeneity of degumming could be evaluated by the variation of crystallinity index for 30 different spots in silk yarns. The homogeneity of degumming was influenced by the total degumming time, the content of surfactant, and the liquor rate. No effect was observed upon changing the number of degumming cycles at the same total degumming time.

  • In our previous studies, dietary supplements of silk protein, sericin, and fibroin, were beneficial for improving epidermal levels of ceramides, which are the major lipids for maintaining the epidermal barrier. In this study, we investigated the dietary effects of silk protein on epidermal levels of free sphingoid bases and their phosphates such as $C_{18}$ sphingosine (So), $C_{18}$ sphinganine (Sa), $C_{18}$ sphingosine-1-phosphate (S1P), and $C_{18}$ sphinganine-1-phosphate (Sa1P), which are either synthetic substrate or degradative metabolites of ceramides. Forty-five male NC/Nga mice, an animal model of atopic dermatitis (AD), were divided into three groups: group CA was an atopic control and fed a control diet, group S was fed a 1% sericin diet, and group F was fed a 1% fibroin diet. Fifteen male BALB/c mice served as group C (control group) and were fed the control diet. All mice were fed with diets and water $ad$ $libitum$ for 10 weeks. Sa in group CA was lower than that in group C, but So in group CA was similar to that in group C. So and Sa were higher in groups S and F than those in group CA; So level was even higher than that in group C, and Sa level was similar to that of group C. The So/Sa ratio in group CA, which is reported to increase in AD, was significantly higher than that of group C. The So/Sa ratio was lower in groups S and F than that in group CA, and decreased further in group F. However, S1P and Sa1P in groups S and F were similar to those in group CA. Taken together, we demonstrated that silk protein, sericin and fibroin dietary supplements, increased So and Sa levels, and decreased the So/Sa ratio.

제사용수의 수질이 견층 세리신(Sericin) 용해에 미치는 영향 (제2보) (Property of Filature Water against the Solubility of Cocoon Layer Sericin Seoul National Silk Conditioning House)

  • 1. 용수의 M-알카리도가 높을수록 견층 sericin의 용해도는 급격히 증가 하였다. 2. 처리후의 시수 조사결과 M-알카리도 25p.p.m. 이상애서 산도는 완전히 중화되었다. 3. 시수의 M-알카리도가 상승함에 따라 처리후의 M-알카리도도 증가 하였다. 즉 견층을 처리한 시수의 M-알카리도가 높을수록 견층내의 M-알카리도 용출량도 증가하는 경향이 있었다. 4. 총경도는 시수의 M-알카리도가 증가 하는데 따라 감소하는 경향을 나타 내었다. .5. 用水의 산도가 높을수록 견층 sericin의 용해도는 현저히 감소되는 경향을 나타내었다. 6. 공시견 처리후의 수질변화를 보면 시수의 산도가 증가함에 따라 처리후 시수의 산도와 경도도 현저히 상승하는 반면 M-알카리도는 시수의 산도 20~40p.p.m. 에서 완전 중화되어 40p.p.m. 이상의 시수에서는 처리후 M-알카리도를 발견할 수 없었다. 7. 용수율의 철분 함유량이 증가하는데 따라 견층 sericin 용해도는 서서히 감소되는 경향이 있었다. 또한 망강에서는 용수중의 망강이 증가하는데 따라 견층 sericin 용해도는 다소 감소 하였으나 그 경향은 매우 완만하였다. 8. 견층 처리후의 수질변화를 보면 산도 M-알카리도 및 총경도가 상당량 견층에서 용출된 것을 볼 수 있었으나 이것은 시수의 pH 및 처리온도에 의한 것으로 보여지며 철분 함유량과는 아무런 관계도 인정할 수 없었다. 또한 망강에서는 시험후 수질변화를 조사한 결과 시수의 망강 함유량이 증가 하는데 따라 처리후의 M-알카리도는 감소 하였으나 산도 및 총경도는 증가되는 경향을 볼 수 있었다. 9. M-알카리도와 총경도를 변화하여 시험을 행한 결과 sericin 용해량이 증대되었다. 10. pH 및 총경도를 고정하고 M-알카리도 만을 변화시킨 경우 M-알카리도가 증가할 수록 sericin 용해도도 증대 되었다. 11. pH 총경도 및 M-알카리도를 고정하고 산도를 변화하여 시험한 결과 sericin 용해가 상당히 억제 되었다. 12. pH(6.8), M-알카리도를 고정하고 총경도만을 변화시킨 경우 sericin의 용해가 상당히 억제 되었다. 13. 각각의 수질 항목별로 sericin 용해에 미치는 작용면을 기본으로 분류하고 다시 "용해력증대" "용해력 보통" "용해력감소"의 구로 복합수질을 조제하여 시험 용해력 증대의 구는 용해력 감소의 구에 비하여 약 2.2%의 sericin 용해가 증대 되었으며 용해력 보통의 구에 있어 sericin 용해량은 2.4~2.9% 정도였다. 14. 4항 및 5항의 시험결과 견층 sericin 변성에 가장 크게 영향을 미치는 제사용수의 성분은 pH, M-알카리도, 산도 및 총경처이므로 앞으로 이상의 4개 항목을 기본으로 한 제사시험을 행하고 이 결과에 따라 최적의 제사용수 수질 조건을 설정코저 한다. 15. 동일 시수를 가지고 견층을 반복처리 하는데 따라 시수내의 완충력이 증가하여 견층 sericin의 용해량은 감소하는 경향이 있었다.견층 sericin의 용해량은 감소하는 경향이 있었다.

