자갈이 포함된 모래의 비배수 전단거동 (Undrained Shear Behavior of Sand with Dispersed Gravels)

  • 풍화 잔류토, 토석류, 산사태, 또는 매립 지반에서 자갈과 같은 굵은 입자가 모래나 점토와 같은 작은 입자로 둘러 쌓여져 있는 경우가 있다. 작은 입자 사이에 굵은 입자가 고립된 상태로 존재하는 혼합토의 강도는 흙에서 대부분을 차지하는 작은 입자 즉 모래나 점토의 역학적 특성에 따라 좌우되지만 흩어져 있는 굵은 입자인 자갈의 크기, 모양, 함유량 등에 의해 영향을 받는 경우도 있다. 본 연구에서는 이와 같이 모래 지반 내에 흩어져 있는 소량의 자갈이 모래의 전단강도에 미치는 영향을 연구하였다. 습윤 상태의 낙동강모래를 이용하여 각층 높이의 중간부분에 굵은 자갈 또는 작은 자갈을 넣고 다음 층을 쌓아 다지는 방식으로 5층으로 된 조밀한 공시체를 제작하였다. 각층 높이의 중간부분에 들어간 굵은 자갈과 작은 자갈의 중량비를 0, 3, 9, 14%로 달리하면서 다양한 공시체를 제작하여 압밀시킨 다음 비배수 삼축압축시험을 실시하였다. 혼합되는 굵은 자갈의 중량비(개수)가 증가할수록 자갈을 포함한 낙동강모래의 최대축차응력은 최대 38%까지 감소하였으며, 이와 같은 굵은 자갈로 인한 최대축차응력 감소는 구속압이 증가할수록 줄어드는 경향을 보였다. 하지만 공시체 내에 포함된 작은 자갈의 중량비가 증가할수록 최대축차응력은 오히려 증가하였으며, 자갈의 중량비가 3, 9%로 작을 경우 최대축차응력의 증가는 미미하였으나 14%로 증가할 경우에는 최대축차응력이 최대 34%까지 증가하였다.

철근콘크리트 특수모멘트골조의 보-기둥 접합부 실험체의 내진성능평가 (Seismic Evaluation of Beam-Column Joint Specimens of RC Special Moment Frames)

  • 본 연구는 철근콘크리트 특수모멘트골조(SMF)내 보-기둥 접합부의 비탄성 회전능력에 대한 연구결과를 조사한 것이다. 모든 실험체들은 ACI 318-02에 정의된 설계 및 세부지침에 따라 특수모멘트골조로 분류되었다. AISC(2002)기준에서 모멘트골조의 접합부에 대한 내진성능의 만족 기준을 이용하여 보-기둥 접합부를 평가하였다. 총 39개의 실험체들에 대해 자세하게 조사되었다. 특수모멘트골조에 대한 내진설계기준을 만족하는 대부분의 접합부들은 3%의 소성회전까지 휨강도의 심각한 감소 없이 연성이 유지되었다. 이는 특수모멘트골조 접합부들에 대한 엄격한 콘크리트 내진설계 규정에 따른 것으로 조사되었다. 접합부내의 횡방향 보의 존재는 보-기둥 접합부의 구속력과 전단에 대한 저항성을 증가시킨 것으로 조사되었다 접합부 전단응력에 대한 ACI 328-02 제한조건을 만족하는 모든 특수모멘트골조의 접합부들은 요구되는 내진성능을 만족하는 것으로 나타났다.

휨항복 후 부착파괴하는 철근콘크리트 부재의 부착 연성 평가 (Evaluation for Deformability of RC Members Failing in Bond after Flexural Yielding)

