A Case of Short-chain Acyl-CoA Dehydrogenase Deficiency Detected by Newborn Screening

  • Short-chain acyl-CoA dehydrogenase (SCAD) deficiency is an autosomal recessive mitochondrial disorder of fatty acid oxidation associated with mutations in the ACADS gene. While patients diagnosed clinically have a variable clinical presentation, patients diagnosed by newborn screening are largely asymptomatic. We describe here the case of a 1-year-old male patient who was detected by newborn screening and diagnosed as SCAD deficiency. Spectrometric screening for inborn errors of metabolism at 72hrs after birth showed elevated butyrylcarnitine (C4) level of 1.69 mol/L (normal, <0.83 mol/L), C4/C2 ration of 0.26 (normal, <0.09), C5DC+C60H level of 39 mol/L (normal, <0.28 mol/L), and C5DC/C8 ration of 7.36 (normal, <4.45). The follow-up testing at 18 days of age were performed: liquid chromatography tandem mass spectrometry (LC-MS/MS), urine organic acids, and quantitative acylcarnitine profile. C4 carnitine was elevated as 0.91; urine organic acid analysis showed elevated ethylmalonic acid as 62.87 nmol/molCr (normal, <6.5), methylsuccinate 6.81 nmol/molCr (normal, not detected). Sequence analysis of ACADS revealed a homozygous missense mutation, c.164C>T (p.Pro55Leu). He is growing well and no episodes of seizures or growth retardation had occurred.

Nutrient Digestibilities and Fecal Characteristics of Diets Including Brown Rice for Miniature Poodle (Miniature Poodle에 있어서 현미 함유사료의 영양소 소화율 및 배설 분 특성)

  • In this experiment, two inclusion levels (15 and 30% of diets, as-fed basis) of brown rice (BR) were tested against a control diet with 0% BR (51% wheat flour of diet). Six female Miniature Poodles (8~9 months age, initial mean body weight of 3.5 kg) were assigned to treatments in replicated $3{\times}3$ Latin square design. Total tract digestibilities of DM, OM, CP, acid hydrolyzed fat and gross energy increased linearly (P<0.05), and observed digestible energy and metabolizable energy values also increased linearly (P<0.001 and P=0.007, respectively) with increasing BR inclusion levels. Wet and dry fecal output decreased linearly (P=0.001) with increasing BR inclusion levels in the diets. Linear (P<0.05) effects was observed in fecal score for dogs fed BR and fecal ammonia concentration increased linearly (P<0.05) in response to increasing BR inclusion level. It seems that the increase in fecal ammonia concentration may be partially related to the decrease in short-chain fatty acid concentration (P<0.01). This study clearly demonstrates that BR improves nutrients digestibility and fecal characteristics of dog.

  • In this experiment, two inclusion levels (15 and 30% of diets, as-fed basis) of brown rice (BR) were tested against a control diet with 0% BR (51% wheat flour of diet). Six female Maltese (8~9 months age, initial mean body weight of $2.8{\pm}0.3$ kg) were assigned to treatments in replicated $3{\times}3$ Latin square design. Total tract digestibilities of DM, OM, acid hydrolyzed fat, CP and gross energy by dogs fed experimental diets responded quadratically (P<0.01 or <0.05) to BR inclusion levels. Similarly, quadratic responses (P<0.001, P=0.015) were observed for digestible energy and metabolizable energy values, respectively. Wet and dry fecal output also exhibited quadratic responses (P=0.006, P=0.014, respectively) but no differences were observed between control and BR 30% diets. Linear (P=0.008) effect was observed for fecal score in dogs fed BR, and fecal ammonia concentration increased linearly (P=0.001) in response to increasing BR inclusion levels. It seems that the increase in fecal ammonia concentration may be partially related to the decrease tendency (P=0.07) in short-chain fatty acid concentration. In this experiment, inclusion of BR in dog diet did not result in pronounced changes in the digestibility but in fecal score. Although inclusion of BR in diets showed profound positive effect on fecal score, it is confounded by the high inclusion level of poultry offal meal.

