Assessment of the Therapeutic Potential of Persimmon Leaf Extract on Prediabetic Subjects

  • Dietary supplements have exhibited myriads of positive health effects on human health conditions and with the advent of new technological advances, including in the fields of proteomics, genomics, and metabolomics, biological and pharmacological activities of dietary supplements are being evaluated for their ameliorative effects in human ailments. Recent interests in understanding and discovering the molecular targets of phytochemical-gene-protein-metabolite dynamics resulted in discovery of a few protein signature candidates that could potentially be used to assess the effects of dietary supplements on human health. Persimmon (Diospyros kaki) is a folk medicine, commonly used as dietary supplement in China, Japan, and South Korea, owing to its different beneficial health effects including anti-diabetic implications. However, neither mechanism of action nor molecular biomarkers have been discovered that could either validate or be used to evaluate effects of persimmon on human health. In present study, Mass Spectrometry (MS)-based proteomic studies were accomplished to discover proteomic molecular signatures that could be used to understand therapeutic potentials of persimmon leaf extract (PLE) in diabetes amelioration. Saliva, serum, and urine samples were analyzed and we propose that salivary proteins can be used for evaluating treatment effectiveness and in improving patient compliance. The present discovery proteomics study demonstrates that salivary proteomic profile changes were found as a result of PLE treatment in prediabetic subjects that could specifically be used as potential protein signature candidates.

주단위 철분보충 방식이 임신부의 철분과 아연 영양상태에 미치는 효과 (The Effect of Weekly Iron Supplementation on Iron and Zine Nutritional Status in Pregnant Women)

  • The purpose of this research is to compare the effdct of the new weekly iron supplementation on maternal iron and zinc nutritional status with the effect of the present daily iron supplementation. To assess the iron ad zinc status of rpegant women visiting public health center in Ulsan, interview for dietary survey and general aspect of each subject was given and biochemical analysis of blood and urine was performed. The study subjects were divided to two groups randomly, but the subjects whose Hgb level was very low were allocated in daily group ethically because theeffect of weekly supplementation was not yet established as safety. Daily group received 80mg elemental Fe(250mg Fe as ferrous sulfate) per day for 100days, while weekly group received 160mg elemental Fe once a week for 15 weeks. After treatement, Hgb(p<0.01), Hct(p<0.01) , serum iron(p<0.05) and serum ferritin(p<0.001) in weekly group but RBC, Hgb , Hct , serum iron and serum iron and serum ferritin increased slightly. After correction for the initial Hgb by including it in the MANIVA, the difference in treatment effects of Hgb and TIBC between groups was statistically significant. The effect of weekly vs, daily iron supplementation program on zinc status was also studied. The difference in treatment effect between two groups was, however, not sifnificant. It is concluded that once weekly rion supplementation program in pregnant women was less effectgive to improve the iron status than daily iron supplementation program. However, because the weekly dose prevented a decline in Hgb as well as in ferritin, itw effect was positive , given the fact that non-supplemented women almost always exhibit a sifnificant drop in Hgb values. It seems that it will improve the iron status well if the amount or frequency of supplements is to be adjusted . Biweekly supplementation of a different dose or antother type of iron supplement, such as through a gastric delivery system, might be better.

자궁내막증 치료 전후 환자의 혈청이 생쥐 난자의 수정률에 미치는 영향 (The Effect of Serum Obtained before and after Treatment for Endometriosis on in vitro Fertilization Rate of Mouse Oocyte)

  • Objective: To evaluate the effect of serum obtained before and after treatment for endometriosis on in vitro fertilization and development of two cell mouse embryo. Design: Pretreatment and posttreatment comparoson of fertilization of mouse oocyte and embryo development in serum supplement from patients with endometriosis; result were compared using Stuent T-test analysis. Method: Infertility Clinic, Department of Obstetrics and Gynecology, Collage of Medicine, Won kwang university, Korea. Patients was chosed eleven consecutive women with endometriosis. Interventions was all patient underwent laparoscopic or conservative surgery. This was followed by a 6-month course of burserelin acetate $900{\mu}g/d$. Main outcome was measured total number of fertilization and embryo that was fertilization after 24 hours and reached blastocyst stage after 72 hours of incubation were compared before and after treatment. Result: Before treatment, 47% of the oocyte were fertilized and 31% of the embryo reached blastocyst stage. After treatment, Significantly more fertilized and Significantly more embryo developed to blastocyst on the stage I and II of endometriosis. Conclusion: The fertilization and embryo toxicity of serum samples from patients with endometriosis is lost after treatment.

