온열처리가 근세포의 분화에 어떤 영향을 미치는 지를 알아보기 위하여 배양한 근세포에 여러가지 온열처리를 한 다음, 단백질 합성, 세포증식, 융합지수, creatine kinase(CK)의 활성 및 cholesterol 함량의 변화를 조사하였다. 배양한지 24시간지나 45$^{\circ}C$에서 1 hr의 온열처리를 하면 근세포의 융합과 CK 활성이 지연되었으며, 55시간지나 같은 온열처리를 하면 세포막내(세포내 양의 95% 이상)의 chloesterol 양이 일시적으로 증가함과 아울러 세포융합이 지연되었다. 그러나 세포증식은 대조군과 뚜렷한 차이가 없었다. 이상과 같은 실험 결과로부터 온열처리가 분화에 미치는 영향은 온열처리를 받게되는 근원세포의 분화정도에 따라 차이가 있으며 온열처리에 따른 chloesterol 양의 일시적인 증가가 근세포 융합에 영향을 미칠 수 있다는 가능성이 제시되었다. 한편, 근세포는 온열처리를 받으면 평소의 단백질 합성 수준이 떨어짐과 더불어 heat shock protein(HSP)합성이 증대 내지는 유도 되었으며 HSP 합성의 유도는 actinomycin D의 처리로 억제되었다. 또한 온열처리로 근세포는 열내성을 얻어 세포융하과 CK 활성은 동일한 온열처리를 4시간 간격으로 두번 주어도 한번 주었을 경우와 차이가 없었으며 두번째 온열처리에 의해서는 새로운 HSP 합성이 유도되지도 않았다.
Steady shear response of a magnetorheological fluid (MRF) system containing porous mono-disperse magnetite ($Fe_3O_4$) spheres synthesized by solvothermal method is demonstrated. In applied magnetic field the interaction between the spherical particles leads to form strong columnar structures enhancing the yield strength and viscosity of the MRFs. The yield strengths of the MRFs also scale up with the concentration of magnetic particles in the fluid. Considering magnetic dipolar interaction between the particles the magneto-mechanical response of the MRFs is explained. Unlike metallic iron particles, the low-density corrosion resistant soft-ferrimagnetic $Fe_3O_4$ spherical particles make our studied MRF system efficient and reliable for shock-mitigation/vibration-isolation applications.

Recent evidence has shown that methionine (Met) supplementation can improve milk protein synthesis under hyperthermia (which reduces milk production). To explore the mechanism by which milk protein synthesis is affected by Met supplementation under hyperthermia, mammary alveolar (MAC-T) cells were incubated at a hyperthermic temperature of 42℃ for 6 h in media with different concentrations of Met. While the control group (CON) contained a normal amino acid concentration profile (60 ㎍/mL of Met), the three treatment groups were supplemented with Met at concentrations of 10 ㎍/mL (MET70, 70 ㎍/mL of Met), 20 ㎍/mL (MET80, 80 ㎍/mL of Met), and 30 ㎍/mL (MET90,90 ㎍/mL of Met). Our results show that additional Met supplementation increases the mRNA and protein levels of BCL2 (B-cell lymphoma-2, an anti-apoptosis agent), and decreases the mRNA and protein levels of BAX (Bcl-2-associated X protein, a pro-apoptosis agent), especially at an additional supplementary concentration of 20 ㎍/mL (group Met80). Supplementation with higher concentrations of Met decreased the mRNA levels of Caspase-3 and Caspase-9, and increased protein levels of heat shock protein (HSP70). The total protein levels of the mechanistic target of rapamycin (mTOR) and the mTOR signalling pathway-related proteins, AKT, ribosomal protein S6 kinase B1 (RPS6KB1), and ribosomal protein S6 (RPS6), increased with increasing Met supplementation, and peaked at 80 ㎍/mL Met (group Met80). In addition, we also found that additional Met supplementation upregulated the gene expression of αS1-casein (CSN1S1), β-casein (CSN2), and the amino acid transporter genes SLC38A2, SLC38A3 which are known to be mTOR targets. Additional Met supplementation, however, had no effect on the gene expression of κ-casein (CSN3) and solute carrier family 34 member 2 (SLC34A2). Our results suggest that additional Met supplementation with 20 ㎍/mL may promote the synthesis of milk proteins in bovine mammary epithelial cells under hyperthermia by inhibiting apoptosis, activating the AKT-mTOR-RPS6KB1 signalling pathway, and regulating the entry of amino acids into these cells.
