Cellular and Molecular Pathology of Fungi on Plants Studied by Modern Electron Microscopy

  • In plant pathology there is an increasing necessity for improved cytological techniques as basis for the localization of cellular substances within the dynamic fine structure of the host-(plant)-pathogen-interaction. Low temperature (LT) preparation techniques (shock freezing, freeze substitution, LT embedding) are now successfully applied in plant pathology. They are regarded as important tools to stabilize the dynamic plant-pathogen-interaction as it exists under physiological conditions. - The main advantage of LT techniques versus conventional chemical fixation is seen in the maintenance of the hydration shell of molecules and macromolecular structures. This results in an improved fine structural preservation and in a superior retention of the antigenicity of proteins. - A well defined ultrastructure of small, fungal organisms and large biological samples such as plant material and as well as the plant-pathogen (fungus) infection sites are presented. The mesophyll tissue of Arabidopsis thaliana is characterized by homogeneously structured cytoplasm closely attached to the cell wall. From analyses of the compatible interaction between Erysiphe graminis f. sp. hordei on barley (Hordeum vulgare), various steps in the infection sequence can be identified. Infection sites of powdery mildew on primary leaves of barley are analysed with regard to the fine structural preservation of the haustoria. The presentation s focussed on the ultrastructure of the extrahaustorial matrix and the extrahaustorial membrane. - The integration of improved cellular preservation with a molecular analysis of the infected host cell is achieved by the application of secondary probing techniques, i.e. immunocytochemistry. Recent data on the characterization of freeze substituted powdery mildew and urst infected plant tissue by immunogold methodology are described with special emphasis on the localization of THRGP-like (threonine-hydrxyproline-rich glycoprotein) epitopes. Infection sites of powdery mildew on barley, stem rust as well as leaf rust (Puccinia recondita) on primary leaves of wheat were probed with a polyclonal antiserum to maize THRGP. Cross-reactivity with the anti-THRGP antiserum was observed over the extrahaustorial matrix of the both compatible and incompatible plant-pathogen interactions. The highly localized accumulation of THRGP-like epitopes at the extrahaustorial host-pathogen interface suggests the involvement of structural, interfacial proteins during the infection of monocotyledonous plants by obligate, biotrophic fungi.

Isolation and Identification of a New Gene Related to Salt Tolerance in Chinese Cabbage (배추에서 신규 염 저항성 관련 유전자 분리 및 검정)

  • This study was conducted to find a salt tolerance gene in Brassica rapa. In order to meet this objective, we analyzed data from a KBGP-24K oligo chip [BrEMD (Brassica rapa EST and microarray database)] of the B. rapa ssp. pekinensis 'Chiifu' under salt stress (250 mM NaCl). From the B. rapa KBGP-24K microarray chip analysis, 202 salt-responsive unigenes were primarily selected under salt stress. Of these, a gene with unknown function but known full-length sequence was chosen to closely investigate the gene function. The selected gene was named BrSSR (B. rapa salt stress resistance). BrSSR contains a 285 bp open reading frame encoding a putative 94-amino acid protein, and a DUF581 domain. The pSL94 vector was designed to over-express BrSSR, and was used to transform tobacco plants for salt tolerance analysis. T1 transgenic tobacco plants that over-expressed BrSSR were selected by PCR and DNA blot analyses. Quantitative real-time RT PCR revealed that the expression of BrSSR in transgenic tobacco plants increased by approximately 3.8-fold. Similar results were obtained by RNA blot analysis. Phenotypic characteristics analysis showed that transgenic tobacco plants with over-expressed BrSSR were more salt-tolerant than the wild type control under 250 mM NaCl for 5 days. Based on these results, we hypothesized that the over-expression of BrSSR may be closely related to the enhancement of salt tolerance.

