Association of the ubiquitin specific peptidase 9X -linked and Afadin expression patterns with sexual maturation in boar testis

  • Closely correlated expression patterns between ubiquitin specific peptidase 9X-linked (USP9X) and adherens junction formation factor (Afadin) in mouse testis development suggests that Usp9x regulates the deubiquitination of Af-6 (also known as Afadin, AFDN), and subsequently, the cell adhesion dynamics during gametogenesis. However, this relationship has not yet been tested in other domestic animals. The study was examined the temporal and spatial expression patterns of porcine USP9X and AFDN from the pre-pubertal to adult stages using real time-PCR and immunohistochemistry. Furthermore, we detected the transcripts of USP9X and AFDN in the testis of 1-, 6- and 12-months old boar, respectively. USP9X and AFDN were found to have similar expressions patterns, with basal expression after 1 month followed by a significant up-regulation from 6 months (puberty) onwards. In addition, neither the AFDN or USP9X proteins were detected in spermatogenic cells but they were expressed in the leydig cells and sertoli cells. USP9X was detected around the basal lamina during pre-puberty, and predominantly expressed in the leydig cells at puberty. Finally, in adult testis, USP9X was increased at the sertoli cell-cell interface and the sertoli cell-spermatid interface. In summary, closely correlated expression patterns between USP9X and AFDN in boar testis supports the previous findings in mice. Furthermore, the junction connections between the sertoli cells may be regulated by the ubiquitination process mediated via USP9X.

  • The testis is composed of four cell types like supporting cells, steroid-producing cells, connective tissue cells and germ cells. Apoptosis is a common phenomenon during spormatogenesis. Apoptosis of germ cells can also be induced by exposure to radiation. Previous studies have shown that most types of germ cells are rather radiosensitive while somatic cells in testis are much more radio-resistant. The somatic cells in testis are divided to mainly Sertoli and Leydig cells. Though somatic cells are more radio-resistant than germ cells, radiation can induce the impairment of their function. This damaged function of somatic cells may accelerates degeneration of germ cell indirectly. Tn the present study, we have examined the apoptotic effect of mouse testis and irradiation effect of steroidogenesis of Leydig cells after irradiation.

카드뮴이 고환에 미치는 영향에 관한 형태학적 및 생화학적 연구 (Morphological and Biochemical Studies on the Effects of Cadmium on the Rat Testis)

  • To investigate the cadmium (Cd) toxicity on the testis, male rats were treated with 1, 2, 4 and 8 mg/kg of Cd by IP. According to histochemical studies, Cd-treated testis tissue showed death of spermatozoa, death of Sertoli cells, death of all the spermatogenic cells, and finally disappearance of basal lamina of seminiferous tubules with increasing doses, and showed decreased ground substances and Leydig cells, increased inflammatory cells and fibroblasts, and fibroblasts, and finally disappearance of ground substances and all the cells except fibroblasts within interstitial tissues with increasing doses. According to biochemical studies, two kinds of proteins, 25 and 45 kDa, were dramatically disappeared from the total protein of rat testis treated with Cd comparing to normal testis. The result of electrophoresis of total protein suggests that actin (45 kDa), presumed on its mmolecular weight and amount, in the testis-cells is the primary target of Cd poisoning. Although its exact mechanism is not clear, the disappearance of two proteins when testis is exposed to Cd should give some clues to understnad the mechanism of necrosis of testis tissue crumbling by heavy metal pollutant such as Cd.

