Leptin 유전자 결핍 동물모델을 이용한 시네트롤(Sinetrol-XPur)의 항비만 효과와 cAMP를 통한 UCP-2 활성화 기전 연구 (Effects of Sinetrol-XPur on Leptin-Deficient Obese Mice and Activation of cAMP-Dependent UCP-2)

  • 본 연구에서는 시네트롤(Sinetrol-XPur)을 유전성 비만 마우스(Obese)에 7주간 식이 투여하여 항비만 효과를 알아보고자 하였다. 시네트롤을 섭취한 유전성 비만 마우스군(Sinetrol low와 Sinetrol high)의 체중증가량은 Obese군에 비하여 유의적으로 감소하였고, 간 무게변화량은 Sinetrol high군에서 Obese군보다 유의적으로 감소하였다. 백색지방의 무게변화량은 Sinetrol high군에서 Obese군보다 유의적으로 감소하였음을 확인하였다. 실험동물의 혈청 지질 함량은 TC, TG, LDL/VLDL-콜레스테롤이 Obese군에 비해 Sinetrol군에서 감소하였고, HDL-콜레스테롤은 유전성 비만군(Obese, Sinetrol low, Sinetrol high) 간에 유의적인 차이가 나타나지 않았다. HDL/LDL ratio를 계산한 결과에서는 Sinetrol군에서 Obese군보다 유의적으로 증가하였음을 확인하였다. 실험동물군의 지방조직에서 비만 관련 유전자들의 발현을 측정한 결과, FAS와 LPL의 발현은 Obese군보다 Sinetrol군에서 감소하였고 HSL의 발현은 유의적인 차이가 나타나지 않았지만, UCP-2의 발현은 Obese군에 비해 Sinetrol high군에서 유의적으로 증가하였음을 확인하였다. 또한 세포 내 cAMP를 측정한 결과 시네트롤 처리 시 농도 의존적으로 cAMP가 증가하였음을 확인하였고, 증가한 cAMP에 의해 UCP-2의 발현이 증가되는 것을 확인하였다. 이러한 결과로부터 시네트롤이 지방 합성과 관련된 유전자 발현의 억제 및 에너지 소비와 관련된 유전자의 발현을 증가시키고 체내 지방의 축적이나 합성을 감소시켜 체중의 증가를 예방하며, 혈중 지질 함량들을 개선하여 항비만 효과가 있는 천연 기능성 식품으로 활용할 수 있을 것으로 기대할 수 있다.

고지방식이로 유도한 비만 흰쥐에서 함초 (Salicornia herbacea L.) 분말의 항비만 효과 탐색 (Anti-obesity effect of Korean Hamcho (Salicornia herbacea L.) powder on high-fat diet-induced obese rats)

  • 본 연구에서는 한국산 함초 (Salicornia herbacea L.)인 퉁퉁마디의 항비만 효과를 알아보고자 동물실험을 실시하였다. 함초 동결건조 분말 및 함초 동결건조 나노화 분말의 항비만 효과를 살펴본 결과는 다음과 같다. 1) 최종체중은 5% 함초 동결건조 분말을 섭취한 5% HS군이 HFC군에 비해 유의하게 감소하였으며, NC군보다도 유의적으로 낮게 나타났다. 식이섭취량은 NC군에 비하여 고지방식이 섭취군 (HFC, 2% HS, 5% HS, 1% HSN)에서 유의적으로 감소하였고, 5% 함초 동결건조 분말을 섭취한 5% HS군은 고지방식이군보다 유의적으로 식이섭취량이 낮았다. 2) 간 무게는 고지방식이의 영향으로 NC군에 비하여 모든 고지방식이 섭취군에서 유의하게 증가하였고, 5% 함초 동결건조 분말 섭취군은 HFC군과 비교해 유의하게 낮았다. 부고환 지방조직의 무게 역시 왼쪽/오른쪽 모두 5% 함초 동결건조 분말 섭취군에서 HFC군에 비해 유의하게 낮았으며, NC군보다도 유의적으로 수치가 낮았다. 3) 혈청 중성지질 농도는 NC군이 모든 고지방식이 섭취군에 비해 유의하게 높았으며, 혈청 총 콜레스테롤 농도는 2% HS군을 제외한 모든 고지방식이군에서 NC군에 비해 유의하게 높았다. 그러나, 혈청 중성지질, 총 콜레스테롤, LDL-콜레스테롤 및 HDL-Cholesterol 농도 모두 HFC군과 비교하여 함초 섭취에 따른 유의한 차이가 없었다. 4) 간 조직의 중성지질과 총 콜레스테롤 농도는 NC군에 비하여 모든 고지방식이 섭취군에서 유의적으로 증가하였다 (p < 0.05). HFC군은 함초 섭취군과 유의적인 차이가 나타나지 않았으나, 5% 함초 동결건조 분말 섭취군의 중성 지질과 총 콜레스테롤 농도는 HFC군에 비해 감소하는 경향을 보였다. 5) 간 세포질 중 FAS 농도의 경우 NC군이 모든 고지방식이 섭취군에 비해 유의적으로 증가하였으며, 함초분말섭취에 따른 유의한 차이는 없었다. 6) 혈청 leptin 농도는 HFC군에 비해 모든 함초 섭취군에서 leptin의 수치가 낮게 나타나는 경향이 보였다. 특히 5% HS군은 HFC군에 비해 leptin 농도가 유의적으로 감소하였다. 혈청 adiponectin 농도는 NC군이 모든 고지방식이섭취군에 비해 유의적으로 높았으며, 5% 함초 동결건조 분말 섭취군의 경우 HFC군 비해 그 농도가 증가하는 경향이 나타났으나 유의적인 차이는 없었다. 이상의 실험 결과에서 고지방식이와 함께 섭취한 5% 함초 동결건조 분말은 식이섭취량을 줄여 체중, 간 및 지방조직의 무게를 감소하였고, 혈중 leptin 농도를 유의적으로 감소시켰으며, 혈중 adiponectin 농도를 증가시키는 경향을 보이는 등 지질대사 개선에 긍정적인 영향을 나타냈다. 이러한 결과는 함초의 식이조절 및 지질개선 건강기능성소재 개발 가능성을 제시한 것으로 생각된다.