비타민 C 보충이 인슐린 비의존형 당뇨병 환자의 혈당 및 혈청지질에 미치는 영향 (Effect of Vitamin C Supplementation on Blood Sugar and Serum Lipid in NIDDM Patients)

  • The purpose of this study is to determine the effect of oral vitamin C supplements on blood sugar and serum lipid level(total cholesterol, triglyceride, high density lipoprotein, low density lipoprotein) in non-insulin independent diabetes mellitus. The study design was a non equivalent control group pre-test post-test design. Data for the study were collected from June 24 to August 31, 2001. The ninty-five research subject were assigned to experimental group(51) and control group(44). Vitamin C(3g/day) was given to 51 subjects for 4 weeks. Following a 12h overnight fasting, blood sample was obstaind at baseline and at the end of 4week - supplementation. Blood samples were taken for plasma vitamin C concentration, fasting blood sugar, HbA1c and serum lipid level. The pre-equivalent test was used by Chi-squre, t-test and two group's pre and post experimental differences were analyzed by t-tset to compare with each other. The results of this study were as follows ; 1. The difference between the two groups in plasma vitamin C concentration was significant(t=-12.950, p=.000). 2. The difference between the two groups in fasting blood sugar was significant(t=5.293, p=.000). 3. The difference between the two groups in HbA1c was not significant(t=1.758, p=.082). 4. The difference between the two groups in total cholesterol, HDL, LDL was significant(t=3.786, p=.000 ; t=-5.515, p=.000 ; t= 4.169, p = .000). These results suggest that megadose vitamin C supplementation be effective in lowering fasting blood sugar, serum lipids and increasing plasma vitamin C. Thus dietary measures to increase plasma vitamin C may be on important health strategy for reducing the compliance of diabetic patients.

Vitamin D in athletes: focus on physical performance and musculoskeletal injuries

  • [Purpose] The aim of this review was to discuss the effects of vitamin D on physical performance and musculoskeletal injuries in athletes and provide information on the field applications of vitamin D. [Methods] A systematic review was conducted to identify studies on vitamin D in athletes that assessed serum vitamin D levels, vitamin D and physical performance, vitamin D and musculoskeletal injuries, and practical guidelines for supplementation of vitamin D. [Results] Several studies reported that a high proportion of athletes had vitamin D insufficiency or deficiency. Low serum levels of vitamin D in athletes were more pronounced in winter than in other seasons, and indoor athletes had lower serum vitamin D levels than outdoor athletes. Low vitamin D levels have been demonstrated to have negative effects on muscle strength, power, and endurance; increase stress fractures and other musculoskeletal injuries; and affect acute muscle injuries and inflammation following high-intensity exercises. Therefore, periodic assessment and monitoring of vitamin D levels are necessary in athletes; the recommended serum level of 25(OH)D is > 32 ng/mL and the preferred level is > 40 ng/mL (-1). In those with low levels of vitamin D, exposure to sunlight and an improved diet or supplements may be helpful. Particularly, 2000-6000 IU of supplemental vitamin D3 can be consumed daily. [Conclusion] Vitamin D is a potential nutritional factor that can significantly affect physical performance and musculoskeletal injuries in athletes. The importance and role of vitamin D in athletes should be emphasized, and the current levels of vitamin D should be assessed. Therefore, it is essential to periodically evaluate and monitor serum vitamin D levels in athletes.

