Genetic Diversity and Population Structure of Spiraea prunifolia for. simpliciflora by Inter-Simple Sequence Repeats (조팝나무의 유전적 다양성과 집단구조 분석을 위한 ISSR 분석)

  • 85 individual Spiraea prunifolia for. simpliciflora (Rosaceae) were sampled to examine the genetic diversity and population structure of S. prunifolia for. simpliciflora populations. Inter-simple sequence repeats (ISSR) produced 65 polymorphic loci and identified 78 ISSR genotypes. Three multilocus genotypes were shared by more than one plant within a population. Total genetic diversity values ($H_T$) and inter-locus variation in the within-population genetic diversity ($H_S$) were 0.293 and 0.183, respectively. On a per-locus basis, the proportion of total genetic variation due to differences among populations ($G_{ST}$) was 0.373. This indicated that about 37.3% of the total variation was among populations. ISSR markers are very effective in classifying natural population levels of S. prunifolia for. simpliciflora in Korea. In addition, insights into the relative gene diversity among and within populations of S. prunifolia for. simpliciflora would be useful in plant breeding and also for the development of strategies for ex situ conservation of plant genetic resources.

Sequence analysis of ORF4 gene of porcine reproductive and respiratory syndrome virus (PRRSV) Korean isolate CNV-1

  • In this study PRRSV was isolated from serum of an infected pig and designated as CNV-1, ORF4 gene was sequenced, and the nucleotide sequence, deduced amino acid sequence and the amino acid sequence of the neutralizing domain was compared with other PRRSV Strains. ORF4 gene of the Korean isolate PRRSV CNV-1 was shown to be 537bp in length, which is the same as US strain ISU55 but 21bp longer than another US strain MN1b, and 15bp shorter than European strain LV. The homologies of the nucleotide sequences between the Korean isolate CNV-1 and the US strains ISU55, MN1b and European strain LV were 91.8%, 88.1%, 67.6%, respectively, and the homologies of the deduced amino acid sequences were 94.4%, 84.4%, 68.5%, respectively. The neutralizing domain of the CNV-1 was shown to be 36 amino acids in length which is the same as ISU55, MN1b, but 4 amino acids shorter than that of the neutralizing domain reported in LV. The homologies of the amino acid sequences of the neutralizing domain between the Korean isolate CNV-1 and the US strains ISU55, MN1b and European strain LV were 92.5%, 85%, 57.5%, respectively. The molecular characteristics of ORF4 gene of the Korean isolate PRRSV CNV-1 shown in this study suggests that the CNV-1 is genetically closer to the US strains. Also the wide variation of the neutralizing domain between the CNV-1 and LV suggests that there is substantial immunogenic variation between the two strains.

Genetic Variation of Korean Masu Salmon (Oncorhynchus masou) Populations Inferred from Mitochondrial DNA Sequence Analysis

  • We analyzed the nucleotide sequences of about 500 bp of the mitochondrial NADH dehydrogenase subunit 3 (ND3) gene to estimate the genetic variation of Korean masu salmon (Oncorhynchus masou) populations. DNA samples were collected from 104 river-only specimens and 52 anadromous specimens from three hatcheries and one river. There are no records of artificial release into the river. We amplified the ND3 gene by polymerase chain reaction, targeting areas that included parts of the cytochrome oxidase III gene and the NADH dehydrogenase subunit 4L gene, and defined 14 haplotypes based on 12 variable nucleotide sites in the examined region. Among the haplotypes, ten were specific to river-only specimens within hatchery populations. Haplotype diversity of river-only populations in hatcheries was higher than that of anadromous and wild populations. Pairwise population $F_{ST}$ estimates and neighbor-joining tree analyses inferred that anadromous and river-only populations were distinct. These results suggest that sequence polymorphism in the ND3 region may be a useful marker for analyzing the genetic variation and population structure of masu salmon.

Geographical Variation and Genetic Diversity of Glhenia littoralis Fr. Schmidt et Miquel based on the Analysis of Internal Transcribed Spacer(ITS) sequence and Random Amplified Polymorphic DNA(RAPD) (멸종위기 희귀식물인 갯방풍 자생지별 유전변이 및 유전적 다양성 연구)

  • Glehnia littoralis Fr. Schmidt et Miquel is an important medicinal plants in East Asian countries. This plant species naturally distributed in Korea, Japan, China, and Taiwan, but it is a rare plants living in the coastal dune in Korea. To investigate the genetic variation, genetic diversity and genetic evolutionary relationships of 14 different geographical G. littoralis, ITS sequence and random amplified polymorphic DNA (RAPD) were analyzed. On the basis of ITS sequences, it was clearly showed that the ITS1 and ITS2 sequences among 14 populations are identical regardless of geographical origin excepting 2 bp in pair-wise comparison of ITS1. Furthermore, RAPD results also showed that 14 different geographical G. littoralis produce various polymorphic patterns without critical relationship among neighboring regions. These combined results suggest that the geographical variation and genetic evolution of G. littoralis is stable and provide important information on genetic diversity, and conservation of this rare plant species in situ and ex situ.

  • The degradation due to channel distortion in a quadrature modulation system from the ideal constant values over the bandwidth of a direct sequence spread spectrum signal are considered. Through using series expansion for the channel gain and phase response, the degradation in the correlator output at the receiver is found as a function of the parameters involved , including phase error, delay error, linear gin variation, quadratic gain variation, and quadratic phase variation.

Distortion Variation Minimization in low-bit-rate Video Communication

  • A real-time frame-layer rate control algorithm with a token bucket traffic shaper is proposed for distortion variation minimization. The proposed rate control method uses a non-iterative optimization method for low computational complexity, and performs bit allocation at the frame level to minimize the average distortion over an entire sequence as well as variations in distortion between frames. The proposed algorithm does not produce time delay from encoding, and is suitable for real-time low-complexity video encoder. Experimental results indicate that the proposed control method provides better visual and PSNR performances than the existing rate control method.

COII Sequence-based Study for Population Genetic Variation of a Ground Beetle, Scarites aterrimus (Coleoptera : Carabidae)

  • The Scarites aterrimus (Coleoptera: Carabidae) dwells exclusively on coastal sandy dunes. Previously, we investigated the nation-wide magnitude and nature of genetic diversity of the species using mitochondrial COI gene and found moderate to low magnitude of sequence diversity, the presence of closely related haplotypes, and relatively high gene flow estimate. Based on these observations we concluded that the species had no historical barriers that bolster genetic subdivision and possible population decline. In this study, we additionally sequenced mitochondrial COII gene from 23 individuals collected from 9 Korean localities to confirm previous findings. Sequencing of 688 bp COII gene provided 5 haplotypes ranging in sequence divergence from 0.145% to 0.291% (1 ~ 2 bp), further confirming low sequence divergence of the species. Gene flow estimates and genetic diversity estimates also support the previous findings that there had been no historical barriers that bolster genetic subdivision.

  • This paper describes an accurate fault location algorithm based on sequence current distribution factors for a double-circuit transmission system. The proposed method uses the voltage and current collected at only the local end of a single-circuit. This method is virtually independent of the fault resistance and the mutual coupling effect caused by the zero-sequence current of the adjacent parallel circuit and insensitive to the variation of source impedance. The fault distance is determined by solving the forth-order KVL(Kirchhoff's Voltage Law) based distance equation. The zero-sequence current of adjacent circuit is estimated by using a zero-sequence current distribution factor and the zero-sequence current of the self-circuit. Thousands of fault simulation by EMTP have proved the accuracy and effectiveness of the proposed algorithm.