An antiserum against the carrot HSP17 (17 KDa heat shock protein) was raised using the HSP17 purified after being expressed in a recombinant E.coli in order to develop an assay system for thermotolerance in crops. The DCHsp17.7 including the coding sequence corresponding to a carrot HSP17 protein was recombined within pET-32(b) vector and achieved a maximum expression in 4 hours after an induction in E.coli. The purified DCHsp17.7 was used as an antigen to generate the corresponding antibody. The polyclonal antiserum was confirmed for it's specificity only to the low molecular weight (1mw) HSP. Besides, the possibilities to use the antiserum to interact with 1mwHSPs from other plants such as rice, cucumber, tomato, and hot pepper were examined to be plausible. To reveal any specific correlation between the amounts of 1mwHSP expressed upon HS conditions and an acquisition of thermotolerance two different approaches have been applied. first, it has been shown that only the pre-HS conditions inducing the synthesis of HSP17 allowed for the seedlings to achieve an thermotolerance and to survive the following lethal condition. Second, a western analysis using 15 different collected lines of hot peppers was performed to distinguish each other in terms of the amount of 1mwHSP. The results indicated that all 14 hot pepper lines were able to synthesize HSPs in response to an exposure to HS conditions and the amounts of the proteins synthesized at different HS temperatures were variable among the lines. There are several different patterns of 1mwHSP synthesized as a function of temperature increase observed and their correlation to physiological aspects of thermotolerance remains to be analyzed.
Several stresses are known to induce synthesis of heat shock protein The present study was performed to see whether pulmonary ischemia, induced by the bronchial artery occlusion, produced HSP70 in cat lung. To This aim, we compared experimental and control groups of cats with respect to the HSP70 production in the lung. Experimental animals were subjected to 10-min bronchial artery occlusion followed by reperfusion. The interval between the end of the occlusion and the end of the reperfusion was 1 hour, 4 hours and 8 hours, whereas control animal was not subjected 10 any manipulation except anesthesia. According to the interval differences, experimental animals were divided into 1HR, 4HRs and 8HRs groups. To determine The induction of HSP70 in each group, total proteins of lung tissues were extracted and separated by PAGE electrophoresis. Immunoblotting with a mouse monoclonal anti -HSP70 IgG antibody revealed that HSP70 was not detected in the pulmonary tissues resected from control, 1HR or 4HRs groups. In contrast. HSP70 expression in 8HRs group was marked. These results suggest that pulmonary ischemia by the bronchial artery occlusion produces HSP70 in a delayed manner.
Although reoxygenation is the best way to salvage hypoxic tissues, reduced oxygen species (ROS) generated during reoxygenation are blown to cause further tissue injuries and the induction of heat shock proteins (HSPs). The present study was undertaken to determine any causal relationship between the severity of hypoxia and the opposite outcomes, either beneficial or detrimental, of the subsequent reoxygenation by measuring the HSP72. To this aim, one group (6 male cats, $2.5{\sim}3.5\;kg$) was subjected to a 5-min episode of hypoventilation (H, ventilation rate: 5/min) for the induction of slight hypoxia and the other group (6 male cats, $2.4{\sim}3.7\;kg$) was subjected to a 5-min episode of apnea (A) for severe hypoxia. Each 3 animals from both groups received a 10-min episode of ventilation with $(95%\;O_2\;(0)$, whereas the remainder did not. After these procedures, all animals were allowed to be ventilated within physiological range for 1, 4, or 8 hours (1H, 1HO, 4H, 4HO, 8H, 8HO, 1A, 1AO, 4A, 4AO, 8A and 8AO groups). Control animals did not receive any manipulation. The arterial blood $pCO_2$ was significantly higher just after apnea than hypoventilation, while $pCO_2$ and pH were significantly lower just after apnea than hypoventilation. Western blot analysis revealed that the magnitude of HSP72 synthesis is larger in 1H, 4H and 8H groups than in 1HO, 4H and 8HO groups, respectively. In contrast, 1AO, 4AO and 8AO groups more induced HSP72 than 1A, 4A and 8A groups, respectively. These results suggest that the reoxygenation is beneficial after slight hypoxia but detrimental after severe hypoxia.