임신기간 중 철 섭취가 모체와 제대 혈청의 미량 무기질 농도에 미치는 영향 (Iron Intake During Pregnancy on Serum Concentrations of Trace Minerals in Mothers and Neonates)

  • It is a common clinical practice to recommend taking iron supplements for pregnant women during gestation. Although it is required to ensure adequate iron stores during pregnancy, there has been some debate over the interference effects of excessive iron load, because it is possible to compete in the transport in the intestine and placenta and in binding to serum proteins of other trace minerals. In this study, maternal and neonatal serum concentrations of Fe, Zn, Cu, Se, Cr, Mn, and Co were assessed along with maternal Fe intakes. A total of 124 pregnant women and their term neonates participated voluntarily in this research. The women were divided into one of the three groups {high Fe intake (HFI), median Fe intake (MFI), and low Fe intake (LFI)} by their total Fe intakes and one of the two groups (Anemic and Normal) by their Fe nutritional status. All the data were compared among the three groups and between the two groups also. Total Fe intakes of HFI, MFI, and LFI groups were 140.8 ${\pm}$ 76.1, 68.0 ${\pm}$ 11.2, and 30.2 ${\pm}$ 8.6 mg/day, respectively. Those of Anemic and Normal groups were 90.1 ${\pm}$ 74.8 and 86.6 ${\pm}$ 46.8 mg/day, respectively. Maternal Hb concentration and Hct were not significantly different among HFI, MFI, and LFI groups but those were significantly different between Anemic and Normal groups. However, neonatal Hb concentration was not significantly different among HFI, MFI, and LFI groups and between Anemic and Normal groups either. Maternal serum Fe concentrations of the three groups, HFI, MFI, and LFI, were similar but that of Anemic group was significantly lower compared to Normal group. However, there was no significant difference in neonatal serum Fe concentrations among the three groups and between the two groups either. Serum concentrations of the other trace minerals in both mothers and neonates were not significantly different among HFI, MFI, and LFI groups and between Anemic and Normal groups. In addition, in the maternal serum, Fe concentration was positively correlated to Zn and Se concentration, respectively. As for the neonatal serum, Fe concentration showed a positive correlation to Zn, Cu, Mn, Se, and Co concentration, respectively. No trace mineral concentration was found to correlate negatively to Fe concentration in both maternal and neonatal serum, The results in this study indicate that Fe intakes of pregnant women, even if it is considerably above the level of estimated average requirement (EAR), may not affect serum Fe concentration in both mothers and neonates. In addition it might not influence adversely on the availability of other trace minerals including Zn and Cu in both mothers and neonates.

  • 본 연구에서는 연골조직으로부터 세포를 분리한 후 10%의 혈청 첨가 배지와 무혈청 배지에서의 세포 성장속도, GAGs 합성 및 Col. II의 발현을 확인하였다. EGF를 첨가한 무혈청 배지를 연골세포의 증식에 매우 효과적이었으나 GAGs 합성 및 Col. II 의 합성을 저해하였다. 또한 무혈청 배지에서 배양한 세포를 차후 신체내로 이식한다면 연골세포의 특성인 Col. II를 재합성할 수 있음을 간접적으로 확인하였다. 이는 무혈청 배지를 이용하여 평판배양 시 짧은 기간 내 연골세포 치료제로서 가능한 세포수를 얻기 위한 모델로서 유용할 것으로 생각된다. 또한 계대배양에 따른 분화 및 형태의 변화 등의 문제점들은 생분해성 지지체와 연계하여 해결할 수 있을 것으로 생각되며, 차후 위의 SFM을 이용한 3D 배양에 대한 연구를 수행할 예정이다.

Change of Insulin-like Growth Factor Gene Expression in Chinese Hamster Ovary Cells Cultured in Serum-free Media

