In vitro Mass Propagation of Ardisia pusilla DC. (산호수 (Ardisia pusilla DC.)의 기내 대량번식)

  • To establish the mass proliferation system of Ardisia pusilla DC, the shoot tips of Ardisia pusilla DC were cultured on the MS and half-strength MS medium supplemented with $0{\sim}5.0$ mg/L BA or $0{\sim}0.5$ mg/L thidiazuron(TDZ), respectively. A few multiple shoot formation observed when the shoots were cultured on MS medium containing TDZ. However, the frequency of multiple shoot formation was reached up to 82.4%, when the shoots were cultured on half-strength MS medium supplemented with 0.5 mg/L BA. Also the number of shoot per explant was 7.1. To promote rooting from multiple shoot, newly formed shoots were transferred to half-strength MS medium containing 0.5 mg/L IBA or 0.5 mg/L NAA, respectively. Regenerated plantlets were grown to normal mature plants in soil.

  • This study was conducted to obtain some basic information needed for the propagational system of high quality garlic trough the culture of healthy tissues. non shoot-tips of bulbil obtained in mid May were cultured on MS medium containing 8% sucrose supplemented with 0.1 mg/L NAA, in vitro bulbli were formed, but the shoots were formed at the early to middle in June. Multiple shoots were induced by the culture of receptacles on MS medium supplemented with 0.1 mg/L NAA and 10mg/L BA..Among the flower bud, bulbil and receptacle, receptacle showed most suitable in terms of shoot formation efficiency, More than 50 shoots per single involucre were produced under the optimum condition. Results indicate that in vitro culture of involucre has a high potential for the micropropagation of high quality seed bulbs.

Multiple shoot induction and plant regeneration from axillary buds of Magnolia 'Vulcan'

  • An efficient protocol for multiple shoot induction and plant regeneration from axillary bud culture of Magnolia 'Vulcan' was developed in the present study. Primary shoots were obtained from axillary bud explants cultured on Murashige and Skoog (MS) medium containing 1.0 mg/L 6-benzylaminopurine (BA). To induce multiple shoots effectively, primary shoot tips were cultured on MS medium supplemented with different concentrations of BA and zeatin at 0, 0.2, 0.5, and 1.0 mg/L. Of these treatments, the MS medium with 0.5 mg/L BA resulted in the highest number of shoots per explant with an average value of 5.9, and it produced the greatest shoot height at 4.8 cm after 12 weeks of culturing. In the rooting of in vitro produced shoots, the greatest percentage of explants forming roots (91.3%), number of roots per explant (9.7), and root length (2.8 cm) were obtained in half-strength MS medium supplemented with 6.0 mg/L indole-3-butyric acid (IBA). Regenerated plantlets were successfully acclimatized and hardened off inside the culture room with 87.5% survival rate. Plants were transferred to a greenhouse with a 97.2% survival rate. The highly efficient shoot multiplication and plant regeneration system reported herein can be used for large-scale clonal propagation of valuable Magnolia species or cultivars.

  • The highest degree of callus formation was obtained from the shoot tips of Dianthus superbus when cultured on the MS medium supplemented with 2.0 mg/L NAA and 0.5 mg/L BAP. Embryogenic calluses were obtained from the seperated friable calluses on MS medium containing 2.0 mg/L 2,4-D after 7-8 wk of culture. For plant regeneration, embryogenic calluses were selected and cultured on te proliferation medium. After 3 wk, somatic embryos appeared on MSK medium (0.5 mg/L NAA, 2.0 mg/L kinetin) and N6 medium (2.0 mg/L kinetin, 0.1 mg/LNAA, 0.1 mg/L 2,4-D and 2.0 g/L casein hydrolysate). When these somatic embryos were kept under continuous illumination, shoots were successfully regenerated on the both media. The shoots were rooted on MS medium supplemented with 2.0 mg/L NAA.