Phylogenetic and Chemical Analyses of Cirsium pendulum and Cirsium setidens Inhabiting Korea (국내에 자생하는 큰엉겅퀴와 고려엉겅퀴의 분자유전학적 및 화학적 분석)

  • Cirsium pendulum plants were collected from Hongcheon, Pyeongchang, Wonju, Yangyang in Kangwondo, Gapyeong in Gyeongkido, and Choongju in Choongcheongbukdo. Cirsium setidens plants were collected from Taebaek in Kangwondo and Bonghwa in Kyeongsangbukdo. Genomic DNA was prepared from those plants and used for the amplification of 18S rDNA, ITS1, 5.8S rDNA, ITS2, and part of 28S rDNA. The PCR products were sequenced, and the sequence was deposited in the GenBank. The comparison of those sequences has revealed that the rDNA sequences are identical for all six C. pendulum plants, but that the ITS1 and ITS2 sequences contain variable nucleotides. The two C. setidens plants had different nucleotides in 18S rDNA, ITS1, and ITS2. The comparison of the DNA sequences of C. pendulum and C. setidens collected in this study with C. pendulum of Hokkaido in Japan and C. japonicum of Anhui in China indicated that the plants of those three species are clearly divided into three distinct groups. The silymarin content of the collected plants was analyzed and turned out to be quite high. Therefore, it has been found that both C. pendulum and C. setidens plants are producing large amounts of silymarin, which has been reported to have various medicinal effects.

Identification of Microorganisms Isolated from Jeju Coastal Sedimentary Layer and Characterization of Their Proteases. (제주 연안 퇴적층에서 분리된 미생물의 동정 및 단백질분해효소 특성)

  • In this study, protease-producing bacteria were isolated from the marine sedimentary layer in coastal Jeju. We isolated 2 protease producing strains (SK-2 and SK-125) and tested their protesase producing activities. Gram staining and BIOLOG of isolated strains revealed that strains SK-2 and SK-125 belong to Bacillus and Pseudoalteromonas families, respectively. The 16S rDNA nucleotide sequences analyses of the isolated strains showed 99% sequence homology with those of Bacillus sp. and Pseudoalteromonas sp.; therefore, the isolated strains SK-2 and SK-125 were named Bacillus sp. SK-2 and Pseudoalteromonas sp. SK-125, respectively. The optimum conditions for the cell growth of protease activities were obtained when the both isolates were cultured at $30^{\circ}C$, 96 hrs and pH $7{\sim}8$.

The First Identified Citrus tristeza virus Isolate of Turkey Contains a Mixture of Mild and Severe Strains

  • The presence of Citrus tristeza virus (CTV) has previously been reported in citrus growing regions of Turkey. All serologically and biologically characterized isolates including I$\breve{g}$d${\i}$r, which was the first identified CTV isolates from Turkey, were considered mild isolates. In this study, molecular characteristics of the I d r isolate were determined by different methods. Analysis of the I$\breve{g}$d${\i}$r isolate by western blot and BD-RT-PCR assays showed the presence of MCA13 epitope, predominantly found in severe isolates, in the I$\breve{g}$d${\i}$r isolate revealing that it contains a severe component. For further characterization, the coat protein (CP) and the RNA-depen-dent RNA polymerase (RdRp) genes representing the 3' and 5' half of CTV genome, respectively, were amplified from dsRNA by RT-PCR. Both genes were cloned separately and two clones for each gene were sequenced. Comparisons of nucleotide and deduced amino acid sequences showed that while two CP gene sequences were identical, two RdRp clones showed only 90% and 91% sequence identity in their nucleotide and amino acid sequences, respectively, suggesting a mixed infection with different strains. Phylogenetic analyses of the CP and RdRp genes of I$\breve{g}$d${\i}$r isolate with previously characterized CTV isolates from different citrus growing regions showed that the CP gene was clustered with NZRB-TH30, a resistance breaking isolate from New Zealand, clearly showing the presence of severe component. Furthermore, two different clones of the RdRp gene were clustered separately with different CTV isolates with a diverse biological activity. While the RdRp-1 was clustered with T30 and T385, two well-characterized mild isolates from Florida and Spain, respectively, the RdRp-2 was most closely related to NZRB-G90 and NZRB-TH30, two well-characterized resistance breaking and stem pitting (SP) isolates from New Zealand confirming the mixed infection. These results clearly demonstrated that the I$\breve{g}$d${\i}$r isolate, which was previously described as biologically a mild isolate, actually contains a mixture of mild and severe strains.