  • Although the sera used in animal cell culture media provide the macromolecules, nutrients, hormones, and growth factors necessary to support cell growth, it could also be an obstacle to the production of recombinant proteins in animal cell culture systems used in many sectors of the biotechnology industry. For this reason, many research groups, including our laboratory, have been trying to develop serum-free media (SFM) or serum-supplemented media (SSM) for special or multi-purpose cell lines. The Chinese hamster ovary (CHO) cell, for example, is frequently used to produce proteins and is especially valuable in the large-scale production of pharmaceutically important proteins, yet information about its genome is lacking. Also, SFMs have only been evaluated by comparing growth patterns for cells grown in SFMs with those grown in SSM or by measuring the titer of the target protein obtained from cells grown in each type of medium. These are not reliable methods of obtaining the type of information needed to determine whether an SFM should be replaced with an SSM. We carried out a cDNA microarray analysis to evaluate MED-3, an SFM developed in our laboratory, as a CHO culture medium When CHO cells were cultured in MED-3 instead of an SSM, several genes associated with cell growth were down-regulated, although this change diminished over time. We found that the insulin-like growth factor (IGF) gene was representative of the proteins that were down-regulated in cells cultured in MED-3. When several key supplements - including insulin, transferrin, ethanolamine, and selenium - were removed from MED-3, the IGF expression was consistently down- regulated and cell growth decreased proportionately. Based on these results, we concluded that when an SFM is used as a culture medium, it is important to supplement it with substances that can help the cells maintain a high level of IGF expression. The data presented in this study, therefore, might provide useful information for the design and development of SFM or SSM, as well as for the design of genome-based studies of CHO cells to determine how they can be used optimally for protein production in pharmaceutical and biomedical research.

이소플라본 섭취 수준이 주폐경기와 폐경 후 여성의 혈청 내 지질 패턴과 Total Antioxidant Status에 미치는 영향 (Dose Respone Relationship of Isoflavone Supplementation on Plasma Lipid Profiles and Total Antioxidant Status in Perimenopausal and postmenopausal Women)

  • There is growing evidence that soy isoflavones play beneficial role in the prevention of cardiovascular disease. However, current data are not sufficient to determine the effective dose of isoflavone on improving plasma lipid profiles and total antioxidant capacity in Korean menopausal women. In the present study, 24 perimenopausal and 23 postmenopausal women were divided into 3 groups and were given soy isoflavone supplements of 100, 150, 200mg/day for 12weeks. Blood samples were taken before and after 6, 12weeks of soy isoflavone supplementation for analysis of total cholesterol, LDL cholesterol, HDL cholesterol, triglyceride and total antioxidant status(TAS). In the perimenopausal women, serum total cholesterol level decreased significantly in all three groups after 12weeks of isoflavone supplementation(p< 0.05). However, serum LDL cholesterol level significantly lowered only in 150 and 200mg groups, and serum triglyceride level decreased significantly in 200mg group(p<0.05). In the postmenopausal women, the 150mg isoflavone supplementation group demonstrated significant effects on lowering serum total cholesterol, LDL cholesterol levels and raising HDL cholesterol level after 12weeks of isoflavone supplementation(p<0.05). Two hundred mg isoflavone supplementation group had a significantly higher TAS than other groups in both perimenopausal and postmenopausal women. (Korean J Nutrition 34(3) : 322∼329, 2001)

Effects of Dietary Lycopene and Vitamin E on Egg Production, Antioxidant Status and Cholesterol Levels in Japanese Quail

  • Japanese Quails were used to evaluate the effects of dietary supplementation with vitamin E (dl-a-tocopheryl-acetate), lycopene, and their combination on egg production, egg quality, concentrations of malondialdehyde (MDA), vitamin E, A and cholesterol in serum and egg yolk. Quails (n = 120; 55 d old) were divided into four groups (n = 30/group) and fed a basal diet or the basal diet supplemented with lycopene (100 mg/kg diet), vitamin E (250 mg dl-${\alpha}$-tocopheryl-acetate/kg diet) or a combination of lycopene and vitamin E (100 mg/kg lycopene plus 250 mg dl-${\alpha}$-tocopheryl-acetate/kg diet). Vitamin E and lycopene did not affect (p>0.05) body weight, feed intake or egg weight. Egg production and Haugh unit were greater (p<0.05) in each supplemental group compared with the control group (p<0.05). Serum and liver MDA levels were decreased in supplemented groups compared with the control group. Separately or as a combination, supplemental lycopene and vitamin E increased serum and egg yolk vitamin E and A but decreased cholesterol concentrations (p<0.05). In general, when a significant effect was found for a parameter, the magnitude of the responses to vitamin and lycopene supplements was greatest with the combination of the lycopene and vitamin E, rather than that observed with each supplement separately. Results of the present study indicate that supplementing with a combination of dietary lycopene and vitamin E reduced serum and yolk cholesterol concentrations and improved antioxidant status.
