Bacillus subtilis 균주로 발효한 죽순발효액의 품질특성 (Quality Characteristics of Bamboo Shoot Liquid Fermented by Bacillus subtilis Strain)

  • To make fermented bamboo shoot liquid, we isolated and classified a microorganism growing in bamboo shoot and investigated its quality characteristics. Crude fiber, crude ash, nitrogen-free extracts, and total sugar contents were higher after fermentation. For free amino acids, only alanine was detected in the control group. Detected 13 kinds of free amino acids were detected in fermented bamboo shoot liquid. In organoleptic test, fermented bamboo shoot beverage containing 20 percent strawberry showed the highest consumer preference.

정아배양에 의한 산돌배나무의 기내번식 (Micropropagation Using Shoot Tip Culture of Pyrus ussuriensis Maximowicz)

  • 산돌배나무의 기내번식 기술을 개발하고자 유묘의 정아를 재료로하여 기내 식물체 증식에 미치는 생장조절제의 효과를 시험하였다. 정아를 MS 배지에 BA 단독처리 혹은 BA+NAA 혼용처리 결과 대부분 줄기는 1-2개로 유도 되었으며 처리에 따른 차이는 나타나지 않았다. 줄기의 생장과정에서 정아괴저가 흔히 관찰되었으며 다경이 유도되는 줄기에서는 과수화 현상이 나타났고 줄기의 기부에는 캘러스가 형성되었다. 줄기의 증식 과정에서 BA+NAA 처리구에서 약 20%의 자발적인 발근이 이루어 졌다. 다경으로부터 얻어진 줄기는 gelrite 경화배지에서 기내발근이 저조하였으며, 기외삽목시 100 mg/L IBA 처리로 50%까지 발근되었다. 발근묘는 순화 후 정상적으로 생장하였다.

Medium compositions reveal potential organogenesis in the diploid and tetrploid Codonopsis lanceolata

  • Medium composition plays a key role on influencing organogenesis in plant tissue culture. This study was carried out to examine the effects of medium composition on organogenesis in diploid and tetraploid Codonopsis lanceolata and obtain in-vitro mass propagation of superior species of C. lanceolata. Diploid C. lanceolata was found to be declined regarding MS medium composition for each concentration. However, shoot and adventitious root formation were suppressed with higher mineral salt concentration, and active growth of shoot and adventitious root was exhibited as 4.9 cm and 3.2 cm respectively in 1/2 MS medium. While in tetraploid C. lanceolata, it showed 2.9 cm and 3.2 cm respectively in 1/4 MS medium. In the case of sucrose concentration, no consistent decrease was observed for growth of shoot and the adventitious root of diploid both at high and low concentration. The growth of shoot (at 3% concentration) and adventitious root (at 7% concentration) was 2.3 cm and 2.0 cm respectively. Although there was no difference in shoot formation of tetraploid C. lanceolata in all concentrations with the range of 1.7~1.8, there was a slight decrease in shoot growth at high concentration. Results revealed that the adventitious root formation was suppressed at high concentration. The concentration of agar exhibited no significant difference in shoot formation of diploid C. lanceolata at all concentrations. The maximum result of adventitious growth (4.1 cm) was observed at 0.8% concentration. Slight inhibition of shoot formation and root formation of tetraploid C. lanceolata was observed at higher concentration. Shoot formation of diploid C. lanceolata also exhibited inhibition at higher concentration. Shoot formation of diploid C. lanceolata was increased at lower pH and shoot growth was the highest (2.3 cm) at pH 3.8. Adventitious root formation was higher at lower pH. However, both the adventitious root formation and growth exhibited comparatively higher result at pH 5.8. Taken together, the levels of pH had an effect on shoot and root formation in diploid and tetraploid of C. lanceolata

Effects of Auxins and Cytokinins on Organogenesis of Soybean Glycine max L.

