흡입물질이 흰쥐 Lactate Dehydrogenase와 Cholinesterase 활성변화에 미치는 영향 (Effects of Volatile Substances on Rat Lactate Dehydrogenase and Cholinesterase)

  • The effects of volatile substances inhalation on lactate dehydrogenase and cholinesterase in rats were investigated. Male Sprague-Dawley rats were exposed to marketed odorant, ethyl acetate and ethyl ether for 15 days. Enzyme activities were measured in serum and several tissues such as liver, lung, brain, heart, kidney and muscle to find differences of effects according to the organ. Cholinesterase activity in serum and most of tissues revealed time-dependent decrease in the case of marketed odorant inhalation. Especially in heart and kidney significant decrease was observed. Ethyl acetate exposure to rats revealed also decrease in serum and all tissues by 40% to 60%. Ethyl ether inhalation showed significant decrease by 30% to 50%. Lactate dehydrogenase activity was markedly increased in serum and similarly in heart, brain and kidney by exposure to marketed odorant. No changes were observed in liver. Ethyl acetate exposure to rats revealed increase in serum by about 200%, compared to normal group and in other tissues by 40% to 70% except in liver and muscle. Ethyl ether inhalation showed significant increase in serum by about 100%. There was no change in 'liver and slight increase in muscle.

  • OBJECTIVES : Schizandra chinensis is well known for its efficacy at liver function reinforcement, relieving thirst and recovery from fatigue. In this study, we examined the effects of sports drink containing Schizandra chinensis on serum metabolic substrate, electrolyte, stress indicators, related-enzyme and exercise performance, rectal temperature, and heat shock proteinb70 (HSP70). METHODS : Elite long-distance runners (male, 21.3yrs, n=16) were selected and divided into two groups; an experimental group (EXP, n=8) and a control group (CON, n=8). A beverage containing Schizandra chinensis was supplemented 3 times per day to EXP for 4 weeks. Serum biochemical elements (glucose, lactate, total cholesterol, triglyceride, high density lipoprotein cholesterol, glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, creatinine, creatine phosphokinase, lactate dehydrogenase, blood urea nitrogen, Na, K, Cl) were analyzed by auto blood analyzer. Exercise performance was measured by treadmill exercise test, HSP70 was detected by electrophoresis and Western blotting, and rectal temperature was measured by rectal temperature probe. RESULTS : Administration of the beverage increased significantly the rest level of blood Na, Cl and glucose and decreased significantly lactate dehydrogenase, glutamate oxaloacetate transaminase. No difference was found in exercise performance, rectal temperature increment or HSP70 concentration between groups. CONCLUSIONS : Administration of a sports drink containing Schizandra chinensis altered blood glucose, lactate dehydrogenase, glutamate oxaloacetate transaminase, Na and Cl levels.

소회향이 허혈성 뇌혈관 질환에 미치는 실험적 연구 (The Experimental Study of FOENICULI FRUCTUS on the Ischemic Cerebrovascular Disease)

  • Objective : This experimental Study was designed to investigate the effects of FOENICULI FRUCTUS(FF) on the change of inhibition lactate dehydrogenase(LDH) activity in neuronal cells and cytokines production in serum of cerebral ischemic rats. Method : FOENICULI FRUCTUS(FF)freeze dry powder and FF on the LDH activity in neuronal cells. Changes of FF on the physiological parameters(PaO2, PaCO2, MABP and HR) in crerbral ischemic rats. Effects of FF on the IL-1beta production, $TNF-{\alpha}$ production, $TGF-{\beta}$ production, and IL-10 in serum of cerebral ischemic rats. MCAO :. cytokines production of serum by drawing from femoral arterial blood after MCAO 1 hr. Reperfusion : cytokines production of serum by drawing from femoral arterial blood after reperfusion 1 hr. Results and Conclusion : 1. FF did not inhibit lactate dehydrogenase(LDH) activity in neuronal cells. 2. In serum by drawing from femoral arterial blood after middle cerebral arterial occlusion(MCAO) 1 hr and reperfusion 1 hr, sample group was significantly decreased $IL-l{\beta}$ production compared with control group 3. In serum by drawing from femoral arterial blood after MCAO 1 hr and reperfusion 1 hr, sample group was significantly decreased $TNF-{\alpha}$ production compared with control group. 4. In serum by drawing from femoral arterial blood after MCAO 1 hr and reperfusion 1 hr, sample group was significantly increased $TGF-{\beta}$ production compared with control group. 5. In serum by drawing from femoral arterial blood after reperfusion 1 hr, sample group was significantly increased IL-10 production compared with control group. This results were suggested that FF had inhibitive effect on the brain damage by inhibited LDH activity, $IL-l{\beta}$ and $TNF-{\alpha}$production, but accelerated $TGF-{\beta}$ production and IL-10 production.