Effects of a High Protein Diet and Aerobic Exercise on Body Weight Changes and Blood Lipids in Slightly Overweight Women

  • To examine the combined effects of a high-protein diet and aerobic exercise on body weight and composition and blood lipid profiles in overweight women, 30 young women were recruited and placed into three groups: The high-protein diet and exercise group (HPE), the exercise-only group (EXO) and the control group (CON) (30$\pm$3%, 27$\pm$2%, and 29$\pm$3% body fat, respectively) for an 8-week experimental period. Daily diet included 25% isolated soybean protein (>90% protein, approximately 400 kcal) combined with each subject s usual diet for the HPE group. The exercise program consisted of aerobic-type exercises undertaken >3 times/wk and for>30 min/session at 50-60% of maximal capacity. Physical fitness, body composition, serum total cholesterol (TC), high-density lipoprotein-cholesterol (HDL-C), triglycerides (TG) and glucose were measured before and after the experiment. Maximal aerobic capacity increased by the end of experiment in both the HPE (from 27.2$\pm$3.5 to 35.l$\pm$5.9 ml/kg/min, p<0.01) and EXO (from 30.3$\pm$5.4 to 33.8$\pm$3.8 mㅣ/kg/min, p<0.05) groups. Percent body fat decreased by 3.3% (p<0.01) in the HPE group and by 1.5% (p<0.05) in the EXO group by the end of the experiment, but not in the CON group. Lower back strength and agility increased only in the HPE group. In the HPE group, TC decreased from 168$\pm$20 to 155$\pm$18 mg/dL and HDL-C increased from 57$\pm$l0 to 61$\pm$9 mg/dL in HPE (p<0.01). But TC and HDL-C did not change in the EXO and CON groups. TG and glucose did not vary among the groups. Although the EXO group showed a similar outcome to that of the HPE group, a favorable change in body composition and blood lipids as well as an improvement in aerobic capacity was more marginal in the latter group.

학령기 아동의 심혈관계 질환 위험인자의 유병률 (Prevalence of Cardiovascular Risk Factors in School-aged Children)

  • Objectives : This study was conducted to assess the distribution of cardiovascular risk factors (serum lipid profiles, BMI, blood pressure, fasting blood sugar) and evaluate the risk profile of CVD by the clustering of the cardiovascular risk factors in school-aged children in the Kyoung-Gi area. Methods : The study sample consists of 208 11 year-old children (51.4% boys, 48.6% girls) who participated in a cross-sectional screening of cardiovascular risk factors. We surveyed their socio-demographic characteristics, measured the anthropometric variables and analyzed the biochemical markers. Results : Of the cardiovascular risk factors, the percentage risk of the BMI, dyslipidememia and hypertension were highest. The prevalence rates of total cholesterol and LDL-cholesterol in girls was higher than in boys. Also, the associations of the BMI, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglycerides, systolic and diastolic blood pressures were highly significant. In addition, the proportion of subjects with 1, 2, 3 and 4 or more risk factors were 29.3, 12.5, 2.9 and 1.9%, respectively; therefore, a total of 97 subjects (46.6%) had at least one more risk factor. Conclusions : From these findings, we concluded that the rates of hypercholesterolemia, cardiovascular risk groups and obesity prevalence of these groups were relatively high. These data provide further evidence that the early intervention for cardiovascular health prevention and promotion in school-aged children is necessary at the population level.