실크 세리신 단백질 가수분해물을 이용한 유기 칼슘제의 제조 및 칼슘 결핍 쥐에서의 생체 이용률 (Manufacturing of Calcium Binding Peptide using Sericin Hydrolysate and Its Bioavailability in Calcium Deficient Rat)

  • Silk sericin protein was hydrolyzed by seven proteolytic enzymes in order to examine the effectiveness of the hydrolysates in binding calcium. The amino acid nitrogen content of hydrolysates from Flavourzyme was higher than that for other enzymes, and its calcium binding capacity showed a dose-dependent increase. We examined the effects of calcium binding peptide from sericin hydolysates on the bioavailability of Ca-deficient rats. Three-week-old male rats were fed an Ca-deficient diet for three weeks. Rats were divided into four groups (DD: non-treated group on calcium deficient diet; DD+MC: milk-calcium treated group; DD+OC: organic calcium made using sericin hydolysates; and DD+IC: inorganic calcium ($CaCl_2$). After oral administration of calcium supplements for one week, the calcium content of the serum and liver were significantly higher in DD+OC ($101.7{\mu}g$/mL and $49.3{\mu}g$/mL) and DD+MC ($83.6{\mu}g$/mL and $42.8{\mu}g$/mL) than DD ($86.3{\mu}g$/mL and $43.4{\mu}g$/mL). The alkaline phosphatase (ALP) content in the treated groups was significantly lower than DD, but no significant difference among groups was shown. Aspartate aminotransferase (AST) levels did not show any significant difference between groups. Alanine aminotransferase (ALT) levels were significantly reduced compared to the DD group. In conclusion, binding calcium to peptides from sericin hydrolysates seems to improve its bioavailability, and to hasten the cure of calcium deficiency in experimental rats.

실크 세리신의 가수분해 특성과 응용 I. 세리신의 분자량 제어 (Hydrolysis characteristics and applications of silk sericin I. Control of molecular weight of sericin)

  • 정련 액에서 수용성 세리신을 얻고, 남은 난용성 세리신을 여러가지 효소로 가수분해하여, 효소 가수분해 특성을 알아보고. 아미노산 조성과 분자량을 조사하여 다음과 같은 결론을 얻었다. 1. 효소 가수분해 시 kojizyme과 flavourzyme의 최적조건은 pH 7, 온도 5$0^{\circ}C$이었고, protamex와 alcalase는 처리 pH의 증가에 따라 활성이 높았고 최적온도는 6$0^{\circ}C$이었다. 2 효소 가수분해 시 용해도 측정에서는 효소 처리농도 증가에 따라 용해도도 높았으며, 4시간 이상의 처리에서는 아주 소폭으로 용해도가 증가하였다. 3. 효소 가수분해 시 흡광도를 이용한 용해도 측정에서는 효소 처리농도 증가에 따라 흡광도도 높았으며. 4시간 이상의 처리에서도 계속하여 흡광도가 증가하였다. 4. 효소 가수분해 시 말단기 정량에 의한 평균중합도 측정에서는 처리시간과 농도의 증가에 따라 중합도가 감소하였다. 5. 아미노산 분석 결과 정련 후 채취한 low와 high의 아미노산 조성이 비슷하고, PK와 PP및 PA의 아미노산 조성이 비슷하였으며, low와 high는 PK, PP, PA보다 세린과 타이로신의 함량이 많았고, 프롤린은 검출되지 않았다. 6. GPC에 의한 분자량 측정결과 중량평균분자량(Mw) 크기는 PK>high>PP>low>PA 순서이었으며, PK와 PA의 Mw는 각각 9,800, 905이었다

