In the present study, mycosporine-like amino acids (MAAs)were extracted from in Chlamydomonas hedleyi, and their function was investigated regarding to protective capacity against UV radiation and a possibility to be developed into functional suncream including MAAs by using natural compounds. In particulr, we assessed UV protective ability and anti-inflammation of shinorine in human skin cells. As a result, shinorine can protect the skin against damage by the absorption of energy from UV radiation and functions as an anti-wrinkling substrate through a increase of collagen synthesis. These data suggest that shinorine can be utilized not only as a substrate to protect UV radiation, but anti-aging material in cosmetic products.
Prions are proteinaceous infectious particles that cause neurodegenerative diseases, such as scrapie in sheep, bovine spongiform encephalopathy in cattle and Creutzfeldt-Jakob disease (CJD) in humans. Although the detailed process, regarding the abnormal conversion of prion proteins (PrP), remains to be fully elucidated, a number of environmental factors appear to affect the formation of misfolded PrP, termed PrPSc. Because oceanic algae contain mycosporine-like amino acids (MAAs), which exhibit cellular defensive activities under a variety of stress conditions, we investigated the level of PrPSc in prion-infected neuroblastoma cells using mycosporine-glycine, porphyra-334 and shinorine. When judged by the level of protease-resistant PrPSc in western blots, porphyra-334 and shinorine increased the level of PrPSc in cells, but mycosporine-glycine did not. The current results indicate that the MAAs tested in this study enhance the formation of PrPSc.
We investigated ultraviolet (UV) absorption materials from Russian seaweeds. First, the UV absorptivities of five seaweeds Gloiopeltis fucatas, Mazzaella sp., Mastocarpus pacificus, Laminaria cichorioides, Saccharina japonica were evaluated by a UV spectrometer. Of these seaweeds, Gloiopeltis fucatas and Mazzaella sp. showed high levels of UV absorption. Column chromatography of active 50% aqueous ethanol extracts from Gloiopeltis fucatas and Mazzaella sp. resulted in the purification of two known compounds. Spectroscopic techniques identified their structures as shinorine and palythine. These materials exhibited UV absorptive capabilities at wavelengths of 333 and 320 nm, respectively. These results suggest that Gloiopeltis fucatas and Mazzaella sp. may be useful as natural cosmeceutical sources.

To examine the effects of a mycosporine-like amino acids (MAAs) mixture from microalgae, Chlamydomonas sp, on the anti-wrinkle activities, the expression levels of genes that are associated with skin aging, including type I procollagen, elastin and involucrin, were analyzed. Asterina 330+palythine (A+P) and shinorine+palythine (S+P) mixtures were purified from Chlamydomonas sp using the following steps: 80% methanolic extraction, column purification, and HPLC analysis. As a result of the MTT assay, A+P and S+P did not induce cellular cytotoxicity with up to 0.1 mg/mL of both MAAs. In addition, the treatment of UV-exposed fibroblasts with A+P (0.05 mg/mL) and S+P (0.01 mg/mL) increased the levels of the PCOLCE mRNAs by 2.7 and 3.6 fold compared to the control group, respectively, The levels of elastin gene expression were elevated 5.59 and 3.1 fold in the A+P and S+P treated (0.01 mg/mL) cells, respectively. In particular, at a concentration of 0.01 mg /mL, the A+P and S+P expression levels of Involucrin mRNAs were increased 3.5 and 2.5 fold in the UV-exposed cells compared to the control, respectively. In conclusion, the MAAs derived from Chlamydomonas sp can be utilized as functional cosmetic materials with anti-wrinkle effects on the skin.