  • A group of 16 $Zn+Pb{\pm}Ag$ deposits distributed in the Pyeongchang-Jucheon area, Kangwon-do, South Korea, were semi-regionally investigated. These deposits are contact metasomatic and/or hydrothermal replacement types hosted in the carbonate-dominated Cambrian Machari Formation and Ordovician Ibtanri Formation, and also in the carbonate interbeds of the Precambrian argillic metasediments. Comparing some key aspects of the individual deposits, it is found that the ore deposits hosted in the Machari and Ibtanri Formations are mostly of steeply-dipping chimneys with or without skarn minerals and are rich in Ag and Pb>Zn in metal grade whereas those occuring in the carbonate interbeds of the Precambrian argillic metasediments are gently-dipping conformable lenticular orebodies mostly with skarn minerals and are generally poor in Ag and Zn>Pb. The skarn mineralization in the area appears to have occurred during the lower Cretaceous (118.7Ma) to mid-Cretaceous (107.8Ma) time assumed from the K-Ar dates of the Dowon and Pyeongchang granites which are closely associated with the skarn ore deposits. The Rb/Ba/Sr ratios of these granites indicate that they are of strongly differentiated anomalous granites, and the Nb vs. Y and Rb vs. Y+Nb plots fall on the field of volcanic arc setting. The contact aureoles are zoned, giving the sequence in order of increasing distance from igneous contact: garnet-wollastonite, granet-wollastonite-clinopyroxene and garnet-clinopyroxene in such as the Pyeongchang and Yeonwol 114 areas. Electron microprobe analyses reveal that garnets and clinopyroxenes are generally low in Fe and Mn. Garnets are grossular to intermediate grandite except for those from the Ogryong exoskarn which are richer in andradite, pyrope and spessartine fractions. This indicates that the oxidation state of skarn-forming environment at Ogryong was higher than at the other deposits. Clinopyroxenes are mostly salitic except for those from the Ogryong exoskarn which involve considerable amounts of hedenbergite and johansenite fractions. The ${\delta}^{18}O$ value of Jurassic biotite granite at Ogryong is higher (+10.21‰) than that of Cretaceous one at Chodun (+8.41‰). The ${\delta}^{13}C$ values of carbonate rocks range from -0.89‰ to 0.68‰ and the ${\delta}^{18}O$ values range from +11.91‰ to + 19.34‰ indicating that these carbonate rocks are of marine origin. However, the ${\delta}^{13}C$ values of skarn calcite and vein calcite are -4.80‰ and -12.92‰, and the ${\delta}^{18}O$ values are +5.56‰ and +10.32‰, respectively, indicating that these calcites are of hydrothermal origin. The ${\delta}^{34}S$ values of sulfide minerals range from +4.4‰ to +8.7‰ suggesting that the sulfurs are of magmatic origin.

Structural Analysis of the Danyang Area, Danyang Coalfield, Korea (단양지역의 지질구조)

  • The Danyang area consists of the thrust and folded sedimentary rocks of Paleozoic and Mesozoic Era. The area is bounded by major tectonic units which are the Gagdong Thrust to the west and the Okdong Fault to the east. According to the structural analyses, the area is affected by polyphase deformation. This study establishes deformational sequence in the area. Mylonite zone along the Okdong Fault corresponds to the first generation of structures ($D_1$). $D_1$-structures are discrete shear zone in the Jangsan Formation and bedding parallel extensional deformation in the Cambro-Ordovician sequences. $D_2$-structures were formed prior to the sedimentation of the Jurassic Bansong Group, which are the NW-trending fold and linear structures. After sedimentation of the Bansong Group, the area is strongly affected by the Daebo Orogeny which produces NE-trending thrusts, folds and linear structures. Earlier structures were tightened and rotated toward NE. Some thrust faults did not propagate into the Bansong Group. It is suggested either the Bansong Group acted as a decoupling horizon or rest on unconformably on the thrust faults. The area is weakly affected by $D_4$-event of which structures are E-W trending folds and faults. The Jugryeong Fault clearly cut the earlier folds and thrust faults. The rocks within the fault zone were sliced and rotated during the strike-slip movements. Block rotation and transpressional features can be commonly observed.