  • 일반적인 내진 설계에서는 구조물의 연성적인 거동을 유도하기 위해서 보-기둥 접합부에 인접한 보에 소성힌지가 발생하도록 한다. 따라서 철근콘크리트 부재의 부착강도와 전단강도가 휨강도보다 큰 값을 가져야 하고, 전단이나 부착파괴가 요구된 연성에 도달하기 이전에 발생하지 않아야 한다. 하지만 전단경간비가 짧은 부재의 경우에는 전단이나 부착 거동의 지배를 받는 경우가 많고, 핀칭 효과로 인해 에너지 소산이 비교적 적게 발생하므로 요구된 연성에 도달하지 못하고 파괴될 수 있다. 이 논문에서는 전단경간비가 짧은 철근콘크리트 부재의 거동 분석과 연성 예측, 특히 부착 연성 능력을 평가하기 위한 방법을 제안하였다. 이것은 반복하중에 의해 저감되는 잠재 전단강도와 잠재부착내력 모델, 그리고 소성힌지 형성에 따른 휨부착응력의 급격한 증대를 도식화하여 나타낼 수 있다. 제안된 해석법은 각 값의 변화 추이를 비교하여 부재의 거동을 파악하고, 부착 거동의 지배를 받는 부재의 경우, 부착내력과 휨부착응력의 값이 만나는 지점까지를 그 부재의 부착 연성으로 평가하는 방법이다. 이 방법은 기존에 수행된 8개의 보, 기둥 시험체를 통해 비교 및 검토하였으며 부재 거동에 대한 예측은 정확히 일치하였으나, 부착 연성 능력에 대해서는 과소평가 되었다. 그 이유는 부재의 부착강도를 실제 부착강도보다 비교적 낮게 예측한 부착강도식에서 찾을 수 있으며, 다른 부착 내력 모델에 대한 부착 연성 평가에 대한 연구가 추후 필요할 것으로 사료된다.

Identification of Muscle Proteins Related to Objective Meat Quality in Korean Native Black Pig

  • This study examined the effects of pre-slaughter fasting, chasing stress and chiller ageing on objective meat quality, and their relations to the proteome profile of longissimus muscle using 20 male Korean native black pigs. Treatments were composed of two levels of pre-slaughter feed withdrawal, two levels of pre-slaughter stress and four chiller ageing times. A 15 min chasing stress immediately prior to slaughter significantly (p<0.05) decreased detectable levels of $\mu$-calpain activity during rigor development and chiller ageing, but did not have any direct effect on objective meat quality. On the other hand, pigs fed until the morning of slaughter resulted in significantly (p<0.05) higher hunter L* value and cooking loss than those which received an 18 h feed withdrawal prior to slaughter. Cooking loss and hunter L* value were constant during 7 d of chiller ageing, followed by significant increases at 14 d. The fed animals showed a significantly (p<0.05) higher hunter a* value at both 3 and 7 d, while the other group maintained a stable redness for 7 d. WB-shear force was not affected by the pre-slaughter treatments, but had significant (p<0.05) linear reduction from 1 to 7 d. A gelbased proteome analysis was performed on selected animals for low and high hunter L* values at 1 d. Ten and five spots had greater than two-fold spot densities for the low and high hunter L* groups, respectively. The ten spots included chain A, deoxyribounclease I complex with actin, heat shock protein 27 kDa, a protein similar to cardiac $Ca^{2+}$ release channel, and myosin heavy chain, while the five spots included chain A aldehyde dehydrogenase, glycerol-3 phosphate dehydrogenase, and hemoglobin alpha chain. In general, feeding until the morning of slaughter resulted in more desirable meat color, but appeared to reduce palatability due to increased cooking loss. Proteome analysis demonstrated that various proteins were concomitantly involved in the determination of final meat color. The most noticeable observation in the current study was that various isoforms for a particular protein differed in degradation and/or expression rate depending on meat quality.

Diatom 혼합토의 공학적 특성 (Engineering Characteristics of Diatom Modified Soil Mixture)