랫드에서 수은이 혈장 DNA와 혈액화학치에 미치는 영향 (Effects of HgCl2 on plasma DNA content and blood biochemical values in rats)

  • Changes of plasma DNA contents and serum biochemical values were measured in rats administered with $HgCl_2$ to investigate the in vivo cytotoxic effects of mercury and examine the usefulness of these changes as indicators of mercury exposure and diagnosis of mercury poisoning. Rats were given once intraperitonealy $HgCl_2$(0.13. 0.32. 0.8 and 2 mg/kg b.w) and the changes of plasma DNA contents and serum biochemical values were measured at the time of 2, 4, 8, 24, 48 and 72 hours after the administration of $HgCl_2$. Plasma DNA contents began to increase from 2 hours after the administration of $HgCl_2$ in all the treatment groups significantly compared to control with dose-dependent pattern. The levels of plasma DNA reached to peak at 48 hours as 2.77, 7.60, 15.46 and 16.51 times higher than control in each treatment group of 0.13, 0.32, 0.8 and 2 mg/kgb.w, respectively and remained to be higher until 72 hours after the administration. The values of creatine kinase, aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, blood urea nitrogen and glucose of serum were increased, however the values of alkaline phosphatase, total protein and triglyceride were decreased. These changes of increase and decrease showed dose-dependent pattern but the starting time, maintenance and magnitude of change were various and characteristic according to serum biochemical indices. Among the changes of serum biochemical values, those of aspartate aminotransferase, lactate dehydrogenase and blood urea nitrogen were apparently and significantly increased compared to control from 2 to 72 hours by the administration of 2 mg/kg $HgCl_2$. This study demonstrates that plasma DNA and serum biochemical values such as aspartate aminotransferase, lactate dehydrogenase, blood urea nitrogen and etc. are valuable as biomarkers for mercury exposure assessment and diagnosis of mercury poisoning.

Sodium Fluoride 함량이 흰쥐의 간 기능 효소활성에 미치는 영향 (Influences of Sodium Fluoride Contents on Hepatic Functional Enzyme Activities in Rats)

  • The purpose of this study was to probe the influences of NaF oral administration on a dose-effect relationship between fluoride levels of serum enzyme activity such as alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) in rats fed experimental diets for 5 weeks. All groups increased the activity of serum ALP, AST, ALT, and LDH levels with increasing NaF. In addition the fluoride levels of serum and organ tissues (liver, brain, heart, lung, kidney) in oral NaF groups (NF3~NF50) were significantly increased by adding sodium fluoride in comparison with normal diet group (ND) (p<0.05). These results, a high concentration of sodium fluoride was determined that the toxicity to various organ tissues.

  • The effects of dichloromethane (DCM) on carbon tetrachloride (CT) toxicity were examined in adult male rats. A concomitant treatment of rats with DCM (0.3, 0.6, 1.2 g/kg, po) significantly potentiated the hepatotoxicity of CT (1.0 g/kg, po) as determined by increase in serum GPT (glutamic pyruvic transaminase), GOT (glutamic oxaloacetic transaminase), and SDH (sorbitol dehydrogenase) activity 24 hr following the treatments. Serum LDH (lactate dehydrogenase) activity was increased by either DCM or CT treatment.

