To find the nutritive values of the edible bamboo shoot (Playllostachys edulis) produced in Korea, the component analysis was conducted using the fresh and canned bamboo shoot. As the principal component ; water, protein, lipid, sucrose, ash, cellulose and calorie content were measured in fresh bamboo shoot and canned bamboo shoot. Also the content of minerals ; K, P, Ca, Fe and Vitamins were analysed and compared between fresh and canned bamboo shoot. The contents of component were devided into upper, middle and bottom bamboo parts. The contents of protein, lipid, ash, cellulose and calories of raw bamboo shoot were higher than those of canned shoot. Generally, the components of bamboo shoot produced in Korea were not significantly different to those of Ueda's results in Japan. But the contents of lipid, calcium and potassium were higher in Korean bamboo shoot.
Culture condition for callus formation and plant regeneration were optimized by the selection of explants and the manipulation of hormonal combination in the culture medium. The calli induced from seed, cotyledon, hypocotyl and mesophyll segments were more vigorously proliferated under dark condition than those under continuous light condition. Hypocotyl-and cotyledon-derived calli were more regenerative as compared with those of seed and mesophyll. Callus formation from hypocotyl and cotyledonary explants was enhanced on MS medium with 1.0 mg/${\ell}$ 2, 4-D and 0.1 to 0.5 mg/${\ell}$ kinetin or BAP. The combination of 0.1 mg/${\ell}$ NAA and 2.0 to 4.0 mg/${\ell}$ kinetin was the most effective for shoot regeneration from the callus. The maximum frequency (24.0%) of shoot regeneration was obtained from the hypocotyl-derived callus transferred to MS medium supplemented with 0.1mg/${\ell}$ NAA and 4.0 mg/${\ell}$ kinetin. The capacities for callus, root and shoot formation from cotyledon and hypocotyl explants were remarkably different among cultivars of B. napus tested. The calli induced from hypocotyl produced more shoots than those from cotyledon.
This study was conducted to investigate the effect of shoot twist on fruitfulness and fruit quality of 'Campbell Early' grapevine. Proper pruning and training are essential to produce a good yield of high-quality fruit and to maintain the balance between vegetative growth and fruiting. The most common problem in spur-pruned 'Campbell Early' cultivar is that vigorous buds has low fruitfulness and thereby the shoot become more vigorous the following spring because of lower crop load. Therefore, shoot twists in very vigorous 'Campbell Early' canes (above 10.0 mm) were performed on the third nodes and the $7^{th}$ nodes of each shoot at 7 days before bloom and full bloom, respectively. Sprouting date, blooming date were not significantly different among the treatments while, harvesting date was delayed approximately 3 days. However, number of berries per cluster, cluster weight and fruitfulness were significantly higher in the shoot twist treatment on the third nodes than the control that was topping alone. Combination treatments of shoot twist and topping had an additive effect on increasing cluster weight resulting in higher increase of yield by 12.1 ㎏ per vine. These results indicated that the shoot twist on very vigorous canes of 'Campbell Early' grapevine for well fruitfulness seemed to be very effective.
Morphological characteristic during formation of tobacco crown gall tumor and genetic tumor, and their differential response to growth regulator were investigated in in vitro culture. Crown gall tumor was induced from tumor tissue transformed by infecting Agrobacterium tumefaciens C58. Genetic tumor was induced from tumor tissue which was induced spontaneously from reciprocal interspecific hybrids between Nicotiana glauca (2n=24) and Nicotiana langsdorffii (2n=18). Morphological characteristic of crown gall tumor, genetic tumor, and teratoma shoot was very similar, and they were actively proliferated on hormone-free medium. Typical tumor callus and teratoma shoot formed from crown gall tumor on the hormone-free medium. On the contrary, tumor callus derived from genetic tumor formed as a crown gall tumor callus on the medium supplemented with 0.5 mg/L of 2,4-D, and lots of teratoma shoots without any root formed on the hormone-free medium. Root development from the teratoma shoots was hardly obtained on the medium with IAA, GA and active carbon. However, teratoma shoots with roots, as normal shoots, were initiated occasionally on the hormone-free medium. These shoots also formed new genetic tumor on the stem, which leaves formed lots of teratoma shoot on the hormone-free medium in in vitro culture.
In vitro culture of shoot tip of garlic (Allium sativium L. cv. Seosan) was carried out to find medium condition of the induction of multiple shoots and bulbing for muliproduction of virus-free seed bulbs. For this work, tank culture was introduced. In shoot tip culture on MS solid medium the induction of multiple shoots and bulbing were better by adding 3% sucrose than 8%. Supplementation with 2mg/L 2ip and 0.2 mg/L IAA in this medium was effective. Three point three shoots including 2.7 bulbs were formed from a shoot tip after cultivation for 30 days on this medium. Bulbing of garlic in liquid culture with plastic water tank of 20L supplied air at the side of the lower part was better by adding 3% sucrose than 8% by subculture for 45 days with shoots obtained from shoot tip culture for 30 days on soid MS medium. Shoot growth was vigorous at 3% sucrose however bulb growth was more effective on the medium of 8% sucrose. Because of the effectiveness on solid medium added 3% sucrose, 2 mg/L 2ip and 0.2 mg/L IAA for initial production of multi-shoot in stem tip culture and the effectiveness in liquid culture with water tank for growth of bulbs, the method of two-step culture could be introduced for the multiple production of seed bulb of high quality. It was more desirable by supply of 0.2 mg/L BA and 0.02 mg/L NAA at tank culture time. But growth of the bulbs became poor by increasing concentration of NAA of the medium.