The Association between the T102C Polymorphism of the HTR2A Serotonin Receptor Gene and HDL Cholesterol Level in Koreans

  • 5-HT2A is one of major serotonin receptor that is involved in the action of serotonin-targeting drugs. Previous clinical studies have shown an unexpected association between lower cholesterol level and psychiatric diseases, in which T102C polymorphism of HTR2A, gene of 5-HT2A serotonin receptor, might be involved. Therefore, we hypothesized a potential association between lower cholesterol level and T102C polymorphism. The effect of the T102C polymorphism on the serum lipid profiles of 646 subjects without specific psychiatric disease was investigated. Genotype was determined by polymerase chain reaction and restriction fragment length polymorphism analysis. There were significantly lower levels of total cholesterol ($193.6{\pm}35.0$ versus $202.1{\pm}45.5\;mg/dl$, p = 0.016) and HDL-cholesterol ($42.7{\pm}11.6$ versus $46.3{\pm}12.7\;mg/dl$, p = 0.004) in CC genotype than non-CC genotypes. Moreover, multivariate analysis showed that the CC genotype is a strong predictor of a lower HDL-cholesterol level (p < 0.001). In conclusion, this study shows that the CC genotype of the HTR2A gene is related to lower HDL-cholesterol level in Koreans. This is the first demonstration showing the potential genetic relationship between the serotonin receptor gene polymorphism and the HDL-cholesterol level.

Efficacy and safety of Panax ginseng berry extract on glycemic control: A 12-wk randomized, double-blind, and placebo-controlled clinical trial

  • Background: Antihyperglycemic effects of Panax ginseng berry have never been explored in humans. The aims of this study were to assess the efficacy and safety of a 12-wk treatment with ginseng berry extract in participants with a fasting glucose level between 100 mg/dL and 140 mg/dL. Methods: This study was a 12-wk, randomized, double-blind, placebo-controlled clinical trial. A total of 72 participants were randomly allocated to two groups of either ginseng berry extract or placebo, and 63 participants completed the study. The parameters related to glucose metabolism were assessed. Results: Although the present study failed to show significant antihyperglycemic effects of ginseng berry extract on the parameters related to blood glucose and lipid metabolism in the total study population, it demonstrated that ginseng berry extract could significantly decrease serum concentration of fasting glucose by 3.7% (p = 0.035), postprandial glucose at 60 min during 75 g oral glucose tolerance test by 10.7% (p = 0.006), and the area under the curve for glucose by 7.7% (p = 0.024) in those with fasting glucose level of 110 mg/dL or higher, while the placebo group did not exhibit a statistically significant decrease. Safety profiles were not different between the two groups. Conclusion: The present study suggests that ginseng berry extract has the potential to improve glucose metabolism in human, especially in those with fasting glucose level of 110 mg/dL or higher. For a more meaningful benefit, further research in people with higher blood glucose levels is required.

Dietary and modifiable factors contributing to hyper-LDL-cholesterolemia prevalence in nationwide time series data and the implications for primary prevention strategies

  • BACKGROUND/OBJECTIVES: A number of studies examined secular trends in blood lipid profiles using time series data of national surveys whereas few studies investigated individual-level factors contributing to such trends. The present study aimed to examine secular trends in dietary and modifiable factors and hyper-LDL-cholesterolemia (HC) prevalence and evaluate their associations using time series data of nationwide surveys. SUBJECTS/METHODS: The study included 41,073 Korean adults aged ≥ 30 years from the 2005, 2007-2009, 2010-2012, 2013-2015, and 2016 Korea National Health and Nutrition Examination Surveys. Stepwise logistic regression analysis was performed to select significant factors associated with HC, which was defined as serum LDL cholesterol levels ≥130 mg/dL. RESULTS: The following factors showed a positive association with HC (P < 0.05): for men having higher body mass index (BMI), being married, having an office job, and consuming higher dairy and vegetable oil products; for women having higher age or BMI, having no job or a non-office job, not in a low-income household, and consuming higher dairy products. In the given model, the 2016 survey data showed that a 2 kg/㎡ reduction in BMI of obese persons resulted in a decreased HC prevalence from 30.8% to 29.3% among men and from 33.6% to 32.5% among women. CONCLUSIONS: Based on these findings, it is suggested that primary prevention programs should advocate having proper BMI for Korean adults with a high-risk of HC. However, whether discouraging consumption of dairy and vegetable oil products can reduce HC prevalence warrants further studies with a prospective longitudinal design.