재활용을 위한 ${\beta}-Cyclodextrin$의 Adipic acid에 의한 가교화와 이를 이용한 크림치즈의 관능적 특성

  • 본 연구결과, 콜레스테롤 제거 크림치즈를 만들기 위한 유지방 함량 36%의 크림의 콜레스테롤 제거 최적 조건은 adipic acid로 가교시킨 ${\beta}-CD$ 첨가량 10%, 교반온도 $20^{\circ}C$, 교반시간 30분, 교반속도 800rpm 이었으며, 이 때 콜레스테롤 제거율은 평균 91.42%로 나타났다. 가교화 ${\beta}-CD$ 처리 크림치즈의 관능검사 결과, 가교화 ${\beta}-CD$ 처리 크림치즈는 control과 powder ${\beta}-CD$ 처리한 크림치즈에 비해 저장 기간 동안 쓴맛의 증가가 거의 없었고, 빵에 발리는 정도도 매우 우수하였으며, 전체적인 기호도 또한 높았다. short-chain fatty acid의 경우 저장 기간이 지남에 따라 가교화 ${\beta}-CD$ 처리 크림치즈는 control과 powder ${\beta}-CD$ 처리한 크림치즈에 비해 저급 지방산 생성에 변화가 거의 없었다. 조직검사에서 가교화 ${\beta}-CD$ 처리 크림치즈는 저장 기간 동안 control에 비해 응집성, 탄력성, 점착성의 변화가 없으며 그 수치가 높게 평가되었다. 또한 가교화 ${\beta}-CD$의 재활용률은 97.82%로 매우 높게 나타났다. 위 실험 결과에 따르면, 가교화 ${\beta}-CD$는 콜레스테롤 제거율이 높으며, 제품에 적용 시 품질이 향상될 뿐만 아니라, 재활용률도 높아 이를 유가공 산업에 다양하게 활용이 가능할 것으로 기대된다.

  • 본 연구는 $\beta$-CD을 이용한 체다. 치즈의 숙성의 촉진을 위한 최적조건을 확립하기 위하여 가염의 양, 가온온도, 가온시간을 변화요인으로 연구하였다. 가염의 농도는 증가할수록 쓴맛이 낮아지는 경향을 확인하였고 hardness는 단단해지는 경향을 확인하였다. Curd의 가온시 온도가 높을수록 hardness가 가장 긍정적인 경향을 나타냈다. 가온시간은 길어질수록 short-chain free fatty acids의 양이 가장 적게 나타났다. 따라서 가염을 3%까지 증가시키고 Curd의 가온온도를 $42^{\circ}C$까지 높이면서 가온시간을 60분 까지 증가시킨다면 $\beta$-CD을 첨가한 체다. 치즈의 부스러지는 조직과 강한 쓴맛을 약화시키는 효과를 가져오면서 숙성을 촉진시킬 수 있다고 사료된다.

한국인 분변에서 분리한 Bifidobacteria의 탈지유에서의 배양특성

  • Wild strains of bifidobacteria isolated from Korean feces were tested for their growth and acid production abilities in 10% skim milk. Growth of bifidobacteria was markedly decreased from the second transfer in the skim milk culture. When two strains, BF5 and BF33, were grown in skim milk with various supplements, the growth was enhanced by supplementation of 0.5% yeast extract, and 0.05% cysteine but not by short chain fatty acids. There was no enhancing effect of CO$_{2}$, substitution in the fermentor on growth. The viable cell counts of bifidobacteria, BF5 and BF33, were 9.76 and 9.98 logCFU/ml, respectively, after 30 hr cultivation and were diminished by 3 and 6 logs during storage at 5$\circ$C for 12 days.

Stunting and Gut Microbiota: A Literature Review

  • Stunting, a condition characterized by impaired growth and development in children, remains a major public health concern worldwide. Over the past decade, emerging evidence has shed light on the potential role of gut microbiota modulation in stunting. Gut microbiota dysbiosis has been linked to impaired nutrient absorption, chronic inflammation, altered short-chain fatty acid production, and perturbed hormonal and signaling pathways, all of which may hinder optimal growth in children. This review aims to provide a comprehensive analysis of existing research exploring the bidirectional relationship between stunting and the gut microbiota. Although stunting can alter the gut microbial community, microbiota dysbiosis may exacerbate it, forming a vicious cycle that sustains the condition. The need for effective preventive and therapeutic strategies targeting the gut microbiota to combat stunting is also discussed. Nutritional interventions, probiotics, and prebiotics are among the most promising approaches to modulate the gut microbiota and potentially ameliorate stunting outcomes. Ultimately, a better understanding of the gut microbiota-stunting nexus is vital for guiding evidence-based interventions that can improve the growth and development trajectory of children worldwide, making substantial strides toward reducing the burden of stunting in vulnerable populations.