The effect of preadaptation at low temperature on cryoprotection was studied for Lactobacillus paraplantarum C7, a bacteriocin producer isolated from kimchi. L paraplantarum C7 cells in their log growth phase were incubated at $15^\circ{C}$, $10^\circ{C}$, or $5^\circ{C}$ for 2, 4, and 6 h, respectively, before being frozen at $-70^\circ{C}$. After 24 h of freezing, viable cells were counted after brief thawing. The freezing-thawing cycles were repeated three more times. Cells preadapted at $10^\circ{C}$ or $5^\circ{C}$ before freezing survived better than control cells, but preadaptation at $15^\circ{C}$ did not confer cryoprotection. Chloramphenicol addition did not destroy the cryoprotection, indicating that protein synthesis was not required for the development of cryoprotection. SDS-PAGE showed induction of a 6.5-kDa protein, a major cold-shock protein, in preadapted cells.
본 연구에서 L. crispatus KLB46을 저온 전처리함으로서 제제화 과정에 받게 되는 얼림과 녹임, 건조 스트레스뿐만 아니라, 여러 다른 환경 스트레스에 대한 내성이 증진된다는 것을 확인하였다. 그리고 내성 증진에 신규 단백질 합성이 필요함을 확인하였으며 나아가, 저온 충격 유전자 (csp)를 확인하였다. 따라서 이 균주를 제제화 하기 위한 방법으로 저온 전처리를 이용할 경우 생균력 유지에 큰 효과를 얻을 수 있다고 사료된다.
고주파 처리는 파밤나방(Spodoptera exigua)의 생리변화를 유발시켜 섭식행동, 발육 및 면역반응의 변화를 초래한다. 본 연구는 이러한 고주파의 영향을 파밤나방의 생화학적 변화를 통해 분석했다. 고주파(5,000 Hz, 95 dB) 처리는 중장 상피세포의 단백질 합성과 분비를 억제시켰다. 또한 이 고주파 처리는 중장의 인지질분해(phospholipase $A_2$) 소화효소의 활성을 현격하게 억제시켰다. 고주파 처리는 세 종류의 열충격단백질과 지질운송단백질(apolipophorin III)의 유전자 발현을 변동시켰고 이러한 변화는 중장 조직에서 뚜렷했다. 혈림프 혈장에 존재하는 지질 및 유리당의 함량이 고주파 처리에 의해 현격하게 증가했다. 이러한 결과는 고주파 처리가 파밤나방의 체내 생화학적 변화를 유발시켜 생리적 교란을 유도하는 스트레스로 작용한다는 것을 제시하고 있다.
유기용매에서 효소를 이용하면 다양한 선택적 반응을 쉽게 수행할 수 있어 산업적 적용 가능성이 매우 높지만, 효소의 안정성 저하는 아직까지 큰 문제 중의 하나로 남아있다. 유기용매에서 효소 반응시 효소의 실활 원인과 효소의 안정화 방법 연구를 위하여 단백질의 folding에 관여하는 molecular chaperone의 일종인 heat-shock protein Hsp90을 이용하여, 대표적인 유기용매 반응시스템에서의 과산화효소 HRP 안정성 향상 연구를 수행하였다. 그 결과 Hsp90은 30% DMSO, 30% 및 50% dioxane 완충용액에서 HRP의 실활 방지 효과를 보였고, 실활된 효소의 재생에도 탁월한 효과를 보였다. 그리고 형광분석과 CD(circular dichroism)에 의한 구조분석을 수행하여 Hsp90이 유기용매에 의해 unfolding되어 있는 효소를 다시 refolding하는 데 기여함을 알았다.