A plant regeneration system was developed through shoot organogenesis from in vitro leaf explants of Chrysanthemum morifolium 'Ohblang'. The effects of different concentrations of plant growth regulators and AgNO3 on efficient shoot regeneration and inhibition of browning were evaluated in chrysanthemum. The explants were cultured on MS shoot induction medium supplemented with 12 combination treatments of 6-benzyladenine (BA) 0.5, 1.0 and 2.0 mg/L, and α-naphthaleneacetic acid (NAA) 0.2, 0.5, 1.0 and 2.0 mg/L in darkness for 6 weeks and cultured under a 16/8 h photoperiod for 6 weeks. The highest shoot regeneration was obtained from the explants cultured on the medium with 1.0 mg/L BA and 1.0 mg/L NAA. Based on this result, AgNO3 was added to a shoot induction medium containing MS salts, vitamins, 1.0 mg/L BA, 1.0 mg/L NAA, 30 g/L sucrose, and 6 g/L agar to reduce browning of chrysanthemum leaf explants. In the control treatment without AgNO3, leaf explants turned brown at the cut edge; however, browning was not observed in AgNO3 treatments. Shoot organogenesis was higher at low concentrations of AgNO3 and decreased with an increase in AgNO3 concentration. The explants cultured on shoot induction medium (MS salts, vitamins, 1.0 mg/L BA, 1.0 mg/L NAA) with 1 mg/L of AgNO3 produced the highest shoot regeneration with 2.6 shoots per explants and a browning index of 0.7. When the regenerated shoots were detached from the explants and cultured on MS medium, the shoots were elongated and rooted successfully.
Plant regeneration protocols for adventitious shoot organogenesis from apple (Malus domestica 'Fuji') leaf explants were developed in the present study. The effects of dark incubation periods in the early stages of culture, pre-treatment methods, the number of explants per culture container, the type of culture containers, and the orientation of the explants on culture media were evaluated to determine the optimal shoot regeneration conditions for 'Fuji' apple leaf explants. Light incubation of explants produced minimal response. However, dark incubation of explants for 4 weeks during the initial culture period enhanced shoot regeneration frequency. Comparing the number of explants per container, a higher percentage of shoot regeneration was obtained with nine explants per container compared with four explants per container. Pre-treatment, before culture, by dipping explants in a liquid regeneration medium containing 40 g/L of sorbitol for 2 hours produced the highest shoot formation rate, and the time of shoot formation was accelerated. The percentage of shoot regeneration and number of shoots per regenerating explant reached a maximum of 87.5% and 4.7, respectively. The regenerated shoots were elongated and rooted on a rooting medium of 1/4 MS with 0.2 mg/L IBA. The plantlets were successfully acclimatized, and the regenerated plants produced normal phenotypes.
A method for induction of multiple shoots using cotyledonary nodes and shoot tips of Macrotyloma uniflorum (Lam.) Verdc. was described. The experiment was conducted in which shoot induction was noticed on Murashige and Skoog (MS) medium supplemented with different concentrations and combinations of four cytokinins (KIN, 2iP, Ads, BAP). These multiple shoots were later developed into normal shoots. The highest rate of shoot proliferation came from MS medium added with BAP 1.5 mg/L. The multiple shoot buds were subcultured into MS medium with BAP (0.5-1.5 mg/L) along with Ads (1.0 mg/L) and GA$_3$ (0.5 mg/L), which gave rise to the highest frequency of shoot proliferation and elongation. The shoots were rooted on MS medium supplemented with 1.75 mg/L IBA.
An effective in vitro multiplication method was developed for clones of Populus davidiana Dode. Ten different media were tested for their effect on shoot multiplication. Both MS and LP medium with 0.2mg/l BAP appeared to be the best for the shoot multiplication with the rate of 9 shoots per explant. There were significant differences among the clones in both multiplication rate and shoot growth. While some clones did not require BAP to promote shoot formation, others did. More than 60% of in vitro shoots rooted on the half-strength GD medium containing 0.2mg/l IBA.
This study was carried out to determine the effects of plant growth regulators on organogenesis from Cymbidium kanran and Cymbidium hybrida. Optimal rhizome formation from Cymbidium kanran was obtained on MS medium with 10 ppm kinetin+2 ppm NAA. and optimal protocorm formation from Cymbidium hybrida was obtained on MS medium with 10 ppm kinetin+0.05 ppm NAA. However, in this study the optimal media for the callus induction from both explants was not identified. Optimal shoot induction from rhizome of Cymbidium kanran was obtained on MS medium with 10 ppm BA+2 ppm NAA and 5 ppm BA+2 ppm NAA. Optimal shoot induction from protocorm of Cymbidium hybrida was obtained on MS medium with 10 ppm kinetin+2 ppm NAA.
