Agronomic Characterization of Sorghum bicolor (L) Moench Germplasm Collected in Gangwon-do for Bio-energy Crop (바이오에너지 자원으로서 강원도 지역에서 수집된 수수의 작물학적 특성 평가)

  • Forty nine accessions of the Sorghum bicolor (L) Moench collected from different agronomical zone of Gangwondo province, Korea were characterized morphologically on field trial. Plant length, stem diameter, tiller number, ear type, ear length, ear width, leaves length, leaves width and leaf color differed significantly in the cultivated accessions. Out of forty nine test accessions, six accessions (12.2%) showed comparatively taller height, measuring more than 300 cm, while, other ten test accessions (20.4%) were measured from 200~300 cm. There were no significant differences in the diameter of stem in the collected test accessions (2~3 cm). Of the total test accession, seven accessions showed leaf length of over 70 cm, and twenty two test accession revealed leaf length between 60-70 cm. Forty nine of the collected test accessions showed narrow and short leaves with white mid vain. These results provided additional information for the breeding programs and conservation of Sorghum bicolor as a bio-energy crop.

Impulse Trafficking in Neurons of the Mesencephalic Trigeminal Nucleus

  • In the primary sensory neuron of the mesencephalic trigeminal nucleus (MTN), the peripheral axon supplies a large number of annulospiral endings surrounding intrafusal fibers encapsulated in single muscle spindles while the central axon sends only a few number of synapses onto single ${\alpha}-motoneurons({\alpha}-MNs)$. Therefore, the ${\alpha}-{\gamma}$ linkage is thought to be very crucial in the jaw-closing movement. Spike activity in a ${\gamma}-motoneuron\;({\gamma}-MN)$ would induce a large number of impulses in single peripheral axons by activating many intrafusal fibers simultaneously, subsequently causing an activation of ${\alpha}-MNs$ in spite of the small number of synapses. Thus, the activity of ${\gamma}-MNs$ may be vital for modulation of jaw-closing movements. Independently of such a spindle activity modulated by ${\gamma}-MNs$, somatic depolarization in MTN neurons is known to trigger the oscillatory spike activity. Nevertheless, the trafficking of these spikes arising from the two distinct sources of MTN neurons is not well understood. In this short review, switching among multiple functional modes of MTN neurons is discussed. Subsequently, it will be discussed which mode can support the ${\alpha}-{\gamma}$ linkage. In our most recent study, simultaneous patch-clamp recordings from the soma and axon hillock revealed a spike-back-propagation from the spike-initiation site in the stem axon to the soma in response to a somatic current pulse. The persistent $Na^+$ current was found to be responsible for the spike-initiation in the stem axon, the activation threshold of which was lower than those of soma spikes. Somatic inputs or impulses arising from the sensory ending, whichever trigger spikes in the stem axon first, would be forwarded through the central axon to the target synapse. We also demonstrated that at hyperpolarized membrane potentials, 4-AP-sensitive $K^+$ current ($IK_{4-AP}$) exerts two opposing effects on spikes depending on their origins; the suppression of spike initiation by increasing the apparent electrotonic distance between the soma and the spike-initiation site, and the facilitation of axonal spike invasion at higher frequencies by decreasing the spike duration and the refractory period. Through this mechanism, the spindle activity caused by ${\gamma}-MNs$ would be safely forwarded to ${\alpha}-MNs$. Thus, soma spikes shaped differentially by this $IK_{4-AP}$ depending on their origins would reflect which one of the two inputs was forwarded to the target synapses.

Influences of DIF on Growth of Capsicum annuum 'Nokkwang' (주야간(晝夜間) 온도차(溫度差)(DIF)처리(處理)에 의한 고추 '녹광' 플러그묘(苗)의 생장반응(生長反應))

  • Capsicum annuum 'Nokkwang' plug seedlings were treated with 9 different day and night temperatures to investigate on growth and differentiation in controlled environment conditions. The plant height was the most affected by day temperature and DIF and stem length was the similler results as plant height. However, Leaf unfolding rate and leaf length and width increased as average daily temperature increased. and especially fresh weight was worst at ADT was $15^{\circ}C$ and increased as ADT increased. In contrast with fresh weight, chlorophyll content calculated highest when ADT was $15^{\circ}C$, and decreased as NT increased respectively. The stem lengths were not significantly different among treatments with exception of DT/NT=15/$15^{\circ}C$ which significantly decreased. The ultimate +DIF condition, DT/NT=30/$15^{\circ}C$, caused high T/R ratio due to abundant top growth. It is concluded that DT is $22.5^{\circ}C$, and NT is ranged from $15^{\circ}C$ to $22.5^{\circ}C$ to be produced short, healthy plug seedlings in Capsicum annuum 'Nokkwang'.