6-Aminonicotinamide가 햄스터의 정소에 미치는 영향 (Effects of 6-aminonicotinamide on the Testes of Golden Hamster)

  • 6-aminonicotinamide (6-AN)의 햄스터 정소에 미치는 영향을 확인하기 위해 실험군에는 체중 kg 당 10 mg의 6-AN을, 대조군에는 동량의 생리식염수를 격일로 복강 투여한 후 정소의 변화를 광학 및 투과전자현미경을 이용하여 관찰하였다. 최초 6-AN 투여 당시의 체중에 비해 7회 투여군부터 유의하게 체중이 감소하였으며, 정소 중량의 감소는 5회 투여시 크게 감소하여 이후 비슷한 양상을 보였다. 정세관상피의 퇴행변화는 5회 투여군부터 나타나기 시작하여 9회 투여군부터는 대부분의 정세관이 심하게 손상을 받은 것으로 나타났다. 손상을 받은 정세관에서 정세관상피를 이루는 정자발생세포 및 지지세포 모두 심한 공포화에 따른 세포의 파괴를 뚜렷하게 관찰할 수 있었으며, 다핵거대세포가 출현하였다. 사이질조직의 부종은 관찰할 수 없었으며, 사이질세포 역시 비교적 온전하게 보존되어 있었다. 따라서 6-AN은 햄스터 정소에서 정자발생세포, 지지세포 등에는 영향을 미치나 사이질세포에는 영향을 주지 않는 것으로 사료된다.

  • Teter와 Boczkowski (1966)는 testicular feminization syndrome을 두 그룹으로 분류했는데, 그것들 중에서 하나는 陰毛가 없고 정상적인 陰核을 갖고 있는 경우 (A group)이며, 다른 한 그룹은 陰毛도 있고 陰核은 팽대된 경우이다 (B group). 그러나 본 연구의 대상자는 위의 두 그룹의 중간형을 나타낸다 (C group). 각 그룹의 차이점을 알기 위하여 우리들은 鼠蹊部에서 제거한 精巢의 미세구조를 관찰하였으며, 精巢培養과 血球培養을 시도하여 染色體를 분석하였다. C 그룹의 細精管은 Leydig세포나 Sertoli세포의 분포나 성숙도 그리고 基底膜의 두께 등에서 A나 B그룹과 상이한 점을 나타낸다. 陰毛나 陰核의 형태상의 차이는 위와 같은 차이점들로 인해 야기된 것이라고 생각된다.

미성숙 돼지 정소에서 pregnancy-associated plasma protein-A의 발현의 세포학적 분석 (Cytological analysis of pregnancy-associated plasma protein-A expression in porcine neonatal testis)

  • 생체 조직 내에서 표지인자의 발견은 해당 세포의 특성과 기능을 이해하는 데 매우 중요하다. 이전의 연구에서 본 연구진은 돼지정원줄기세포에서 특이적으로 IGFBP 3가 발현되어 표지인자로의 가능성을 보고한 바 있다. 본 연구에서는 IGFBP 3이외의 다른 family member가 정원줄기세포에서 특이적으로 발현하는 지와, 이의 발현을 조절하는 PAPP-A의 조직학적인 측면에서의 발현 양상을 5일령 돼지 정소에서 확인하였다. 그 결과 IGFBP 1, 2, 3, 4, 6의 발현은 정소 전체에서 발현되는 수준보다 돼지 정원줄기세포에서 더 높은 수준에서 발현하고 있음을 확인하였다. PAPP-A는 sertoli cell에서 특이적으로 발현하며, 정원줄기세포에서는 발현하지 않는 것을 PGP9.5와의 동시-조직염색으로 확인하였다. 이러한 결과는 Sertoli cell에서 발현하는 PAPP-A 단백질은 미성숙 돼지 정소에서 IGFBP family를 통해 정소 세포의 발달과 분화를 조절할 것으로 판단된다.