Effects of the Zizyphus jujuba Seed Extract on the Lipid Components in Hyperlipidemic Rats

  • The purpose of this study was designed to observe the effects of Zizyphus jujuba seed extract on the concentrations of the lipids and blood glucose in the S.D. rats fed the experimental diets for 4 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride (TG), phospholipid (PL) and blood glucose in serum were significantly higher in the cholesterol administration groups (group 2 (cholesterol+water), group 3 (cholesterol+Zizyphus jujuba seed extract)) than those in the control group (group 1, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum ware remarkably lower in the group 3 than those in the group 2. In the ratio of HIDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Zizyphus jujuba seed extract administration group was higher percentage than in the group 2. The activities of aspartate aminotransferase (AST), alanine amino-transferase (ALT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Zizyphus jujuba seed extract administration group (group 3) than in the cholesterol diet group (group 2). From the above research, Zizyphus jujuba seeds were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic rats. And particularly, Zizyphus jujuba seeds were more effective as a therapeutic regimen for the control of metabolic derangements in adult disease.

Analysis of Lactate Dehydrogenase Levels of COVID-19 Patients in a Korean Hospital According to Sex and Age

  • This study aimed to determine whether lactate dehydrogenase (LDH) levels in coronavirus disease (COVID-19) patients in Korea were higher than those in patients without COVID-19, and the effect of sex and age on LDH levels. A retrospective, observational study was conducted to measure LDH levels in 247 and 225 female and male COVID-19 patients, respectively, who were admitted to the study hospital between April 1 and October 30, 2020. Serum LDH levels were measured using an automated analyzer. Results: LDH levels were elevated in both male and female patients with COVID-19. Among patients with COVID-19, LDH levels were higher in males than in females, and LDH levels were higher in patients with COVID-19 than in patients in the control group. In the analysis of differences in LDH levels by age, LDH levels in patients with COVID-19 increased statistically significantly with age in males and females (males: p=0.001, females: p=0.001). By examining the differences in LDH levels according to sex and age, this study contributed to the basic biochemical data available in Korea, particularly regarding patients with COVID-19. Further research may be needed to examine confounding variables.

Anti-Fatigue Properties of Cultivated Wild Ginseng Distilled Extract and Its Active Component Panaxydol in Rats

  • Objectives: Cultivated wild ginseng (cWG), called SanYangSanSam, has been used clinically in patients with chronic fatigue in Korea. Little is known about effects of the ginseng distilled (volatile) components produced during evaporizaiton. Recently, we first identified one major component from cWG distilled extract, panaxydol, by using mass spectrometry. However, functional properties of cWG distilled extract and panaxydol remains elusive. Therefore, the present study evaluated the effect of cWG distilled extract or panaxydol on exercise-induced fatigue in rats. Methods: Fatigue was induced by forced swimming and the immobility time was analyzed in male Sprague-Dawley rats. The animals received intraperitoneally either vehicle, cWG distilled extract, or panaxydol 10 min prior to beginning of the forced swimming test (FST) once daily for 5 days. After the FST on day 5, we also analyzed fatigue-related biochemical levels including blood urea nitrogen (BUN), lactate acid (LAC), and lactate dehydrogenase (LDH) in serum and levels of glycogen in liver and soleus muscle. Results: The forced swimming time in cWG distilled extract (0.6 mL/kg)-treated group was significantly longer than that of control group on day 4 and 5. Panaxydol (0.1 and 0.25 mg/kg)-treated groups showed significantly enhanced performance in the forced swimming, compared to control. In addition, a significant decrease in serum LDH level was found in panaxydol-treated group, while there were no alternations in levels of serum BUN and LAC and glycogen in liver or soleus muscle. Conclusion: The present study demonstrated cWG distilled extract and its active component panaxydol have a function of anti-fatigue.