Occurrence and Geochemistry of Argyrodite, a Germanium-Bearing Mineral(Ag8GeS6), from the Weolyu Ag-Au Hydrothermal Vein Deposits (월류(月留) 은(銀)-금(金) 열수광상(熱水鑛床)에서 산출된 함(含) Ge 광물(鑛物)인 Argyrodite의 산상(産狀)과 지구화학(地球化學))

  • Ag-Au vein ores from the Weolyu mine, Youngdong district, contain significant germanium (up to 145g/t, average 34.9g/t), in the form of argyrodite ($Ag_8GeS_6$). Mineral chemistries of argyrodite and its associated minerals were determined by electron probe microanalysis. Twenty eight elements in thirteen ore samples were analyzed using an ICP mass spectrometer. Argyrodite occurs in the paragenetically later mineral assemblage consisting of carbonates+quartz+native silver+argentite+Ag-sulfosalts, indicating that the germanium mineralization represents the culmination of a complex mineral sequence which includes early gold and late silver deposition. The mean formula of the argyrodite is $Ag_{7.90}\;(Ge_{0.76}Sn_{0.04})S_6$, with minor amounts of Cu, Fe, Sb, As, Sn, and Zn. The Weolyu argyrodite shows systematic substitutions of Ag by Cu, and of Ge by Sb. Chemical analyses of vein ores indicate that metals were precipitated in the order of $Fe{\rightarrow}Pb$, $Zn{\rightarrow}Cu{\rightarrow}Ag$, Sb, As, Ge. Germanium has a strong geochemical affmity with As and Sb, and Cu, Pb, Zn, Mo, and Sr show weak positive correlations with Ge. Germanium deposition at Weolyu was mainly a result of cooling of hydrothermal fluids (down to $175^{\circ}C{\sim}210^{\circ}C$, due to increasing involvement of cooler meteoric waters in the epithermal system.

  • Foward-swept wings are known to possess superior aerodynamic performance compared to the conventional straight wings. However major concerns regarding forward-swept wings include divergence at lower airspeeds which require careful consideration at the design stage. As an endeavor to overcome such drawbacks, aeroelastic tailoring is attempted. In order to find an optimal ply sequence, recursive aeroelastic analyses is conducted and one-dimensional beam analysis coupled with simple aerodynamics is used for the improved computational efficiency and modelling convenience. The analysis used in this paper, DYMORE and analytic formula, both use one-dimensional beam model for the structure. Cross-sectional analysis for multi-cell NACA0015 airfoil section is conducted using VABS and oblique function is used for the sweep angle. Throughout the present aeroelastic tailoring, the maximum divergence speed of 290.2m/s is achieved which is increased by approximately 43% than that for the conventional ply configuration.

KMTNET SUPERNOVA PROGRAM VARIABLE OBJECTS I. NGC 2784 FIELD

  • We present analyses of ~1250 variable sources identified in a 20 square degree field toward NGC 2784 by the KMTNet Supernova Program. We categorize the variable sources into three groups based on their B-band variability. The first group consists of 31 high variability sources with their B-band RMS variability greater than 0.3 magnitudes. The second group of medium variability contains 265 sources with RMS variability between 0.05 and 0.3 magnitudes. The remaining 951 sources belong to the third group of low variability with an RMS variability smaller than 0.05 magnitudes. Of the entire ~1250 sources, 4 clearly show periods of variability greater than 100 days, while the rest have periods shorter than ~51 days or no reliable periods. The majority of the sources show either rather irregular variability or short periods faster than 2 days. Most of the sources with reliable period determination between 2 and 51 days belong to the low-variability group, although a few belong to the medium-variability group. All the variable sources with periods longer than 35 days appear to be very red with B - V > 1.5 and V - I > 2.1 magnitudes. We classify candidates of 51 Cepheids, 17 semi-regular variables, 3 Mira types, 2 RV(B) Tauri stars, 26 eclipsing binary systems and 1 active galactic nucleus. The majority of long-term variables in our sample belong to either Mira or semi-regular types, indicating that long-term variability may be more prominent in post-main sequence phases of late-type stars. The depth of the eclipsing dips of the 26 candidates for eclipsing binaries is equivalent to ~0.61 as the average relative size of the two stars in the binary system. Our results illustrate the power of the KMTNet Supernova Program for future studies of variable objects.