  • Diatom 성분이 함유된 자연 퇴적토층의 공학적 특성은 대상층의 변형 및 강도의 특이한 특성 때문에 다수의 연구자에 의해 수 십년간 연구되어 왔다. 원반형 혹는 원통형 모양을 한 diatom의 존재는 재료의 낮은 압축성, 높은 투수성, 높은 전단강도 특성을 나타내게 하며, 이러한 특이한 차이점들은 큰 내부입자 간극과 모난 입자의 결합력으로 diatom의 특성을 나타낸다. 이러한 현상은 흙의 공학적 특성을 변화시켜서 지반개량 재료로써 diatom을 이용할 수 있음을 의미한다. 본 논문은 diatom 개량토의 공학적 특성을 연구하기 위해 diatom-kaolin 혼합토의 공학적 특성을 통상적인 지반공학 실험 및 탄성파를 이용한 미소변형 실험, 전자기파를 이용한 전기저항 측정실험 등을 통하여 평가하였다. 실험 결과에 따르면, diatom 함유량에 따라 흙의 공학적 개선이 이루어진 것을 확인할 수 있었으며, 높은 응력에서 diatom 입자의 결합이 파괴되어 일부 공학적 특성이 저감되는 것을 확인 할 수 있었다.

형질전환 벼 현탁세포 배양에서 hCTLA4Ig의 in situ 회수 (In situ Recovery of hCTLA4Ig from Suspension Cell Cultures of Oryza sativa)

  • In this research, recombinant human cytotoxic T-lymphocyte antigen 4-immunoglobulin (hCTLA4Ig) was produced by transgenic rice cells. RAmy3D promoter was used for overcome the limitation of low expression level in transgenic plant cells, and the secretion of target protein was accomplished by signal peptide. However, the RAmy3D promoter system which can be induced only by sugar starvation causes the decrease of cell viability. As a result, cell death promotes the release of protease which degrades the target proteins. The protein stability and productivity can be significantly influenced by proteolysis activity. Therefore, development of new strategies are necessary for the in situ recovery of target proteins from cell culture media. In this study, in situ recovery was performed by various strategies. Direct addition of Protein A resin with nylon bag leads to cell death by increased shear stress and decrease in production of hCTLA4Ig by protease. Medium exchange through modified flask could recover hCTLA4Ig with high cell viability and low protease activity, on the other hand, the productivity was lower than that of control. When in situ recovery was conducted at day 7 after induction in air-lift bioreactor, 1.94-fold of hCTLA4Ig could be recovered compared to control culture without in situ recovery. Consequently, in situ recovery of hCTLA4Ig from transgenic rice cell culture could enhance productivity significantly and prevent degradation of target proteins effectively.

변형률에 따른 모래-고무 혼합재의 거동 특성: 실험적 관찰 (Characteristics of Sand-Rubber Mixtures under Different Strain Levels: Experimental Observation)

  • 다양한 모래부피비($sf=V_{sand}/V_{total}$)를 가지는 시료를 조성하여 변형률 크기에 따른 모래-고무 혼합재($D_{sand}/D_{rubber}=1$)의 거동을 분석하였다. 공진주시험, 압밀시험, 그리고 직접전단시험을 실시하였다. 변형률 크기에 따라 모래와 고무는 혼합재 전체 거동을 서로 다르게 제어한다. $sf{\geq}0.4$의 혼합재는 비선형 전단강성의 감소가 관찰되는 반면, $sf{\leq}0.2$의 낮은 모래부피비를 가지는 혼합재는 상당히 높은 탄성한계변형률을 보인다. 고무 입자가 force chain의 역할을 수행할 때 수직변형은 급격한 증가를 보인다. 혼합재 내의 고무부피비가 감소함에 따라 혼합재의 강도는 증가하는 경향을 보이며 $sf{\leq}0.8$의 혼합재는 전단변형에 따라 부피 수축 거동을 보인다. 고무 입자는 변형률 크기에 따라 혼합재 내에서 서로 다른 역할을 수행한다: 미소변형률 영역에서는 혼합재 내의 접촉수 증가 및 소성의 제어; 중간변형률 영역에서는 force chain의 좌굴 방지; 그리고 대변형률 영역에서는 혼합재의 부피수축 거동을 이끈다.