Differential Expressions of Aquaporin Subtypes in the Adult Mouse Testis

  • Many efforts have been made to study the expression of aquaporins (AQP) in the mammalian reproductive system, but there are not enough data available regarding their localized expression to fully understand their specific roles in male reproduction. The present study investigated the expression and localization patterns of different AQP subtypes in the adult mouse testes and testicular spermatozoa using an immunofluorescence assay. All the studied AQPs were expressed in the testes and revealed subtype-specific patterns in the intensity and localization depending on the cell types of the testes. AQP7 was the most abundant and intensive AQP subtype in the seminiferous tubules, expressing in Leydig cells and Sertoli cells as well as all stages of germ cells, especially the spermatids and testicular spermatozoa. The expression pattern of AQP3 was similar to that of AQP7, but with higher expression in the basal and lower adluminal compartments rather than the upper adluminalcompartment. AQP8 expression was limited to the spermatogonia and Leydig cells whereas AQP9 expression was exclusive to tails of the testicular spermatozoa and elongated spermatids. Taken together, the abundance and distribution of the AQPs across the different cell types in the testes indicating to their relavance in spermatogenesis, as well as in sperm maturation, transition, and function.

Sleep deprivation induces structural changes in the adult rat testis: The protective effects of olive oil

  • Objective: Sleep deprivation (SD) is a common problem in today's stressful lifestyle and have physiological consequences, including reproductive dysfunction and infertility. As an antioxidant, olive oil may be effective in reducing testicular and spermatological damage by decreasing the production of free radicals. Methods: This study investigated the effects of olive oil on sperm quality and testicular structure using stereological methods to assess rats with SD. Results: When comparing SD group to grid floor+distilled water (GR) group, we found that the sperm count and motility, as well as the percentage of slow progressive sperm was significantly lower in SD group (p<0.05), but the percentage of immotile sperm was higher (p<0.01). However, no improvement was observed in sperm count or motility after concomitant treatment of SD group with olive oil. Stereological examinations revealed no significant change in the total volumes of the seminiferous tubules, interstitial tissue, and germinal epithelium in the study groups. Conversely, the total number of testicular cell types was significantly lower in SD group than in GR group. Although the total number of Sertoli and Leydig cells was significantly higher in the S +olive oil group than in the untreated SD group, no significant difference in the total number of other testicular cell types was observed between the two groups. Conclusion: SD potentially induced structural changes in testis that affected sperm count and motility. However, olive oil only improved the total number of Sertoli and Leydig cells in the animals with SD and did not improve sperm count and motility.

흰쥐 생식소에서 GnRH-like mRNA의 발현과 세포내 분포 (Expression and Cellular Localization of Gonadotropin-Releasing Hormone (GnRH)-like Messenger Ribonucleic Acid in the Rat Gonad)

  • 시상하부에서 합성, 분비되는 gonadotropin releasing horrnone (GnRH)의 면역반응성이 생식소를 비롯한 여러 부위에서도 검출됨이 알려졌으나, 이 펩타이트가 과연 생식소에서 국부적으로 합성되는 지에 관해서는 아직 밝혀지지 않았다. 본 연구에서는 흰쥐 생식소에서 GnRH유전자발현을 연구하기 위하여 GnRH-like mRNA와 GnRH펩타이트의 발현과 세포내 분포 양상을 조사하였다. GnRH 방사면역측정법과 GnRH를 크로마토그라피 방법으로 분리한 결과,시상하부에서 합성되는 GnRH와 유사한 GnRH 면역반은이 흰쥐 생식소 추출물에서 상당량 검출되었다. GnRH-면역반응이 흰쥐 난소의 다양한 세포군에서 나타냄에 반하여, GnRH-like mRNA는 granulsa,theca 그리고 luteal 세포에서만 주로 발현되었다. 또한 흰쥐 정소에서 GnRH면역반응성은 원시정세포, Sertoli,Leydig 세포에서만 검출된 반면에, GnRH-like mRNA는 정세관내의 Seertoli세포에서만 발현되었다. 따라서 이 연구는 생식소에 존재하는 GnRH는 생식소 내에서 국부적으로 합성, 발현되는 결과라고 사료되며, 생식소 내에서 생성된 GnRH는 생식소내 세포군간의 정보교환의 매개자로서 역활을 수행하고 있다고 추정된다.