Total Lipid and Total Fatty Acid Composition and Vitamin E Content of Human Mature Milk, Infant Formulas and Market Milk (인유, 조제 분유 및 시유의 총지방질 조성, 총지방산 조성 및 비타민E 함량의 비교)

  • The lipid and fatty acid compositions and vitamin E content were analyzed in 48 human mature milk, 3 infant formulas (modified milk powder) and 8 market milk samples. The total lipid content in modified milk powder was similar those in human milk and market milk when total solids content of modified milk powder was corrected to that of human milk. In comparison with human milk, modified milk powder contained a lower proportion of triglycerides and higher proportions of phospholipids, free fatty acids and cholesterol esters. The ratios of phospholipids/triglycerides, total cholesterol / triglycerides and total tocopherol/total lipids in modified milk powder were significantly higher than those in human milk and market milk. The American recommendation for linoleic acid (0.7 IU/g) could be satisfied with human milk, modified milk powder and market milk. The proportions of short- and medium-chain even numbered saturates were higher and the proportions of long-chain derivatives of linoleic (${\omega}6$ series) and linolenic (${\omega}3$ series)acids were lower in modified milk powder and market milk than in human milk. It is concluded that in view of their levels in breast milk, the polyenoic derivatives of linoleic and linolenic acids must be taken into account when assessing infant foods.

Gut microbiome-produced metabolites in pigs: a review on their biological functions and the influence of probiotics

  • The gastrointestinal tract is a complex ecosystem that contains a large number of microorganisms with different metabolic capacities. Modulation of the gut microbiome can improve the growth and promote health in pigs. Crosstalk between the host, diet, and the gut microbiome can influence the health of the host, potentially through the production of several metabolites with various functions. Short-chain and branched-chain fatty acids, secondary bile acids, polyamines, indoles, and phenolic compounds are metabolites produced by the gut microbiome. The gut microbiome can also produce neurotransmitters (such as γ-aminobutyric acid, catecholamines, and serotonin), their precursors, and vitamins. Several studies in pigs have demonstrated the importance of the gut microbiome and its metabolites in improving growth performance and feed efficiency, alleviating stress, and providing protection from pathogens. The use of probiotics is one of the strategies employed to target the gut microbiome of pigs. Promising results have been published on the use of probiotics in optimizing pig production. This review focuses on the role of gut microbiome-derived metabolites in the performance of pigs and the effects of probiotics on altering the levels of these metabolites.

Reactivity of Phospholipase D toward Phosphatidylcholines with Different Length of Acyl Chains (길이가 틀린 아실사슬을 갖는 콜린 인지질에 대한 포스포리파제 D의 반응성)

  • In order to explore a substrate specificity for cabbage phospholipase D, we examined the PLD reactivity toward the phosphatidylcholines with different chain length of acyl groups. The selected acyl chains were the saturated fatty acid of $C_8:0,\;C_{12}:0,\;C_{16}:0,\;C_{20}:0$. The reactivity of these phospholipids were dependent largely on the ratio of PC : SDS. The PC : SDS ratio showing the optimal PLD activity were found to be 1:1.4, 1:2.2, 1:2.5, and 1:3.6 respectively as the increase of the acyl chain length. Likewise the optimum temperature for the maximal PLD activity were altered markedly to 25$^{\circ}C$, 30$^{\circ}C$, 35$^{\circ}C$, 45$^{\circ}C$ when the length of acyl chains increased. On the contrary the pH and concentration of $Ca^{2+}$ necessary for the optimum PLD activity were not altered significantly. The kinetic parameter $V_{max}$ for short acyl chain substrate was greater than the values for the longer acyl chain, which indicates the fastest rate of hydrolysis. By the same token, the reactivity of longer chain substrate became slower for the hydrolysis activity.