Effect of Lactic Acid Bacteria-Fermented Mulberry Leaf Extract on the Improvement of Intestinal Function in Rats

  • This study examined the laxative effects of mulberry leaf extract (MLE) fermented by lactic acid bacteria (LAB), which contains high levels of polyphenolic and flavonoid compounds, against loperamide-induced constipation in rats. Sprague-Dawley rats were divided into a normal group (N) and three experimental groups; loperamide treated group (C), loperamide and LAB-fermented MLE 300 mg/kg treated group (MLEL), and loperamide and LAB-fermented MLE 600 mg/kg treated group (MLEH). After 33 d, fecal pellet amount, fecal weight, water content of fecal, gastrointestinal transit time and length, and serum lipid profiles were measured. Constipation was induced via subcutaneous injection of loperamide (2.0 mg/kg b. w., twice a day) for the final 5 d of the experiment. After loperamide administration, the LAB-fermented MLE groups showed a significantly increase in the fecal pellets number, wet weight, and water content in rats compared with the C group. Moreover, increases in the intestinal length and viable Lactobacillus numbers in the feces were observed in the LAB-fermented MLE groups. The intestinal transit time was shorter in the LAB-fermented MLE groups than in the C group. In addition, the LAB-fermented MLE groups showed a significant decrease in triglyceride and total cholesterol levels and an increase in HDL-cholesterol level. These results indicated that oral administration of LAB-fermented MLE shows laxative effect in loperamide-induced constipated rats.

태음인 처방을 이용한 비만 연구에 대한 문헌연구 - 국내 문헌을 중심으로 - (Literature Review of Studies of Obesity Using Tae-eumin Prescriptions and Herbs - Based on Korean Literature -)

  • Objectives: This study analyzed literature published in Korea using Tae-eumin prescriptions and herbs for obesity for future research. Methods: We conducted a computerized search at journal web sites and manual searches. A total of 25 sites were included. Obesity, obese, over weight, herb, herbal, taeum, taeyuem, and taeeum were selected keywords and they were combined properly. Results:A total of 28 studies were analyzed. The percentage of clinical and case report studies was 35.7% and experimental studies 64.3%. The most frequently used prescription was Taeeumjowui-tang (太陰調胃湯) and herbs were Raphani semen(蘿葍子) and Platycodon grandiflorum(桔梗). Weight (kg), body mass index (BMI) and side effects and symptoms were used as indices in clinical and case report studies. Weight, weight gain percentage and blood serum lipid profiles were frequently used as indices in experimental studies. Conclusion: Further well-designed randomized controlled studies and clinical reports of side effects and symptoms are needed for development of obesity studies for Tae-eumin prescriptions.

송기 분말이 Loperamide로 유도된 흰쥐의 변비 완화에 미치는 효과 (Effect of Pine Inner Bark (Song-gi) Powder on Relief from Constipation in Loperamide-induced Rats)

  • This study investigated the effects of insoluble dietary fibers from pine (Pinus radiata D. Don) inner bark powder (PIBP) on loperamide constipation in rats. Male Sprague-Dawley rats were administered PIBP in their diets at concentrations of 5% and 10% for 4 weeks. Rats were divided in to four groups, normal diet group (Cont), a normal diet and loperamide group (Lop), 5% PIBP experimental diet and loperamide group (Lop+PIBP 5%), and 10% PIBP experimental diet and loperamide group (Lop+PIBP 10%). Constipation was induced by oral administration of loperamide (2 mg/kg, twice per day) for the last three days of the experiment. Food intake, body weight, properties of feces, gastrointestinal transit time, and serum lipid profiles were measured. When compared with the normal diet and loperamide group (Lop), there were increases in fecal pellet number (p<0.05), wet weight (p<0.001), and water content (p<0.001). Positive results were derived from relevant indicators to improve constipation. In addition, the number of fecal pellets in the colon was not significant, and decreased as PIBP content increased. PIBP had a concentration-dependent effect on reduction of total cholesterol, LDL-cholesterol, and triglycerides and elevation of HDL-cholesterol. These results indicate that PIBP may effectively prevent constipation.