Characterization of Odontoblasts in Supernumerary Tooth-derived Dental Pulp Stem Cells between Passages by Real-Time PCR (과잉치 치수유래 줄기세포의 Real-time PCR에 의한 계대간 상아질모세포 발현 특성)

  • The aim of this study is to compare the properties of odontoblast gene of early passage cells and late passage cells derived from impacted maxillary supernumerary teeth. Impacted supernumerary teeth with maxilla were extracted from 12 patients (8 males, 4 females) between 6 - 9 years old without medical history. Real-time polymerase chain reaction (PCR) was conducted to compare characterization of odontoblast cell in the 3rd and 10th passage, and between with bone inducing additive group and without additive group. Genes for odontoblasts characteristics are osteonectin (ONT), alkaline phosphatase (ALP), osteocalcin (OCN), dentin matrix protein 1 (DMP-1) and dentin sialophosphoprotein (DSPP). The level of gene expression was in a decreasing order of ONT, ALP, OCN, DMP-1 and DSPP in the 3rd passage, and in decreasing order of ONT, DMP-1, OCN, ALP, and DSPP in the 10th passage in the undifferentiation and differentiation group. The order of ONT, DMP-1, and OCN did not changed. ALP and DMP-1 were switched in order. ALP and DMP-1 may be used as important markers for differentiating between the 3rd passage and 10th passage cells. Considering that supernumerary tooth was extracted young age and the time required to cultured 10th passage was short, supernumerary tooth can be considered a useful donor site of dental pulp stem cells.

Gene Expression of Supernumerary Dental Pulp Related to the Subculture Speed: A Pilot Study (계대 배양 속도가 다른 과잉치 치수유래 줄기세포 간 유전자 발현 특성)

  • The purpose of this study was to investigate the odontoblast gene expression related to the subculture speed of supernumerary dental pulp stem cells (sDPSCs). The stem cell is undifferentiated cells which has the ability to differentiate into various cells. Specific stimulation or environment induces cell differentiation, and these differentiation leads to bone or muscle formation. 20 sDPSCs were obtained from 20 children under aseptic condition. During the culture through the 10th passage, the third passage cells which showed short subculture period and 10th passage cells which showed long subculture period were earned. Each cell was divided into differentiated group and non-differentiated group. Quantitative real-time polychain reaction (q-RT-PCR) was performed for each group. The genes related to odontoblast differentiation, Alkaline Phosphatase (ALP), Osteocalcin (OCN), Osteonectin (ONT), Dentin sialophosphoprotein (DSPP) and Dentin matrix acidic phosphoprotein 1 (DMP-1), were measured. Differentiated cells showed more gene expression levels. Undifferentiated cells showed higher gene expression level in 10th passages but differentiated cells showed higher gene expression level in 3rd passages. Cells that showed faster subculture period showed relatively lower gene expression level except for OCN and DSPP.

A Shinpung Typed Large Grain and High-Yielding Peanut Variety 'Daan' (단경 소분지 대립 다수성 땅콩 신품종 '다안')

  • A new peanut variety 'Daan'(Arachis hypogaea ssp. hypogaea L.) was developed at the Department of Southern Area Crop Science, National Institute of Crop Science (NICS) in Milyang 2014. This was developed from the cross between 'Iksan 31' with Virginia typed short stem and 'Dakwang' with Shinpung-typed larger grain. 'Daan' which is a Shinpung plant type had 13 branches per plant and its length of main stem was 44cm. Each pod has two grains with brown testa and long ellipse-shaped large kernel. Its yield components showed 34 pods per plant, 127 g of 100-seed-weight and 75% of pod shelling ratio in the regional yield trials (RYT). Seed quality showed 47.8% of crude oil and 28.3% of protein content. This variety showed resistant to early leaf spot and had more resistant to late leaf spot, stem rot and lodging, compared to reference variety. In the regional yield trials for 3 years 'Daan' was more productive than reference variety by 16% with 5.00 MT/ha for grain production.

Anti-inflammatory Effect of Conditioned Medium From an Immortalized Adipose-derived Stem Cell Line by SV40 T Antigen (SV40의 T항원으로 불사화한 지방줄기세포주로부터 생산한 배양액의 항염증 효능)

  • Adipose-derived stem cells (ADSCs) are capable of differentiation into multiple lineages of cells, which has attracted attention for clinical therapy. However, ADSCs have poor proliferation capacity and a short life span in culture, which is an impediment in the application to clinical use. Previously, to overcome growth disadvantages, we had established an immortalized ADSC line (ADSC-T) by introducing the SV40 T antigen coding gene into primary human ADSC. In the present study, we evaluated the differentiation potential of this cell line and assessed the anti-inflammatory effect of its conditioned medium (CM). ADSC-T appeared to maintain the differentiation potential into adipocyte and chondrocyte. The CM of ADSC-T suppressed the NF-κB activity and its target gene expression of COX-2 and iNOS. Furthermore, the phosphorylations of MAPKs, including ERK, JNK and p38, were suppressed by the ADSC-T CM. The expressions of pro-inflammatory cytokines such as TGF-β, TNF-α, IL-6, and IL-13 were also suppressed by the CM of ADSC-T. In the Nc/Nga atopic model mice, the CM showed therapeutic effect on DNCB-induced atopic dermatitis. These results indicate that the immortalized ADSC-T maintains the beneficial properties of primary ADSC and could be a versatile cell source for not only research into ADSC but also for production of CM suitable for clinical application.