Enzymatic hydrolysis of insoluble silk sericin by Alcalase

  • This study was undertaken to figure out the effects of hydrolysis conditions on the solubility of insoluble sericin, molecular weight distribution and thermal characteristics of hydrolysates in enzymatic hydrolysis by Alcalase 2.5L. It was indicated that the optimum treatment temperature and pH for the insoluble sericin were 50$\^{C}$ and 11, respectively. When the insoluble sericin was hydrolyzed with a various treatment conditions, the solubility of all hydrolysates were represented above 85% at given conditions. As the enzyme concentration increased, the solubility increased roughly, but the solubility increasement ratio was less above 2% enzyme concentration. As the treatment time increased, the solubility was also increased. It was showed in the molecular weight distribution of hydrolysates treated various enzyme concentrations and treatment times that when enzyme concentrations were 0.5, 2, 3%, the peaks of the distribution curve were shifted to left side which meant low molecular weight and was distributed much quantity with shifted to be left side, but treatment time was 6 hr. the peak was shifted to right side. When enzyme concentration was 5% and treatment time was below 2 hr., the peaks were shifted to right side, but treatment time was above 4hr. the peak was shifted to left side. The number-average molecular weights were distributed from 300 to 800 and those were decreased when treatment time was up to 4 hr., but increased a little when treatment time was 6hr. It was showed in the DSC curves of hydrolysates treated with treatment time of 0.5, 1, 2, 4, 6 hr. fixed 1% o.w.s enzyme concentration and control that the endothermic peak was observed near at 200$\^{C}$. The denaturation peak of the hydrolysates depending on treatment times had a tendency to shift to higher temperature. But, when the treatment time was 6 hr., the peak was shifted to lower temperature comparing another hydrolysates.

  • Realizing the scope of utilizing by-products of silk cocoons by applying appropriate methods is the immediate crave to optimize returns. The nutritive value of pupae suits for human diet, feed for poultry, carps, fish, rabbits, piggery and dogs. The pupal skin, fat, oil, cocoon palade have applications in oleo chemical, soap, glycerin, cosmetic, artificial fibres, membranes and n-triacontanol isolation. The pupal proteins Chitin, Shinki fibroin, Serrapeptidase, glucosamine are latent precursors of post surgical, anti-carcinogenic, anti-inflammative, anti-bacterial, anti-histaminic, gastric, hepatitis, pancreatitis, leukocytopenia, neurological, ophthalmic, blood pressure, cardiac and diabetic medicines and for preparation of vitamins A, E and K. The silk and its proteins sericin and fibroin are potentially used for wound healing, diabetes, impotence, sinusitis, arthritis, edema, cystitis, epididymitis, tissue regeneration, cancer, post-surgical trauma and used as anti-oxidatives, bio-adhesives, ultra violet screens and bio-active textiles. The waste cocoons can be used in making art crafts like garlands, carpets, overcoats, decoratives and greeting cards. The in-depth research towards utility optimization and make aware this reality to sericulturists, reelers, weavers, traders, entrepreneurs, policy makers etc., is the upright want of the today's Sericulture industry.

Bacillus licheniformis 단백질 분해 효소에 의한 정련 견사의 특성 II. 정련 견사의 열처리 영향 (Physicochemical Characteristics of Silk Fibroin Degummed by Protease in Bacillus licheniformis II. Effect of Heat Treatment onto Degummed Silk Fiber)

  • 비누 정련 견사와 효소 정련 견사의 결정 구조는 정련 구조 중 처리온도의 차이에 의하여 효소 정련 견사의 결정성이 비누 견사의 결정성 보다 높다고 전보(김·남, 1992)에서 보고하였는데 이와같은 정련 견사의 결정성의 차이가 정련 견사를 열처리를 하였을 때 어떻게 변화하는지를 밝히기 위하여 두가지 방법으로 정련한 견사에 대하여 습열 처리와 동결 처리한 후 특성을 분석한 결과는 다음과 같다. 1. 정련 견사를 습열 처리한 경우 강력과 신도는 모두 저하되었으며 비누 정련 견사는 습열 처리온도의 상승과 함께 열 분해 온도도 고온측으로 이동하고 IR crystallinity도 증가하였으나 효소 정련 견사는 습열 처리에 의하여 열 분해 온도와 IR crystallinity의 큰 변화가 없었다. 이와 같은 결정 구조의 차이는 비누 정련 견사는 습열 처리로 재배향 결정화가 일어나 결정화도는 증가되었으나 효소 정련 견사는 습열 처리에 의한 재배향 결정화보다 습열 처리에 의하여 팽윤되어 수축하는 과정에서 배향성이 저하되기 때문에 결정성은 더 이상 증가되지 않은 것으로 추측된다. 2. 정련 견사를 동결 처리하였을 때 강력과 신도는 약간씩 감소하는 경향이었으나 열 분해 온도와 IR crystallinity는 비누 정련 견사와 효소 정련 견사 모두 큰 변화가 없었다.