  • To select the section with shoot formation ability, the calli and shoot formation from three sections (first leaf including cotyledonary node, hypocotyl and cotyledon explants) of 5-days-seedlings of soybean were induced on MS medium supplemented with 1.0 mg/L BAP, 3% sucrose, and 0.3% gelrite for one month. The first leaf section exhibited the highest shoot formation rate (51%), followed the hypocotyl section (10%) and the cotyledon section (0%). The shoot formation rates and shoot number of the four excised sections (whole first leaf, a half of the first leaf, a third of the first leaf and only node) of the first leaf were also investigated on the same medium. A half of the first leaf explant and the third of the first leaf explant had higher shoot formation rates (76-80%) and numbers (3-4 / explants) than those in other two explants. Effects of six cytokinins (kinetin, zeatin, BAP, 2iP, PBA, and TDZ) on shoot formation were determined, using the half of the first leaf explants. Zeatin (1.0 mg/L) exhibited the highest in shoot formation rate (94%) and numbers (8 / explant). In addition, the combined effects of three cytokinins (zeatin, BAP, and TDZ; 0.5, 1.0, 2.0 mg/L, respectively) and an auxin (IAA; 0.0, 0.5, 1.0, 2.0 mg/L) were determined. The combination (1:1, v/v) of zeatin (1.0 mg/L) and IAA (1.0 mg/L) exhibited the highest in shoot formation rate (96%) and numbers (16 / explant), twice more than zeatin (1.0 mg/L) alone. The shoot cuttings were transferred and cultivated on the rooting media supplemented with only auxin, IBA at various concentrations. The highest root formation (8 / shoot) was achieved on the medium supplemented with 1.5 mg/L. After 4 weeks of cultivation, the plantlets with an extensive root system were transplanted in pots with a soil mixture of vermiculite and fine sand. Transferred to field, about 75% of the plantlets survived.

Shoot Rot of Spikenard Caused by Rhizoctonia solani AG-2-1

  • In January 2021, unusual outbreak of shoot rot symptoms was observed in young spikenard (Aralia cordata) plants growing in vinyl greenhouses located in Chuncheon and Yanggu, Gangwon Province, Korea. The symptoms initially appeared on young shoots of the plants at or above the soil surface level. Later, the infected shoots wholly rotted and blighted. The incidence of diseased plants in the vinyl greenhouses investigated ranged from 5% to 30%. Eight isolates of Rhizoctonia sp. were obtained from shoot lesions of the diseased plants. All the isolates were identified as Rhizoctonia solani AG-2-1 based on the morphological characteristics and anastomosis test. Three isolates of R. solani AG-2-1 were tested for pathogenicity on young shoots of spikenard plants using artificial inoculation. All the tested isolates induced shoot rot symptoms on the inoculated plants. The symptoms were similar to those observed in spikenard plants from the vinyl greenhouses investigated. This is the first report of R. solani AG-2-1 causing shoot rot in spikenard.

Practical Factors Controlling in vitro Multiplication and Rooting in Empetrum nigrum var. japonicum, an Endangered Woody Species

  • The plant Empetrum nigrum, valued in the traditional system of medicine, is well known for its antibacterial, antifungal, and antioxidant properties. In the present work, the effect of removal of shoot apical meristem (SAM) on shoot proliferation was studied. It was observed that removal of SAM promoted shoot proliferation whereas intact tip resulted in higher survival percentage. Further, the effect of different concentrations of BA on above was also studied. During root formation the effect of light quality after treatment with IBA was investigated. For rooting, continuous red light without IBA resulted in maximum rooting percentage. The above factors when taken into consideration during micropropagation of this endangered plant can result in healthier plantlets. The results show that the species could be successfully conserved by in vitro propagation system.