Flowering and Fruiting of Characteristics of Short Flowering Period Lines in Peanut (땅콩 단기개화성 선발 계통의 개화 및 결실 특성)

  • To breed high quality and yield peanut variety according to select the short flowering duration, fifteen germplasms (1 virginia,7 spanish,6 valencia types and var, Daekwang) were investigated the flowering habit and agronomic characteristics from 1998 to 1999. Emergence date of the selected short flowering duration germplasms (SPFGs) was earlier 1-3 days and middle or small seed than that of var, Daekwang. Main stem length was longer 57cm but the number of branches, pods, 100-seed weight, and pod weight per plant was reduced 25%, 23%, 42%, 46%, respectively, in SPFCs comparing to var, Daekwang. The flowering date in SPFGs was similar but the flowering duration was earlier 5-16 days than that of var, Daekwang (52 days). Varieties that flowered shorter duration than 50 days were 18.8% among the SPFGs. The number of total flowers in SPFGs was fewer 50% than that of var, Daekwang. The rate of flowering inhibition were 50-52% than that of var Daekwang. The frequencies of flowering duration (under 50 days) were 7.7% in virginia, 46.2% in spanish, 53.9% in valencia. The effect of shading treatment on rate of flowering inhibition were 11%, but number of branches and pods were reduced by 27-31% in valencia type compared to non-shade. Correlation coefficient was significant positively ($r=0.9314^*$ virginia, $r=0.9551^*$ spanish, $r=0.9551^*$ valencia) between the air temperature and flower number, The frequency of peg and pod number on 1st to 2nd nodes in SPFGs were more 3-12%, 21-26% than that of var. Daekwang. The rate of mature pods at 80 days after flowering were higher 12-17% than that of var, Daekwang (68%). Correlation coefficient was high significant negatively between date of first flower and flowering date, the ratio of mature pod.

The Application of Early-maturing Corn to Cropping System of Forrage Crop I. The late-sowing adaptability of corn varieties requiring the short period to maturing (조숙 옥수수에 의한 사료작물 작부체계 구성 I. 조숙 옥수수 품종의 만파적응성)

  • For the constitution of forage cropping system including the double-cropping of corn, attention has been directed towards the early and short maturing varieties of corn such as Comet 80, Comet 85 and Linda as a component forage crop of forage cropping system. Four corn varieties, Comet 80, Comet 85, Linda and Suwon 19 were planted delayedly by each of 45, 55 and 65 days from the recommended sowing date of suwon 19. Under these condition, growth characteristics and late-sowing adaptability of 4 corn varieties were investigated, and the results are summarized as follows; 1. The variety of suwon 19 did not reach the stage of silking in the occasion of late sowing by 50 days from the optimum sowing time for mono cropping of suwon 19. However early varieties such as Comet 80, Comet 85 and Linda silked under the condition of 65 days-delayed sowing from the recommended sowing time of Suwon 19 for mono-cropping. 2. Early varieties of Comet 80, Comet 85 and Linda were so grown by approximately 92% in plant height, 66% in ear height, 88% in stem diameter and 81% in leaf number respectively compared to Suwon 19 at each of three delayed sowing time that exhibited the dwarfish appearance of canopy. 3. Days from sowing to silking decreased as the sowing time was delayed and the average days from sowing to silking in the sowing treatments of 55 days-delayed of Suwon 19, Comet 80, Comet 85 and Linda were 61, 46, 47 and 51 days, respectively. 4. The size of variations of plant height and ear height influenced by delayed-sowing time was comparatively larger in the varieties of Suwon 19 and Linda than in those of Comet 80 and Comet 85. 5. As sowing time was delayed from the optimum sowing time of Suwon 19 for mono-cropping, ear content decreased from 0% of Suwon 19 at 55 day-delayed sowing treatment to 3540% of Comet 80 and Comet 85 at 65 days-delayed sowing treatment.

Estimation of Air Travel Demand Models and Elasticities for Jeju-Mainland Domestic Routes (제주-내륙 간 국내선 항공여객수요모형 및 탄력성의 추정)

  • Jeju-Mainland demand for air passenger is variated by the season because most of the demands stem from the leisure travel. This research is to estimate the econometrics demand models(A simple time series model and the partial adjustment model) and elasticities of each models for the Jeju-Mainland domestic routes air travel market using the time series aggregate data between the year 1996 and 2005. As the result of estimating, income elasticity was evaluated to be elastic(1.55) and fare elasticity was inelastic(-0.49${\sim}$-0.59) for A simple time series models. In the partial adjustment model's case, income elasticity was evaluated to be inelastic(0.51) in short-run whereas it was evaluated to be elastic(1.88) in long-run. Fare elasticity was evaluated to be inelastic in short-run(high-demand season: -0.13, slack season: -0.20) and long-run(high-demand season: -0.48, slack season: -0.72).