The UV-B sensitivity was tested for the intertidal species Chondrus ocellatus from Korea, by measuring photosynthesis estimated as effective quantum yield ($\Phi_{PSII}$) of photosystem II (PS II), growth and content and composition of photosynthetic pigments and UV-absorbing pigments (UVAPs). The $$\Phi_{PSII}$ of the alga decreased with increasing time of exposure to UV-B radiation, followed by fast and nearly full recovery indicating dynamic photoinhibiton. Fresh weight-based growth and pigment contents of C. ocellatus were not seriously affected by UV-B radiation. A single broad peak at 327 nm was obtained from methanol extracts of C. ocellatus, and the absorbance peak increased with increasing UV. The single peak was resolved into three peaks (311, 330 and 336 nm) by the fourth -derivative, and quantitative change in response to UV-B radiation occurred only at 330 nm. High performance liquid chromatography (HPLC) analysis of purified extracts indicated that three MAAs (mycosporine-like amino acids) are present, asterina 330, palythine and shinorine. Field observations during three growing months showed that C. ocellatus exhibit the highest amount of UVAPs in May followed by July and little trace in September, coinciding with the species' phenology. In an ecological context, dynamic photoinhibition as well as accumulation of UVAPs may enable the shallow water red alga C. ocellatus to be well adapted to high UV-B environments.
Cyanobacteria have been widely reported to produce a variety of UV-absorbing mycosporine-like amino acids (MAAs). Herein, we reported production of the unusual MAA, mycosporine-glycine-alanine (MGA) in the cyanobacterium Sphaerospermopsis torques-reginae ITEP-024 using a newly developed UHPLC-DAD-MS/HRMS (ultra-high performance liquid chromatography-diode array detection-high resolution tandem mass spectrometry) method. MGA had previously been first identified in a red-algae, but S. torques-reginae strain ITEP-024 is the first cyanobacteria to be reported as an MGA producer. Herein, the chemical structure of MGA is fully elucidated from one-dimensional / two-dimensional nuclear magnetic resonance and HRMS data analyses. MAAs are unusually produced constitutively in S. torques-reginae ITEP-024, and this production was further enhanced following UV-irradiance. It has been proposed that MAA biosynthesis proceeds in cyanobacteria from the pentose phosphate pathway intermediate sedoheptulose 7-phosphate. Annotation of a gene cluster encoded in the genome sequence of S. torques-reginae ITEP-024 supports these gene products could catalyse the biosynthesis of MAAs. However, addition of glyphosate to cultures of S. torques-reginae ITEP-024 abolished constitutive and ultra-violet radiation induced production of MGA, shinorine and porphyra-334. This finding supports involvement of the shikimic acid pathway in the biosynthesis of MAAs by this species.
Growth and pigment responses to different levels of solar radiation with or without ultraviolet (UV)-B component $({\lambda}=280-315nm)$ were investigated in Antarctic rhodophytes, Kallymenia antarctica and Palmaria decipiens, collected around King George Island during the summer of 2000. In K. antarctica specific growth rate, based on thallus area or fresh weight, decreased with increasing solar irradiances while P. decipiens were relatively insensitive to the effects of light. It is noticeable that the presence or absence of UV-B had no significant effect on growth for either species. However, K. antarctica showed a more pronounced reduction in chlorophyll (Chl a) concentrations at higher irradiances in the presence of UV-B. In P. decipiens, Chl a concentrations did not differ despite radiation level fluctuations being lower albeit than initial measurements. Thallus thickness was greater in K. antarctica than in P. decipiens. There were higher relative amounts of UV-absorbing pigments (UVAPs) in P. decipiens than in K, antarctica. The single absorbance peak obtained from the methanol extracts was resolved into three (316,332 and 346nm) in K. antarctica and four peaks (315,326,333 and 349 nm) in Palmaria as a result of the fourth-derivative. After 7 days exposure to solar radiation, the amount of UVAPs in K. antarctica was significantly reduced to a similar degree at all light levels, whereas that of P. decipiens remained unchanged except at 5% of surface irradiance. High performance liquid chromatography (HPLC) analysis of purified extracts indicated that P. decipiens possesses porphyra-334 in addition to three other mycosporine-like anlino acids (MAAs; asterina-330, palythine, shinorine), which are commonly present in K. antarctica. Significantly lower tolerance of K. antarctica to high levels of solar radiation may be connected with its usual absence in the eulittoral, while the active growth and elastic pigment responses of P. decipiens over a wide range of solar irradiance levels up to full sunlight seems to correspond well with its wide vertical distribution from rock pools down to 25-30m.
