Studies on Toxic Components of Auricularia polytricha (털목이버섯의 독성(毒性)에 관한 연구(硏究))

  • To screen biologically active components of the higher fungi of Korea, the dried carpophores of Auricularia polytricha were extracted with water. The extract was examined for acute toxicity in ICR mice. A low molecular weight toxin of this fungus was purified by acetone precipitation followed by cellulose, silica gel and LH-20 Sephadex column chromatography. Major symptoms of this toxin were eye extrusion, hair erection, trembling of head, paralysis, rapid running or moving before death and depression of respiration. The median lethal doses of the total extract were 1.25 g/kg and 4.18 g/kg by i.p. and p.o. administrations, respectively. The amounts of one mouse lethal unit (MLU) of the total extract and final fraction that killed a 20-g mouse within 30 minutes were 28.5 mg/mouse and 12 mg/mouse, respectively.

  • The microencapsulation of droplets or particles within a solid shell leads to the formation of core-shell particles. Microencapsulation provides protection and controlled release of core materials such as drugs, vitamins, enzymes, perfumes, and the like. Such particles have, therefore, found a diverse range of applications in the pharmaceutical, agricultural, cosmetic, and food industries. UV absorbers are widely used for cosmetics to screen out ultra violet (UV) rays which have side effects on human skin. The absorbers are made generally from synthetic organic compounds, which can stimulate the human skins to develop allergic phenomena.

한국산 도꼬마리 추출물의 항균효과 및 분리 정제

  • Antimicrobial activity of various extracts of Xanthium strumarium L. was tested against 25 strains of bacteria, yeast and fungus. The crude ethylacetate extract exhibited strong growth inhibition to the tested strains with the exception of partial Gram-negative bacteria. The property of antimicrobial compound was very stable under heat treatment at $120^{\circ}C$, but it was unstable in acid (pH 3.0) and alkali (pH 10.0) treatment. The antimicrobial compounds were purified by boiling water extraction, ethylacetate extraction, charcoal column chromatography, silica gel column chro- matography and reverse phase HPLC. The purified compound A and B were detected in a single peak (each above 98% purity) through the HPLC analysis. The compound A and B showed a strong growth inhibition against Gram-negative and positive bacteria in the agar diffusion method. When tested by the FDA method using the esterase, compound A mainly inhibited the growth of bacteria and compound B showed the growth inhibition of both bacteria and yeasts.

Isolation and Identification of Antitumor Promoters from the Seeds of Cassia tora

  • A methanol extract of Cassia tora seeds was successively partitioned with diethyl ether, chloroform, ethyl acetate, and water, and the antitumor-promoting activity of the solvent fractions was determined by inhibition of Epstein-Barr virus early antigen (EBV-EA) activation induced by teleocidin B-4 in Raji cells. The diethyl ether (68.7%) and chloroform (91.2%) fractions and the hydrolysate (94.3%) of the ethyl acetate fraction had strong inhibitory activities. The chloroform and ethyl acetate fractions were chromatographed on silica gel and further purified by HPLC. Three active compounds, obtusifolin-2-glucoside (75.0%), chryso-obtusin-6-glucoside (56.8%), and norrubrofusarin-6-glucoside (39.4%), were obtained from the ethyl acetate fraction, and two active compounds, questin (97.9%) and chryso-obtusin (53.8%), were isolated from the chloroform fraction.

  • A chiral stationary phase (CSP) prepared by bonding (18-crown-6)-2,3,11,12-tetracarboxylic acid (18-C-6-TA) to aminopropyl silica gel by HPLC was used in resolving several racemic drugs containing primary amino moiety. Most compounds used in this study were resolved on the CSP using 80% methanol in water (V/V) containing 10mM sulfuric acid as a mobile phase. These results on the CSP were compared to those on the similar CSP derived from 18-C-6-TA of the same chiral selector by different connecting method.

  • Dried roots of Paeonia japonica were extracted with dichloromethane, methanol and water aerially. Silica gel column chromatography and thin layer chromatography were used to separate the fractions with antimicrobial activities, and mass spectrometry was used to determine the mass. Dichloromethane extract showed the highest antimicrobial activity. Dichloromethane extract from Paeonia japonica could be a candidate for a new antimicrobial agent against MRSA.

  • The essential oils of Lycopus lucidus have been studied. The essential oil obtained from the dried herba by steam distillation followed by fractionation by column chromatography was analysed by gas chromatography-mass spectrometry(GC-MS). The compounds identified by GC-MS were carvacrol, 2,5-dimethoxy-p-cymene, trans-caryophyllene, spathurenol and $trans-{\beta}-farnesene$. Two compounds, thymol and caryophyllene were isolated by silica gel column chromatography and analyzed by TLC, IR, Mass and NMR. And the weak diuretic effects of essential oil and water extract from the dried drugs were observed in rats.

(+)-Catechin, an Antioxidant Principle from the Leaves of Pinus densiflora that Acts on 1,1-Diphenyl-2-picrylhydrazyl Radical

  • The antioxidant activity of Pinus densiflora was determined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals. The methanol extract of P. densiflora showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate>n-butanol>water>dichloromethane> n-hexane fraction. The ethyl acetate soluble fraction exhibiting strong antioxidant activity was further purified by repeated silica gel column chromatography. Antioxidant (+)-catechin was isolated as one of the active principles from the ethyl acetate fraction, together with the inactive components, dihydrokaempferol and 1-O-benzoylglucose. The radical scavenging effect of (+)-catechin on DPPH radical exceeded to that of L-ascorbic acid which is a well known antioxidant.

Antioxidative Activity on Human Low Density Lipoprotein(LDL) Oxidation by 2,6-Dimethoxyphenol Purified from Bacillus sp. KS-96

  • This study was designed to develope the antioxidative activity on oxidation of human low density lipoprotein(LDL) from marine microbials. Bacillus KS-96 producign antioxidant have been isolated and identified from seawater, Bacillus sp. KS-96. The optimal medium pH was 7.0 and incubation temperature was 30$^{\circ}C$. The antiosidant of potential substance produced extracellularly in the culture broth by Bacillus sp. KS-96 was obtained by elution of silica gel culumn chromatography with hexane, ethylacetate and water. The ethylacetate faction are shown at highest level of antioxidant activity using thiocyanate method among them. By IR, NMR, and GC/MS, antioxidant purified from ehtylacetate fraction was identified and named as 2,6-dimethoxyphenol. 2,6-dimethoxyphenol inhibited the metal mediated oxidation of human LDL at concentration of 50∼100 ${\mu}$g/mL in the presence of 5uM CuSO4 with macrophage or J774 cells.

Identification of Cinnamon Components and Quantitative Determination of Cinnamlc Acid from Crude Drug Drink Preparations (생약복방 드링크제 중 계피성분의 확인 및 계피산의 분리정량)

  • For the quality control of crude drug drink preparations, methods for identification of cinnamon components and quantitative determination of cinnamic acid were established. Cinnamon components were identified by TLC with benzene/ethyl acetate (1:1, v/v) on silica gel plate by spraying p-anisaldehyde-sulfuric acid. Cinnamic acid contents were determined at UV 280 nm by HPLC on $\mu$-Bondapak Cls column with acetonitrile/water/acetic acid (40:60:2, v/v). Recoveries of cinnamic acid in three crude drug drink preparations were between 84.1-90.2% compared to the content of the cinnamon extract.