Elimination of SPFMV from Virus-infected Sweet Potato Plants through Apical Meristem Culture

  • Sweet potato infected with a viral disease (SPFMV) showed irregular chlorotic patterns, so called feathering associated with faint or distinct ring spots that have purple-pigmented borders. SPFMV was eliminated from sweet potato plants using meristem tip culture. MS medium supplemented with BAP (2mg/L) and NAA (0.05 mg/L) was used for shoot proliferation and 1/2 MS medium for rooting of the plants. Highest percentage of regenerated plants (60%) was obtained from the optimum size (0.3-0.5mm) meristem tips. Of these, 60% plants were found negative for SPFMV by RT-PCR. Virus detection by RT-PCR was found to be a reliable method. Meristem-tip culture to produce SPFMV-free quality sweet potato and virus detection by RT-PCR is an efficient, time saving and reliable method for production of SPFMV-free tissue culture raised plants.

Effect of Plant Growth Regulators and Medium Salt Strength on In Vitro Propagation of Belamcanda chinensis DC (범부채의 기내증식에 미치는 식물생장조절물질 및 무기염류농도의 효과)

  • The effects of plant growth regulators on callus formation and organogenesis in shoot tip explant of Belancanda chinensis were examined. Shoot tip explants cultured in full salt strength of MT(Murashige and Tucker) medium containing 2,4-D 1.0 or 2.0mg/l were vigorously formed callus. Full salt strength of MT medium and 1/2 MT medium supplemented with zeatin 1.0mg/l were more effective than that with combination treatments of 2,4-D on the formation of shoots from calli. When shoots regenerated from shoot tips were transplanted into 1/2 MT medium added with 1.0mg/l, 41% of shoots formed roots.

  • The shoot tips of Delphinium cv. Princess Caroline were cultured on the MS medium supplemented with cytokinin and auxin alone or in combination. Among cytokinins, BA was most effective in shoot multiplication, adequqte concentrations being 1.0-5.0 mg/L. Shoot multiplication was very favorable on the media with 1.0-3.0 mg/L BA and 0.1-0.5 mg/L IAA. Additions of BA and IAA did not stimulate shoot multiplication, but increased a little fresh weight. Shoots were scarcely rooted on the media with IBA or NAA, and were not done utterly on the media containing activated charcoal. Therefore, shoots were treated by Rootone and planted in the cultural media for in vivo rooting. The highest rate of rooting was 68% in the mixed cultural medium composed of Perlite 1 and Vermiculite 1.

  • An efficient protocol has been developed for rapid mass propagation of sweetpotato from shoot-tips derived embryogenic callus. Optimal embryogenic callus was induced from shoot apical meristem explants on Murashige and Skoog (MS) medium supplemented with 1mg/L 2,4-D. The addition of casein hydrolysate in the media increased the embryogenesis efficiency of sweetpotato. Somatic embryos were easily induced from the embryogenic callus on MS basal medium containing 300-500mg/L casein hydrolysate without phytohormon. Treatment of casein hydrolysate (100∼300mg/L) with 1mg/L 2,4-D also improved the secondary embryonic efficiency from somatic embryos below 2mm in length. Plant regeneration was achieved via somatic embryogenesis and direct organogenesis. Regenerated planlets with well developed shoots and roots on MS basal medium were successfully transferred to soil.

Effect of Plant Growth Regulators on Multiple Shoot Formation and Elongation from Shoot Tip Cultures of Grape Species (포도의 경정배양에 의한 다아체형성 및 신장에 미치는 생장조절제의 영향)

  • Shoot tips of grape were cultured in uitro and tried to identify optimal culture conditions for regeneration, multiple shoot formation from meristemoid tissue and those subsequent elongation of multi-shoots. Healthy growing shoots were taken in early May, rinsed with running tap water, soaked in a neutral detergent and washed with soft brushing, and washed out with tap water, then sterilized with 10g Ca(ClO)$_2$/140 mL distilled water (Wilson's solution) for 5 min. Survival percentage of the cultures which were sterilized as above procedures was highly increased, compared with the other sterilized method. Propagation of multi-shoots from meristemoid showed a good response in 3/4 strength MS medium enriched with 0.1 mg/L NAA and 3.0 mg/L BA. Shoot elongation from multi-shooting clump well occurred in 3/4 strength MS medium supplemented with 80 mg/L adenine sulfate, 0.1 mg/L NAA and 1.0~2.0 mg/L BA.