Occurrence of black shoot blight in apple and pear trees in Korea

  • Erwinia pyrifoliae, which causes black shoot blight in apple and pear trees, was first identified in Korea in 1995. Extensive measures are typically used to control the disease by eradicating trees in diagnosed orchards, owing to the detrimental impact of the disease on apple and pear production. However, despite governmental efforts, the disease has continuously spread. In this study, we analyzed the current status of the black shoot blight occurrence in apple and pear orchards between 1995 to 2022. Our findings reveal that over the past 28 years, black shoot blight has occurred in 26 cities and districts across five Korean provinces. The affected regions are predominantly concentrated in the northern part of Korea, including the Gangwon and Gyeonggi provinces. Furthermore, black shoot blight has gradually expanded to the northern provincial regions of Chungbuk, Chungnam, and Gyeongbuk, which are centrally situated in Korea. Furthermore, the occurrence pattern of black shoot blight differed between apple and pear orchards; in apple orchards, black shoot blight occurred consistently each year, with a sudden increase in cases in 2020; however, in pear orchards, it has considerably decreased since 2007. To the best of our knowledge, this is the first comprehensive report on the occurrence of black shoot blight in apple and pear trees in 28 years, and the results will provide valuable insights for future disease management strategies.

Peace 포플러(Populus koreana X P. trichocarpa)의 줄기형성에 미치는 Thidiazuron 효과 (Effect of TDZ (Thidiazuron) on Shoot Proliferation of Peace Poplar)

  • 기공이 잎의 앞뒤 양면에 존재하는 생리ㆍ형태적 특징을 가지고 있는 외래 교잡종인 Peace (Populus koreana ${\times}$ P. trichocarpa)포플러의 효율적인 기내증식법을 개발하고자 액아와 엽육 절편을 이용하여 기내배양을 실시하였다. 몇 종류의 배지 시험을 통해 1/2MS배지를 적정배지로 선정할 수 있었고, 이 배지에 TDZ를 처리하여 multiple shoot 및 adventitious shoot유도 효과를 조사하였다. 절간액아 및 엽육을 절편으로 TDZ의 농도별 처리로 배양 2주 후부터 multiple shoot 및 adventitious shoot가 유도되었으며 절편 및 TDZ처리 농도에 따라 차이를 나타냈다. 액아로부터는 TDZ 처리로 절편당 5-7개의 multiple shoot가 유도된 반면 엽육 절편에서는 2-5개의 adventitious shoot가 유도되었다. 한편 부정아로부터 정상적 인 줄기의 성장에 있어서도 절편에 따른 차이를 나타냈다. 이상의 결과는 저농도의 TDZ의 처리로 peace포플러의 액아 혹은 엽육 절편의 기내배양을 통해 대량증식이 가능함을 보여주는 결과이다.

Evaluation of Cancer Chemopreventive Potential of Various Grape Shoot Extracts and Refined Materials Using in vitro Bioassay Systems

  • Since reactive oxygen species, prostaglandins, and nitric oxide are closely involved in various pathological conditions anti play important roles in the initiation, promotion, and progression of carcinogenesis, agents that modulate the production or activity of them might be considered as cancer chemopreventive agents. In the present study, we evaluated chemopreventive potential of some grape shoot extracts and their refined materials using various in vitro assay systems. As a result, both grape shoot extracts and refined materials possessed effective radical scavenging activities about 70~80% at the concentration of 500 ${\mu}$g/ml, and especially the Sheridan shoot extract showed the most potent 1, 1-diphenyl-2-picrylhydrazyl radical scavenging activity that was similar to resveratrol. In addition, refined materials from grape shoot extracts suppressed lipopolysaccharide-induced nitric oxide production in macrophage cells, anti refined materials from Kyoho and Campbell shoot extracts exhibited similar inhibitory activities with $IC_{50}$ value of 224 ${\mu}$g/ml and 285 ${\mu}$g/ml, respectively. In addition, at the concentration of 50 ${\mu}$g/ml, all of refined materials inhibited cell proliferation against various human cancer cells about 30~40% compared to control. These findings suggest that grape shoot extract and their refined materials might be useful sources for the development of chemopreventive agents and/or functional foods.