절리암반의 변형률 의존적 전단탄성계수 및 감쇠비 특성을 고려한 터널의 내진 해석 (Seismic analysis of tunnel considering the strain-dependent shear modulus and damping ratio of a Jointed rock mass)

  • 암석과는 달리 절리암반은 변형률 의존적 변형특성(탄성계수 및 감쇠비)을 나타낸다. 탄성파를 이용한 현장실험을 통해 미소변형률 수준에서 암반의 최대탄성계수를 얻을 수 있으며 이를 내진 설계에 반영하고 있으나, 미소 변형률 이상의 중변형률($10^{-4}{\sim}0.5%$) 영역의 동적거동에 대한 실험적인 규명과 이에 대한 수치적 적용은 전무한 실정이다. 본 연구에서는 변형률 의존적 전단탄성계수 및 감쇠비의 비선형 거동 특성을 반영하여 동적해석을 수행할 수 있는 FLAC3D 해석 모듈을 개발하였다. 리커 웨이브의 파동 변화를 분석하여 개발된 모듈에 대한 검증을 수행하였다. 절리 암반의 탄성파 전파특성과 동적 거동특성을 모사할 수 있는 절리암반 공진주 시험장비를 통하여 현장에서 채취한 절리암반의 변형률 의존적 전단탄성 계수의 감쇠 특성과 감쇠비의 증폭 특성을 획득하였다. 개발된 비선형 해석 모듈에 실험으로부터 획득된 거동 특성을 반영하여 수직구와 사갱의 접속부에 대한 내진 안정성 평가를 수행하였다. 내진해석 결과, 비선형 해석이 선형 해석보다 더 큰 연직변위와 수평변위 결과를 나타냈다. 라이닝의 휨압축응력은 수직구과 사갱의 접속부에서 집중되는 것으로 나타났으며 비선형해석의 경우 라이닝에 더 큰 휨압축응력이 발생되는 것으로 나타났다. 본 연구를 통하여 변형률 의존적 절리암반의 비선형 거동특성을 보다 깊이 있게 이해하고 해석 및 설계시 고려할 수 있을 것으로 사료된다.

인산염 농도에 따른 물성 변화로 발생하는 황색포도상구균 바이오필름 제거 현상 (Phosphate Concentration Dependent Degradation of Biofilm in S. aureus Triggered by Physical Properties)

  • 본 연구는 인체 친화적인 소재로 균을 제거하는 기술을 만들기 위해서 진행하였다. 균총 상호 균형에 중요한 역할을 하는 황색포도상구균이 바이오필름을 생성시킬 때 다양한 농도의 인산염 투입시 나타나는 물성변화를 조사하였다. 원자현미경을 이용해서 인산염 5 mM 처리시 황색포도상구균 바이오필름의 크기와 경도가 통계적으로 유의차가 있게 최소값을 가짐을 관찰하였다. 염료 태깅법으로 흡광도를 관찰한 결과 인산염과 함께 성장한 전체 바이오필름의 농도도 감소한 것을 발견하였다. 이것이 카운터 이온으로서 작용하는 염에 의한 영향인지 확인하기 위하여 소금을 같은 조건에서 처리해보았는데 이때는 바이오필름의 농도 감소가 관찰되지 않았고 이를 통해 인산염이 특별한 생리적인 작용에 관여함을 알 수 있었다. 비행시간형 이차이온 질량분석기를 통해서 이온 검출량을 평가하여 바이오필름 구성성분을 분석한 결과 인산염을 투입하기 전과 후의 모든 바이오필름 외곽에서 세균막만 감지되었는데 특별히 인산염 5 mM에서 이 세균막의 농도가 가장 낮음을 확인하였다. 바이오필름 내부에 어떤 물성 변화가 일어났는지 관찰하기 위해서 시어 응력을 조절하는 유변기기로 바이오필름의 점탄성 특징을 측정을 하니 인산염 5 mM에서 바이오필름의 점도는 변화하지 않았으나 탄성률 감소가 일어난 것을 관찰하였다. 이것을 통해 인산염이 5 mM인 환경에서 균은 내부 탄성률 감소를 통해서 세균막을 탈피시키는 것을 알 수 있었다. 인산염 농도 5 mM에서 관찰되는 세균막 농도 감소는 균이 더 많은 성장을 하기 위해 다른 곳으로 이동하기 쉽게 하기 위해서 스스로 표면에서 탈착하는 것과 연관이 있음을 제시하였다. 마침내 인산염을 투입하면 균의 세균막 제거가 유도되어 결론적으로 황색포도상구균이 쉽게 제거될 수 있음을 밝혀내었다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.
